Objectives Ethanol is a potent inhibitor of muscle protein synthesis. Muscle mass is regulated by the balance between rates of protein synthesis and protein breakdown. Both acute and chronic alcohol consumption inhibits synthesis to a greater extent than degradation. Protein synthesis is more intensely decreased in type II fibers than in type I fibers. Apoptosis has been shown to occur frequently in a variety of tissues in response to chronic alcohol feeding. Increased muscle fiber apoptosis has been shown in alcoholics with myopathy. Pueraria radix has been used for many disorders such as fevers, gastrointestinal disorders, muscle aches, allergies, respiratory problems, skin problems, high blood pressure, migraine headaches, lowering cholesterol and treating chronic alcoholism. We therefore tested the hypothesis that oral treatment with Pueraria radix could reduce the ethanol-induced muscle atrophy. Methods Young male Sprague-Dawley rats were orally given 25% ethanol (5 ml/kg, body weight) daily with Ethanol for 4 weeks. Normal group was similarly administrated with saline. The Rats of Pueraria radix treated group (EtOH+PR) were orally administrated Pueraria radix water extract, and rats of EtOH group were given with the vehicle only. After 4 week, the morphology of gastrocnemius and plantaris muscles were assessed by hematoxylin and eosin staining. The immunoreactivities of pre-apoptotic BAX and anti-apoptotic Bcl-2 proteins were also measured. Results The muscles from rats of EtOH group represented a significant reduction in average cross section area compared to Normal group. EtOH+PR group had increased fiber compared to the EtOH group. Moreover, to investigate the ethanol-induced muscular apoptosis, the immunohistochemical analysis of Bax and Bcl-2 was carried out. The treatment with Pueraria radix (EtOH+PR) significantly decreased BAX expression and increased Bcl-2 expression 4 weeks after ethanol administration when compared with Normal group. Conclusions These results suggest that Pueraria radix water extract has protective effects on chronic alcohol induced myopathy.
본 연구는 만성적인 에탄올 급여에 의한 조직내 항산화 영양소 함량의 변화에 대하여 비타민 A의 형태별 급여효과를 조사하고자 실시되었다. 실험동물은 Sprague-Dawley종 숫컷 흰쥐를 사용하였으며 에탄올 공급과 동시에 비타민 A 결핍군(FE), ${\beta}-carotene$ 섭취군(${\beta}E$), retinyl acetate 섭취군(RE) 그리고 13-cis-retinoic acid 성취군(RAE)과 에탄올 대신 동일 열량을 sucrose으로 공급 시킨 각각의 pair-fed군 등 모두 8군으로 나누어 7주 동안실섬 식이를 공급하였다. 항산화 영양소로 비타민 E, C, glutathione 그리고 무기질 함량의 분석을 통해서 에탄올에 의해 변화되는 생체내 항산화 영양소 상태와 이에 비타민 A의 형태별 급여 효과를 조사하여 다음과 같은 결과를 얻었다. 혈장내 ${\alpha}-tocopherol$ 함량은 에탄올 급여로 감소되었으나 간 조직내에서는 ${\beta}-carotene$ 섭취군만이 낮은 함량을 보였다. 그러나 간 조직내 tocopheryl acetate 함량은 pair-fed군에 비해서 에탄올 급여로 현저히 저하되었다. 혈장과 간 조직내에서 에탄올 급여에 의한 비타민 E 함량 감소는 retinyl acetate 공급으로 그 감소 정도가 가장 저하되었다. 혈장내 총 비타민 C 함량은 에탄올 급여와 동시에 ${\beta}-carotene$이나 retinyl acetate 섭 취군이 pair-fed군에 비해서 낮았으며 환원형과 산화형 비타민 C 함량비는 에탄올 급여로 낮았으나 ${\beta}-carotene$ 섭취시킨 군만이 높게 나타났다. 간 조직내 총 비타민 C 함량은 13-cis-retinoic acid 섭취군을 제외한 전 군에서 에탄올 급여로 증가하였으여 환원형과 산화형 비타민 C 비는 에탄올 급여군 중 ${\beta}-carotene$을 섭취시켰을 때 가장 높았다. 간 조직내 총 glutathion 함량은 에탄올 공급군과 pair-fed군 사이에 유의적인 차이를 보이지 않았으나 환원형과 산화형의 함량비에서는 에탄올과동시에 retinyl acetate나 13-cis-retinoic acid 섭취군에서 pair-fed군에 비해 높았다. 혈장내 Zn, Cu, Se, Fe 함량은 에탄올 급여군과 pair-fed군 사이에 일정한 경향을 보이지 않았으나 간 조직내 Zn, Cu 함량은 에탄올 급여로 감소된 반면 Fe 함량은 증가되었다. 이러한 결과들로 보아 에탄올은 체내 항산화 영양소 상태의 변화를 초래하며 비타민 A의 공급여부 뿐만 아니라 형태별 섭취에 의해서 항산화 영양소 상태가 변화될 수 있는 것으로 나타났다. 그러나 생체내 항산화 영양소들은 서로 상쇄, 상승 혹은 길항작용을 통해서 상호영향을 미치므로 각각의 항산화 영양소 상태 변화에 대한 정확한 기전 규명을 위해서는 다각적인 연구가 요구된다고 볼 수 있다. 그러나 지질과산화물의 함량과 체내 항산화 영양소 상태는 상호관련성이 있었으며 본 실험에서 에탄올 공급에 의한 항산화 영양소 함량의 감소는 대체로 비타민 A를 retinyl acetate 형태로 섭취시켰을 때 다소 경감되는 것으로 나타났다.
Effects of Bupleuri Radix, Paeonie Radix or Uncariae Ramulus et rncus on experimental yperlipidemia were studied. Hyperlipidemia was induced on male Wistar rats by feeding them on high holesterol diet for one week, as previously described by the authors. Blood lipid profile was verified on these ats by measuring total cholesterol(TC), triglyceride(TG), high density lipoprotein cholesterol(HDL) and low ensity lipoprotein cholesterol(LDL). Then, the rats were followed by feeding normal diet. At the same time, ethanol extracts of the three individual herb medicines were orally administered to the rats for 4 weeks and the parameters above mentioned were monitored. Methanol extract of 5Bupleuri radix reduced the TC value significantly at 2 week point and Paeoneae Radix reduced the TC value at 4 week point in compare to control group, suggesting the antihyperlipidemic effect of the two herbal medicines in vivo. The extract of Uncariae Ramulus et Uncus however did not show antihyperlipidemic effect in our experiment.
Fed-batch culture was designed to increase cell concentration and astaxanthin content by mutant AJ-6 of Phaffia rhodozyma. Fed-batch culture was performed in the continuous feeding with manual adjustment of flow rate to control glucose concentration. When the final glucose concentration was 100 g/L, the cell and astaxanthin were 38.3 g/L, 34.8 mg/L, respectively. Addition of ethanol(10 g/L), when glucose was depleted, the cell and astaxanthin concentration were 37.2 g/L and 45.6, respectively, 5 g/L of acetic acid supplied, 40.6 g/L, 43.9 mg/L were obtained. Ethanol and acetic acid enhanced the astaxanthin content act as precursor of carotenoid synthesis.
Effect of glucose addition to xylose medium on xylitol production was investigated by using Candida parapsilosis ATCC 21019 mutant. With increasing the ratio of glucose to xylose in total amount of 50 g/l as glucose and/or xylose, xylitol production was decreased but ethanol and glycerol production were increased. Ethanol and glycerol concentration were maxmum in 10 g/l of xylose and 40 g/l of glucose medium as 21.5 g/l and 3.6 g/l, respecti- vely. No xylitol was formed in glucose medium without xylose because xylitol could be not produced from glucose. With increasing the ratio of glucose to xylose, the activity of xylose reductase which converted xylose to xylitol were decreased. The activities of xylitol dehydrogeiiase which converted xylitol to xylulose and then cell materials were found to be constant regardless of the ratio of glucose to xylose. This results indicated that glucose addition to xylose medium on cell growth was not affected. In order to prevent the inhibitory effect of glucose on xylitol production, glucose in a fermentor was fed with low concentration and then ethanol and glycerol was critically decreased and the xylitol yield from xylose of the culture with glucose feeding was recovered the almost same as that with only 50 g/l of xylose. However, the xylitol yield from total sugars (xylose and glucose) was decreased and glucose was not contributed to xylitol production. Therefore, the fermentation at high concentration of xylose without glucose was carried out. A final xylitol concentration of 242 g/l which corresponding 80.7% of xylitol yield was obtained from 300 g/l of xylose for 273 hours.

The current study examined the effect of quercetin supplements on the lipid-lowering and antioxidant metabolism in ethanol-fed rats. The control $group(E_8)$ received ethanol only diet for 8 wks, whereas the other group$(E_8Q_4)$ received a diet including quercetin supplementation(0.05% wt/wt) for 4 wks while on the ethanol diet for 8wks. The hepatic alcohol dehydrogenase activity was significantly higher in the $E_8Q_4$ group than in the $E_8$ group. Supplementation with quercetin significantly elevated the HDL- cholesterol concentration, the HDL-C/total-C ratio, and lowered the atherogenic index(AI) compared to the $E_8$ group. The hepatic triglyceride and cholesterol contents were significantly lowered by the quercetin supplement compared to those of the control group. The hepatic HMG-CoA reductase and ACAT activities of the $E_8Q_4$ group were significantly lower than those of the $E_8$ group. The overall potential for antioxidant defense was significantly enhanced by the quercetin supplement, as indicated by a decrease in plasma and hepatic TBARS levels. The hepatic GSH-Px and G6PD activities were significantly higher in the $E_8Q_4$ group compared to the $E_8$ group. The current results suggest that dietary quercetin leads to the inhibition of HMG-CoA reductase and ACAT, which in turn lowers cholesterol levels and normalizes antioxidant enzyme activities.
Xylophagous termites are capable of degrading lignocellulose by symbiotic gut microorganisms along with the host's indigenous enzymes. Therefore, the termite gut might be a potential niche to obtain natural yeasts with celluloytic, xylanolytic and ethanologenic traits required for bioethanol production from lignocellulosic biomass. In this study, we cultured 79 yeasts from three different termites viz. Coptotermes heimi, Odontotermes javanicus and Odontotermes obesus. After suitable screening methods, we identified 53 yeasts, which belonged to 10 genera and 16 different species of both ascomycetous and basidiomycetous yeasts. Most yeasts in the present study represent their first-ever isolation from the termite gut. Representative strains of identified yeasts were evaluated for their cellulolytic, xylanolytic, and ethanologenic abilities. None of the isolates showed cellulase activity; 22 showed xylanolytic activity, while six produced substantial quantities of ethanol. Among xylanolytic cultures, Pseudozyma hubeiensis STAG 1.7 and Hannaella pagnoccae STAG 1.14 produced 1.31 and 1.17 IU of xylanase. Among ethanologenic yeasts, the strains belonging to genera Candida and Kodamaea produced high amount of ethanol. Overall, highest ethanol level of 4.42 g/L was produced by Candida tropicalis TS32 using 1% glucose, which increased up to 22.92 g/L at 35 ℃, pH 4.5 with 5% glucose. Fermentation of rice straw hydrolysate gave 8.95 g/l of ethanol with a yield of 0.42 g/g using the strain TS32. Our study highlights the gut of wood-feeding termites as a potential source of diverse yeasts that would be useful in the production of xylanase and bioethanol.
에탄을 투여로 인한 뇌조직의 지질과산화에 미치는 식이성 Met과 Se의 효과를 구명코자 Se을 결핍시킨 횐쥐를 대상으로 Met을 수준(0, 3, 9g/kg diet)별로 급여 하였으며, 실험기간은 5주와 10주로 사육하여 뇌조직의 알코을 대사효소와 유리기 제거 효소계의 활성을 관찰하였다. 알코을 대사효소인 ADH 활성은 5주와 10주군에서 에탄을 투여시 증가되었고 Met정상급여군이 결핍과 과량급여군에 비해 유의적인 증가를 나타내었다. AIDH 활성은 에탄을 투여로 유의하게 감소되었고 특히 Met와 Se 동시 결핍시 감소가 가장 현저하였다. 투여기간이 길수록 활성은 감소하는 경향이었다. MAO활성은 에탄올 투여군이 대조군에 비해 유의적으로 증가하였으며, Met급여에 의해 활성은 억제되는 것으로 나타났다. SOD 활성은 에탄올 투여로 유의적인 감소를 보였으며 5주 및 10주 모두 Met 과량급여군이 결핍과 정상급여군에 비해 유의적으로 증가하였다. GSH-Px 활성은 에탄을 투여로 유의적으로 감소되 었으며, Met과 Se 동시 결핍군에서 가장 감소정도가 큰 것으로 나타났다. Met 급여수준이 증가할수록 그 활성은 유의적으로 증가하였으며, 10주군이 5주군에 비해 활성이 증가하였다. 에탄올 투여로 증가된 GST 활성은 Met결핍에 의해 더욱 가중되었으며 10주군이 5주군에 비해 GST 활성이 유의적으로 증가된 것으로 보아 장기간의 Met 결핍은 에탄을 중독을 심화시킬 것으로 생각된다. Catalase 활성은 에탄을 투여로 유의적인 증가를 나타내었고 Met 결핍군이 정상급여군 및 과량 급여군에 비하여 유의적인 증가를 보였다. GSH함량은 에탄올에 의해 유의적으로 감소되었는데 Met 급여 수준이 증가할수록 에탄올로 인해 감소된 GSH함량이 유의적으로 증가되었으며, 10주군이 5주군에 비해 증가 정도가 현저하였다. 과산화의 지표인 LPO 함량은 에탄을 투여시 증가되었고 Met 결핍군은 정상급여군에 비해 유의적으로 증가되었으며, 특히 Met 과량급여시 5주군에 비해 10주군이 유의적으로 증가하였다. 이 상의 결과에서 Se 결핍시 Met 급여로 인해 에탄올 대사효소의 활성 증가 및 에탄올에 의해 증가된 뇌의 지질과산화를 줄이는 것으로 보아 Met이 생체의 노화를 방지할 수 있을 것으로 생각되며, Se 결핍으로 증가된 지질과산화 반응에 대한 보호와 적응 수단으로써 방어 효소의 활성도가 높아진 것으로 사료된다.
The effect of the addition of the white powder separated from Sogi (Gyrophora esculenta) by the extraction methods with ethanol and acetone was checked by the feeding experiment in rats fed with a high cholesterol diet. From this experiment, it was confirmed that this material, which was assumed to be the mixture of gyrophoric acid and the decomposition products, could lower both liver and plasma cholesterol levels.
본 연구는 천마가 혈압에 미치는 영향을 평가하기 위하여 본태성고혈압쥐(SHR/NCrj)를 공시하여 고지방식이(lard, 10%)를 급여하면서 천마 분말과 50% 에탄올과 열수추출물이 수축기 혈압(systolic blood pressure)에 미치는 영향을 조사하였다. 고지방식이 4주와 고지방식 이와 천마식이 4주 등 총 8주간 시험 후 종료체중은 대조군(A)이 통계적으로 가장 높았고(p<0.05), 천마분말 1%군(C)이 가장 낮았다. 평균 성장률은 식이군간 차이가 없었고 대조군이 천마식이군에 비해 다소 높았다. 식이섭취량은 천마분말 10%군(D)과 열수추출물 2 brix군(G)이 가장 많았고 천마분말 5%군(C)과 열수추출물 10 brix군(H)이 유의하게 낮았다(P<0.05). 장기중 간장과 신장무게는 열수추출물군(H)이 유의하게 낮았고(p<0.05), 비장무게는 차이가 없었다. 정소무게는 천마분말군(C,D)이 유의하게 높았고, 열수추출물군(G, H)이 가장 낮았다(p<0.05). 혈청지질중 TC 함량은 대조군(A, B)이 가장 높았고, 천마식이군이 유의하게 낮았다(p<0.05). TG 함량은 식이군간 통계적인 차이가 없었다. HDL 함량은 대조군(A)과 천마분말 5%군(D)을 제외한 천마식이군이 유의하게 높은 반면(p<0.05), LDL 함량은 대조군(A, B)이 가장 높았고, 열수추출물군(E, G)이 유의하게 낮았다(p<0.05). 천마식이를 급여하기 전(생후 약 8주령)모든 SHR의 기준혈압은 176.7∼195.1 mmHg으로, 평균혈압은 185.7 $\pm$ 5.8 mmHg이었다. 천마식이를 급여한 후 14일이 경과시 50% 에탄올추출물군(F)과 열수추출물군(G, H)에서 혈압이 각각9.5, 19.0, 10.9 mmHg 감소하였고, 28일에는 특히 50% 에 탄올추출물군(E, F)에서 각각 13.1, 10.5 mmHg으로 유의하게 감소하였다. 천마식이 급여 전 기준혈압 대비 감소율은 50%에탄올(E, F)과 열수추출물군(G)에서 각각 16.8, 20.2, 11.7 mmHg으로써 유의한 혈압변화로 인정되었다. 이상의 결과에서 고지방식이를 급여한 SHR 실험쥐의 혈압강하에 천마의 50% 에탄올추출물(F)이 천마의 분말식이 및 열수추출물에 비해 수축기혈압을 유의하게 감소시킨 것으로 사료된다.

② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
