Antioxidant Activity of Cercis chinensis and Its Protective Effect on Skin Aging

  • Reactive oxygen species are capable of damaging biomolecules such as lipids, proteins, and DNA, which can not only lead to various diseases, but also oxidative damage resulting aging. In our previous study, Cercis chinensis (Leguminosae) showed a potent antioxidant activity. Nineteen compounds were isolated through antioxidant activity-guided fractionation. The C. chinensis extract and some of the constituents exhibited a potent antioxidant activity on the free radicals and lipid peroxidation and a notable protective effect on the t-BuOOH induced oxidative damage. In vivo test of skin damage induced by UVB irradiation, the extract of C. chinensis and a constituent, piceatannol, exhibited a significant protective effect. The life-span of the HEK-N/F cells were extended by 1.21-2.12 fold as a result of the continuous administration of 3 $\mu\textrm{g}$/ml of the C. chinensis extract and the active constituents compared to that of the control. These observations were attributed to the inhibitory effect of the C. chinensis extract and its constituents on the age-dependent shortening of the telomere. Thus, C. chinensis was demonstrated to protect the skin cells against oxidative stress and inhibit thereby the cellular aging, followed by expectation as anti-aging cosmetic ingredient.

백두옹엑스 분획물의 소염진통작용 (The Anti-inflammatory and Analgesic Actions of the fractions from Pulsatilla koreana Root Extract)

  • From our previous report, the water extract of Pulsatilla koreana root was found to have potent anti-inflammatory and analgesic actions in intravenous administration in animals. Among chloroform, ethyl acetate, butanol and water fractions which were obtained through successive fractionation of the extract, only the water fraction was found to have the antiinflammatory and analgesic actions. The fraction did not affect normal body temperature at the effective doses in mice and showed low acute toxicity of which $LD_{50}$ was less than 500 mg/kg i.v. in mice. It is interesting that its anti-inflammatory action might be attributed in part to inhibition of cyclooxygenase-1 and -2.

  • The vascular relaxant effets of Alpinia officinarum(Zingiberaceae) were evaluated on isolated thoracic aorta from rats. The methanolic extract of rhizome of Alpinia officinarum caused a concentration-dependent relaxation of rat aortic preparations precontracted with $0.3{\mu}M$ phenylephrine, The intensive investigation of the extract by way of activity-guided fractionation led us to yield three kind of active components, galangin(1), kaempferide(2), and kaempferol(3), which were responsible for the vasodilating property of the extract. Stuctures of the isolated active components were established by chemical and spectroscopic means.

식중독 유발 세균의 증식에 미치는 겨우살이 추출물의 영향 (Antimicrobial Effect of Viscum album var. coloratum Extracts on Food-Borne Pathogens)

  • This study was performed to investigate the antimicrobial effect of the Viscum album var. coloratum extracts against food-borne pathogens. First, the Viscum album var. coloratum was extracted with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol, respectively. The antimicrobial activity of the Viscum album var. coloratum extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The petroleum ether extracts of Viscum album var. coloratum showed the highest antimicrobial activity against Bacillus cereus and Shigella dysenteriae. Synergistic effect in inhibition was observed when Viscum album var. coloratum extract was mixed with Perillae folium extract as compared with each extract alone. Finally, the growth inhibition curves were determined by using petroleum ether extracts of Viscum album var. coloratum against Bacillus cereus and Shigella dysenteriae. The petroleum ether extract of Viscum album var. coloratum had strong antimicrobial activity against Bacillus cereus at the concentration of 5,000 ppm. At this concentration, the growth of Bacillus cereus was retarded more than 24 hours and up to 12 hours for Shigella dysenteriae. In conclusion, the petroleum ether extracts of Viscum album var. coloratum inhibit efficiently Bacillus cereus and Shigella dysenteriae.

Anti-allergic Actions of the Leaves of Castnea crenata and Isolation of an Active Component Responsible for the Inhibition of Mast Cell Degranulation

  • The anti-allergic actions of the leaves of Castanea crenata (Fagaceae) were studied. The water extract demonstrated potent anti-allergic actions in in vivo and in vitro experiments. The oral or intraperitoneal administration of the extract (100 or 200 mg/kg) caused a significant inhibition of the 48 hr-PCA (up to 90%) and the vascular permeability induced by histamine or serotonin in rats (about 80%). The anaphylactic release of ${\beta}$-hexosaminidase for RBL-2H3 cells was also significantly inhibited by the extract in as dose-dependent manner with an IC50 value of 230 $\mu\textrm{g}$/ml. The activity-guided fractionation of the extract, based on the determination of inhibitor effect upon the release of ${\beta}$-hexosaminidase, led to the isolation of quercetin as an active principle responsible for the inhibition of degranulation.

왕겨추출물의 쥐소장점막 ${\alpha}-glucosidase$에 대한 in vitro에서의 저해효과 (In vitro Inhibitory Activity on Rat Intestinal Mucosa ${\alpha}-glucosidase$ by Rice Hull Extract)

  • 벼 가공공정의 부산물인 왕겨를 당뇨병 환자의 혈당상승을 억제하는 식품소재로 응용할 가능성을 검토하기 위하여, 왕겨 ethanol 추출물의 쥐 소장 점막의 ${\alpha}-glucosidase$에 대한 저해효과와, 저해성분의 정제에 관하여 연구하였다. 왕겨 ethanol 추출물은 0.8 mg/mL의 농도에서 6 mM p-nitrophenyl ${\alpha}-D-glucopyranoside$를 기질로 한 쥐소장 점막 1.4 mU/mL ${\alpha}-glucosidase$에 의한 효소반응을 30% 저해하였다. Ethanol 추출물을 n-hexane, chloroform, ethyl acetate, n-butanol, 물로 분획하였을 때 ethyl acetate 분획물의 ${\alpha}-glucosidase$에 대한 저해활성은 ethanol 추출물의 경우와 같은 조건에서 65%로서, 다른 네 분획물보다 높았다. Ethanol 추출물이 ${\alpha}-glucosidase$ 반응을 50% 저해하는 농도$(IC_{50})$는 162 mg/mL 인데 비하여, ethyl acetate 분획물의 $(IC_{50})$은 0.14 mg/mL 이었다. Ethyl acetate 분획물을 silica gel column chromatography에 의하여 분획하였을 때 chloroform : methanol=90 : 10 분획물의 ${\alpha}-glucosidase$에 대한 저해율은 90%이었으며, 이는 ethyl acetate 분획물의 저해율 65%에 비하여 증가한 것이다. Chloroform : methanol=90 : 10 분획물을 Sephadex LH-20 column chromatography에 의하여 분획하였을 때 중간 정도의 시간에서 용출된 분획물의 저해율은 ethanol 추출물과 같은 조건에서 88%이었다.

번행초 추출물의 항산화 활성 (Antioxidant Activities of the Solvent Extracts from Tetragonia tetragonioides)

  • 본 연구에서는 예로부터 위장병의 예방 및 치료 효과가 뛰어난 작물로 알려진 번행초(Tetragonia tetragonioides)의 각종 용매 추출 분획물의 항산화 효과를 명확히 하기 위해 총 폴리페놀과 플라보노이드 함량, 비타민 E함량, DPPH free radical 소거활성, Superoxide 음이온 소거활성, 과산화물생성 억제 활성, 환원능력 그리고 금속 킬레이트 효과를 측정하였다. 먼저 총 폴리페놀 화합물의 함량은 DCM 분획, EA 분획에서 높게 나타났고, 플라보노이드 함량은 HX 분획, EA 분획 순으로 나타났다. 비타민 E의 함량은 HX 분획, DCM 분획 순으로 높게 나타났다. DPPH radical 소거활성 효과는 DCM과 EA 추출 분획에서 $IC_{50}$ 값이 각각 $554.25{\mu}g/ml,\;394.96{\mu}g/ml$으로 산화방지제로 널리 이용되고 있는 BHT의 $784.7{\mu}g/ml$보다 우수하게 측정되었다. 과산화물 생성 억제 활성에서 번행초 분획은 지질단백질을 산화로부터 보호한다고 알려진 ${\alpha}$-tocopherol과 유사한 능력을 보였으나, 페놀계 합성 항산화제인 BHT, BHA에 비해서는 억제능이 다소 낮음이 확인되었다. 환원능력을 통한 항산화 활성 측정에서는 1.5 mg/ml농도에서 EA 분획의 활성이 ${\alpha}$-tocopherol보다 유의하게 높게 나타나 좋은 항산화 소재임이 확인되었다.

어류 알로부터 Protease Inhibitors의 단백질 용해도 차이에 의한 분획 특성 (Fractionation and Characterization of Protease Inhibitors from Fish Eggs Based on Protein Solubility)

  • A protease inhibitor was fractionated from fish eggs using methods based on protein solubility. Fractionation efficiency was evaluated with regard to percent recovery and total inhibitory activity (U). The fractionation of protease inhibitor (PI) from egg extracts of skipjack tuna (ST, Katsuwonus pelamis), yellowfin tuna (YT, Thunnus albacores), and Alaska pollock (AP, Theragra chalcogramma) was performed by precipitation with cold acetone or ammonium sulfate (AS). Fractions exhibiting the strongest inhibitory activity contained 20-40% (v/v) cold acetone or 40-60% saturated AS fractions. AS fractionation was more effective in isolating PI than was precipitation with acetone. The total inhibitory activity and percent recovery of fraction obtained with AS 40-60% toward trypsin and $N{\alpha}$-benzoyl-L-arginine-p-nitroanilide (BAPNA) were 4,976 U and 24.2% for ST, 3,331 U and 38.1% for YT, and 4,750 U and 43.8% for AP, respectively. In comparisons against six commercial proteases, 40-80% AS fractions, made by combining the 40-60% and 60-80% AS fractions from fish egg extract, exhibited the strongest inhibition of trypsin when using a casein substrate. These results suggest that fish eggs act as serine protease inhibitors and may be useful for protease inhibition in foodstuffs.

Acetylcholinesterase Inhibitors from the Roots of Angelica dahurica

  • In the course of finding Korean natural products for acetylcholinesterase (AChE) inhibitory activity, we found that a methanolic extract of the roots of Angelica dahurica showed significant inhibitory effects on AChE. Bioassay-guided fractionation of the methanolic extract resulted in the isolation of three furanocoumarins, isoimperatorin (1), imperatorin (2) and oxypeucedanin (3), as active principles. These compounds inhibited AChE activity in a dosedependent manner, and the $IC_{50}$ values of compounds 1-3 were 74.6, 63.7 and 89.1 uM, respectively.
