Gallate group이 포함된 친수성 안의료용 렌즈의 광물리적 특성 (Optophysical Properties of Hydrogel Ophthalmic Lenses Containing Gallate Group)

  • 본 연구는 ethyl 및 propyl gallate를 첨가제로 사용하여 대표적인 친수성 렌즈 재료로 사용되는 HEMA (2-hydroxyethyl methacrylate)와 교차결합제인 EGDMA (ethylene glycol dimethacrylate), MMA (methyl methacrylate), AA (acrylic acid) 그리고 개시제인 AIBN (azobisisobutyronitrile)과 함께 공중합 하였다. 생성된 고분자의 물리적 특성을 측정한 결과, 굴절률 1.433-1.435, 함수율 38.71-38.99%, 가시광선 투과율 85.4-88.8%, 인장강도 0.2468-0.2740 kgf 그리고 접촉각의 경우 $49.77^{\circ}$에서 $36.29^{\circ}$ 범위의 분포를 나타내었다. Ethyl 및 propyl gallate를 0.1-1.0% 첨가한 친수성 콘택트렌즈의 경우, UV-B 영역에서 49.0-7.4%, UV-A 영역에서 71.0-43.4%의 투과율을 나타내어 자외선 차단 효과가 있는 것으로 나타났다. 본 실험 결과 중합된 ethyl gallate와 propyl gallate를 첨가한 친수성 안의료용 콘택트렌즈 재료의 경우, 기본적인 의료용 렌즈의 물성을 만족하였으며 함수율의 큰 변화를 나타내지 않으면서도 우수한 습윤성과 자외선 차단효과를 증가시키는 결과를 보였다.

Phenolic Compounds from the Bark of Acer barbinerve Max.

  • The bark of Acer barbinerve was extracted with 70% aqueous acetone and the organic extracts were concentrated to small volume using rotary evaporator and then fractionated successively with n-hexane, dichloromethane, ethyl acetate and water. The chromatographic separation of ethyl acetate soluble fraction led to the isolation of five phenolic compounds. By means of spectroscopic method, the structures of these compounds were identified to methyl gallate (1), methyl gallate-4-O-${\beta}$-D-glucose (2), (+)-catechin (3), (-)-epicatechin (4) and (-)-epicatechin-3-O-gallate (5). These compounds (1-5) have not been reported in this plant yet.

  • Four ursane-type triterpenoids, 2${\alpha}$,3${\alpha}$,19${\alpha}$-trihydroxy-24-norurs-4(23),12-dien-28-oic acid (1), 4(R),23-epoxy-2${\alpha}$,3${\alpha}$,19${\alpha}$-trihydroxy-24-norurs-12-en-28-oic acid (2), myrianthic acid (3), and tormentic acid (4), and a phenolic compound, ethyl gallate (5), from an EtOAc-soluble extract of the stems of Rumex japonicus, were subjected to in vitro bioassays to evaluate advanced glycation end products (AGEs) and rat lens aldose reductase (RLAR) inhibitory activity. Compounds 1 and 5 exhibited a significant inhibitory activity on AGEs formation with $IC_{50}$ value of 87 ${\mu}M$ and on RLAR with $IC_{50}$ value of 14.3 ${\mu}M$, respectively. Ethyl gallate (5) was isolated for the first time from this plant.

  • Methyl gallate isolated from bark of the tree Rhus verniciflua Stokes has significant antimicrobial activity against the fish pathogenic bacteria Edwardsiella tarda and Vibrio anguillarum. To evaluate the antimicrobial activity of gallate derivatives, eight alkyl gallates were tested. Ethyl gallate and propyl gallate had the highest activities, with MICs of $15.6-31.3{\mu}g/mL$ against E. tarda. For V. anguillarum, propyl gallate and butyl gallate were highly effective, with MICs of $7.81-31.3{\mu}g/mL$. When used in combination with antibiotics, methyl gallate exhibited synergistic effects with oxytetracycline against E. tarda and with norfloxacin against V. anguillarum. These results suggest that short-chain alkyl gallates can be used as alternatives to antibiotics against the fish pathogenic bacteria.

  • In the course of searching for BACE1 (${\beta}$-secretase) inhibitors from natural products, the ethyl acetate soluble fraction of green tea, which was suspected to be rich in catechin content, showed potent inhibitory activity. (-)-Epigallocatechin gallate, (-)-epicatechin gallate, and (-)-gallocatechin gallate ware isolated with IC$\_$50/ values of 1.6${\times}$10$\^$-6/ M, 4.5${\times}$10$\^$-6/ M, and 1.8${\times}$10$\^$-6/ M, respectively. Seven additional authentic catechins were tested for a fundamental structure-activity relationship. (-)-Catechin gallate, (-)-gallocatechin, and (-)-epigallocatechin significantly inhibited BACE1 activity with IC$\_$50/ values of 6.0${\times}$10$\^$-6/ M, 2.5${\times}$10$\^$-6/ M, and 2.4${\times}$10$\^$-6/ M, respectively. Free Radical Scavengers from the Fruits of Paeonia suffruticosa

  • The antioxidant activity of Paeonia suffruticosa (Paeoniaceae) was determined by measuring the radical scavenging effect on DPPH (1,1-diphenyl-2-picrylhydrazyl). The methanolic extract of P. suffruticosa showed the strong antioxidant activity. The antioxidant activity of ethyl acetate soluble fraction was stronger than that of the others ($IC_{50}$, $1.2\;{\mu}g/mL$), and the fraction was subjected to purification by repeated silica gel and Sephadex LH-20 column chromatography. Suffruticosol A, suffruticosol B, methyl gallate, and gallic acid, were isolated from the ethyl acetate soluble fraction. Their structures were elucidated by physico-chemical and spectroscopic studies.

Protective effect of gallic acid derivatives from the freshwater green alga Spirogyra sp. against ultraviolet B-induced apoptosis through reactive oxygen species clearance in human keratinocytes and zebrafish

  • In the present study, we enhanced the phenolic content of 70% ethanol extracts of Spirogyra sp. (SPE, $260.47{\pm}5.21$ gallic acid equivalent $[GAE]mg\;g^{-1}$), 2.97 times to $774.24{\pm}2.61GAE\;mg\;g^{-1}$ in the ethyl acetate fraction of SPE (SPEE). SPEE was evaluated for its antiradical activity in online high-performance liquid chromatography-ABTS analysis, and the peaks with the highest antiradical activities were identified as gallic acid derivatives containing gallic acid, methyl gallate, and ethyl gallate. Isolation of ethyl gallate from Spirogyra sp. was performed for the first time in this study. In ultraviolet B (UVB)-irradiated keratinocytes (HaCaT cells), SPEE improved cell viability by 8.22%, and 23.33% and reduced accumulation of cells in the sub-$G_1$ phase by 20.53%, and 32.11% at the concentrations of 50 and $100{\mu}g\;mL^{-1}$, respectively. Furthermore, SPEE (50 and $100{\mu}g\;mL^{-1}$) reduced reactive oxygen species generation in UVB-irradiated zebrafish by 66.67% and 77.78%. This study suggests a protective activity of gallic acid and its derivatives from Spirogyra sp. against UVB-induced stress responses in both in vitro and in vivo models, suggesting a potential use of SPEE in photoprotection.

항균활성을 갖는 천연물 개발과 화장품 응용에 관한 연구 (Development of Antimicrobial Plant Extracts and its Application to Cosmetics)

  • 본 연구는 항균 활성을 지니는 식물추출물을 찾고 화장품에서 방부제로서 적용하기 위해 진행되었다. 디스크 확산법(disk diffusion method)을 통해 3가지 식물 추출물, 함박꽃나무(천녀목란, Magnolia sieboldii), 오배자(Rhus chinensis), 메타세콰이어(Metasequioa glyptostroboides)가 항균활성을 지니고 있음을 확인하였다. 세 물질의 최소저해농도(MIC)를 측정한 결과 메타세콰이어는 0.3 ~ 0.35 %, 함박꽃나무는 0.35 ~ 0.4 % 농도에서 곰팡이의 생장을 억제하고 오배자는 0.45 ~ 0.5 % 농도에서 세균의 생장을 억제함을 확인하였다. 또한 추출물 내의 항균 활성을 지니는 성분을 분리하여 분석한 결과 메타세콰이어에서 분리한 caryophyllene oxide와 caryophyllene, 함박꽃나무에서 분리한 costunolide와 dehydrocostus lactone, 오배자에서 분리한 ethyl gallate, ethyl-3-gallate 등이 항균활성을 지닌 물질임을 확인하였다. O/W 에멀션 제형에서 식물 추출물을 넣고 방부력을 확인한 결과 혼합사용 시 세균과 곰팡이 모두에 대한 방부효과 가 있음을 확인하였다. 따라서 함박꽃나무, 오배자, 메타세콰이어 추출물의 혼합물은 기존의 화학 방부제를 대체할 천연 방부제로서 화장품에서 응용할 수 있을 것으로 기대된다.

녹차로부터 EGCG(Epigallocatechin Gallate)의 추출 및 정제 (Extraction and Purification of EGCG(Epigallocatechin Gallate) from Green Tea)

  • 실험에 사용된 녹차는 전남 보성에서 재배된 것으로 국내시장에서 구입하였다. 녹차 5g을 순수한 물 5$0^{\circ}C$에서 추출한 후 클로로포름과 에틸아세테이트로 분배하였다. 추출액을 크로마토그래픽 컬럼 (4.6$\times$250 mm, 15$\mu\textrm{m}$, Lichrospher 100RP-18)을 이용하여 정제하였다. 정제된 추출액으로부터 $\mu$-Bondapak $C_18$(3.9$\times$300mm, 10$\mu\textrm{m}$) 컬럼을 이용하여 녹차에 포함된 카테킨 화합물을 분리하였으며, EGC(Epigallocatechin, C(catechin), EC(Epicatechin), EGCG(Epigallocatechin Gallate) and ECG(Epicatechin Gallate)의 순으로 용출되었다. 본 연구의 결과로서는 목적물질인 EGCG를 분리하기 위해 이동상의 조성은 0.1%의 아세트산이 포함된 물/아세토나이트릴, 87/13 %(v/v)이었다. 유량은 1.0 $m\ell$/min, UV 검지기는 280nm로 고정하였다. 위의 실험조건으로 5g의 전남 보성산 녹차로부터 순도 98% 이상의 121.3mg EGCG를 얻을 수 있었다.

Protocatechuic acid 및 Gallic acid 유도체들의 항 혈전작용 (Anti-thrombotic Effects of Analogs of Protocatechuic Acid and Gallic Acid)

  • Platelet anti-aggregating activities were tested with analogs of protocatechuic acid and gallic acid. Six of them which showed comparable inhibitory effects with aspirin against collagen induced platelet aggregation were selected and their anti-thrombotic effects were evaluated in the mouse thrombosis model and compared with those of aspirin and paeonol. At the dose of 50 mg/kg, p.o., ethyl gailate(13) treated group showed higher % of recovery within 6 min of thrombotic challenge and lower mortality within 5 min than aspirin treated group.
