Chronic cold stress-induced myocardial injury: effects on oxidative stress, inflammation and pyroptosis

  • Background: Hypothermia is a crucial environmental factor that elevates the risk of cardiovascular disease, but the underlying effect is unclear. Objectives: This study examined the role of cold stress (CS) in cardiac injury and its underlying mechanisms. Methods: In this study, a chronic CS-induced myocardial injury model was used; mice were subjected to chronic CS (4℃) for three hours per day for three weeks. Results: CS could result in myocardial injury by inducing the levels of heat shock proteins 70 (HSP70), enhancing the generation of creatine phosphokinase-isoenzyme (CKMB) and malondialdehyde (MDA), increasing the contents of tumor necrosis factor-α (TNF-α), high mobility group box 1 (HMGB1) interleukin1b (IL-1β), IL-18, IL-6, and triggering the depletion of superoxide dismutase (SOD), catalase (CAT), and glutathione (GSH). Multiple signaling pathways were activated by cold exposure, including pyroptosis-associated NOD-like receptor 3 (NLRP3)-regulated caspase-1-dependent/Gasdermin D (GSDMD), inflammation-related toll-like receptor 4 (TLR4)/myeloid differentiation factor 88 (MyD88)-mediated nuclear factor kappa B (NF-κB), and mitogen-activated protein kinase (MAPK), as well as oxidative stressinvolved thioredoxin-1/thioredoxin-interacting protein (Txnip) signaling pathways, which play a pivotal role in myocardial injury resulting from hypothermia. Conclusions: These findings provide new insights into the increased risk of cardiovascular disease at extremely low temperatures.

LPS 주사한 BALB/c 마우스에서 Genistein의 산화적 스트레스 억제효과 및 항염증 효과 (Anti-oxidative and Anti-inflammatory Effects of Genistein in BALB/c Mice Injected with LPS)

  • 본 연구에서는 내독소인 LPS로 산화적 스트레스를 유발시킨 BALB/c mice에 genistein을 투여하였을 때 TNF-$\alpha$, TBARS, superoxide anion 농도와 GSH, 항산화 효소계 활성, 그리고 NF-${\kappa}B$ transactivation에 미치는 영향을 조사하여 genistein이 내독소에 의한 산화적 스트레스와 염증반응을 억제하는 효과가 있는지 알아보고자 하였다. 평균체중 20 g인 BALB/c mice 암컷 120수를 30수씩 완전임의배치로 4군으로 분류하여 PBS군(대조군)은 PBS를 복강 속으로 투여 후 30분경과 후에 다시 PBS를 투여하였으며, genistein군은 genistein을 체중 kg당 200 mg으로 투여한 30분 후 PBS를 투여하였다. LPS군은 LPS를 처리한 군으로 PBS 투여 30분 후 LPS를 체중 kg당 100 mg 농도로 복강 투여하였고, genistein+LPS군은 체중 kg당 genistein 200 mg을 투여 30분 후 LPS를 체중 kg당 100 mg을 투여하였다. 마지막 투여 1시간, 4시간, 8시간 경과 후 mouse의 안와정맥으로부터 혈액을, 복강으로부터 복강대식세포와 간을 취하였다. LPS 투여 8시간 경과 후 LPS군의 ${O_2}^-$ 생성량은 현저하게 증가하였으며 geinstein+LPS군은 genistein 대조군, PBS군과 비슷한 수준을 유지하였다. 반면 SOD 활성은 geinstein+LPS군이 LPS군에 비해 유의적으로 높은 수준이었다. 혈장에서의 TNF-$\alpha$ 수준은 LPS 투여 8시간 후 genstein+LPS군이 LPS군보다 유의적으로 낮은 수준을 보였다. LPS 투여는 항산화 효소계 활성과 GSH의 수준을 감소하였으나, genistein을 투여한 genistein+LPS군은 LPS군에 비해 GSH 농도와 catalase, GSH-px, GSH-reductase 활성이 모두 유의적으로 증가하는 결과를 보였다. 간에서의 NF-${\kappa}B$ transactivation 정도는 LPS 투여 후 1시간, 4시간 경과 후에 PBS군에 비해 LPS군과 genistein+LPS군에서 유의적으로 높은 수준이었으나 8시간 경과 후 LPS군은 증가하는 반면 genistein+LPS군은 변화하지 않았다. 이상의 결과를 요약해보면 LPS의 투여는 혈장과 간의 산화적 스트레스와 염증반응을 촉진하는 것으로 나타났으며 LPS 투여 전 공급한 genistein은 LPS로 유도된 산화적 스트레스와 염증반응을 항산화 효소계 활성 증가와 NF-${\kappa}B$ transactivation 억제, TNF-$\alpha$ 생성 저하 등의 기작으로 세포내의 과산화수준을 수준을 낮추고 GSH를 증가시켜 산화적 스트레스를 억제하는 것으로 사료된다.

LPS로 유도된 RAW 264.7 세포와 마우스 귀 조직에 대한 주름까막살 에탄올 추출물의 항염증 효과 (Anti-Inflammatory Effect of Ethanol Extract from Grateloupia crispata on Lipopolysaccharide-Induced Inflammatory Responses in RAW 264.7 Cells and Mice Ears)

  • 본 연구는 해조류 중에서도 홍조류인 주름까막살 에탄올 추출물(GCEE)의 항염증 효과를 확인하기 위한 실험으로 proinflammatory cytokines의 분비량 및 iNOS, COX-2, $NF-{\kappa}B$와 MAPKs의 발현량을 관찰하고 마우스모델에서 항염증 효과를 확인하였다. GCEE가 세포 생존율에 있어 독성을 나타내지 않음을 MTT assay를 통해 확인한 후 같은 농도로 추후실험을 진행하였다. NO 분비량이 GCEE에 의해 농도 의존적으로 감소하였으며 전염증성 매개물질인 사이토카인(IL-6, $TNF-{\alpha}$$IL-1{\beta}$)의 분비량 또한 유의적으로 감소하였다. 이러한 결과가 전염증성 매개인자의 전사인자인 $NF-{\kappa}B$와 MAPKs 경로에 의한 것인지 확인하기 위하여 발현량을 관찰한 결과, GCEE가 LPS 처리로 현저히 증가한 염증 관련 효소인 iNOS와 COX-2의 단백질 발현을 농도 의존적으로 유의성 있게 억제하였고 전사인자인 $NF-{\kappa}B$ 및 MAPKs의 발현을 억제하였다. 또한, GCEE는 croton oil로 부종을 유발한 마우스모델에서 귀 부종 억제 효과를 나타내었고 250 mg/kg 농도에서 조직의 경피 및 진피 두께의 발달을 prednisolone 50 mg/kg 처리구와 유사한 정도까지 현저히 억제하고 염증성 세포인 mast cell의 침윤 억제 효과도 확인하였다. 이를 통해 주름까막살 에탄올 추출물은 염증반응의 전사인자인 $NF-{\kappa}B$와 MAPKs의 발현을 조절함으로써 iNOS와 COX-2의 발현을 억제하고 그에 따라 전염증성 매 개인자인 NO, IL-6, $TNF-{\alpha}$$IL-1{\beta}$의 분비를 억제하여 항염증 활성을 가지는 것을 확인하였으며, 이 결과를 종합해 볼 때 주름까막살 에탄올 추출물이 염증 치료제의 소재로 이용될 가치가 충분할 것으로 생각한다.

Ginsenosides from Korean Red Ginseng ameliorate lung inflammatory responses: inhibition of the MAPKs/NF-κB/c-Fos pathways

  • Background: Korean Red Ginseng (steamed and dried white ginseng, Panax ginseng Meyer) is well known for enhancing vital energy and immune capacity and for inhibiting cancer cell growth. Some clinical studies also demonstrated a therapeutic potential of ginseng extract for treating lung inflammatory disorders. This study was conducted to establish the therapeutic potential of ginseng saponins on the lung inflammatory response. Methods: From Korean Red Ginseng, 11 ginsenosides (Rb1, Rb2, Rb3, Rc, Rd, Re, Rf, Rg1, Rg2, Rg3, and Rh2) were isolated. Their inhibitory potential and action mechanism were evaluated using a mouse model of lung inflammation, acute lung injury induced by intranasal lipopolysaccharide administration. Their anti-inflammatory activities were also examined in lung epithelial cell line (A549) and alveolar macrophage (MH-S). Results: All ginsenosides orally administered at 20 mg/kg showed 11.5-51.6% reduction of total cell numbers in bronchoalveolar lavage fluid (BALF). Among the ginsenosides, Rc, Re, Rg1, and Rh2 exhibited significant inhibitory action by reducing total cell numbers in the BALF by 34.1-51.6% (n = 5). Particularly, Re showed strong and comparable inhibitory potency with that of dexamethasone, as judged by the number of infiltrated cells and histological observations. Re treatment clearly inhibited the activation of mitogen-activated protein kinases, nuclear factor-${\kappa}B$, and the c-Fos component in the lung tissue (n = 3). Conclusion: Certain ginsenosides inhibit lung inflammatory responses by interrupting these signaling molecules and they are potential therapeutics for inflammatory lung diseases.

골 대사 및 phytochemicals의 estrogen 효과 (Bone Metabolism and Estrogenic Effect of Phytochemicals)

  • 전 세계적인 인구 고령화 현상으로 인하여 골다공증은 주요한 질병으로 대두되고 있다. 골다공증은 뼈의 질량과 강도가 감소하여 골절의 위험이 증가하는 질환으로 조골세포의 골 형성 및 파골세포의 골 흡수의 불균형으로 인해 발생하는 질환이다. 조골세포에 의한 골 형성은 BMP, RUNX2, $Wnt/{\beta}-catenin$ 경로 등을 통하여 활성화 되며, 파골세포에 의한 골 흡수는 RANKL과 RANK의 결합에 의해서 시작된다. 폐경기 여성은 호르몬 불균형에 의해 여러 질병의 위험에 처해 있으며, 폐경기 여성의 약 30%에서 관찰되는 골다공증은 폐경기 여성에게서 발생되는 가장 흔한 대사성 질환이기도 하다. Estrogen이 부족할 때 파골세포의 골 흡수가 촉진되므로, 특히 폐경 여성에서 골다공증의 발생위험이 증가하게 된다. 호르몬대체요법은 폐경기 증후군의 증상을 경감시키거나 치료하기 위해 널리 사용되어 왔으나, 호르몬 치료를 장기간 실시할 경우 유방암, 난소암, 자궁암 등의 부작용 위험성이 매우 높은 것으로 알려져 있다. 따라서 최근 들어 여러 부작용을 보완하기 위해 폐경기 증후군 증상에 대처할 수 있는 estrogen과 유사한 활성을 지닌 식물성 estrogen인 phytoestrogen에 대한 연구가 활발히 진행되고 있다. 따라서, 본 총설에서는 조골세포 및 파골세포의 분화 기전에 대한 선행연구를 알아보고 골 대사에서의 estrogen의 역할 및 phytoestrogen과 관련한 연구들에 대해서도 살펴보았다.

카드뮴으로 유발된 산화적 스트레스에 대한 침 자극의 간 보호 효과 (The Protective Effects of Acupuncture on the Liver in the Oxidative Stress Caused by Cadmium)

  • This study was performed to investigate the protective effects of acupuncture on the liver in the oxidative stress caused by cadmium accumulation. Sprague-Dawley male($150{\pm}30g$) rats were stabilized for a week and divided into 5 groups which is normal, control, $LR_3$ acupuncture, $BL_{23}$ acupuncture and sham acupuncture group. For three days experimental groups were received oral doses of cadmium 2 mg/kg twice a day. Acupuncture was given bilaterally at each point 10 times for two weeks. The depth of stimulation was 1 mm at right angles and torsion of acupuncture was produced 2 times per second for 1 minutes. The liver was shipped off and taken weight at the last day of two weeks, and hepatic functions was confirmed through alanine transaminase(ALT) and aspartate amino-transferase(AST). We measured reactive oxygen species of serum, liver and kidney, and compared expression levels of superoxide dismutase(SOD), catalase, glutathione peroxidase(Gpx), nuclear factor erythroid derived 2-related factor 2(Nrf-2), heme oxygenase-1(HO-1), nuclear factor-${\kappa}B$ (NF-${\kappa}B$), cyclooxygenase-2(COX-2), inducible nitric oxide synthase(iNOS), Bax and Cytochrome c. $BL_{23}$ acupuncture group significantly increased liver weight and decreased ALT compared to control group. For the oxidative stress, $LR_3$ acupuncture group significantly reduced reactive oxygen species, and $BL_{23}$ acupuncture group significantly reduced reactive oxygen species and inflammation-related protein compared to control group. But $LR_3$ acupuncture group and $BL_{23}$ acupuncture group didn't significantly reduce apoptosis-related protein. Therefore $LR_3$ and $BL_{23}$ acupuncture showed the effects of antioxidant and anti-inflammatory, especially $BL_{23}$ acupuncture was more effective than $LR_3$ acupuncture on the protection of liver in the oxidative stress.

파골세포 분화에 목향 물 추출물의 효과 (Effect of Water Extract of Saussureae Radix in RANKL-induced Osteoclast Differentiation)

  • Osteoporosis is social problem around the world, because fracture of old age may lead to critical medical condition. Osteoclast is a main target for prevention and treatment of osteoporosis due to their responsibility for bone resorption. Saussureae Radix has been known that has gastro-protective, bronchodilatory effect and has a anti-biotic effect. Saussureae Radix has been widely used in Oriental medicine. However, the effect of extract of Saussureae Radix in osteoclast differentiation remains unknown. Thus, we examined the effect of Saussureae Radix in receptor activator of nuclear factor-${\kappa}$B ligand (RANKL)-induced osteoclast differentiation. From the results of our study, Here we found that Saussureae Radix significantly inhibited osteoclast differentiation induced by RANKL. Saussureae Radix suppressed the activation of NF${\kappa}$B in bone marrow macrophages (BMMs) treated with RANKL. Also, Saussureae Radix significantly inhibited the mRNA expression of c-Fos, tartrate-resistant acid phosphatase (TRAP), osteoclast-associated receptor (OSCAR), nuclear factor of activated T cells (NFAT)c1 and cathepsin K in BMMs treated with RANKL. Particularly, Saussureae Radix greatly inhibited the protein expression of c-fos and NFATc1. especially in the case of NFATc1 expression, a master transcription factor of the differentiation of osteoclasts is very important step for osteoclastogenesis. These results demonstrate that Saussureae Radix may be useful treatment option of bone-related disease such as osteoporosis and rheumatoid arthritis.

Effect of the Inhibition of Platelet Activating Factor on Oxidative Lung Injury Induced by Interleukin-$1\;{\alpha}$

  • In order to know the pathogenesis of adult respiratory distress syndrome (ARDS) in association with the oxidative stress by neutrophils, the role of platelet activating factor (1-0-alkyl-2-acetyl-snglycero-3-phosphocholine, PAF) was investigated during acute lung injury induced by interleukin- $1{\alpha}$ (IL-1) in rats. An insufflation of IL-1 into the rat's trachea increased the acetyltransferase activity in the lung and the increase of PAF content was followed. As evidences of acute lung injury by neutrophilic respiratory burst, lung leak index, myeloperoxidase activity, numbers of neutrophils in the bronchoalveolar lavage fluid, neutrophilic adhesions to endothelial cells and NBT positive neutrophils were increased after IL-1 treatment. In addition, a direct instillation of PAF into the trachea caused acute lung leak and the experimental results showed a similar pattern in comparison with IL-1 induced acute lung injury. For the confirmation of oxidative stress during acute lung leak by IL-1 and PAF, a histochemical electron microscopy was performed. In IL-1 and PAF treated lungs of rats, the deposits of cerrous perhydroxide were found. To elucidate the role of PAF, an intravenous injection of PAF receptor antagonist, WEB 2086 was given immediately after IL-1 or PAF treatment. WEB 2086 decreased the production of hydrogen peroxide and the acute lung leak. In ultrastructural study, WEB 2086 mitigated the pathological changes induced by IL-1 or PAF. The nuclear factor kappa B (NFkB) was activated by PAF and this activation was inhibited by WEB 2086 almost completely. Based on these experimental results, it is suggested that the PAF produced in response to IL-1 through the remodeling pathway has the major role for acute lung injury by neutrophilic respiratory burst. In an additional experiment, we can also come to conclude that the activation of the NFkB by PAF is thought to be the fundamental mechanism to initiate the oxidative stress by neutrophils causing release of proinflammatory cytokines and activation of phospholipase $A_2$.

Differential gene expression profiles of periodontal soft tissue from rat teeth after immediate and delayed replantation: a pilot study

  • Purpose: In dental avulsion, delayed replantation usually has an uncertain prognosis. After tooth replantation, complex inflammatory responses promote a return to periodontal tissue homeostasis. Various types of cytokines are produced in the inflammatory microenvironment, and these cytokines determine the periodontal tissue response. This study aimed to identify the gene expression profiles of replanted teeth and evaluate the functional differences between immediate and delayed replantation. Methods: Maxillary molars from Sprague-Dawley rats were extracted, exposed to a dry environment, and then replanted. The animals were divided into 2 groups according to the extra-oral time: immediate replantation (dry for 5 minutes) and delayed replantation (dry for 60 minutes). Either 3 or 7 days after replantation, the animals were sacrificed. Periodontal soft tissues were harvested for mRNA sequencing. Hallmark gene set enrichment analysis was performed to predict the function of gene-gene interactions. The normalized enrichment score (NES) was calculated to determine functional differences. Results: The hallmark gene sets enriched in delayed replantation at 3 days were oxidative phosphorylation (NES=2.82, Q<0.001) and tumor necrosis factor-alpha (TNF-α) signaling via the nuclear factor kappa light chain enhancer of activated B cells (NF-κB) pathway (NES=1.52, Q=0.034). At 7 days after delayed replantation, TNF-α signaling via the NF-κB pathway (NES=-1.82, Q=0.002), angiogenesis (NES=-1.66, Q=0.01), and the transforming growth factor-beta signaling pathway (NES=-1.46, Q=0.051) were negatively highlighted. Conclusions: Differentially expressed gene profiles were significantly different between immediate and delayed replantation. TNF-α signaling via the NF-κB pathway was marked during the healing process. However, the enrichment score of this pathway changed in a time-dependent manner between immediate and delayed replantation.

Therapeutic effects of paeoniflorin on irritable bowel syndrome in rats

  • Background: Irritable bowel syndrome (IBS) is a functional bowel disorder (FBD). Objectives: To assess the therapeutic effects of paeoniflorin (PF) on IBS in rats. Method: Sixty male Sprague-Dawley rats were randomly divided into normal, model, positive drug, low-dose PF, medium-dose PF and high-dose PF groups (n = 10). After gavage for 2 consecutive weeks, the effect of PF on abdominal pain symptoms was assessed based on the abdominal withdrawal reflex (AWR) score, fecal water content and pathological changes in colon tissues. D-lactate, interleukin-1β (IL-1β), transforming growth factor-β (TGF-β) and tumor necrosis factor-α (TNF-α) were detected by enzyme-linked immunosorbent assay, and phosphorylated nuclear factor kappa B (p-NF-κB) p65 was detected by Western blotting. The abundance and diversity changes of intestinal flora were explored using 16S ribosomal RNA sequencing. Result: In PF groups, the mucosal morphology of colon tissues was intact, and the glands were arranged neatly and structured clearly, without obvious inflammatory cell infiltration. Compared with the model group, PF groups had significantly elevated pain threshold, and mRNA and protein levels of zonula occludens-1 (ZO-1) and occludin, decreased AWR score at 20 mmHg pressure, fecal water content, mRNA levels of IL-1β, TGF-β, and TNF-α, protein level of p-NF-κB p65 and level of serum D-lactate, and reduced levels of serum IL-1β, TGF-β, and TNF-α (p < 0.05, p < 0.01). PF groups had higher abundance of Lactobacillus, Akkermansia, Alistipes, and Bacteroides, but lower abundance of Desulfovibrio, Parasutterella, and Enterococcus than those of the model group. Conclusions: PF exerts therapeutic effects on IBS in rats probably by regulating the intestinal flora, and then up-regulating the expressions of ZO-1 and occludin in colon tissue while down-regulating the levels of IL-1β, TGF-β, TNF-α, D-lactate and p-NF-κB p65.
