  • In order to investigate what kind of response anaerobic bifidobacteria has on oxygen stress, five oxygen-tolerant bifidobacteria were isolated from human fecal samples. All were temporarily identified as Bifidobacterium longum through an analysis of carbohydrate utilization patterns and cellular fatty acid profiles. In the presence of oxygen, the lag phase became extended and the cell growth was suppressed. Bifidobacterial cell was able to remove dissolved oxygen in an early stage of growth and to overcome oxygen stress to a certain extent. The cell became long n size and showed a rough surface containing many nodes which were derived from abnormal or incomplete cell division. Cellular fatty acid profiled changed remarkably under a partially aerobic condition, so that the carbon chain of cellular fatty acid became short. All the dimethyl acetals originated from plasmalogen were reduced, any cyclopropane fatty acid, 9, 10-methyleneoctadecanoic acid ($C_{19:0}cyc9,10$), was increased remarkably. Oxygen stress induced a 5.5 kD protein in B. longum JI 1 of the oxygen-teolerant bifidobacteria, that was named Osp protein, and its N-terminal amino acid sequence was as follows: unknown amino acid-Thr-Gly-Val-Arg-Phe-Ser-Asp-Asp-Glu. Therefore, the oxygen-tolerant bifidobacteria seemed to defend against oxygen stress byincreasing the content of short fatty acid and cyclopropane fatty acid, and induction of an oxygen stress protein, but not the plasmalogen.

  • The content analysis of fatty acids in Perilla cultivars and commercial oils is conducted through gas chromatography with a flame ionization detector. Results show that Perilla cultivars, such as Deulsaem and Daesil, contain high amounts of ${\alpha}-linolenic$ acid (262.22 and 261.97 mg/g, respectively). Among commercial oils, Perilla oil contains a higher amount of ${\alpha}-linolenic$ acid (515.20 mg/g). Accordingly, ${\alpha}-linolenic$ acid is a major fatty acid of Perilla cultivars and oil. Therefore, Perilla cultivars could be used as a food supplement for nutritional and pharmaceutical purposes.

  • Seakso 5, a maize hybrid, was applied for variety in 2021 by Gangwon Agricultural Research and Extension Services in Korea. This variety was bred to produce a purple extract of com husk. It shows purple color in the husk and cob and contains abundant anthocyanins. In this study, to obtain basic data on purple com 'Seakso 5' husk extract (PCHE), a husk extract was prepared and the contents of vitamins, amino acids and fatty acids were analyzed. The Seakso 5 husks were extracted with 30% alcohol and concentrated, after adding dextrin to the concentrate, it was spray-dried to prepare an extract. The PCHE were analyzed for vitamins, amino acids, and fatty acids by referring to the Food Code. As a result, three types of vitamins were detected, and their contents were vitamin B1 0.14 mg/100g, vitamin B2 2.30 mg/100g, and vitamin C 11.32 mg/100g. A total of 15 amino acids were detected in the PCHE, including aspartic acid and glutamic acid, and the content of aspartic acid was the highest at 3.02 mg/g. Simultaneous analysis of fatty acids Among 37 types, saturated fatty acids were palmitic acid (C16:0), stearic acid (C18:0), monounsaturated fatty acids were oleic acid (C18:1(n-9)), polyunsaturated fatty acids were linoleic acid(C18:2(n-6)c) and a-Linolenic Acid (C18:3(n-3)) were detected. Among the detected fatty acids, the content of palmitic acid was the highest at 40.75 mg/100g. • A total of 48 different volatile oils were identified form P. brevitarsis larvae by gas chromatography/mass spectrometry (GC/MS). Acids (48.67%) were detected as the major group in P. brevitarsis larvae comprising the largest proportion of the volatile compounds, followed by esters (19.84%), hydrocarbons (18.90%), alcohols (8.37%), miscellaneous (1.71%), aldehydes (1.35%) and terpenes (1.16%). The major volatile constituents were 9-hexadecenoic acid (16.75%), 6-octadecenoic acid (14.88%) and n-hexadecanoic acid (11.06%). The composition of fatty acid was also determined by GC analysis and 16 fatty acids were identified. The predominant fatty acids were oleic acid ($C_{18:1}$, 64.24%) followed by palmitic acid ($C_{16:0}$, 15.89%), palmitoleic acid ($C_{16:1}$, 10.43%) and linoleic acid ($C_{18:2}$, 4.69%) constituting more than 95% of total fatty acids. The distinguished characteristic of the fatty acid profile of P. brevitarsis larvae was the high proportion of unsaturated fatty acid (80.54% of total fatty acids) versus saturated fatty acids (19.46% of total fatty acids). Furthermore, small but significant amounts of linoleic, linolenic and ${\gamma}$-linolenic acids bestow P. brevitarsis larvae with considerable nutritional value. The novel findings of the present study provide a scientific basis for the comprehensive utilization of the insect as a nutritionally promising food source and a possibility for more effective utilization.

  • Abdominal obesity is an important factor of metabolic syndrome and is known as an independent risk factor of cardiovascular disease. The purpose of this study was to examine the fatty acid intake pattern as well as the association between fatty acid intake and abdominal obesity and hyperglycemia in Korean adults. This study was based on data from the Korea National Health and Nutrition Examination Surveys (KNHANES) I, II, III, and IV. After excluding subjects who had missing data for dietary intakes, anthropometric measurments, and laboratory tests, a total of 12,320 subjects aged 20~64y (5,266 men and 7,054 women) were included in the study. Trends in individual consumption of dietary fatty acids by year were analyzed by One-way ANOVA test, and the association between fatty acid intake and abdominal obesity and hyperglycemia was analyzed by logistic regression analysis after adjusting for age, energy intake, marital status, job, education level, alcohol intake, and smoking status. In men, intakes of TFA, SFA, MUFA, FUFA, and n-6 PUFA significantly increased with year. On the other hand, intakes of TFA, SFA and FUFA significantly increased with year in women. Logistic regression analysis showed that the risk of abdominal obesity was significantly associated with intakes of PUFA, LNA and n-3 PUFA in women. However, there was no association with any fatty acid intakes in men. The risk of hyperglycemia was significantly and positively associated with intakes of TFA, SFA, PUFA, and LNA in men, but, there was no significant association with any fatty acid intakes in women. These results show that abdominal obesity and hyperglycemia are significantly associated with individual intake of fatty acids, suggesting that fatty acid type may be associated with risk of abdominal obesity and hyperglycemia. Futher, the macronutrients of measl must be properly balanced.

  • Fabrea salina is a hypersaline ciliate having importance as a live food source for juvenile stages of aquatic animals including smaller invertebrates. The analysis of this ciliate for proximate and biochemical composition was carried out. The moisture, protein, fat, carbohydrate and ash content of F. salina from natural sources were 86.66$\pm$0.380, 56.66$\pm$0.494%, 36.66$\pm$0.614%, 1$\pm$0.073% and 4$\pm$0.182%, respectively. Gas chromatographic analysis (percent area below the curve) revealed that the presence of oleic acid was higher over other fatty acids in both natural and cultured F. salina. The absolute content of oleic acid was higher in natural (18.91% area) than in the cultured (10.74% area) F. salina. Linoleic and linolenic acids were also among major fatty acids with the percentage area of 16.29 and 14.58, respectively. The number of fatty acids in cultured Fabrea was less as compared to the natural ones and the oleic acid was followed by palmitic acid, palmitoleic acid, linoleic acid and stearic acid.

  • An ${\omega}-3$ fatty acid desaturase gene which is involved in de novo synthesis of -Iinolenate was isolated from cDNA library of Perilla frutescens. A cDNA library was constructed with mRNA extracted from perilla seeds of 12 DAF. The cDNA clone consisting of 1317-bp open reading frame encoding 438 amino acids with a relative MW of 50kDa, was isolated and showed 65-83% similarities to other known genes. This cDNA is deduced to encode a plastidal ${\omega}-3$ fatty acid desaturase based on the fact that it has higher homology to plastidal ones than to microsomal ones and its N-terminal sequence shares several characteristics of transit peptides of chloroplast proteins. Southern blot analysis of genomic DNA indicated that more than one gene or alleles for ${\omega}-3$ fatty acid desaturase are present in the genome of perilla. Northern blot analysis showed that the ${\omega}-3$ fatty acid desaturase gene is mainly revealed in early developing seeds and has different expression patterns depending on tissue types compared to the microsomal ones.
