Rapid Identification of the ${\alpha}$-Glucosidase Inhibitory Compounds from Geranium thunbergii Sieb. et Zucc. by HPLC-micro-fractionation and HPLC-UV-$MS^n$

  • In this study ethanol extracts of aerial part of Geranium thunbergii Sieb. et Zucc. was investigated for their ability to inhibit a-glucosidase, and thus was fractionated using two organic solvents, including dichloromethane, ethyl acetate. The ethyl acetate-soluble fraction, which manifested potent enzyme inhibitory properties, was then followed by tracking down the active compound by combining HPLC micro-fractionation to an enzyme assay in 96-well plate. The ${\alpha}$-glucosidase inhibitory activity profile showed that two peaks exhibited potent inhibitory activity, and then the structural analyses of the two peaks were carried out by HPLC-UV-MS. The main ${\alpha}$-glucosidase inhibitory compounds in the ethyl acetate-soluble fractions of ethanol extracts of Geranium thunbergii Sieb. et Zucc. were tentatively identified as geraniin and kaempferol-7-rhamnoside.

  • The experiment was carried out to study the bothanical characteristics of Aconitum charmichaeli Sieb. et Zucc in Korea.The results obtained are summarized as follows ; maximum flowering date was 15 Sep. and plantheight was 65cm, stem diameter was 8.7mm, No. of node was 25 and node of flowering initiation was15th.In flowering, No. of anothers were 52 and No. of flowers were 49.In leaf, length and width was same size, lower leaf size was 2-3 times than upper leaf, lower leafarea was 8.4 times than upper leaf.In tuber, No. of tuber per plant was 8, fresh weight per tuber was 25.4g, total fresh weight per plantwas about 200g.

  • A model was designed to identify wood species between Pinus densiflora for. erecta Uyeki and Pinus densiflora Sieb. et Zucc. using the near-infrared (NIR) spectroscopy in combination with principal component analysis (PCA) and partial least square discriminant analysis (PLS-DA). In the PCA using all of the spectra, Pinus densiflora for. erecta Uyeki and Pinus densiflora Sieb. et Zucc. could not be classified. In the PCA using the spectrum that has been measured in sapwood, however, Pinus densiflora for. erecta Uyeki and Pinus densiflora Sieb. et Zucc. could be identified. In particular, it was clearly classified by sapwood in radial section. And more, these two species could be perfectly identified using PLS-DA prediction model. The best performance in species identification was obtained when the second derivative spectra was used; the prediction accuracy was 100%. For prediction model, the $R_p{^2}$ value was 0.86 and the RMSEP was 0.38 in second derivative spectra. It was verified that the model designed by NIR spectroscopy with PLS-DA is suitable for species identification between Pinus densiflora for. erecta Uyeki and Pinus densiflora Sieb. et Zucc.

Inhibitory Activity of IL-6 Production by Flavonoids and Phenolic Compounds from Geranium thunbergii

  • Three flavonoids (1 - 3) and three phenolic compounds (4 - 6) were isolated from the whole plant of Geranium thunbergii Sieb. et Zucc (Geraniaceae). Their structures were determined by chemical and spectral analysis. These compounds were examined for the inhibitory activity of IL-6 production in $TNF-{\alpha}$ stimulated MG-63 cell. Among the isolated compounds, gallic acid (4) and gallic acid methyl ester (6) showed potent inhibitory activity.

  • The dried ground needles (2.0 kg) of Chamaecyparis pisifera (Sieb. et Zucc.) Endlicher were extracted with acetone-$H_2O$ (7:3, v/v), concentrated, and fractionated with a series of n-hexane, methylene chloride, ethyl acetate and water on a separation funnel. Each fraction was freeze dried, then a portion of ethyl acetate soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-n-hexane mixture as eluents. The isolated compounds were identified by cellulose TLC, $^1H$-, $^{13}C$-NMR, COSY, HETCOR, FAB and EI-MS. (+)-catechin, taxifolin-3-O-${\beta}$-D-xylopyrano-side, quercetin-3-O-${\alpha}$-L-rhamnopyranoside were isolated from the ethyl acetate soluble fraction of Chamaecypairs pisifera needle. Antioxidative tests on the isolated compounds indicated that all of the compounds showed similar values to ${\alpha}$-tocopherol and BHT as controls.

  • To develop natural food preservative, antimicrobial activity of Pinus densiflora Sieb. et Zucc. (PD) ethanol extract against pathogens were investigated. The growth of pathogens such as Staphylococcus aureus ATCC 29737, Bacillus subtils KCTC 1021, Bacillus megaterium KCTC 3007 and Vibrio parahaemolyticus ATCC 17802 was inhibited as much as 4 to 6 log cycle in tryptic soy broth containing 40mg/㎖ of PD extract but Salmonalla typhimurium ATCC 14028 and Pseudomonas fluorescens ATCC 21541 were not inhibited. The total, gram negative and psychrotrophic bacterial count were inhibited in minced pork containing 40mg/㎖ of PD extract for 20 days at 4$\^{C}$. The sensory quality of hamburger patty such as taste, flavor and overall acceptability were not decreased by the addition of 40mg/㎖ PD(p<0.05).

Optimum Condition for Acetic Acid Fermentation Using Mume (Prunus mume Sieb. et Zucc) Fruits (매실을 이용한 초산 발효의 최적 조건)

  • To produce acetic acid from Mume (Prunus mume Sieb. et Zucc) fruits, fermentation conditions were optimized by a response surface methodology (RSM) using the fractional factorial design with 3 variables and 5 levels. The coefficient of determination ($R^2$) of response surface regression equation for acetic acid production was 0.9462. Optimum conditions for acetic acid production were involved with 8.76% of alcohol content 26.27$^{\circ}C$ of fermentation temperature and 8.42% of sugar content. Finally, predicted level of acetic acid production at these conditions was 3.23%.

  • This experiment was carried out for the improvement of drying schedules for 50 mm-thick Japanese red pine (Pinus densiflora Sieb. et Zucc.) lumber. Drying rates were analyzed comparatively after drying to 15% of final moisture content through the applications of conventional kiln drying schedule, time schedule, continuously rising temperature schedule, and drying rate schedule. As results drying rate of 50 mm-thick red pine lumber was 0.53%/hr with conventional kiln drying schedule, 0.9%/hr with time schedule, 2.29%/hr with continuously rising temperature schedule, and 1.52%/hr with drying rate schedule, respectively.

  • This study was performed to anticancer activities and immune modulatary activities according to the parts of the S. japonica Sieb. et Zucc. The cytotoxicity on human kidney cell (HEK 293) was showed below 27.4% in adding the methanol extracts. The anticancer activity were increased in over 60% by barks extracts in AGS and MCF-7 cells. The immune cell growth using human immune B and T cells was improved by the barks extracts of S. japonica Sieb. et Zucc. in adding 1.0mg/ml concentration. The secretion of the IL-6 and TNF-${\alpha}$ from human immune B and T cells was showed secretion for the amount of cytokines by bark extracts of S. japonica Sieb. et Zucc. NK cell growth was increased against control all of the extracts of S. japonica Sieb. et Zucc. Densitometric analysis of Bcl-2 revealed that possible to decrease potentialities of taking cancer in adding of extracts from S. japonica Sieb. et Zucc. From the results, the roots and barks extracts of S. japonica Sieb. et Zucc. were showed useful biological activities.
