Screening for Antimicrobial Activity from Korean Plants (국내산 식물의 항균활성 검색)

  • 141 methanol extracts from 125 plant species which populate in Korea were screened for antimicrobial activity against various food-borne pathogens and food spoilage microorganisms. Those plants were selected from 3 different plant groups: traditional herbs, edible plants and flowers. The methanol extracts were tested by using the disk diffusion assay against five bacteria: Bacillus subtilis, Staphylococcus aureus, Pseudomonas aeruginosa, Enterobacter aerogenes, Escherichia coli. From the evaluation of the inhibition zone diameter of microbial growth, the most significant antimicrobial activity against Bacillus subtilis, Staphylococcus auresus, Pseudomonas aeruginosa, Enterobacter aerogenes, Escherichia coli was observed from the extract of Schizandra chinensis (Turcz.) Baill., Rheum officinale Baill., Schizandra chinensis (Turcz.) Baill., Koelreuteria paniculata Lax and Crataegus pinnatifida Bunge, respectively. The extract from many plants - Koelreuteria paniculata Lax, Chaenomeles sinensis Koehne, Scutellaria bacicalensis Georgi, Castanea crenata Sieb. et Zucc., Rosa centifolia L., Allium fistulosum L. var. giganteum Makino, Crataegus pinnatifida Bunge, Schizandra chinensis (Turcz.) Baill., Lonicera japonica - showed antimicrobial activity all four tested bacteria.

  • Manuka oil sometime named New Zealand's tea tree oil is soluble in oil and come from nature. The $\alpha$-pinene extracted from Manuka oil and R-limonene which is one of the component of extracted Citrex from Grapefruit were used to estimate the antimicrobial activity and to improve the capability of antiseptic. Disk diffusion and broth dilution methods were used to measure the antimicrobial activity. Escherichia coli which is gram-negative bacteria and Staphylococcus aureus which is gram-positive bacteria were used as strain. The antimicrobial activity of Manuka oil and $\alpha$-pinene for Escherichia coli, Staphylococcus aureus is similar when the concentration of Manuka oil and $\alpha$-pinene is $10{\mu}l$. However, Antimicrobial activity of Manuka oil for Escherichia coli, Staphylococcus aureus is better than that of $\alpha$-pinene when the concentration of Manuka oil and $\alpha$-pinene is low. Antimicrobial activity of Citrex is superior to that of R-limonene. The proper ratio of Maunka oil and Citrex can improve the antimicrobial activity. The proper ratio obtained from studies was 75% of Maunka oil and 25% Citrex for Escherichia coli, 25% of Maunka oil and 75% Citrex for Staphylococcus aureus.

  • Microbes have been used extensively in various fields of researches and industries but has not been used widely for microfluidic biosensor applications because it is difficult to immobilize properly to a small space. Therefore, we developed a microbial immobilization method for microfluidic devices using single-walled nanotubes and dielectrophoretic force. Single-walled nanotubes and Escherichia coli were aligned between two cantilever electrodes by a positive dielectrophoretic force resulting in a film of single-walled nanotubes with attached Escherichia coli. The optimal condition of film formation without a cell lysis was investigated. Diameter of single-walled nanotubes and electric field (intensity and duration of application) had an effect on the cell viability. On the other hand, the cell concentration of the suspension did not affect the cell viability. Paraoxon was detected using single-walled nanotubes film with attached Escherichia coli that expressed organophosphorus hydrolase. This film which is suspended from the substrate showed faster response time than sensors that are not suspended from the substrate.

  • The purpose of this study was to examine O serotypes, colicin and hemolysin production. antibiotic susceptibility and haemagglutinating ability to animal erythrocytes among Escherichia coli strains isolated from pigeons in Taegu province. Of the 166 strains isolated, 28 strains (16.9%) were classified into 6 O serotypes and their types were O20(42.9%), O15(17.9%), O139(14.3%), O101(10.7%), O149(7.1%) and O8(7.1%). Of the 166 strains isolated, none was hemolytic and 3(1.8%) were colicinogenic. Antibiotic susceptibility test of Escherichia coli isolates was performed by the agar dilution method, using ampicillin, chloramphnicol, gentamicin, rifampicin, streptomycin (Sm), nalidixic acid, sulfadimethoxine and tetracycline (Tc). Forty four strains (26.5%) were resistant to one or more drugs and the most common resistance patterns were SmTc (27.3%). Of the 44 drug resistant strains, 6 strains haemagglutinated erythrocytes of chicken, guinea pig and 2 of the 6 strains agglutinated goose erythrocytes.

  • Shiga-like toxin-II(SLT-II)-producing Escherichia coli 0157 : H7 strain B2387이 분비하는 SLT-II가 gnotobiotic자돈에서의 뇌혈관 병변을 일으키는 pathogenesis에 관해서 실험을 했다. 제왕절개 수술로 태어난 자돈들을 두 그룹으로 나누어서, 한 그룹에는 SLT-II 중화항체를 포함한 혈청을 구강을 통해서 수동면역을 시키고, 또다른 한 그룹에는 SLT-II 중화항체가 포함되어 있지 않은 혈청을 구강을 통해서 수동면역시켰다. 24시간후 두 그룹 모두에게 SLT-II producing Escherichia coli O157 : H7 strain B2387를 구강으로 접종했다. SLT-II 중화항체가 포함되어 있지 않은 혈청으로 수동면역시킨 그룹의 자돈들은 설사와 맹결장염, 신경증상, 뇌혈관병변을 일으키고, plasma의 prostacyclin의 level이 증가했다. 하지만 SLT-II 중화항체가 포함되어 있는 혈청으로 수동면역시킨 그룹의 자돈들은 설사와 맹결장염은 유발했지만, 신경증상과 뇌혈관병변은 관찰되지 않았고, prostacyclin의 level도 증가하지 않았다. 이런 실험결과는 SLT-II 중화항체는 뇌혈관병변은 방어하지만 맹결장염은 방어하지 못한다는 의미를 나타내며, prostacylin의 증가는 뇌혈관의 endothelium의 병변을 의미한다.

  • In this study, we coated the polypropylene filter with titania sol and investigated various characteristic such as its deodorization efficiency and sterilization effect of gram negative bacillus Escherichia coli(ATCC 25922) and gram positive bacillus Staphylococcus aureus(ATCC 6538). As the results, in case of titania sol coated polypropylene filter, the deodorization rate of ammonia, trimethylamin and acetic acid which cause indoor air pollution was 66%, 90% and 16% respectively. Also. the sterilization rate of gram negative bacillus Escherichia coli(ATCC 25922) and gram positive bacillus Staphylococcus aureus(ATCC 6538) was 99.9% after 24 hours.

Repeated-Batch Operation of Immobilized ${\beta}$-Galactosidase Inclusion Bodies-Containing Escherichia coli Cell Reactor for Lactose Hydrolysis

  • In this study, we investigated the performance of an immobilized ${\beta}$-galactosidase inclusion bodies-containing Escherichia coli cell reactor, where the cells were immobilized in alginate beads, which were then used in repeated-batch operations for the hydrolysis of o-nitrophenyl-${\beta}$-D-galactoside or lactose over the long-term. In particular, in the Tris buffer system, disintegration of the alginate beads was not observed during the operation, which was observed for the phosphate buffer system. The o-nitrophenyl-${\beta}$-D-galactoside hydrolysis was operated successfully up to about 80 h, and the runs were successfully repeated at least eight times. In addition, hydrolysis of lactose was successfully carried out up to 240 h. Using Western blotting analyses, it was verified that the ${\beta}$-galactosidase inclusion bodies were sustained in the alginate beads during the repeated-batch operations. Consequently, we experimentally verified that ${\beta}$-galactosidase inclusion bodies-containing Escherichia coli cells could be used in a repeated-batch reactor as a biocatalyst for the hydrolysis of o-nitrophenyl-${\beta}$-D-galactoside or lactose. It is probable that this approach can be applied to enzymatic synthesis reactions for other biotechnology applications, particularly reactions that require long-term and stable operation.

  • The role of enteropathogenic Escherichia coli(EPEC) was evaluated in a group of children with endemic diarrhea admitted to Hanyang University Hospital in Seoul, Korea. EPEC was detected in fecal samples of 23% of 100 cases and 4.5% of 44 concurrent control children. The most commonly isolated EPBC strains were serogroups $O_{18a}O_{18c}:K_{77},\;O_{86a}:K_{61},\;O_{119}:K_{69},\;and\;O_{128}:K_{70}$. On testing for enterotoxin production, 6(26%) strains were isolated from 17% of the 100 diarrheal children and in 4.5% of the 44 well controls(P<0.05). Our study supports the concept that EPEC may be an important cause of endemic diarrheas in Korea.

  • A phylogenetic comparison of homologous protein can often supplement genetic and biochemical analysis by revealing conserved structures that are critical for function(Waugh et al., 1989). I therefore isolated a secY homologue from B. subtilis, a gram positive bacterium evolutionary distant from E. coli. The comparison and interplay between these two bacterial systems should contribute greatly to our understanding of the functions and interactions within systems evolved for protein translocation in both prokaryotic and eukaryotic organisms.

Virulence factors, antimicrobial resistance patterns, and genetic characteristics of hydrogen sulfide-producing Escherichia coli isolated from swine

  • Escherichia (E.) coli is commensal bacteria found in the intestine; however, some pathogenic strains cause diseases in animals and humans. Although E. coli does not typically produce hydrogen sulfide ($H_2S$), $H_2S$-producing strains of E. coli have been identified worldwide. The relationship between virulence and $H_2S$ production has not yet been determined. Therefore, characteristics of $H_2S$-producing isolates obtained from swine feces were evaluated including antibiotic resistance patterns, virulence gene expression, and genetic relatedness. Rates of antibiotic resistance of the $H_2S$-producing E. coli varied according to antibiotic. Only the EAST1 gene was detected as a virulence gene in five $H_2S$-producing E. coli strains. Genes conferring $H_2S$ production were not transmissible although the sseA gene encoding 3-mercaptopyruvate sulfurtransferase was detected in all $H_2S$-producing E. coli strains. Sequences of the sseA gene motif CGSVTA around Cys238 were also identical in all $H_2S$- producing E. coli strains. Diverse genetic relatedness among the isolates was observed by pulsed-field gel electrophoresis analysis. These results suggested that $H_2S$-producing E. coli strains were not derived from a specific clone and $H_2S$ production in E. coli is not associated with virulence genes.
