We investigated the optimal condition for the production of extracellular pretense(a cuticle-degrading pretense) from entomopathogenic fungus Beauveria bassiana(ATCC7159) in liquid medium by adding of gelatin, bovine serum albumin(BSA), casein and polypeptone. The optimal induction medium for production of extracellular pretenses is composed of 0.5% polypeptone, trace elements and 50 mM potassium phosphate(pH 6.0). In this condition, the production of extracellular pretenses increased rapidly after the 24hrs, peaking at the third day and there was little inductive effect in culture broth more than pH 7.0. The pretenses were inhibited by phenyl methyl sulfonyl fluoride(PMSF). High activity of pretense was showed both range of pH 8.5 and 11.5 and also detected by three different portions of slice gel derived from non-denaturing isoelectricfocusing gel. At least three different extracellular pretenses are produced in optimal production medium when polypeptone is used as the sole carbon and nitrogen source.
From the culture supernatant of the psychrotrophic strain of Bacillus amyloliquefaciens an extracellular serine protease was purified to apparent homogeneity by successive purification steps using QAE-Sephadex, SP-Sephadex and Sephacryl S-100 column chromatography. The pretense is monomeric, with a relative molecular mass of 23,000. It is inhibited by the serine protease inhibitor phenylmethylsulfonyl fluoride, but not by EDTA. The enzyme is most active at pH 9-10 and at $45^{\circ}C$, although it is unstable at $60^{\circ}C$.
The purpose of this study was to investigate the practical availability of pretense production that can be used at home after isolating Serratia sp.2000-1 which produced extracellular pretense from clinical specimen. Basic production conditions and partial enzymatic characteristics of pretense produced by Serratia sp. 2000-1 was as follows: The kind and concentration of carbohydrate, nitrogen and metal salts for optimal enzyme production condition were each identified as the concentration of 1.5% glucose, 2.0% CSP, and 0.1% CaCl$_2$, and the optimal temperature, time and initial pH for culture were each 3$0^{\circ}C$, 72 hours, and pH 8.0. The final enzymatic yeild that was purified by 3 steps with ammonium sulfate precipitation (45~80%), DEAE-cellulose column chromatography, and Sephadex G-200 gel chromatography was 14.4%, and enzyme inactivity rate increased approximately 291314s. The optimal temperature and pH for purified pretense activity were 35$^{\circ}C$ and pH 7.0~8.0, and purified pretense activity was relatively stable by 4$0^{\circ}C$ at pH 6~10 for 30 min, however heating at 6$0^{\circ}C$ for 30 min, it liminated detectable pretense activity. The pretense activity was activated by $Mg^{2+}$, $Ba^{2+}$, $Ca^{2+}$, Mn$^{2+}$, but inactiviaed by Hg$^{2+}$, Ag$^{2+}$, Cu$^{2+}$, and the pretense activity was inhibited strongly by SDS among enzyme activity inhibitors. Further study is required to evaluate the practical availability of pretense production that can be used at home by isolating Serratia sp. from more clinical specimen and examining pretense more in details.
An extracellular pretense of Bacillus amyloliquefaciens S94 was purified to apparent homogeneity. The enzyme activity was strongly inhibited by general inhibitor for serine protease, PMSF, suggesting that the enzyme is a serine pretense. The purified enzyme activity was inhibited by leucine peptidase inhibitor, bestatin, suggesting that the enzyme is a leucine endopeptidase. The maximum proteolytic activity against different protein substrates occurred at pH 10, 45$^{\circ}C$ (protein substrate) and pH 8, 45$^{\circ}C$ (synthetic substrate). The purified enzyme was specific in that it readily hydrolyBed substrates with Leu or Lys residues at P$_1$ site. The pretense had characteristics of a cold-adapted protein, which was more active for the hydrolysis of synthetic substrate in the range of 15$^{\circ}C$ to 45$^{\circ}C$, specially at low temperature.

In order to develope a new pretense applicable to industries, a bacterium which produces a remarkable amount of extracellular pretense were isolated from soil. About 10 bacterial strains producing pretense were isolated from samples of soil, and strain PANH765 showed the highest activity of pretense production among them. The strain was identified as Bacillus subtilis according to the Bergey's Manual of Systematic Bacteriology based on its morphological, cultural and physiological characteristics. B. subtilis PANH765 showed the maximal production of pro-tease in the medium containing 2.0% glucose, 1.0% yeast extract, 0.2% ammonium nitrate, 0.02% ferrous sulfate and 0.02% dipotassium hydrogen phosphate. Under the optimal condition with temperature of 3$0^{\circ}C$, initial pH of 7.0 and shaking speed of 150 rpm, the pretense production reached a maximum level with 36 hr cultivation (6.34 U).
Proteases are degradative enzymes which hydrolyze a peptide bond between amino acids and they are abundantly applied to commercial field. In order to screen new source of pretense, bacteria secreting extracellular pretense were isolated by enrichment culture from deep sea water samples of East Sea, Korea. A bacterium, named as HJ19, showed the best growth and the largest clear zone in plates supplemented skim milk at $30^{\circ}C$. The partial DNA sequence analysis of the 16S rRNA gene, phenotypic tests and morphology identified that this strain was In genus Micrococcus. The strain HJ19 could not grow at $10^{\circ}C$ but it started growth and showed pretense activity at $20^{\circ}C$. The optimal growth was at $37^{\circ}C$ and the maximal protease activity at $30^{\circ}C$ was about 480unit/ml.
V parahaemolyticus possessed an extracellular alkaline pretense activity during the stationary growth phase. Various factors such as nitrogen sources, the concentration of NaCl and metal ions were investigated for optimizing the production of alkaline pretense from V. parahaemolyticus ATCC 17802. Among the nitrogen sources tested skim milk showed the distinct increase of the activity and the activity was the highest at 2% in final concentration after 60 hours incubation. The addition of NaCl and metal ions did not increase the alkaline pretense activity. CoC$_2$, CuC1$_2$, and HgCl rather highly inhibited alkaline protease production.
In this study, the biochemical characterization of strong proteolytic bacteria and their extracellular pretense isolated from fermented fish paste were experimented for the purpose of industrial large scale-production by accelerated fermentation. The results obtained were as follows: Among 4 strains isolated from fermented fish paste, B. subtilis p-4 and B. licheniformis p-5, which grow well at $40^{\circ}C$, pH 7.0 and $1\%$ of salt contents, were the best proteolytic bacteria and were shown $0.48hr^{-1}$, $0.49hr^{-1}$ of specific growth rate in TPY medium, respectively. Maximum enzyme activity of B. subtilis p-4 was 335n mole-Tyr/min.ml after 30 hrs and that of B. licheniformis p-5 was 300n mole-Tyr/min.ml after 28 hrs of shaking culture. Purified pretense produced by B. subtilis p-4 and B. licheniformis p-5 showed maximum activity at $50^{\circ}C$, pH 7.0 and molecular weight were estimated to be 18,000, 30,000 by sephadex G-100 gel filtration, respectively. These were supposed to be a kind of metal chelator sensitive neutral pretense from the result of high sensitivity against EDTA, o-phe-nanthroline and metal ions such as $Cu^{2+},\;Ni^{2+},\;Zn{2+}$.
Extracellular protease (bassiasin I), from the culture filtrate of entomopathogenic fungus Beauveria bassiana ATCC7159, was successively purified by precipitation with ammonium sulfate followed by DEAE-Sephadex A-50, CM-cellulose and Hydroxyapatite column chromatography. A typical procedure provided 41-fold purification with 13.6% yield. The molecular weight of the purified pretense (bassiasin I) was found to be approximately 32,000 by SDS-PAGE. Isoelectric-focusing analysis of the enzyme showed a pI of 9.5. $NH_2-terminal$ sequence of the pretense showed homology with those of the fungal proteases. The enzyme has an optimal pH for activity at 10.5 and is stable over pH 5.0-11.0. The maximum activity of the enzyme was at $60-65^{\circ}C$, and approximately 20% activity remained at $60^{\circ}C$ after 120 min. The pretense was inhibited by phenylmethylsulfonyl fluoride (PMSF) and diisopropyl fluorophosphate (DIPF).
The optimum temperature and pH for mycelial growth of B. bassiana DGUM 34001 were $24^{\circ}C$ and pH 7.0, respectively. Among the complex media used, mushroom complex medium (MCM) was the most favorable for mycelial growth. When Czapek-Dox medium was used as a minimal medium, glucose was an excellent source for carbon and energy. Soytone and sodium phosphate were favorable constituent for culture medium as a source of organic nitrogen and phosphorus, respectively. When the fungus was grown in MCM broth, the specific activity of extracellular enzyme of ${\alpha}$-amylase, lipase, chitinase, CMCase and pretease were 297.0, 0.058, 0.33, 0.21 and 22.8 units/mg protein, respectively. When various sources of organic nitrogen and chitin were supplemented to determine the production of enzymes, casein and soluble chitosan enhanced the production of extracellular protease and chitinase.
