Changes of Endoplasmic Reticulum of Rat Intestinal Epithelium Induced by Ethionine Feeding (Ethionine 투여로 인한 소장흡수상피세포의 소포체변화)

  • The author studied ed the effect of ethionine upon the absorption epithelium of ileum with particular ettention to the endoplasmic reticulum. Five tenth per cent of DL-ethionine was added to the diet of the experimental group rats and they were sacrificed after 1, 2, 3, and 4 weeks. respectively. The ileum were observed by the electron microscope. The results obtained were summarized as follow: The changes were detachment of membrane bound ribosome, dilatation of endoplasmic reticulum, decrease of polysome with reciprocal increase of monosome, and dilatation of Golgi complex. These changes were occured at 2 nd to 3 rd week from start of ethionine feeding and more severe at fouth week. These observation suggested that ethionine induced degenerative changes of the organelles.

Studies on the Toxicity of Dietary Ethionine and Methionine to Pancreas and Liver (취장 및 간장에 미치는 식이성(食餌性) Ethionine 과 Methionine 의 독성에 관한 연구)

  • It has been demonstrated repeatedly that the administration of ethionine, a methionine analogue, will produce destruction of pancreatic tissue and liver cells. Destruction of acinar cells of pancreas by the administration of excess methionine similar to that seen after feeding diets supplemented with ethionine was also reported, but the liver was not involved by this amino acid. In an attempt to reproduce the results of these investigators the tissue damages were found to be slight and seen only irregulary in rats receiving ethionine on a complete diet and the result of tissue damages by excess methionine were also controversial. The present studies describe the toxicity of dietary supplemented ethionine and methionine to liver and pancreas in rats fed a low protein diet. Hundred five albino rats weighing around 120 gm were divided into three groups as follows; 1) Control group: A low protein diet containing 8% casein was fed throughout the experimental period. 2) Methionine group: A low protein diet (7% casein) supplemented with 1% methionine was used. 3) Ethionine group: A similar diet as methionine group except the supplementation of 1% ethionine instead of methionine was used. Five animals per wee from each group were killed for 6 weeks. The liver and pancreas were fixed in 10% formalin and histologic sections were prepared and stained with hematoxylin eosin. Serum amylase was expressed as much of glucose liberated from a starch substrate. The glucose was determined by the method of Nelson (1944). Serum glutamic pyruvic transaminase (SGPT) and glutamic oxaloacetic transaminase (SGOT) were determined by the technique described in Sigma Bulletin.

Protective Effect of Soybean against Hepatocarcinogenesis Induced by DL-Ethionine

  • There has been increasing interest in the value of using soybean to delay or reduce the tumor incidence. This study was undertaken to investigate the possible protective effects of soybean against hepatocarcinogenesis induced by DL-ethionine. Accordingly, we measured biochemical changes occurring in serum and liver of rats treated with DL-ethionine in the presence or absence of soybean. Male albino rats were fed a control diet containing the hepatocarcinogen, DL-ethionine, or the control diet plus soybean 30%, or the control diet plus soybean plus DL-ethionine 0.25% for three months and then returned to a control diet for up to nine months. Rats fed a control diet plus DL-ethionine showed a gradual decrease in liver DNA, RNA, total protein, and liver weight and enzyme activites of liver transaminases (GOT and GPT) and alkaline phosphatase over the 7-month study period. This was followed by a large increase in the liver parameters at the end of the $9^{th}$ month, except for 5'-nucleotidase and glucose-6-phosphatase that showed a large decrease. On the other hand, a gradual increase in the serum enzyme activities of GOT, GPT, 5-nucleotidase, alkaline phosphatase, and in the albumin/globulin (A/G) ratio is observed in the group of rats fed a control diet plus DL-ethionine compared to the control group over 8 months, and this was followed by a large increase in all serum parameters studied at nine-months. The administration of 30% soybean to the rat diet in addition to DL-ethionine maintained all parameters studied at near control values until the end of the $9^{th}$ month. This study suggests that soybean has a protective effect against the hepatocarcinogenesis induced by DL-ethionine.

Effect of Pretreatments and Holding Solutions on Vase Life and Quality of Cut 'Saphir' Rose (전처리와 보존용액이 절화장미 'Saphir'의 수명과 품질에 미치는 영향)

  • This experiment was carried out to investigate the effect of pretreatments and holding solutions on quality and vase life of cut 'Saphir' rose (Rosa hybrida L.). Pulsing with aluminum sulfate or NaOCl prolonged vase life held in HQS+sucrose+ethionine. Solution absorption held in distilled water were more increased than held in HQS+sucrose+ethionine. Holding solution with 8-hydroxyquinoline sulfate (HQS)+sucrose+ethionine increased fresh weight compared with distilled water except pulsing with STS+sucrose. Pulsing with aluminium sulfate delayed bent-neck held in HQS+sucrose+ethionine. Flower diameter and dry weight of petal held in HQS+sucrose+ethionine were more increased than held in distilled water except pulsing with STS+sucrose. There were no significance in Hunter color value 'L' and value 'a', but value 'b' was decreased held in distilled water.

  • Liver and serum xanthine oxidase(XOD) activity were determined in rats treated with DL-ethionine. Concomitantly, the enzyme activity was compared with that of rats treated with CCl$_{4}$, actinomycin D and those fed a protein depleted diet. The activity of XOD in liver was inhibited by injection of ethionine to the rats. But, there were no differences in activity of serum XOD between control group and ethionine-treated rats. And the pattern of changes in enzyme activity of serum and liver in ethionine-treated rats, was similar with actinomycin D treated rats or those fed a protein depleted diet. On the other hand, the activity of XOD was rather elevated both in serum and liver by injection of CCl$_{4}$ to rats.

  • 새로운 간장질환 치료제로 개발중인 YH439가 orotic acid, nicotinamide 및 ethionine에 의해 유발되는 지방간에 대한보호 및 치료효과를 관찰하였다. 웅성 SD계 rat에 orotic acid(1%) 또는 nicotinamide(2%)가 첨가된 사료를2주간 섭취시켜 유발된 지방간에 대한 YH439의 보호 및 치료효과와 ethionine(100mg/kg, i.p.)투여로 유도되는 지방간에 대한 YM439의 보호효과를 관찰하기 위하여 간 조직 중 triglyceride, cholesterol 및 phospholipid의 함량을 측정하여 비교하였다. 그 결과, erotic acid 또는 nicotinamide의 섭취로 인해 랫드의 간 조직 중 triglyceride, cholesterol 및 phospholipid의 함량은 1.5-3배정도 증가하였으며, 이 증가된 지질들은 YH439 100, 200mg/kg 투여에 의해 유의성 있게 억제되었다. 또한 ethionine투여에 의해서도 랫드의 간 조직 중 triglyceride, cholesterol 및 phospholipid의 함량은 약 2배정도 증가되었으며, YH439 전저치에 의해 이 현상들이 억제되는 효과를 나타내었다.

  • The ethionine resisconferring gene (ERCI) was constitutively expressed under the control of the alcohol dehydrogenase gene promoter (ADClp) and introduced into the chromosomes of an industrial polyploid strain of Saccharocerevisiae by using the 8-sequences of the Tyl retrotransposon as the recombination site. 8-Integrative cassette devoid of bacterial DNA sequences containing the ampicillin resistance gene was constructed that had the aureobasidin A resistance gene (AURl-C) as the selection marker and ERCl gene. The ERCl gene was also employed as the selection marker in the 8-integrative cassette lacking the A URl-C gene. Industrial Saccerevisiae transformed with these integrative cassettes exhibited strong resistance to DL-ethioncompared with nontransformants.

The Hepatoprotective Effect of Acanthopanax senticosus Fermentation Products in Fatty Liver Model (지방간 모델에서 가시오가피 발효물의 간 기능 개선 효과)

  • We demonstrated that Acanthopanax senticosus fermentation products (FM-5111 and FM-5131) administered to rats functionally protect against DL-ethionine-induced and ethanol-induced fatty liver models. In DL-ethionine-induced fatty liver models, the serum concentrations of aspartate aminotransferase (AST), as well as liver concentrations of triglyceride and total lipid against the control decreased in FM-5111 and FM-5131 treated rats. In ethanol-induced fatty liver models, FM-5111 and FM-5131 treated rats showed a decrease in the liver concentrations of triglyceride and total lipid in ethanol-induced fatty liver models. There were no significant differences in the serum concentrations of AST and alanine aminotransferase in FM-5111 and FM-5131 treated rats. Additionally, FM-5111-, or FM-5131-treated rats showed no significant differences in the body weight gain between the control. These results indicate that Acanthopanax senticosus fermentation products might have protective effects against DL-ethionine-induced and ethanol-induced fatty liver models.

Protective Effect of ACTIValoe N-931 Complex, a Mixture of Aloe vera and Silybum marianum, on Experimental Acute Liver Injury

  • The aim of this study was to investigate the hepatoprotective effect of $ACTIValoe^{(R)}$ N-931 complex, a mixture of Aloe vera and Silybum marianum, against acute liver injuries. Acute liver damages were induced by intraperitoneal injection of galactosamine (GalN, 700 mg/kg), naphthylisothiocyanate (ANIT, 40 mg/kg) and ethionine (500 mg/kg). $ACTIValoe^{(R)}$ N-931 (85, 170 and 340) was administered orally 48 h, 24 h, 2 h before and 6 h after the injection of hepatotoxins. At 24 h after GalN treatment the levels of serum aminotransferases and hepatic lipid peroxidation were significantly elevated, whereas hepatic glutathione, serum triglyceride (TG) and total cholesterol were decreased. These changes were attenuated by $ACTIValoe^{(R)}$ N-931 complex. The serum aminotransferase activities and total bilirubin significantly increased at 48 h after ANIT treatment, but were attenuated by $ACTIValoe^{(R)}$ N-931 complex. The bile flow was lower after ANIT treatment, which was restored by $ACTIValoe^{(R)}$ N-931 complex. $ACTIValoe^{(R)}$ N-931 complex reduced the ethionine-induced elevated hepatic TG contents. Histopathological analysis revealed that signs of liver injury were prominent at 24 h as result of ethionine injection, demonstrated by extensive areas of fatty change and microvesicular steatosis were observed around cells. These changes were attenuated by $ACTIValoe^{(R)}$ N-931 complex. Our results suggest that the $ACTIValoe^{(R)}$ N-931 complex has a protective effect on acute liver injury.

Effects of Pretreatments and Storage Conditions on Quality and Vase Life of Cut 'Red Sandra' Rose (전처리와 저장조건이 절화장미 'Red Sandra'의 품질과 수명에 미치는 영향)

  • This experiment was conducted to investigate the effect of pretreatments and storage conditions on quality and vase life of cut 'Red Sandra' rose (Rosa hybrida L.). 'Red Sandra' rose flowers were harvested and pulsed with distilled water, 0.2% RVB or 200ppm aluminum sulfate+3% sucrose+50ppm $AgNO_3$+0.1mM ethionine for 22 hours and then stored in wet or dry condition for 12, 24, 48 hours at low or room temperature. Pulsing with aluminum sulfate+sucrose+$AgNO_3$+ethionine delayed bent-neck, increased solution absorption and flower diameter, and prolonged vase life rather than pulsing with distilled water or RVB. Storage in wet condition or low temperature also delayed bent-neck, increased solution absorption and flower diameter, and prolonged vase life compared with storage in dry or room temperature condition. As the storage hours prolonged, solution absorption, flower diameter, and vase life were decreased especially pulsing with distilled water. Pulsing with aluminum sulfate+sucrose+$AgNO_3$+ethionine prolonged vase life compared to pulsing with distilled water or RVB regardless of storage conditions and shipping hours.
