Inhibitory Effects of Doen-jang(Korean Fermented Soybean Paste) and Soybean Extracts on the Growth of KB Cells (한국 전통 된장 및 콩 추출물의 KB 세포에 대한 증식 억제효과)

  • The present study was designed to investigate whether traditional Korean fermented soybean paste(doen-jang) and soybean extracts have inhibitory effects on the growth of KB cell, an oral epithelioid cancer cell. When KB cell ATCC CCL-17 was cultivated for 48 flours with the addition of 0.5% of the five types or doen-jang extract, the growth of KB cell was inhibited by all types of extract, and ethyl acetate extract showed the highest inhibitory effect. In case of soybean extract, all types of extract also showed KB cell inhibitory effects, however, generally less than those of doen-jang extract. When ethyl acetate extract of doen-jang was added in different concentrations and KB cell was cultivated for 24 hours and 45 hours, strong inhibitory effect began to appear from the concentration of 1.25 mg/ml. Although soybean extract showed such a tendency, its effect was lower than that of doen-jang extract. These results indicate that doen-jang extract has inhibitory effect against KB cell, and particularly ethyl acetate extract has the highest effect. The effect of doen-jang extract might be possibly enhanced by the fermentation of soybeans. It is assumed that doen-jang extract may be used to develop nontoxic medicines for preventing and treating oral diseases.

Antioxidant Activity of Rhododendron brachycarpum D. Don Extracts and Its Skin Hydration Effect Measure (만병초 추출물의 항산화활성과 보습효과 측정)

  • In this study, the antioxidative effects, inhibitory effects on tyrosinase and elastase of Rhododendron brachycarpum D. Don extracts were investigated. And the moisturizing effect of cream containing R. brachycarpum D. Don extract were investigated by clinical trial. The ethyl acetate fraction of R. brachycarpum D. Don extract (1.83 ${\mu}g/mL$) showed the most prominent the free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of R. brachycarpum D. Don extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The 50 % extract fraction (0.064 ${\mu}g/mL$) showed the most prominent ROS scavenging activity. The protective effects of extract/fractions of R. brachycarpum D. Don on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The R. brachycarpum D. Don extracts suppressed photohemolysis in a concentration dependent manner (1 ~ 10 ${\mu}g/mL$). The inhibitory effects ($IC_{50}$) of R. brachycarpum D. Don extracts on tyrosinase were determined with ethyl acetate fraction of R. brachycarpum D. Don extract (70.5 ${\mu}g/mL$) and aglycone fraction of extract (122.40 ${\mu}g/mL$). The inhibitory effects ($IC_{50}$) on elastase were determined with ethyl acetate of R. brachycarpum D. Don extract (43.50 ${\mu}g/mL$) and aglycone fraction of extract (20.73 ${\mu}g/mL$). The cream containing the ethyl acetate fraction of R. brachycarpum D. Don extracts was formulated for skin hydration effect and transepidermal water loss (TEWL). The cream containing R. brachycarpum D. Don extract was applied to the right lower arm. After 180 min, the water contents in skin were increased by 1 ~ 4 % than the placebo cream. And TEWL of parts was decreased as 7.7 $g/m^2h$ (experimental cream) and 8.9 $g/m^2h$ (placebo cream) respectively. These results indicate that extract/fractions of R. brachycarpum D. Don can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. And inhibitory activity on tyrosinase of the aglycone fraction could be applicable to new functional cosmetics for whitening and anti-wrinkle products. Also the increase of skin hydration of the cream containing extract could be applicable to new functional cosmetics for antiaging.

The Effects of Sedum sarmentosum Bunge on Serum Lipid Concentration in Ovariectomized Rats (돌나물이 난소 절제한 흰쥐의 혈 중 지질 함량에 미치는 영향)

  • The aim of this study was to investigate in an in vivo model the effects of ethanol extract and ethyl ether and ethyl acetate fractions of Sedum sarmentosum Bunge (SS). To investigate the effects of ethanol extract and ethyl ether and ethyl acetate fractions, Sprage-Dawley female rats were randomly assigned to groups as follows : Sham, OVX-control and ovariectomized rats supplemented with SS at 50 mg/kg bw/day, with SS at 100 mg/kg bw/day, with SS at 200 mg/kg bw/day, with ethyl ether fraction of SS at 10 mg/ka bw/day and with ethyl acetate fraction of SS at 10 mg/kg bw/day. Food intake and food efficiency ratio were not significantly different in groups. But body weight gain was decreased by supplementation of ethyl acetate fraction of SS compared to OVX-control. The levels of total cholesterol and triglyceride in serum were higher in ovariectomized control rats than Sham-operated rats, but supplementation of SS ethanol extracts at 200 mg/kg bw/day decreased the level of the lipid in serum. The level of HDL-cholesterol in serum increased by supplementation of SS ethanol extracts at 200 mg/kg bw/day (p<0.01). The levels of total cholesterol and triglyceride in serum were decreased by supplementation of ethyl acetate fraction of SS (p<0.01). On the other hand, the level of HDL-cholesterol in serum was increased by supplementation of ethyl acetate fraction of SS (p<0.05). On conclusion, it might be expected that ethanol extract and ethyl acetate fraction of SS is believed to be a possible protective effects for the fatty serum increasing serum lipid.

In vitro Inhibitory Activity on Rat Intestinal Mucosa ${\alpha}-glucosidase$ by Rice Hull Extract (왕겨추출물의 쥐소장점막 ${\alpha}-glucosidase$에 대한 in vitro에서의 저해효과)

  • In order to search for the way to utilize rice hull as a renewable resource, the inhibition on ${\alpha}-glucosidase$ and the fractionation of rice hull extract was investigated. An ethanol extract of rice hull from Japonica-type rice seeds exhibited 30% inhibitory activity on rat intestinal brush border ${\alpha}-glucosidase$ (1.4 mU/mL) in vitro at the concentration of 0.8 mg/mL using 6 mM p-nitrophenyl ${\alpha}-D-glucopyranoside$ as a substrate $(IC_{50}\;162\;mg/mL)$. Among the fractions obtained by partitioning the ethanol extract successively with solvents, the ethyl acetate fraction at the concentration of 0.8 mg/mL was found to exhibit the most potent inhibitory activity i.e. 65% inhibition of ${\alpha}-glucosidase\;(IC_{50}\;0.14\;mg/mL)$. Silica gel column chromatography of the ethyl acetate fraction exhibited slightly higher (90%) inhibitory activity, and its subsequent fractionation by Sephadex LH-20 column chromatography did not improve inhibitory activity. Considering the inhibitory activity and yield, the ethyl acetate fraction obtained by the solvent-partitioning process would be a candidate for the hypoglycemic food if it has in vivo effectiveness.

Flavonoids and Antimicrobial Activity of the Ethanol Extract of Korean Cherry (Prunus tomentosa Thunberg) (앵두 Ethanol 추출물의 항균력과 Flavonoid)

  • Quantitaties of flavonoids were determined by HPLC in three different ethanol extract (50, 70 and 95%, v/v) from Korean cherry. Antimicrobial activity of the ethanol extract and its solvent fraction (ethyl acetate and n-butanol fractions) against Staphylococcus aureus (ATCC 6538) and Staphylococcus epidermidis (ATCC 12228) was evaluated using MIC and disk test. Additionally, UV protective effect was also determined. The highest flavonoid quantity was found in 70% ethanol extract of Korean cherry. The quantities of quercitrin were 12.7 mg% and 2.1 mg% in ethyl acetate fraction and in buthanol fraction, respectively. In addition, the flavonoid in 70% (v/v) ethanol extract of Korean cherry included 14.9 mg% of quercitrin, 6.0 mg% of tannic acid and 5.2 mg% of catechin. Therefore, it was confirmed that main flavonoid of Korean cherry was quercitrin. For antimicrobial activity of ethanol extract of Korean cherry against Staphylococcus aureus (ATCC 6538) and Staphylococcus epidemidis (ATCC 12228), MIC and disk test using ethyl acetate fraction and butanol fraction were carried out. As a result, ethyl acetate fraction with higher quantity of flavonoid showed higher antimicrobial activity. Therefore, it was confirmed that the higher antimicrobial activity was brought about by flavonoid in Korean cherry. For UV protective effect of Korean cherry, ethanol extract showed the absorption zone of ultraviolet rays in 210∼380 nm including 290∼320 nm that is the wave length of UV-B harmful to the skin of human body. The pattern is the same as the absorption zone of 220∼300 nm in catechin and 210∼400 nm in quercitrin. Then, the functionalities of catechin and quercitrin were anticipated. It is expected that quercitrin in Korean cherry has the antimicrobial effect and UV protective effect related to the skin.

Centella asiatica and Asiaticoside Regulate H2O2-induced Cellular Inflammation via Mitochondrial Respiration and the TLR4 Pathway (병풀(Centella asiatica) 및 아시아티코사이드는 미토콘드리아 호흡 및 TLR4 경로를 통해 H2O2 유도 세포염증 조절)

  • This study determined the effects of Centella asiatica leaf on H2O2 induced cell cycle arrest, mitochondrial activity, and proinflammatory cytokine production in human dermal fibroblast (HDF) cells. We used an 80% methanol extract of C. asiatica, its ethyl acetate fraction, and asiaticoside, the major constituent of C. asiatica. The C. asiatica extract, its ethyl acetate fraction, and asiaticoside attenuated G1 cell cycle-arrest and the apoptotic effect caused by H2O2-induced oxidative stress. The cells treated with C. asiatica extract, its ethyl acetate fraction, and asiaticoside secreted lower levels of TNF-α and IL-6. The antioxidant effect of asiaticoside was higher than that of C. asiatica extract and its ethyl acetate fraction. Treatment with C. asiatica extract, its ethyl acetate fraction, and asiaticoside also increased the mitochondrial membrane potential and restored normal mitochondrial morphology. Following H2O2 stress induction, cells treated with C. asiatica extract, its ethyl acetate fraction, and asiaticoside showed increased mitochondrial oxygen consumption rates and decreases in the TLR4-MyD88-TRAF6-p65 pathway activity. These findings suggest that C. asiatica extract, its ethyl acetate fraction, and asiaticoside have antioxidant and anti-inflammatory effects, as well as the ability to control the mitochondrial activities of HDF cells.

Antioxidant and Nitrite Scavenging Activities of Solvent Extracts from Rhus verniciflua Stokes (용매별 옻나무 추출물의 항산화 활성 및 아질산염 소거능)

  • This study explores the antioxidant activities of solvent extracts from Rhus verniciflua, and the results were as follows. Total phenolic compound and flavonoid contents of Rhus verniciflua were $4.32{\pm}0.17$ mg/100 g and $0.16{\pm}0.02$ mg/100 g, respectively. Electron-donating abilities of solvent extracts from Rhus verniciflua proportionally increased with concentration, and ethyl acetate extract ($95.4{\pm}0.4%$) showed higher activity than that of BHT ($93.3{\pm}1.7%$) at a concentration of 1,000 ${\mu}L$/mL. Reducing power of the solvent extracts from Rhus verniciflua was the highest in ethyl acetate extract ($2.45{\pm}0.03$) at a concentration of 1,000 ${\mu}L$/mL. Nitrite scavenging ability of the solvent extracts from Rhus verniciflua (pH 1.2, 1,000 ${\mu}L$/mL) was the highest in ethyl acetate extract ($95.5{\pm}0.7%$). Therefore, ethyl acetate extract from Rhus verniciflua has similar antioxidant activity as that of BHT.

Study on Antioxidant Activity of Thermal Treatment Yam (Dioscorea batatas DECNE.) by n-Butanol and Ethyl Acetate Extracts

  • In Korea, yam with thermal treatment is widely used to produce tea and beverage, which appeals to all age groups. To better understand the antioxidative activity of thermal treatment yam (Dioscorea batatas $D_{ECNE.}$), n-butanol and ethyl acetate extracts were evaluated with different antioxidant testing system. Their antioxidant activities, including ABTS [2,2' -azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, reducing power, and ferric reducing antioxidant power as well as total phenol contents were assessed in vitro. Results showed ethyl acetate extract ($51.63{\pm}2.51mg\;CAE/g$) possessed similar total phenol content compared with n-butanol extract ($53.93{\pm}1.00mg\;CAE/g$). Ethyl acetate extract also exhibited similarly stronger antioxidant activity compared with n-butanol extract apart from ABTS radical scavenging activity assay.

  • This study aimed at evaluating the antioxidant activity of raw yam (Dioscorea batatas $D_{ECNE.}$) extracted by n-butanol and ethyl acetate. The antioxidant activities, including ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, reducing power, and ferric reducing antioxidant power as well as total phenol contents were studied in vitro. Results showed ethyl acetate extract ($111.88{\pm}0.66mg\;CAE/g$) possessed higher total phenol content than n-butanol extract ($78.68{\pm}0.58mg\;CAE/g$). Ethyl acetate extract exhibited stronger antioxidant activity compared with n-butanol extract apart from reducing power assay.

Antioxidative Activity and Tyrosinase Inhibition Effect of Ethanol Extract and Its Fractions from the Branch of Rhododendron schlippenbachii (철쭉 가지 에탄올 추출물 및 용매별 분획물의 항산화 활성과 Tyrosinase 저해 활성)

  • This study was conducted to investigate the antioxidative activity and tyrosinase inhibitory activity of 50% ethanol extract and its fractions from the branch of Rhododendron schlippenbachii. In DPPH radical scavenging ability, butanol and ethyl acetate fractions showed 59.98% and 55.17% of relative activity compared with positive control (ascorbic acid), but the 50% ethanol extract showed relatively low activity. In nitric oxide (NO) scavenging ability, the ethyl acetate and butanol fractions showed 141.80% and 131.55% relative activity compared with ascorbic acid as used for positive control. On the other hand, tyrosinase inhibitory activity of the ethyl acetate and butanol fractions showed about twice higher activity than positive control (arbutin). It means that the ethyl acetate and butanol fractions from the extract of R. schlippenbachii branch has ability for used as effective radical scavenger and tyrosinase inhibitor.
