Antioxidant Activities of Agrimonia pilosa Ledeb: In Vitro Comparative Activities of Its Different Fractions

  • The antioxidant potentials of the methanol extract and its various fractions from Agrimonia pilosa Ledeb were measured as 1,1-diphenyl-2-picrylhydrazyl (DPPH), and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging abilities as well as the reducing power decreasing NO in vitro. The methanol extract of Agrimonia pilosa Ledeb showed significantly strong scavenging effects on free radicals of DPPH, ABTS, and NO. Hexane fraction (HF), ethyl acetate fraction (EF) and n-butyl alcohol fraction (BF) were prepared by solvent fractionation. By comparison of polyphenol contents among BF and HF, EF with high polyphenol contents showed the highest DPPH, ABTS, and NO scavenging effects with the value of 50.2%, 80.4%, and 65.7%, respectively, at the highest tested dose. HPLC analysis revealed the presence of various phenolic compounds in EF of Agrimonia pilosa Ledeb. These results suggest that EF could be considered as natural antioxidant sources and dietary nutritional supplements to prevent oxidation-related diseases.

Cytotoxic Polyacetylenes from Aralia cordata (독활의 세포독성 폴리아세틸렌 성분)

  • An n-hexane extract of the roots of Aralia cordata Thunb. (Araliaceae) was found to show significant in vitro cytotoxic activity against P388D$_{1}$ lymphocytic leukemia cell in culture. Bioactivity-directed fractionation of this extract led to the isolation of four polyacetylenes, falcarindiol (1), dehydrofalcarindiol (2), falcarindiol-8-acetate (3) and dehydrofalcarindiol-8-acetate (4). Cytotoxicity of compounds 1 and 3 was found to be better than that of compounds 2 and 4 when these compounds were tested against eight in vitro tumor cell lines, namely, A549, HCT15, DLD1, MCF7, SKOV3, HL60, K562 and P388D$_{1}$. The fact that the cytotoxicity of compounds 1 and 3 against series of tumor cell lines was much stronger than that of compounds 2 and 4 suggested that the saturated carbon chain at the termial and the hydroxyl group at the C-3 are important for the activities. The requirement for the activity was further confirmed by synthesizing and assaying the acetate derivatives of compounds 1 and 2.

  • The methanol extract of Arachis hypogaea shell showed antioxidative and antimicrobial activity. The methanol extract was successively purified by solvent fractionation, silica gel adsorption column chromatography, Sephadex LH-20 column chromatography and octadecylsilane column chromatography. The purified active substances were isolated by high performance liquid chromatography, and were identified as 3-methoxy-4-hydroxybenzoic acid and 4-hydroxybenzoic acid by LC-MS and GC-MS. The amount of 3-methoxy-4-hydroxybenzoic acid and 4-hydroxybenzoic acid were 3.8mg and 9.8 mg per kg of shell, respectively.

  • The methanolic extract of the roots of Lithospermum erythrorhizon (Boraginaceae) was found to show inhibitory activity towards farnesyl protein transferase (FPTase). Bioassay-guided fractionation of the methanolic extract resulted in the isolation of three naphthoquinone compounds, as inhibitors of FPTase. These compounds inhibited the FPTase activity in a dose-dependent manner.

  • Prunus mandshurica var. glabra Nakai (Rosaceae) is widely distributed in South Korea and bears a fruit with a bitter and astringent taste. An ethyl acetate-soluble extract of Prunus mandshurica was found to exhibit significant cytotoxicity against human leukemia cell lines. Bioassay-directed fractionation of this extract using an MTT(3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) cell proliferation assay as a monitor led to the isolation of the bioactive compounds. Two compounds, 1 and 2 were subsequently found to mediate cytotoxicity against U937, human monocytic leukemia cells. The 50% growth inhibitory concentrations ($IC_{50}$/) of compounds 1 and 2 on U937 were 40 and 62 $\mu\textrm{g}$/mL, respectively.

  • The methanol extract from the whole parts of the flowers of Chrysanthemum boreals was found to exhibit a significant inhibition of a rat lens aldose reductase (RLAR) activity in vitro. Bioassay guided systematic fractionation of the methanol extract led to the isolation of four flavonoids which were identified as acacetin (1), apigenin (II), luteolin (III) and linarin (IV). Compounds I-III were demonstrated to exhibit a significant inhibition of RLAR. Luteolin (II) was found to be the most potent AR inhibitor with $IC_{50}$ value of $5{\times}10^{-7}M$.

  • In the course of finding Korean natural products with acetylcholinesterase (AChE) inhibitory activity, we found that a methanolic extract of the twigs of Vaccinium oldhami significantly inhibited AChE. Bioassay-guided fractionation of the methanolic extract resulted in the isolation of two compounds, taraxerol (1) and scopoletin (2), as active constituents. These compounds inhibited AChE activity in a dose-dependent manner, and the $IC_{50}$/ values of compounds 1 and 2 were 33.6 (79 $\muM$) and 10.0 (52 $\muM$) $\mu$g/mL, respectively.

  • As an attempt to search for bioactive natural constituents exerting antinociceptive and antiinflammatory activities, we examined the potency of the extract of R. coreanus fruits by the activity-guided fractionation. The EtOAc- and BuOH fraction and those alkaline hydrolysates showed significant antinociceptive effects as assessed by writhing-, hot plate- and tail flicks tests in mice and rats as well as antiinflammatory effect in rats with carrageenan-induced edema. BuOH extract was subjected to column chromatography to obtain a large amount of niga-ichigoside F$_1$ (1, 23-hydroxytormentic acid 28-O-glc), which was again hydrolyzed in NaOH solution to yield an aglycone 23-hydroxytormentic acid (1a).   • These experiments were conducted to purify the glucoamylase produced by Rhizopus oryzae. Two forms of glucoamylase (GI and GII) from Phizopus oryzae were purified by $(NH_2)_2SO_4$ fractionation, acetone fractionation and successive column chromatography on DEAE-cellulose and CM-cellulose. The specific activities of GI and GII toward soluble starch were 157.6 U/㎎. protein (37.5 fold of crude extract), and 164.7 U/㎎. protein (39.2 fold of curde extract), respectively, and the yields of them were 4.3% and 3.8%, respectively. The two purified enzymes have shown a single band by polyacrylamide disc gel electrophoresis and SDS-polyacrylamide gel electrophoresis. The protein bands of their electrophoresis gel were revealed to have glucoamylase activity by iodine staining and were proved to be glycoprotein by periodic acid Schiff's staining.
