Successful genetic transformation of plants requires non-chimeric selection of transformed tissues and their subsequent regeneration. With rare exceptions, most transformation protocols still rely heavily on antibiotics for selecting transgenic cells that contain an antibiotic-degrading selectable marker gene. Here, the morphogenic capacity of in-vitro explants of chrysanthemnum and tobacco stems and leaves (control and transgenic) changed with the addition of aminoglycoside antibiotics (AAs), In a test of 6 AAs, phytotoxicity occurred at concentrations of 10 to 25 and 50 to 100$\mu\textrm{g}$$mL^{-1}$ in chrysanthemum and tobacco explants, respectively. Light conditions as well as explant source and size also had significant effects. The use of transverse thin cell layers (tTCLs), in conjunction with high initial AA selection levels, supported the greatest regeneration of transgenic material (adventitious shoots or callus) and the lowest number of escapes. Flow-cytometric analyses revealed no endodu-plication in chrysanthemum, even at high AA levels. However, this phenomenon was observed in tobacco calli(8C or more), even at low AA concentrations (i.e., 5 to 10 $\mu\textrm{g}$ mL$^{-1}$ ).

Seeds of neem were collected from different parts of India and analyzed for their azadirachtin content by High Performance Liquid Chromatography (HPLC). In order to assess the effects of genotypic and geographical variation on azadirachtin content in cell cultures, callus development was attempted in the seeds containing high and low concentration of azadirachtin. The concentration of azadirachtin in callus cultures was significantly affected by the explant source. Seed kernels with higher azadirachtin content produced higher azadirachtin content in callus cultures and lower azadirachtin content was seen in callus cultures produced from seed kernels with low azadirachtin content. The protocol for development of elite stock culture of Azadirachta indica was established with the objective of selecting a high azadirachtin-producing cell line. The highest azadirachtin-producing cell line was selected and the effects of different media and illumination conditions on growth and azadirachtin production were studied in shake flask suspension culture. Detailed batch growth kinetics was also established. These studies provided elite starter culture and associated protocols for cultivation of A. indica plant cell culture in the bioreactor.
This experiment was conducted to identify the optimal in vitro propagation condition of Cnidii rhizoma (Cnidium officinale Makino). It was effective to reduce contamination and improve regeneration of shoot when shoot tips taken in July were cultured in 1/2 strength Murashige and Skoog medium supplemented with 500 mg/L carbenicillin disodium 1.0 mg/L BA and 1.0mg/L $GA_3$followed by surface sterilization of explant source in solution of 1% sodium hypochlorite for 20 minutes. When shoot tips were 쳐cultured in 1/2 strength MS medium with 0.5 mg/L BA and 60 g/L sucrose, shoot elogation and subsequent multiplication of the formed shoot were favorable than in other media. Regenerants were well rooted in 1/2 strength MS medium containing 3.0 mg/L NAA.
Protocols for in vitro conservation was developed for Coleus forskohlii. Plants maintained both in field served as explant source. Shoot tips and single node cuttings were used to optimize protocols for in vitro multiplication. MS basal medium supplemented with $0.54\;{\mu}M$ naphthalene acetic acid (NAA) and $8.87\;{\mu}M$ benzy-ladenine (BA) induced multiple shoots in shoot tips and nodes. Shoot multiplication was amplified with a gradual decrease of BA concentration, leading to its final omission after 4 months. Concomitant rooting on multiplication media enabled successful establishment extra vitrum. For in vitro conservation studies, experiments were carried out with 2-3 week maintained in vitro plants under standard and reduced culture conditions (SCC, RCC). In vitro plants could be successfully conserved in full strength MS medium (FMS) under SCC for 6 months without subculture with full potential to regenerate, producing viable shoots and nodes. The root production remained unaffected due to conservation, showing high rooting activity in mannitol and low temperature treatments. Preset low temperature (15 and $10^{\circ}C$) and reduction in media constituents does not appear to favour conservation, although the former accomplished conservation levels equal to (FMS) under SCC.

An efficient and reproducible plant regeneration system was established using transverse thin cell layers (tTCLs) in five cultivars of Brassjca juncea L. The effects of medium conditions, explant types (tTCLs of hypcotyl and cotyledonary petiole) on shoot regeneration were examined in this study. The maximum shoot regeneration frequency was obtained in Murashige and Skoog (MS) medium supplemented with 4 mg/L 6-benzylaminopurine (BA) and 0.2 mg/L 1-naphthaleneacetic acid (NAA). The hypocotyls derived tTCL explants had more shoot regeneration frequency (52%) than the cotyledonary petiole derived tTCL explants. Shoot induction was further improved by the addition of silver nitrate ($AgNO_3$) in the regeneration medium. A significant genotypic effect was also observed between the five cultivars; Rai-5 displayed higher capacities to produce shoots than other cultivars. Regenerated shoots were rooted on MS basal medium without PGRs which induced 90% of roots. The plantlets established in greenhouse conditions with 99% survival, flowered normally and set seeds. The regenerated plants were fertile and identical to source plants.
Calli were induced from the cotyledon and the hypocotyl of alfalfa and the callus lines were tested for the resistance to glyphosate in the liquid medium containing 0.01-3.00 mM glyphosate. Some resistant cell lines were selected from the gradual increase of glyphosate concentration and the lines resistant to 10 mM glyphosate were analyzed with EPSPS activity. Vigorous callus proliferation from the cotyledon and the hypocotyl was observed from the MS medium containing 1 mg/L 2,4-D and 0.5 mg/L kinetin. The hypocotyl was thought to be better explant source for callus induction than the cotyledon. $ID_{50}$ (Inhibition Dosage of 50%) to glyphosate was between 0.1 mM and 0.2 mM level. A49-10G and A58-10G cell lines selected as resistant to 10 mM glyphosate had 8.0 and 9.1 fold increased EPSPS activity to those of the control lines, respectively.

Callus and cell suspension cultures of Eurycoma longifolia Jack were initiated from leaves of different trees. The leaf explant of tree Eu9 produced the most calli and also induced high cell biomass in the cell suspension culture. Optimum production of cell biomass could be initiated in proliferating culture medium with a pH of 5.75 prior to autoclaving. The effects of macronutrient inorganic salts of Murashige and Skoog (MS) liquid medium supplemented with X on production of cell biomass of Eurycoma longifolia were also investigated. The highest cell biomass was produced in MS medium containing macronutrients of $21\;mM\;NH_4NO_3,\;12.25\;mM\;KNO_3,\;3.00\;mM\;CaCl_2.2H_2O,\;0.575\;mM\;MgSO_4.7H_2O$, and $1.83\;mM\;KH_2PO_4$. A new medium labeled as TAM was formulated for the production of Eurycoma longifolia cell biomass in the cell suspension culture.
Codonopsis lanceolata ("Deoduck" in Korea) is a perennial herb, and belongs to family, Campanulaceae. Its taproot is used a good source of a wild vegetable as well as an herbaceous medicine. In this study, to develop a bialaphos-resistant transgenic Codonopsis, seed germination mechanism and somatic embryogenesis of the plant were investigated, and Agrobacterium-mediated transformation with bar gene encoding phosphinothricin acetyltransferase (PAT) was performed. Attempt were made to regenerate plant from cells via somatic embryogenesis. When the cotyledons, nodes and leaf disks were cultured on MS medium containing 2,4-D and zeatin, embryogenic calli were induced. Upon transferring the somatic embryos to N6 solid medium without plant growth regulators, they developed into plantlets under continuous illumination. All plants were dead on MS basal medium containing 10 mg/L phosphinothricin (PPT) and Basta, respectively. The explants did not produce calli in the medium containing 200 mg/L kanamycin. The explants were cocultured with Agrobacterium tumefaciens for 2 days, and transformants were selected in MS basal medium containing 1.0 mg/L 2,4-D, 100 mg/L kanamycin and 500 mg/L carbenicillin. After the selection, embryogenic calli were induced and then somatic embryos were produced by subsequent subculturing. The somatic embryos were germiated on N6 basal medium containing 200 mg/L kanamycin and 500 mg/L carbenicillin. PCR analysis showed that nptII and bar genes were introduced in the Deoduck transformants. After the confirmation of bar gene expression in RNA and protein level, the transgenic Deoduck will be used to study the genetics of filial generation with the herbicide control gene, bar.gene, bar.

An efficient and reliable Agrobacterium transformation procedure based on TDZ (thidiazuron)-induced organogenesis was established and applied to six Brazilian eggp1ant varieties. Optimum transgenic plants recovery was achieved upon the study of the following parameters affecting transformation efficiency, using F-100 variety as a model: i) explant source; ii) pre-culture period; iii) physical state of the pre-culture medium and iv) coculture conditions. The highest frequency of kanamycin-resistant calli derived from leaf explants (5%) was obtained without a pre-culture period and co-cultivation for 24 h in liquid medium followed by five days on solid RM (regeneration medium). For cotyledon explants, best results were achieved upon a pre-culture of 24 h in liquid RM and a co-cultivation period of 24 h in liquid RM followed by three days in solid RM, resulting in a transformation Sequency of 22.7%. Kanamycin-resistant organogenic calli were also obtained from cultivars Emb, Preta Comprida, Round nose Shaded, Campineira and Florida Market. The expression pattern of an epidermis-specific promoter was studied using transformants expressing a chimaeric construct comprised by the promoter Atgrp-5 transcriptionally fused to the coding region of the gus gene. The expression pattern was similar to that previously observed in tobacco and Arabidopsis thaliana, with preferential expression at the epidermis and the stem phloem. These results support the idea that the Atgrp-5 promoter can be used to drive defense genes in these tissues, which are sites of pathogen interaction and spread, in programs for the genetic improvement of eggplant.

Embryogenic calli were formed on zygotic pro-embryos together with their endosperms when they were cultured on Ms media containing 1.0mg/L 2,4-D. Embryogenic calli were also formed on the intact surface in vitro grown stem or petiole segmentsafrer 6-8 weeks of culture, whereas non-embryogenic calli were formed on cut surfaces of the stem and petiole after 2 weeks of culture.
