Identification of Dinitrotoluene Selective Peptides by Phage Display Cloning

  • Biomolecules specific to explosives can be exploited as chemical sensors. Peptides specific to immobilized dinitrotoluene (DNT) were identified using a phage display library. A derivative of DNT that contained an extended amine group, 4-(2,4-dinitrophenyl)butan-1-amine, was synthesized and immobilized using a self-assembled monolayer surface on gold. Filamentous M13 phages displaying random sequences of 12-mer peptides specific to the immobilized DNT-derivate were isolated from the M13 phage library by biopanning. A common peptide sequence was identified from the isolated phages and the synthesized peptides showed selective binding to DNT. When the peptide was immobilized on a quartz crystal microbalance (QCM) chip, it showed a binding signal to DNT, while toluene barely showed significant binding to the QCM chip. These results demonstrate that peptides screened by biopanning against immobilized DNT can be useful for quick and accurate detection of DNT.

Development of a Single Chain Antibody Using a Phage Display Cloning Method for the Detection of 2,4-Dinitrotoluene

  • Single-chain variable fragments of antibodies (scFv) specific to 2,4-dinitrotoluene (DNT) were isolated from a phage library displaying synthetic human scFv fragments with 6 diversified complementary determining regions (CDRs). A DNT derivative that contained an extended amine group was synthesized and conjugated to the NHS-group that was linked to magnetic beads. Phages specific to the immobilized DNT derivatives were isolated from the library after 4 rounds of sequential binding and elution processes. The displayed scFv fragments from the isolated phages showed consensus CDR sequences. One DNT-specific scFv was expressed in E. coli and purified using Ni-affinity chromatography. The purified DNT-specific scFv binds specifically to the immobilized DNT-derivative with $K_D$ value of $6.0{\times}10^{-7}$ M. The scFv and DNT interaction was not disrupted by the addition of 4-nitrotoluene or benzoic acid. These data demonstrate that the screened scFv from the phage displayed library could be used for selective and sensitive detection of explosives such as TNT.

  • The performance characteristics of aluminized high explosive are considered by varying the aluminum(Al) concentration in a two-phase model. Since the time scales of the characteristic combustion process of high explosives and Al particles differ, the process of energy release behind the leading detonation wave front occurs over an extended period of time. Two cardinal observations are reported: a decrease in detonation velocity with an increase in Al concentration and a double front detonation (DFD) feature when anaerobic Al reaction occurs behind the front. In the current study, a series of confined rate sticks are considered for characterizing the performance of aluminized HMX with a maximum Al concentration of 50%. The simulated results are compared with the experimental data for 5%-25% concentrations.

Shifts in Biochemical Environments and Subsequent Degradation of Explosive Compounds (TNT and RDX) by Starch Ball Addition in the Benthic Zone of Bench Scale Settling Basins (전분 환 투입에 의한 실험실 규모 침전지 저부에서의 생화학적 환경 변화와 화약물질(TNT 및 RDX) 분해)

  • A starch ball was devised to conveniently supply carbon source to indigenous microorganisms and to enhance biotransformation of explosive compounds(TNT and RDX) in the sediments of settling basins installed in military shooting ranges. To identify optimum dose/sediment ratio for degradation of explosives in the basin, a series of bench scale settling basin experiments were performed for 30 days while monitoring supernatant pH, DO, concentrations of nitrite, nitrate, sulfate, explosive compounds, and acute toxicity measured by bacterial luminescence. Addition of starch ball induced changes in oxidation conditions from oxic to anoxic in the benthic zone of the basin, which resulted in subsequent reductive degradation of both TNT and RDX in the liquid and solid phase of basin. However, fermentation products of excess starch, acetic acid and formic acid, caused acute toxicity in the liquid phase. The optimum ratio of starch ball/sediment for explosive compounds degradation by inducing changes in bio-geochemical environments without increase in acute toxicity, was found to be 0.009~0.017.

  • A bench-scale anoxic membrane bioreactor (MBR) system, consisting of a bioreactor coupled to a ceramic crossflow ultrafiltration module, was evaluated to treat a synthetic wastewater containing alkaline hydrolysis byproducts (hydrolysates) of RDX, The wastewater was formulated the same as RDX hydrolysates, and consisted of acetate, formate, formaldehyde as carbon sources and nitrite, nitrate as electron accepters. The MBR system removed 80 to 90% of these carbon sources, and approximately 90% of the stoichiometric amount of nitrate, 60% of nitrite. The reactor was also operated over a range of transmembrane pressures, temperatures, suspended solids concentration, and organic loading rate in order to maximize treatment efficiency and permeate flux. Increasing transmembrane pressure and temperature did not improve membrane flux significantly. Increasing biomass concentration in the bioreactor decreased the permeate flux significantly. The maximum volumetric organic loading rate was $0.72kg\;COD/m^3/day$, and the maximum F/M ratio was 0.50 kg N/kg MLSS/day and 1.82 kg COD/kg MLSS/day. Membrane permeate was clear and essentially free of bacteria, as indicated by heterotrophic plate count. Permeate flux ranged between 0.15 and $2.0m^3/m^2/day$ and was maintained by routine backwashing every 3 to 4 day. Backwashing with 2% NaOCl solution every fourth or fifth backwashing cycle was able to restore membrane flux to its original value.

  • In the present research paper large scale blasting was conducted on two different firing patterns, namely, straight V type and skewed V type pattern on the same sandstone overburden bench with similar explosives. The post-blast fragmentation assessments were made by use of digital imaging technique. The total cycle time of 10 $m^3$ rope shovels was also recorded in the field. The results reveal improvements in the fragmentation and excavator performance results for the blasts fired on skewed V type pattern. The paper discusses the skewed V firing pattern and the reasons for its superior performance vis-$\grave{a}$-vis the straight V type pattern.

  • The thermal property is one of the most important characteristics in the field of energetic materials. Because energy materials release decomposition heat, differential scanning calorimetry (DSC) is frequently used for thermal analysis. However, thermodynamic events, such as melting can interfere with DSC kinetic analysis. In this study, we use isothermal mode for DSC measurement to avoid thermodynamic issues. We also merge accelerating rate calorimetry(ARC) data with DSC data to obtain a robust prediction results for small scale samples and for large scale samples as well. For the thermal property prediction, advanced kinetics and technology solutions(AKTS) programs are used.

  • The development of explosive detection technology in a security environment and fear of terrorism at homeland and abroad has been one of the most important issues. Moreover, research works on the explosive detection are highly required to achieve domestic production technology due to the implementation of aviation security performance certification system. Traditionally, explosives are detected by using classical chemical analyses. However, in the view of high sensitivity, rapid analysis, miniaturization and portability electrochemical methods are considered as promising. Most of electrochemical explosive detection technologies are developed in USA, China, Israel, etc. This review highlights the principle and research trend of electrochemical explosive detection technologies carried out overseas in addition to the research direction for future exploration.

Fabrication and Characterization of Highly Reactive Al/CuO Nano-composite using Graphene Oxide (산화그래핀을 적용한 고반응성 Al/CuO 나노복합재 제조 및 분석)

  • The aluminum (Al)/copper oxide (CuO) complex is known as the most promising material for thermite reactions, releasing a high heat and pressure through ignition or thermal heating. To improve the reaction rate and wettability for handling safety, nanosized primary particles are applied on Al/CuO composite for energetic materials in explosives or propellants. Herein, graphene oxide (GO) is adopted for the Al/CuO composites as the functional supporting materials, preventing a phase-separation between solvent and composites, leading to a significantly enhanced reactivity. The characterizations of Al/CuO decorated on GO(Al/CuO/GO) are performed through scanning electron microscopy, transmission electron microscopy, and energy dispersive X-ray spectroscopy mapping analysis. Moreover, the functional bridging between Al/CuO and GO is suggested by identifying the chemical bonding with GO in X-ray photoelectron spectroscopy analysis. The reactivity of Al/CuO/GO composites is evaluated by comparing the maximum pressure and rate of the pressure increase of Al/CuO and Al/CuO/GO. The composites with a specific concentration of GO (10 wt%) demonstrate a well-dispersed mixture in hexane solution without phase separation.

Social Disaster Adaptation Experiences of Railroad Workers: Focused on the Iri Station Explosion of 1977 (철도종사자의 사회 재난 적응 경험: 1977년 이리역 폭발 사고를 중심으로)

  • Purpose: The Iri station explosion that occurred in 1977 was a major social disaster in Korea, caused by a fire in a train equipped with explosives. The purpose of this study was to investigate the social disaster adaptation experiences of railroad workers. Methods: This study was based on qualitative research using phenomenological methodology. Participants were six railroad workers who experienced the Iri station explosion. Data were collected through in-depth interviews with individual workers from March to June, 2018. The data analysis method was based on Colaizzi's approach. Results: Experiences of railroad workers were categorized into 12 themes and the following 6 theme clusters: (1) Anxiety due to the extreme vibration and crash, (2) Terror regarding the horrible situation that one cannot face, (3) Anger about the cause of the explosion and a sense of relief about survival, (4) Confusion regarding different rumors, (5) Various efforts to return to daily life, and (6) Trauma that continues to exist. Conclusion: The findings of this study recommend that railroad organizations and managers should pay attention to enhance disaster preparedness and develop organizational disaster coping guidelines for members. The results of this study can help us to better understand the various aspects of the Iri station explosion of 1977.
