Evaluation of Resistance in Hot Pepper Germplasm to Phytophthora Blight on Biological Assay (생물검정을 통한 고추 유전자원의 역병저항성 평가)

  • Phytophthora blight of pepper is the most economically important disease in the world cultivation regions. We investigated the phytophthora blight resistance of 300 accessions of Korean landrace of hot pepper germplasms collected from 83 local regions. The disease incidence rate was checked from 7 days to 28 days at an interval of 7 days after inoculation under greenhouse conditions. Among 300 accessions, the disease incidence rate of phytophthora blight of 67 accessions of pepper germplasm was more than 60.1%, while no disease was observed in 37 accessions at 7 days after inoculation. At 28 days after inoculation, five and eleven accessions of pepper germplasm were resistance and moderate resistance to $P.$ $capsici$, respectively. Two hundred forty four susceptible accessions (81.3%) of pepper were scored as having more than 60.1% of disease incidence of phytophthora blight. This result suggests that five candidate pepper germplasm might be used as breeding resources for the phytophthora blight resistance breeding program. Also, further genetic studies should be carried out to verify this result, with the overall focus of providing information on important characteristics of pepper germplasm.

Bactericidal Efficacy of a Disinfectant Spray Containing a Grapefruit-seed Extract, Citric acid, Malic acid and Benzalkonium Chloride against Salmonella Typhimurium and Brucella ovis

  • Salmonella spp. and Brucella spp. can cause considerable diseases on both humans and animals. In addition, these microorganisms cause the economic loss in animal farming and food industry. In this study, the disinfection efficacy of a disinfectant spray, composed to grapefruit seed extract, citric acid, malic acid and benzalkonium chloride, was evaluated against S. Typhimurium and B. ovis. A bactericidal efficacy test by broth dilution method was used to determine the lowest effective dilution of the disinfectant following exposure to test bacteria for 30 min at $4^{\circ}C$. The disinfectant and test bacteria were diluted with hard water (HW) or organic matter suspension (OM) according to treatment condition. On HW condition, the bactericidal activity of the disinfectant spray against S. Typhimurium and B. ovis was 5 and 4 fold dilutions, respectively. On OM condition, the bactericidal activity of the disinfectant spray was 2 and 1 fold dilutions against S. Typhimurium and B. ovis, respectively. As the disinfectant spray possesses bactericidal efficacy against foodborne pathogens such as S. Typhimurium and B. ovis, the disinfectant spray can be used to control the spread of bacterial diseases.

  • Antimicrobial resistance has been a well-recognized problem ever since the introduction of penicillin into clinical use. History of antimicrobial development can be categorized based on the major antibiotics that had been developed against emerging resistant $pathogens^1$. In the first period from 1940 to 1960, penicillin was a dominating antibiotic called as a "magic bullet", although S.aureus armed with penicillinase led antimicrobial era to the second period in 1960s and 1970s. The second stage was characterized by broad-spectrum penicillins and early generation cephalosporins. During this period, nosocomial infections due to gram-negative bacilli became more prevalent, while those caused by S.aureus declined. A variety of new antimicrobial agents with distinct mechanism of action including new generation cephalosporins, monobactams, carbapenems, ${\beta}$-lactamase inhibitors, and quinolones characterized the third period from 1980s to 1990s. However, extensive use of wide variety of antibiotics in the community and hospitals has fueled the crisis in emerging antimicrobial resistance. Newly appeared drug-resistant Streptococcus pneumoniae (DRSP), vancomycin-resistant enterococci (VRE), extended-spectrum ${\beta}$-lactamase-producing Klebsiella, and VRSA have posed a serious threat in many parts of the world. Given the recent epidemiology of antimicrobial resistance and its clinical impact, there is no greater challenge related to emerging infections than the emergence of antibiotic resistance. Problems of antimicrobial resistance can be amplified by the fact that resistant clones or genes can spread within or between the species as well as to geographically distant areas which leads to a global concern$^2$. Antimicrobial resistance is primarily generated and promoted by increased use of antimicrobial agents. Unfortunately, as many as 50 % of prescriptions for antibiotics are reported to be inappropriate$^3$. Injudicious use of antibiotics even for viral upper respiratory infections is a universal phenomenon in every part of the world. The use of large quantities of antibiotics in the animal health industry and farming is another major factor contributing to selection of antibiotic resistance. In addition to these background factors, the tremendous increase in the immunocompromised hosts, popular use of invasive medical interventions, and increase in travel and mixing of human populations are contributing to the resurgence and spread of antimicrobial resistance$^4$. Antimicrobial resistance has critical impact on modem medicine both in clinical and economic aspect. Patients with previously treatable infections may have fatal outcome due to therapeutic failure that is unusual event no more. The potential economic impact of antimicrobial resistance is actually uncountable. With the increase in the problems of resistant organisms in the 21st century, however, additional health care costs for this problem must be enormously increasing.

  • This study was conducted from July to December 2004, in order to find measures to improve the water quality and the function of pond ecosystems which are often negatively affected by phytoplankton and suspended particle matters. Most of the time, the management of ponds in Korea does not consider the ecological concepts such as self-purification and nutrient recycling. Instead, conventional methods depend on other factors such as fish farming and the function of fountains and waterfalls. Sustainability of pond ecosystems must be studied with the highest emphasis given to water quality. Water discharges of inflow and outflow as well as balance adjustment for different factors must be thoroughly studied. There is a great need for studies on sustainability because it can be realized through the horizontal or vertical balances of an ecosystem. Our current research offers useful information to the academia and public on maintaining sustainability in terms of structure and function of the pond.

Induction of Physiological Sex-Reversal and Gynogenetic Diploid in Rainbow Trout, Oncorhynchus mykiss (무지개송어의 생리학적 성전환과 자성발생 2배체 유도)

  • This study was conducted to increase the efficiency of farming practice in rainbow trout, Oncorhynchus mykiss, by sex reversal and chromosome-set manipulation techniques. To obtain phenotypic males, hormonal sex reversal was carried out using an exogenous hormone treatment method. 5 mg of 17 alpha-methyltestosterone per kg diet was supplied for 82 days after first feeding at $10^{\circ}C$ and $13^{\circ}C$. More than 93% of the male population was produced by this method and growth of hormone-treated fish at $13^{\circ}C$ was faster than that of untreated bi-sexual groups. Induced diploid gynogenesis was carried out using artificial insemination of UV-irradiated sperm into haploid eggs. Based on the appearance of the rate of haploid syndrome and survival of embryo, a UV ray dose of at least $3,600\;erg/cm^2$ was required to inactivate rainbow trout sperm genetically. Haploid embryos were restored to diploid by blocking the extrusion of the second polar body using heat shock treatment at $28^{\circ}C$ for 20 min, 10 min post insemination. Gynogenetic diploid sex ratios were confirmed after maturation of the fish erythrocyte measurements and chromosome counts.

Antibacterial Effect of Bacteria Isolated from the Plant Rhizosphere against Pathogenic Bacteria of Fish (식물근권에서 분리한 세균의 어류질병세균에 대한 항균활성 효과)

  • Olive flounder (Paralichthys olivaceus) is an important aquaculture fish species in Jeju Island, South Korea. Due to the intensification of flounder fish farming, huge amounts of chemical antibiotics are used against several fish diseases. This has many harmful side effects on fish, as well as human consumers. Hence, an alternative to chemical antibiotic agents is needed for disease control. In this study, three strains of rhizobacteria (BRH433-2, TRH415-2, and THJ609-3) were isolated from the rhizosphere of plants. Assays of their antibacterial activity against fish pathogens, such as S. iniae, S. parauberis, V. anguillarum, and E. tarda, were performed with untreated broth culture (without cell separation), supernatant, and precipitated pellets separated by centrifugation. Among these, the cell suspension prepared from the precipitated pellet showed significant antimicrobial activity when compared with that of the untreated broth culture and centrifugal supernatant. These results indicate that the three isolated rhizobacterial strains exhibit antibacterial activity. Analysis of the 16S rDNA sequences of the BRH-433-2, THJ609-3, and TRH415-2 strains showed the highest similarity to Burkholderia gladioli (99.5%), Pseudomonas baetica (97.7%), and P. koreensis and P. baetica (98.4%), respectively. We suggest that the strains hold promise in disease management of fish.

Community Structure and Health Assessment of Macrobenthic Assemblages during Spring and Summer in the Shellfish Farming Ground of Wonmun Bay, on the Southern Coast of Korea (원문만 패류양식어장에 서식하는 대형저서동물의 춘계와 하계의 군집구조 및 건강도 평가)

  • This study investigated the community structure and health assessment of macrobenthic assemblages in Wonmun Bay, in the southern coast of Korea. The total number of species and mean density were 170 species and $1,269\;indivuduals/m^2$, respectively. The major dominant species were the bivalve Theora fragilis ($230{\pm}446\;indivuduals/m^2$) and the polychaetes Lumbrineris longifolia ($180{\pm}339\;indivuduals/m^2$), Sigambra tentaculata ($124{\pm}153\;indivuduals/m^2$), Paraprionospio patiens ($104{\pm}293\;indivuduals/m^2$) and Capitella capitata ($82{\pm}223\;indivuduals/m^2$). Community statistics [cluster analysis and nonmetric multidimentional scaling (NMDS) ordination] revealed that the macrobenthic community structure in the study area was classified into three station groups for both seasons. Several biotic indices based on macrobenthic communities have been developed to assess the ecological quality of coastal ecosystems. In this study, the ecological status was assessed by four biotic indices Shannon's H', the AZTI Marine Biotic Index (AMBI), multivariate-AMBI (M-AMBI) and the Ecological Conservation Index (ECI). The ecological statuses of the macrobenthic communities in Wonmun Bay were poorer in the summer than in the spring. The results of the present study indicate that three biotic indices ( i.e.; Shannon's H', M-AMBI and ECI ) were better correlated with bottom dissolved oxygen than the AMBI in Wonmun Bay.

In consideration the education level were evaluated the college grauated ; 48.8% and High School graduated: 30.1% shows that more educated level has significant higher rate value were appearenced. 2) On the subject of criticism to evaluation for adapted personnel by native place such as larger citypeople ; 29.6%, Farming and fishing village people were 28.4%, although the classfication of job analysis for estimation appearence rate were ; students 27.4%, Sales and service field personnel rate ; 15.1% and expert technique field personnel were ; 90% comprehensively appearenced. Although consideration for ecnomics situation review to the middle level shows of rate ; 61.7% and lower level was appearenced rate ; 23.9% of each others. 3) The people of resident in cities were appearenced nutrition intake has more than 2 weeks continuousely practiced by experiance rate shows ; 72% thus intake has experianced rate were 74.6% and female was 68.5% affact is comprehensively male has more many higher rate valued. In the classfication by age group 60 year old has 100% they have experiaced of nutrition intake and age group 50 to be appearenced its rate ; 76.4 % and age group 40 were 75.5% Thus result of survey for comparative rate shows in generally say more aged group has intake significant higher than younger age of group. Nevertheless, age group 10 years old be experienced of nutrition of intake rate ; 63.2% and age group 20 shows 73.9% was comparative more significant higher than age group 30 years old level adolescents.

Estimating Groundwater Level Variation due to the Construction of a Large Borrow Site using MODFLOW Numerical Modeling (대규모 토취장 개발 예정 지역의 수치모델을 이용한 지하수위 변동 예측)

  • A numerical model and field monitoring data are used to estimate a change in groundwater level at a borrow site, which will be constructed at the mountainous area with a large ground excavation in the study area, Hwaseong city. Lithologic data and hydraulic coefficients are collected at 9 boreholes and also groundwater levels are measured at these boreholes and existing wells in the study area. Additionally, groundwater recharge rate for the type of land cover is estimated using water budget analysis; 133.34mm/year for a mountainous area, 157.68mm/year for a farming area, 71.08mm/year for an urbanized area, and 26.06mm/year for a bedrock exposure area. The change in groundwater level in and around a borrow site is simulated with Modflow using these data. The result of a transient model indicates that a removal of high ground (over 40El.m) by an excavation will produce a decrease in groundwater levels, up to 1 m, around a borrow site in 10 years. It also explains that this ground excavation will bring about the decreases of 9.4% and 7.0% for groundwater recharge and surface runoff, respectively, which are the factors causing groundwater level's change. This study shows that it is required to construct the groundwater monitoring wells to observe the change of groundwater near a borrow site.

Biogeochemistry of Metal and Nonmetal Elements in the Surface Sediment of the Gamak Bay (가막간 표층퇴적물 중의 금속 및 비금속 원소의 생지화확적 분포특성)

  • In order to evaluate die geochemical characteristics of sediment in a semi-enclosed bay used as shellfish and fish farming area, the concentrations of metallic(V, Cr, Mn, Fe, Co. Ni, Cu, Zn, Ag, Cd, Hg, Ph, As) and non-metallic(P, Se) elements and uranium were measured in the surface sediment samples collected from 19 stations of Gamak Bay in April 2010. Metal contamination status in the sediments were also evaluated using the sediment quality guidelines(SQGs) proposed by the National Oceanic and Atmospheric Administration(NOAA) and the enrichment factor(EF). The concentrations of elements in sediment were mainly controlled by quartz-dilution effect(V, Cr, Fe, Co and Ni), the dilution effect of organic matter(Cd and U), and metal redistribution by the decomposition of organic matter(Mn, Ag, As, and Se). The concentrations of metals, except As and Ni, in sediments from all sampling stations were lower than ERL values of NOAA. Conclusively, the surface sediment of Gamak Bay was slightly polluted with Ni, Ag, Cd, and Cd but was not polluted with other elements on the basis of EF results. Our results suggest that the surface sediment in Gamak Bay is not polluted by metallic elements.
