HEALING PROCESS OF THE CALVARIAL DEFECT FILLED WITH HYDROXYLAPATITE AND TGF-β IN RAT (백서 두개골 결손부에 Hydroxylapatitie와 TGF-β 매식 후 치유과정에 관한 연구)

  • The purpose of this study was to evaluate the healing process of the calvarial defect filled with hydroxylapatite(HA) and $TGF-{\beta}$ in Rat. 72 Sprague-Dawly rats were divided into 3 groups, control and two experimental groups. Bony defect were artificially prepared in the calvaria of all 72 rats and followed by implantation of HA (experimental group of 24 rats) and HA+$TGF-{\beta}$(another experimental group of 24 rats) into the defects. Sequential sacrifice was performed at 1, 2, 4, 6, 8, 12 weeks of experiment. Obtained specimen was stained with Hematoxylin and Eosin, Masson's Trichrome and Immunohistochemistry. The results were as follows, 1. Granulation tissue was prominent on control group in 1 and 2 weeks. Bony defects were filled with dense fibrous tissue through the whole experimental period and osteoinduction could not be observed in all groups. 2. Inflammatory cell infiltration was prominent on control group in 1 and 2 weeks and osteoclastic activity was high in HA implanted experimental group at 1 and 2 weeks. 3. Inflammatory cell infiltration was less and maturation of fibrous tissue could be found on HA+$TGF-{\beta}$ implanted experimental group at 1 and 2 weeks. 4. Osteoconduction activity was high in HA+$TGF-{\beta}$ implanted experimental group at 2 and 4 weeks but there was no difference after 6 weeks among 3 groups. 5. In grafted site of HA+$TGF-{\beta}$ implanted group, osteonectin expression was slightly increased from 1 week to 6 weeks. In the host site, it was increased from 1 to 4weeks. 6. In grafted site of HA+$TGF-{\beta}$ implanted group, osteocalcin expression was high at 4 weeks. In the host site, we could find the difference among 3 groups. From above results, the HA with mixture of $TGF-{\beta}$ has the potentiality of promoting bone formation in the bony defect area in the rat.

Effects of Hyangsayangyi-tang Aqueous Extracts on the Hypothyroidism Related Hepatic Damages induced by PTU in Rats (香砂養胃湯이 PTU로 유발된 Rat의 갑상선기능저하와 간손상에 미치는 영향)

  • To evaluate the benefits of Hyangsayangyi-tang aqueous extracts (HSYYT) on the propylthiouracil (PTU)-induced Rat hypothyroidism. 6 groups, each consisting 8 Rats were used in the present study - Intact vehicle control, PTU control, LT4, HSYYT 500, 250 and 125 ㎎/㎏ treated groups. HSYYT was given, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 500, 250 and 125 ㎎/㎏(body weight), and for 28 days while the PTU 10 ㎎/㎏ by daily subcutaneous treatment induced hypothyroidism. Compared the results with LT4 0.5 ㎎/㎏ intraperitoneally treated rats in this experiment. Results of the PTU treatment included; decreases of body weight, increase in thyroid weight, decrease in liver weight, in serum T3, and T4 level decrease with increase of serum TSH levels, in serum HDL increase and in TG content decrease, decrease in liver antioxidants defense system, increase of serum AST levels were observed. However, these PTU induced hypothyroidism and related hepatic damages were dose-dependently inhibited by treatment of HSYYT 500 and 250 ㎎/㎏, and they also effectively regulated the PTU-induced abnormal antioxidant defense system changes in liver. Therefore, in comparison with the PTU control group, it was effective and advantageous changes were not observed in HSYYT 125 ㎎/㎏ treated rats on the PTU induced hypothyroidism and related hepatic damages. In this experiment, HSYYT 500 and 250 ㎎/㎏ dose-dependently inhibited PTU-induced hypothyroidism and related liver damage in rats but not in HSYYT 125 ㎎/㎏.

Effects of Vitamin E Supplementation on Antioxidation and Lipid Profiles of Rats on Diets Supplemented with Cholesterol and Olive Oil

  • Lipid peroxidation (LPO) has been identified as an important component of atherosclerosis. In this study, the effects of supplementation with cholesterol (0.5%), olive oil (5%) and vitamin E (0.05%) on erythrocyte glutathione (GSH), plasma malondialdehyde (MDA), total cholesterol, HDL-LDL cholesterol and triacylglycerol, brain and liver MDA and GSH concentrations of rats were investigated. A total of 50 Sprague-Dawley male rats aged 6 months, and of equal body weight were used and fed a standard ration ad libitum. Animals were housed in the University of Selcuk, Veterinary Faculty Experimental Animals Unit. The experiment lasted 60 days and there were five experimental groups as follows: 1. Control, 2. Cholesterol (0.5%), 3. Olive oil (5%), 4. Cholesterol plus vitamin E (0.05%), 5. Olive oil plus vitamin E (0.05%). At the end of the experiment, blood samples were taken by cardiac puncture and erythrocyte GSH, plasma MDA, cholesterol, HDL-LDL cholesterol, triacylglycerol and also GSH and MDA concentrations in brain and liver tissue of rats were spectrophotometrically determined. Supplementation of olive oil and cholesterol into rat diets (groups 2 and 3) caused significant differences in lipid parameters; HDL cholesterol concentrations were increased in the olive oil group and LDL cholesterol was lower than in the cholesterol fed group. Moreover, these decreases in LDL and triacylglycerol concentrations were more significant with vitamin E supplementation. The high plasma MDA concentrations showed that lipid peroxidation occurred in the olive oil group and the highest brain MDA concentrations were determined also in the olive oil group. These findings suggest that vitamin E addition may decrease the sensitivities of several oils to oxidation and that monounsaturated fatty acids in olive oil may decrease the incidence of atherosclerosis by regulating blood lipid profiles.

Articular Cartilage Protective Effects of Kangwhaldoche-tang(Qianghuodaozhi-tang) Aqueous Extracts on the Adjuvant-induced Rat Rheumatoid Arthritis (강활도체탕(羌活導滯湯) 물 추출물의 Adjuvant 유발 류마티스 관절염 랫트에 대한 관절연골 보호효과)

  • Objectives : This study was conducted to determine whether or not Kangwhaldoche-tang(Qianghuodaozhi-tang) (KDT) aqueous extracts can protect articular cartilage losses on the Freund's complete adjuvant(FCA)-induced rat rheumatoid arthritis. Methods : 520, 260 or 130 mg/kg of KDT were orally administered once a day for 14 days from 14 days after FCA treatments, and 15 mg/kg of dexamethasone was intraperitoneally administered as reference drug in this experiment. Changes on the body weight, knee circumferences, gross arthritis score, inflammatory tissue prostaglandin(PG) $E_2$ levels were monitored with cartilage collagen components and glucosaminoglycans(GAGs) compositions - chondroitin sulphate, heparin sulphate and hyaluronic acid in the present study. Results : As results of FCA treatment, classic rheumatoid arthritis featuring dramatic decreases of the body weights, cartilage collagen and GAGs contents with increases of the knee circumferences, gross arthritis scores and inflammatory tissue $PGE_2$ levels, were demonstrated in the present study. However, these changes from FCA - induced rheumatoid arthritis were clearly inhibited by treatment of dexamethasone and all three different dosages of KDT. Although overall anti-inflammatory effects of KDT 520 mg/kg were lowered than those of dexamethasone 15 mg/kg treated rats, KDT 520 mg/kg showed more favorable preserve effects on the cartilage GAGs and KDT 260 mg/kg treated rats showed similar preserve effects as compared with dexamethasone 15 mg/kg in this experiment. Conclusions : The present results supported that over 75 mg/kg of KDT showed favorable anti-arthritic effects on the FCA-induced arthritis mediated by suppression of $PGE_2$, representative inflammatory mediator, and may help improve rheumatoid arthritis.

The Study on the Sochungyong-tang's Effect of Rat on Allergic Rhinitis (小靑龍湯이 알레르기 비염 유발 白鼠의 과립구 및 조직학적 변화에 미치는 효과)

  • Objective: The study is performed to assure the effect of Sochungyong-tang against the wide-spread syndrome allergic rhinitis. For this purpose, the Sochungyong-tang is administered to the rat, which was induced the allergic rhinitis, then the histological change is observed. And the change of the number of neutrophil and eosinophil is also observed. Methods: The Sprague-Dawley male rats, about 200g of body weight, by Saemtako BIO Korea were divided into normal, control, and experimental group. The rats were adapted under the constant temperature and humidity for a week. The solid feed and the water were provided ad lid during the whole experiment By the Levine & Vaz method, the allergic reaction induced with ovalbumin as antigen by intraperitoneal route. As the Sochungyong-tang is administered, the histological changes of neutrophil, eodinophil, and mucosal findings were observed. Results: 1. Number of neutrophil was significantly decreased in the experimental group as compared with the control group. 2. Number of eosinophil was significantly decreased in the experimental group as compared with the control group. 3. The mucosa and submucosa of the nose in experimental group were appeared as normal histological findings. Conclusion: According to the above results, it is supposed that the Sochungyong -tang has considerable effect on allergic rhinitis.

A Study on Effect of Electric Field for Carcinogenesis of Strontium 90 (Strontium 90의 골수 발암성에 대한 전계장의 영향에 관한 실험적 연구)

  • Sprague Dawley rats were exposed to electric fields (6,000 V, 10 kV/m, 30 min/day, 6 days per week) and injected strontium 90 (681 kbq/rat, one time) through abdominal cavity (strontium 90 and electric field complexed exposure group). In parallel, series with the electric field exposure only, strontium 90 injection only and control groups were run. Every group was consisted of 110 rats (55 male and 55 female). This animal experiment was performed from May to December in 1993. This results were conducted to investigate the effect of electric field for 11 weeks. The results are summarized as follows: 1. Ornithine decarboxylase (ODC) activity in rat's bone marrow cells: The ODC values was significantly increased in Sr$^{90}$ injection group and Sr$^{90}$ and electric field complexed exposure group as compared with that of control group (p<0.05). The ODC value was significantly decreased in electric field and Sr$^{90}$ complexed exposure group in comparison with Sr$^{90}$ injection group (p<0.05). The ODC values of electric field only exposure group was not different to that of control group (p>0.05). 2. The amount of Sr$^{90}$ accumulation in the femur, kidney and spleen:The accumulation amount of Sr$^{90}$ in the femur of Sr$^{90}$ injection group represented higher value than that of electric field and Sr$^{90}$ complexed exposure group (p<0.05). In the kidney and spleen, the difference between electric field and Sr$^{90}$ complexed exposure group and Sr$^{90}$ injection group wasn't observed. 3. The counts of white cells in blood of Sr9?injection group was decreased as compared with the value from control group and electric field and Sr$^{90}$ complexed exposure group (p<0.05). The rat's body weight, red blood cell counts and the weight data of liver, kidney and spleen did not show differences among four groups.

Effect of Hyeonsamdansameum on Hypertensive Rat Induced Monocrotaline (현삼단삼음(玄蔘丹蔘飮)이 Monocrotaline으로 유발된 고혈압 흰쥐에 미치는 영향)

  • This experiment was performed to investigate the effects of Hyeonsamdansameum(HDE) on hypertension. For the study of HDE, we had divided Sprague-Dawley rats to three groups-normal, control, HDE. The control group was injected subcutaneous with monocrotaline(50 mg/kg). The treatment group was injected subcutaneous with monocrotaline(50 mg/kg) and orally administered with HDE extract for 4 weeks(once a day, 208 mg/kg). Then we measured blood pressure, heart rate, on the plasma aldosterone, catecholamine, electrolyte, uric acid, BUN, creatinine, and observed the lung, cardiac muscle. liver. etc. The results of these were as follows: 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity and superoxide dismutase(SOD) - like activity were increased. reactive oxygen species (ROS) was decreased. Angiotensin converting enzyme(ACE) inhibitory activity was increased in a concentration-dependent by HDE. HDE significantly increased body weight in monocrotaline hypertensive rat, so supported nearly normal group. HDE significantly decreased blood pressure and heart rate in monocrotaline hypertensive rat. HOE significantly decreased aldosterone in adrenocortical hormones. HDE significantly decreased dopamine. norepinephrine, epinephrine. Na+. Cl- were intended to decrease. K+ was decreased significantly by HDE. Uric acid. BUN were significantly decreased and creatinine was intended to decrease by HDE. HDE inhibited lung, liver and heart injury connected with hypertension. These results suggest that HDE is usefully applied in treatment and prevention of hypertension.

IRRADIATION EFFECT ON SECRETING FUNCTION, AMYLASE ACTIVITY AND NUCLEIC ACID CONTENTS OF RAT PAROTID GLAND (방사선 조사가 이하선 기능에 미치는 영향에 관한 연구)

  • This experiment was performed to clarify the effects of /sup 60/Co gamma irradiation on secretory function, amylase activity and contents of nucleic acids of parotid gland in rat. Experimental animals were divided into 6th hours, 3rd, 7th, 14th and 28th days after irradiation and control. The experimental animals are singly irradiated with 20Gy (2,000rad) through protective lead block. Secretory function of parotid gland was evaluted by uptake and clearance of /sup 99m/TcO₄. /sup 99m/TcO₄. 0.2μ ci/gm, was injected into peritonium in uptake groups. Rats were sacrified with cervical dislocation after 30 minutes and gland was excised. In the clearance group. pilocarpine nitrate (8㎎/㎏) was intraperitoneally injected at 30 minutes after /sup 99m/TcO₄ injection and rats were sacrified 30 minutes after pilocarpine injection. Radioactivity of excised parotid gland was measured by using of gamma counter and stimulation-secretion coefficients, uptake radioactivity divided by clearance radioactivity, was calculated. Amylase activity and contents of DNA and RNA were determined by spectrophotometry. The results obtained were as follows: 1. In the uptake test, the radioactivity of /sup 99m/TcO₄ per unit weight increase in experimental group except 6th hours group, compared with control groups and showed a peak at 3rd days after irradiation. 2. In the clearance test, the radioactivity of /sup 99m/cO₄per unit weight rose to a peak at 3rd days after irradiation and gradually recovered thereafter. 3. Stimulation-secretion coefficient of parotid gland decreased at 6th hours, 3rd and 7th days after irradiation, and gradually increased. 4. Amylase activity of parotid gland decreased in 3rd and 7th days group, and especially lowest in 3rd days after irradiation. 5. The contents of DNA showed no definite difference in all the experimental groups, but RNA was seemed to decrease with time after irradiation.

The effects of lead poisoning on the accumulation of lead, zinc, copper, iron and calcium in tissues of rats (Rat에 있어서 실험적 납중독이 체조직내 납 및 무기질함량에 미치는 영향)

  • This experiment was carried out to clarify the effect of lead poisoning on the lead and essential trace elements homeostasis of tissues in rats. Fifty female Wistar rats, 7 weeks old, were divided into four experimental groups(B, C, D, and E) and a control(A). The four experimental groups received diets contaminated artificially with 10 (B group), 100 (C group), 1,000 (D group) and 5,000 (E group) ${\mu}g/g$ of lead as lead acetate, respectively. The control group received normal diet. Rats were necropsied on the 4th or 8th week of experimental periods and heart, liver, spleen, kidney, muscle, lung and hair were taken. The tissues were analysed for lead, copper, zinc, iron and calcium content by atomic absorption spectrophotometry. From these experiments following results ware obtained ; In C group, lead content of the heart, liver, kidney and hair were significantly increased both on the 4th and 8th week of experiment. In D and E group, lead content of all tissues including heart, liver, spleen, kidney, muscle, hair and lung were significantly increased in accordance with the lead contamination both on the 4th and 8th week of experiment. In C group, copper content of tissues were significantly increased in the heart, liver, kidney and lung on the 4th week and all tissues on the 8th week of experimental periods. In D and E group, copper content of the liver were significantly increased both on the 4th and 8th week of experiment. But the copper content of tissues were gradually decreased compared with C group in accordance with the lead contamination. Especially the content in spleen, heart or kidney were significantly decreased compared with the control group. The zinc content of kidney, heart, or spleen were decreased in D and E groups whereas the iron and calcium content showed no significant change.

Effects of Recombinant Human Epidermal Growth Factor (rhEGF) on Experimental Radiation-Induced Oral Mucositis in Rats (Rat의 방사선 조사성 구내염에 대한 Recombinant Human Epidermal Growth Factor (rhEGF)의 효과)

  • Purpose: Oral mucositis is a common toxicity of radiation or chemotherapy, which is used a treatment for head and neck cancer. We investigated effects of recombinant human epidermal growth factor (rhEGF) on radiation-induced oral mucositis in rat model. Materials and Methods: Spraque-Dawley rats (7 per group) exposed to a single dose of 25 Gy (day 0) on their head, except for one group, were randomly divided into un-treated, vehicle-treated, and two rhEGF-treated groups. Rats were topically applied with rhEGF (15 or $30{\mu}g/oral$ cavity/day) or vehicle to their oral mucosa. Survival rate of rats, weight changes, and food intakes were examined from day 0 to 18 after radiation. Histology study was performed from oral mucosa of rats at day 7 and 18 after radiation. Results: rhEGF-treated groups (15 or $30{\mu}g/oral$) showed all survival rate 33%, whereas un-treated and vehicle-treated groups showed all survival rate 0% at the end of experiment. rhEGF-treated groups statistically had less weight loss compared to vehicle-treated group from day 2 to 7 after radiation. Food intake of rats with rhEGF treatment turned to increase at day 14 after radiation. At 7 day after radiation, un-treated and vehicle-treated groups showed severe pseudomembraneous or ulcerative oral mucositis. On the other hand, rhEGF-treated groups had no more than cellular swelling and degeneration of epidermal cells in oral mucosa of rats. Conclusion: These results suggest that rhEGF has significantly positive effects on radiation-induced oral mucositis in rats. rhEGF display a therapeutic potential on a clinical level.
