This study was performed to determine the antimutagenic and cytotoxic effect of Aster scaber root ethanol extract on Salmonella typhymurium TA98, TA100 and cancer cell lines using Ames test and cytotoxicity assay, respectively. Cancer cell lines include chronic myelogenous leukemia(K562), human gastric carcinoma(KATOIII), human hepatocellular carcinoma(Hep3B) and human breast adenocarcinoma(MCF-7). Futher fractionations with hexane, chloroform, ethyl acetate, butanol and water from ethanol extract of Aster scaber root were performed to obtain effective fraction. Ethanol extract and ethyl acetate fraction showed 79% and 82% inhibitory effect on the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) against TA100, while 48% and 60% inhibition was observed on the mutagenesis induced by 4-nitroquinoline-l-oxide(4NQO) against TA98. In the meanwhile, ethyl acetate fraction showed 78% and 85% inhibitory effect on the mutagenesis induced by benzo(${\alpha}$)pyrene[B(${\alpha}$)P] against TA98 and TA100, respectively, while 83% inhibition was observed on the mutagenesis induced by 3-amino-l,4-dimethyl-5H-pyrido(4,3-b) indole(Trp-P-1) against TA98. Ethyl acetate fraction (0.125 mg/ml) showed the strongest cytotoxic effect against K562, KATOIII, Hep3B and MCF-7 at the same concentration compared to those of other fractions. Ethanol extract and water fraction showed the least inhibitory effect.
The antibacterial effect of Quercus salicina Blume extract was investigated and then the stability of a cream containing it best performing fraction, the ethyl acetate fraction, was evaluated. The antibacterial effect was evaluated on the skin microorganisms Staphylococcus aureus, Bacillus subtilis, Propionibacterium acnes, Escherichia coli, and Pseudomonas aeruginosa. Among the Q. salicina Blume extract fractions, the ethyl acetate fraction demonstrated the lowest minimum inhibitory concentration against S. aureus (1,200 ${\mu}g/ml$), B. subtilis (2,500 ${\mu}g/ml$), P. acnes (1,200 ${\mu}g/ml$) and P. aeruginosa (312 ${\mu}g/ml$). Therefore, a cream containing 0.25% ethyl acetate fraction of Q. salicina Blume extract was prepared and evaluated for stability. The pH, viscosity, and absorbance of the cream were measured under various temperatures (4, 20, 37, $45^{\circ}C$) and sun light during a 12 week period. The changes in viscosity, absorbance and pH of the cream did not change significantly during the term of the experiment when compared with a placebo cream. In addition, any change in color or odor of the cream was not observed during the 12 weeks. These results indicate that the ethyl acetate fraction of Q. salicina Blume extract has a high antibacterial effect and is stable as a cream. There is therefore some potential for its use in cosmetic materials.
The leaves and stems of Dendropanax morbiferus were separated from organic solvents with methanol. The organic solvent fractions were fractionated with dichloromethane, ethyl acetate and butanol according to the systematic fractionation method. Oxidation in the body induces aging, and antioxidant activity has attracted the attention of many people as a preventive component to suppress negative reactions in the body. To investigate the antioxidant activity of Dendropanax morbiferus were subjected to DPPH free radical assay. In addition, acetyl cholinesterase inhibitions were performed for Alzheimer's disease as an aging neurological disease. As a result, it was confirmed that the antioxidant effect of DPPH was generally good in the antioxidant test. The ethyl acetate fractions of Dendropanax morbiferus stems and leaves were $IC_{50}=30{\mu}g/m{\ell}$. Acetyl cholinesterase inhibition experiments were carried out at a concentration of $250{\mu}g/m{\ell}$. Dendropanax morbiferus stems fractions showed dichloromethane fraction of 57.68%, which significantly inhibited the activity of acetyl cholinesterase.

To search for antioxidant compounds from plant resources, ethanol extracts of 10 plants species were investigated using DPPH method and SOD activity by NBT method. The highest activity was shown in the ethanol extracts of Schizandra Chinesis Baillon. And than, antioxidant effects and total polyphenol concentration were investigated in solvent fraction of ethanol extracts from Schizandra Chinesis Baillon. The DPPH inhibitory activity and SOD activity of ethyl acetate fraction (DPPH 51.6%, SOD 66.2%) and butanol fraction(DPPH 60.7%, SOD 67.4%) showed strong biological activities.
This study was conducted to investigate the changes in saponin content and antioxidant activity of crude ginseng and extruded ginseng by using different solvent extraction methods. Each of the fractions was first extracted by 80% ethanol followed by ether treatment to remove the lipid components. Water soluble components were separated by ethylacetate and water saturated butanol. Four fraction, including 80% ethanol, ethylacetate, butanol and water were obtained from crude and extruded ginsengs to analyze saponin content and antioxidant activity. Saponin content and antioxidant capacity of each of the four fractions were measured by LC/MS analysis and ORAC(Oxygen Radical Absorbance Capacity) assay, respectively. It was found that a major portion of saponin was present in ethyl acetate and water saturated butanol fractions. When extracted by 80% ethanol, ginsenoside Rb1 and Rg1 were mostly found in crude ginseng, while ginsenoside Re and Rb1 were detected in extruded ginseng. Even though Rh1 and Rg3 were found in a very small quantity in crude ginseng, there was a significant quantity of both in extruded ginseng when extracted by 80% ethanol. Similar tendency was also observed in extruded ginseng fraction when extracted with ethyl acetate and butanol. In crude ginseng, the level of Rg1 was the highest among other ginsenosides upon extraction by ethyl acetate, while Rh1 and Rg3 were predominantly found by employing similar solvent extraction in the extruded ginseng. Also, Rg1, Re and Rb1 were also found in the extruded ginseng with small quantity. Rg1, Re and Rb1 were found in crude ginseng by butanol extraction, while Rb1 and Re were extracted from the extruded ginseng. Overall, there was no difference in the saponin content between crude ginseng and extruded ginseng when extracted by butanol and water, but twice as much of saponin was obtained by 80% ethanol extraction and 6 times more saponin were obtained in ethyl acetate fraction in the extruded ginseng. Antioxidant capacity of crude ginseng as determined by ORAC assay was higher in 80% ethanol(high in many different kinds of biological compounds) and water saturated butanol(high in polar saponin) fractions than the ethyl acetate and water fractions. No difference in antioxidant capacity was observed between crude and extruded ginseng. However, antioxidant capacity of ethyl acetate and water fractions in extruded ginseng was significantly higher than crude ginseng($P$ >0.05). All the fractions in both, crude and extruded ginseng possessed antioxidant capacity and even water fractions that contained almost no saponin had some antioxidant capacity. While determining correlation coefficient between fractions in extruded ginseng by Pearson correlation, it was observed that 80% ethanol fraction was in correlation with ethyl acetate($P$ >0.01) and ethanol($P$ >0.001) and in the case of ethylacetate, correlation was observed only with butanol fraction($P$ >0.05).

In this study, the antibacterial, antioxidative effect and component analysis of Pinus koraiensis leaf extracts were investigated. MIC values of the ethyl acetate fraction from P. koraiensis leaf extracts on P. acnes, S. aureus, P. ovale, and E. coli were 0.06 %, 0.25 %, 0.13 % and 0.50 %, respectively. The results showed that the antibacterial activity of the ethyl acetate fraction on P. acnes, P. ovale. and S. aureus was more prominent. The aglycone fraction of P. koraiensis leaf extracts ($22.93\;{\mu}g/mL$) showed more higher free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of P. koraiensis leaf extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. The 50 % ethanol extract ($0.70\;{\mu}g/mL$) showed the most prominent ROS scavenging activity. Also the ethyl acetate ($1.04\;{\mu}g/mL$) and the aglycone fraction ($1.43\;{\mu}g/mL$) showed very high antoxidant activity. The protective effects of extract/fractions of P. koraiensis leaf extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The P. koraiensis leaf extracts showed cellular membrane protective effects in a concentration dependent manner ($5{\sim}50\;{\mu}g/mL$). TLC and HPLC chromatogram of the ethyl acetate fraction obtained from hydrolysis of P. koraiensis leaf extracts revealed 2 main bands (PK-4, PK-6) and peaks (peak 1, peak 2), which were identified as kaempferol-3-O-glucoside (PK-6, peak 1) and kaempferol-3-O-arabinoside (PK-4, peak 2) by LC/ESI-MS/MS, respectively. These results indicate that extract/fractions of P. koraiensis can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging ROS, and protect cellular membrane against ROS. Extract/fractions of P. koraiensis can be applicable to new cosmeceuticals for antioxidant, antiaging, and antibacterial activity.

In this study, the antioxidative effects, inhibitory effects on tyrosinase and elastase and components of Fagopyrum esculetum extracts were investigated. Fagopyrum esculetum used for this study is hulls and dehulled seeds. The free radical(1,1-diphenyl-2-picrylhydrazyl radical, DPPH) scavenging activities($FSC_{50}$) and reactive oxygen species(ROS) scavenging activities($OSC_{50}$) of extract/fractions from Fagopyrum esculetum were measured. The aglycone($3.5{\pm}0.0{\mu}g/mL$) and ethyl acetate fractions($0.2{\pm}0.1{\mu}g/mL$) of hulls showed the most effective scavenging activities. The protective effects on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction exhibited the most prominent cellular protective effect(${\tau}_{50},\;771.7{\pm}9.9 min$ at 10 ${\mu}g/mL$). The inhibitory effect of aglycone fraction of hull and dehulled seeds on tyrosinase were examined ($53.6{\pm}0.5{\mu}g/mL,\;35.6{\pm}0.4{\mu}g/mL$, respectively). And The inhibitory effect of aglycone fraction of hull and dehulled seeds on elastase were investigated($3.7{\pm}0.4{\mu}g/mL\;and\;6.0{\pm}0.7{\mu}g/mL$ respectively). But 50% ethanol extract rarely exhibited the inhibitory activity on tyrosinase and elastase. (+)-Catechin and (-)-epicatechin were contained in ethyl acetate fraction of dehulled seeds. And ethyl acetate fraction of hull contains flavonoids of hyperin, quercetin, rutin and so on. These results indicate that extract/fractions of Fagopyrum esculentum can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Fagopyrum esculentum extract could be used as a new cosmeceutical for whitening and anti-wrinkle products.

In this study, the antioxidative effects and active component analysis of 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from Prunella vulgaris L. were investigated. The free radical scavenging activities ($FSC_{50}$) was investigated at 50% ethanol extract ($15.25{\mu}g/mL$), ethyl acetate fraction ($8.68{\mu}g/mL$), and aglycone fraction ($8.25{\mu}g/mL$) respectively. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay was investigated at 50% ethanol extract ($4.68{\mu}g/mL$), ethyl acetate fraction ($1.00{\mu}g/mL$), and aglycone fraction($1.02{\mu}g/mL$) respectively. In the cellular protective effect against $^1O_2$ induced cellular damage of human erythrocytes, extract/fractions of P. vulgaris L. were increased in a concentration dependent manner($1{\sim}25{\mu}g/mL$). Especially, ${\tau}_{50}$ of aglycone fraction at concentrations of $25{\mu}g/mL$ showed the most protective effects at 337.9 min. It's showed nine times higher (+)-${\alpha}$-tocopherol (${\tau}_{50}=38.7min$) as typical antioxidant in the $^1O_2$-induced photohemolysis of human erythrocytes. TLC and HPLC were used to analyse active components in the ethyl acetate fraction and aglycone fraction of P. vulgaris L. In ethyl acetate fraction, caffeic acid, rosmarinic acid, quercetin 3-${\beta}$-D-glucoside, rutin, kaempferol-3-O-rutinoside, astragalin (kaempferol-3-O-glucoside) were identified. In aglycone fraction, caffeic acid, rosmarinic acid, quercetin, kaempferol were identified. These results indicated that extract/fraction of P. vulgaris L. is may be used in cosmetics industry as natural antioxidants by quenching and/or scavenging $^1O_2$ and other ROS, and protecting cellular membranes.

Youngia koidzumiana is an endemic plant growing in Mt. Chiri. In our ongoing research for endemic species in Korea, we investigated the chemical constituents from the MeOH extract of Y. koidzumiana whole plants. The MeOH extract was partitioned with hexane, ethyl acetate and BuOH, successively. Four known compounds were isolated from ethyl acetate fraction by repeated column chromatography. Their structures were elucidated by the physicochemical and spectral data as germanicol acetate (1), oleanolic acid (2), brachynereolide (3) and ixerin Y (4).
