Headspace법에 의한 솔향의 분석 (Comparisons of Volatile Compounds Extracted from Pinus densiflora by Headspace Analysis)

  • Pinus densiflora(2엽송) 1년생을 채취하여 가지, 잎, 새순 부분으로 구분하고, 시료병에 넣어 밀봉한 뒤, solide phase microextraction (SPME)과 dynamic headspace analysis(DHA)를 이용하여 휘발성 성분을 분리, 동정하였다. SPME와 DHA의 결과를 terpenes의 구성 성분에 따라 monoterpenes과 sesquiterpenes, oxygenated terpenes으로 나누어 조성비를 살펴보면, monoterpenes의 조성은 잎의 경우 각각 66.7%와 14.3%, 새순은 90.6%와 0.7%, 가지에서는 90.6%와 1.2%로 나타났다. Sesquiterpenes의 함량은 SPMe의 결과에서만, 잎이 25.8%, 가지 4.4%, 새순 1.5%로 나타났다. 산소를 함유하고 있는 terpenes의 함량은 새순 부위가 4.7%, 79.0%로 나타났으며, 가지 부위는 3.7%와 70.4%, 잎에서 1.0%와 50.7%를 각각 차지하였다. SPME의 결과는 이전의 용매추출을 통한 결과와 다소 비슷한 양상을 보였으며, DHA의 결과는 10$0^{\circ}C$이상의 고비점 화합물들이 나타나지 않은 반면, 5$0^{\circ}C$이하의 저비점 화합물이 15개 분리되었다.

건강한 성인 지원자를 대상으로 한 필로칼핀 저작정의 타액분비 유도 및 타액중 용출패턴 평가 (Evaluations on Salivary Flow Induction and Dissolution Patterns in Saliva of Pilocarpine Chewing Tablet in Healthy Human Volunteers)

  • Xerostomia is caused by organic or functional changes affecting the salivary system at different levels. Patients suffering from xerostomia may also complain of an oral burning sensation, ulceration or soreness, difficulty in swallowing, and poor denture retention. And pilocarpine is administered orally to induce salivary secretion. In Seoul National University Hospital(SNUH) pharmacy, the pilocarpine chewing tablets are prepared and supplied to patients of xerostomia in request of the dental hospital in SNUH. And we tested the salivary flow induction and the dissolution patterns of these products in saliva by a double-blind, sequential cross-over trials to eight healthy human volunteers with placebo. The pilocarpine chewing tablet contained 5 mg of pilocarpine, and placebo consisted of same materials as test drug, but didn't contain pilocarpine. In vivo experiment, all subjects were instructed to chew as 60-80 times/min. Mixed saliva was collected in the ranges of intervals such as 0-2, 2-5, 5-10, 10-15, 15-20, 20-30, 30-45 and 45-60 min after pilocarpine chewing tablet or placebo administration. Saliva volume was measured in each collecting time interval, and saliva pilocarpine concentrations were determined by reversed phase HPLC. The 82.5 percent $(4.13{\pm}0.69\;mg)$ of pilocarpine was extracted from chewing tablets during mastication of 60-80 times per minute for 60 minutes. Among these dissolved amounts, 90 percent was extracted within 20 minutes. The salivary flow rates were more increased in a group who administered pilocarpine chewing tablet at the interval of 5-10, 10-15, 20-30 and 45-60 min rather than a placebo-group, but only extracted amount of pilocarpine at 45-60 min interval is significanly different between two groups (p<0.05). But total amounts of saliva secreted for 1 hour in two group-pilocarpine and placebo treated- were $46.36{\pm}9.72\;ml\;and\;39.09{\pm}7.81\;ml$, respectively, and were not significantly different between two groups.

홍삼박으로부터 산성다당체의 최적 추출 조건 분석 (Conditioning of the Extraction of Acidic Polysaccharide from Red Ginseng Marc)

  • This study was carried out to investigate the optimum conditions for extraction of acidic polysaccharides from red ginseng marc produced by manufacturing alcoholic extract from red ginseng. Method of carbazole-sulfuric acid was applied to determine the amount of acidic polysaccharides in red ginseng marc. The amounts of acidic polysaccharides in water extract of red ginseng marc were increased with increasing extraction temperature. The contents of acidic polysaccharides were not significantly different despite of the extraction time increasing from 6 hours to 48 hours. The contents of starch in water-extract of red ginseng marc were increased with increasing extraction temperature. The starch amounts in water extract of red ginseng marc extracted for 48 hours were increased. The yields of polysaccharide precipitated from water-extract of red ginseng marc were increased with increasing extraction temperature. The hydration rate of acidic polysaccharides and starch from water-extract of red ginseng marc were decreased with increasing extraction temperature. The contents of starch were not significantly different despite of the extraction time increasing from 6 hours to 48 hours at $8^{\circ}C$. However, the rehydration rate of acidic polysaccharide for 48 hours were decreased at $8^{\circ}C$. The rehydration rate of acidic polysaccharide and starch extracted from 6 hours to 24 hours at $25^{\circ}C$ were not significantly different, but those extracted for 48 hours were increased. From the above results, we suggest that by altering the extraction conditions in red ginseng marc it is possible to develop optimum conditions for extraction that modulate the proportions of acidic polysaccharide and starch.

솔 부위 및 추출 용매를 달리한 솔향의 분석 (Comparisons of Volatile Compounds of Pinus densiflora on Kinds of Extraction Solvent and Parts of Pinus)

  • Volatile compounds from twigs, needles and sprouts in Pinus densiflora were extracted with n hexane, diethyl ether or ethanol for 24 hours, and the extracted compounds were separated and identified by gas chromatography and mass selective detector. The kinds and amount of volatile compounds extracted from three parts of Pinus densiflora were different in solvent extraction and the extraction by the modified Liken Nickerson apparatus. The contents of volatile compounds of twigs contained more than those of needles and sprouts, and the volatile compounds were extracted more in n hexane than the others. In the extraction with hexane, the main volatile compounds of twigs were 18.5% pinene, 14.5% limonene, 12.7% pinene and 3.2% myrcene. Sprouts were 16.8% limonene, 4.4% pinene, 4.3% pinene and 1.7% myrcene. Needles contained 14.7% pinene, 5.4% pinene, 2.2% limonene and 0.8% myrcene. The highest yield for pine aroma was shown in the extraction from pine twigs with n hexane, and in this extraction the amounts of pinene, pinene and limonene were 742 g, 1108 g and 922 g per gram sample, respectively.

추출조건 및 방법에 따른 인삼내의 panaxydol과 panaxynol의 추출율 비교

  • The extraction efficiencies of panaxynol and panaxydol according to extraction temperature were the highest at $80^{cdot}C$ among 65, 80, $95^{cdot}C$ with soxhlet and increased with shaking method when temperature increased from $25^{cdot}C$ to $45^{cdot}C$. The amounts of panaxynol and panaxydol were determined by gas chromatography. In time dependence of extracted amounts of panaxynol and panaxydol using shaking method, the efficiencies of panaxynol and panaxydol were increased during 12 hour. The effect of water swelling on panaxynol and panaxydol extraction efficiency using soxhlet and shaking methods, the efficiencies of panaxynol and panaxydol were decreased as swelling time was increased.

Amylase를 이용한 인삼엑기스의 제조 (The Studies on the Production of Ginseng Extract by Amylase)

  • It order to extract the soluble organic substances of Korean ginseng effectively, the ginseng extract have been made by using amylase. The investigation on the optimum condition of enzyme reaction was carried out, and the amounts of gained extract and its saponin pattern were compared among the ethanol extract, water extract and enzyme extract. The results obtained are summerized as follows. 1. The gaining ratio or ginseng extract was the highest value when the raw ginseng and dried ginseng were extracted in the concentration of 7.5% and 5% with 0.3%∼0.6% enzyme for 25 hour. 2. The amounts of ethanol extract, water extract and enzyme extract were 9.14%, 17.23% and 23.73% in case of raw ginseng and 64.09%, 72.52% and 74.36% in case of dried ginseng, respectively. The amount of enzyme extract was increased as much as 6∼14% in case of raw ginseng, and 2∼10% in case of dried ginseng compared with that of ethanol and water extract. 3. The absolute content of saponin was nearly constant in spite of the different extraction method and all of the ginseng saponin pattern of thin-layer chromatograms were almost same.

추출 및 건조방법이 다시마 (Laminaria japonica) 알긴산의 물성에 미치는 영향 (Effects of Extracting and Drying Method on Physical Properties of Alginates from Sea Tangle, Laminaria japonica)

  • In order to choose the manufacturing method for extracting alginates from sea tangle, Laminaria japonica, three methods were applied. In Method I, alginates were extracted with NaOH solution from sea tangle powder and extracted alginates were precipitated and converted to alginic acid by $CaCl_2$ and HCI solution. Then alginic acid was converted to sodium alginates with $Na_2CO_3$ solution. Sodium alginates were precipitated with methyl alcohol and were resolved with hot water and this step was repeated three times. Method II was same to Method I except final step including that sodium alginates were precipitated and washed with methyl alcohol three times. Method III included that sodium alginates were extracted with $Na_2CO_3$ solution from sea tangle powder then sodium alginates were precipitated and washed with methyl alcohol three time. Extracting time increased with Increasing extracted alginates amounts but increasing rates were below $0.4\%/h.$ Alginates amounts recovered by Method III showed above 2 times more to those by Method I and II. Extracting time increased with increasing ash amount of sodium alginates but increasing rates were below $0.1\%/h.$ and that of sodium alginates extracted by Method III showed higher value $(5\%)$ than those by Method I and II. In the sodium alginates prepared by Method III, the amount of ash in alginates dried by air was $34.4\%,$ that by vacuum freeze drying was $47.8\%.$ Extracting time increased with decreasing average molecular weight (MW) and degree of polymerization (DP) of sodium alginates, MW and DP of alginates prepared by Method III were higher than those by Method I and II In same extracting time. Extracting time increased with decreasing rate of apparent viscosity change (SAV) of alginates solution, and SAV of alginates prepared by Method III showed higher value than those by Method I and II in same extracting time. SAV of alginates dried by air was higher than that by vacuum freeze drying. Relating equation among SAV MW and DP were MW = 60.066 (SAV) -93.950, DP =309.760 (SAV) -485.084 and MW = 0.914 (DP)+0.213.

비정질 Ba-Ferrite로부터 추출한 BaFe12O19 입자의 특성 (Characteristics of BaFe12O19 Particles Extracted from Amorphous Ba-Ferrite)

  • Amorphous ferrites with composition B2O3-mBaO-nFe2O3 were prepared by an ultra rapidquenching technique. X-ray patterns and SEM reveal the products to be amrophous at room temperature. BaFe12O19 fine particles extracted from amorphous Ba-ferrites which additived Al2O3, SiO2, TiO2 and CoO have narrow size distribution, average 0.2$\mu\textrm{m}$, and coercive force, average 1400 Oe, increased with increasing TiO2 amounts. The obtained BeFe12O19 fine particles would be appreciated as perpendicular magnetic recording media.

표준시료를 이용한 토양중 중금속 분석방법 비교

  • The heavy metal analysis (Cd, Pb, Cu, Cr, Zn, Ni) with various extraction methods was performed using the certified sample and real soil sample. In case of the certified samples, $10.5{\sim}118%$ of recovery was showed with various ranges depend on the metal kinds and extraction methods. Also, the alkali-digestion method was showed the proper results by applying in hexavalent chromium. In case of real sample, compared to amounts of heavy metals extracted using 0.1N-HCl, those extracted using acid digestion are higher by $3{\sim}24$ times in Cu, $1.1{\sim}1.5$ times in Cd, $2{\sim}23$ times in Pb, $3{\sim}104$ times in Zn, $12{\sim}101$ times in Ni, $30{\sim}202$ times in Cr. There is no considerable difference between four acid digestion methods.

  • A sensitive method for the quantitation of oxandrolone in urine was developed using GC/MS. After oral administration of 10mg oxandrolone, oxandrolone excreted in urine as unchanged form was extracted in ether and derivatized to its O-TMS. Oxandrolone excreted in urine as glucuronide conjugated form was extracted after enzymatic hydrolysis and derivatized to its O-TMS. The amounts of oxandrolone-O-TMS was measured in GC/MS with selected ion monitoring. Calusterone, a structurally similar anabolic steroid, was employed as internal standard.
