독소루비신 생합성 유전자의 발현을 촉진시키는 유전자인 dnrI와 다나루비신으로부터 독소루비신으로의 생전환에 관여하는 유전자인 doxA를 ermE 프로모터가 포함된 pSE34에 도입하였을 때 각각 5.5배, 2.5배의 독소루비신 생산성 증가가 이루어졌다. 독소루비신 생합성 유전자군의 발현패턴 분석을 위한 DNA microarray system을 구축하였고, 고생산 균주의 독소루비신 생합성 유전자 발현 패턴을 DNA microarray를 통해 확인하였다. 독소루비신 생합성 유전자군의 세포성장에 따른 발현패턴을 분석한 결과, 독소루비신 생산성 증가에 따라 생합성 유전자의 발현도 증가함을 확인할 수 있었고, pSE34를 통해 도입해준 donA, dnrI 유전자의 경우 전체 생합성 유전자의 평균보다 높은 수준의 발현량을 보여줌으로써, ermE 프로모터에 의해 발현이 극대화되었음을 확인할 수 있었다. 독소루비신 내성 유전자의 경우 다른 독소루비신 생합성 유전자들에 비해 발현정도가 크게 증가했고, DnrI 의해 조절을 받는 다른 유전자들의 발현 수준과 비교하였을 때 TDP-daunosamine을 생합성의 첫 번째 단계에 관여하는 dnmL 유전자는 그 발현양의 증가가 크지 않았다. 따라서 DNA microarray 시스템 분석 결과, 독소루비신 생산성 극대화를 위해서는 dnrI, doxA, drrA, drrB, drrC, dnmL 등의 유전자들의 안정적 발현이 매우 중요하고도 핵심적인 인자임이 확인되었다.
Streptococcus sp. TR-1 which has inducible resistance to MLS antibiotics was isolated from soil samples in Korea. Streptococcus sp. TR-1 was cultured in Lysis broth, then a plasmid was isolated by modified Elliker method. Bacillus subtilis UOTO277 was transformed with that plasmid. This result showed that the plasmid has the gene relating with inducible resistance to MLS antibiotics. It was named pMB4 and its size was determined about 2.4 Kb by results of digestion with various restriction enzymes. Restriction endonuclease cleavage site map of pMB4 plasmid was made by double digestion of the plasmid. pMB4 plasmid has different restriction endonuclease site map from the other plasmids that have been discovered in Streptococcus sp. so far. And it could be identified that pMB4 plasmid does not have homology with ermK of Bacillus licheniformis EMR but has homology with ermC of Staphylococcus aureus from the results of Southern hybridization.
In order to evaluate the pollution of heavy metals in offshore surface sediments around shipyards in Korea, surface sediment samples were collected at eleven stations around four major shipyards located in the southeastern coast of Korea in summer 2010 and nine kinds of heavy metals such as copper(Cu), zinc(Zn), cadmium(Cd), lead(Pb), chrome(Cr), arsenic(As), mercury(Hg), iron(Fe) and aluminum(Al) in sediments were analyzed. The concentrations of Cu at all sampling stations were in the range of 47.10~414.96 mg/kg and exceeded TEL(Threshold Effects Level) 20.6 mg-Cu/kg of Korean marine environmental standards for offshore sediments and ERL(Effect Range-Low) 34.0 mg-Cu/kg. The concentrations of Cu at seven stations around four shipyards were 65.18~414.96 mg/kg and exceeded PEL(Probable Effects Level) 64.4 mg-Cu/kg of Korean marine environmental standards for offshore sediments. The concentration of Cu at one station around B-shipyard was 414.96 mg/kg and exceeded ERM(Effect Range-Median) 270.0 mg-Cu/kg. The concentrations of Zn at all stations were in the range of 135.09~388.79 mg/kg which exceeded ERL 150.0 mg-Zn/kg. The concentrations of Zn at seven stations around four shipyards were 157.57~388.79 mg/kg and exceeded PEL 157.0 mg-Zn/kg. The concentration of Zn at one station around B-shipyard was 388.79 mg/kg and was approaching ERM 410.0 mg-Zn/kg. The concentrations of Cd at all stations were in the range of 0.11~0.54 mg/kg and were below TEL 0.75 mg-Cd/kg and ERL 1.2 mg-Cd/kg. The concentrations of Pb at all stations were in the range of 18.04~105.62 mg/kg. The concentrations of Pb at two stations around B-shipyard were 73.87~105.62 mg/kg which exceeded TEL 44.0 mg-Pb/kg and ERL 46.7 mg-Pb/kg, and were below PEL 119.0 mg-Pb/kg and ERM 218.0 mg-Pb/kg. The concentrations of Cr at all stations were in the range of 51.26~85.39 mg/kg. The concentration of Cr at one station around B-shipyard was 85.39 mg/kg and exceeded ERL 81.0 mg-Cr/kg. The concentrations of As at all stations were in the range of 8.70~22.15 mg/kg which exceeded ERL 8.2 mg-As/kg and were below ERM 70.0 mg-As/kg. The concentrations of As at eight stations around A-shipyard, B-shipyard and D-shipyard were 14.93~22.15 mg/kg which exceeded TEL 14.5 mg-As/kg and were below PEL 75.5 mg-As/kg. The concentrations of Hg at all stations were in the range of 0.02~0.35 mg/kg. The concentrations of Hg at three stations around A-shipyard were 0.11~0.13 mg/kg which were almost equal to TEL 0.11 mg-Hg/kg. Those at two stations around B-shipyard were 0.27~0.35 mg/kg which exceeded TEL 0.11 mg-Hg/kg and ERL 0.15 mg-Hg/kg, and were below PEL 0.62 mg-Hg/kg and ERM 0.71 mg-Hg/kg. The concentrations of Fe and Al at all stations were in the range of 2.90 3.66 % and 3.12 6.80 %, respectively. These results imply that heavy metals such as copper, zinc, lead, arsenic and mercury were likely to be transferred to marine environment from shipyards, especially from B-shipyard.
Objectives : In this study, an essential oil fragrance from Danggwi was administrated into the neural stem cell and the effect of the essential oil on the differentiation and proliferation of the neural stem cells were observed. Methods : The establishment of the neural stem cell was identified via Nestin, DAPI dye. An essential oil fragrance from Danggwi was administrated with a proved optimum level for the survival of the cell through MTT assay. Also, according to the analysis of Western blot, the essential oil fragrance from Danggwi promotes the phosphorylating of Akt, Erk, ERM protein. Results : MTT assay showed increased in GFAP. The result indicates that the differentiation to astrocyte is promoted. The phosphorylation levels of ERM, Erk and Akt were increased at 60 min after addition of 5 ug/ml of essential oil fragrance from Danggwi and sustained to 48 hours. These imply that essential oil fragrance from Danggwi may induce the survival and the proliferation of the differentiated cells. Conclusions : These results suggest that the essential oil fragrance from Danggwi can be effective for the in vivo study of degenerative neuronal disease using neural stem cell.
When an emergency situation happens in buildings, the top priority is to ensure the occupant from danger as soon as possible. Achieving that goal is a multifaceted and difficult task. However, current evacuation systems have many deficiencies in dealing with the emergency in multi-level structures. The shortage of abilities to continuously update database, predict the future situation and provide the information to users with contextual information is the limit in current systems. Thus, it is very crucial to introduce Evacuation Information System (EIS), which is able to respond quickly to the emergency, and transfer the information to both the administrator and the occupant. The main purpose of this paper is to build EIS on the basis of the indoor Geographical Information System (GIS). When the emergency happens, EIS gives the instruction to Emergency Response Model (ERM) at once. ERM carries out the order and calculates the optimal evacuation routes, then sends the result to EIS. At last, EIS transmits evacuation messages to the occupant who implements evacuation plan. This paper highlights the benefits of EIS in two aspects. One is that EIS can update the data continuously to support evacuation strategy-making. The other is that it can transmit evacuation messages to both the administrator and the occupant.
구강악취 원인 물질인 $H_2S$를 발생시키는 NaHS로 RAW 264.7 세포를 자극시킨 결과, VSCs 생성에 관련된 CSE 유전자 발현증가와 함께 IL-6, COX-2, iNOS와 같은 염증매개 cytokine이 증가하며, 아울러 본 대식세포나 백혈구 활성에 관련된 세포내 골격단백질인 ERM의 발현증가가 유도되었다. 이러한 사실은 이전의 연구결과인 VSCs의 생성이 위점막 염증 및 손상과 연관되어 있다는 이전의 연구를 증빙해주는 사실인데, 더욱 흥미로운 사실은 고려 홍삼의 전처치를 통하여 CSE의 억제는 물론 염증 연관 cytokine 및 ERM의 발현억제를 유도할 수 있으며, 이는 세포내 신호전달계인 p38 및 ERK의 억제, 주로는 ERK 비활성화를 통하여 작동됨을 규명하였다. 고려홍삼이 구강악취 환자에게서 증상 완화내지는 소멸을 위하여 매우 유용하다는 이전의 연구결과에 본 연구결과가 추가됨으로 고려홍삼에 의한 VSC 생성억제 기전이 관련된 유전자의 약화는 물론 cytokine 억제 및 백혈구 활성에 관련된 인자억제를 통한 항염증 작용에 근거한다는 기전을 추가할 수 있었다.

The prevalence of resistance to a range of macrolides was determined for clinical isolates of common respiratory pathogens using NCCLS testing methods and interpretative criter ia.71.4% of Streptococcus pneumoniae, 62.3% of Staphylococcus aureus, 50.8% of coagulase-negative staphylococci and 4.4% of Strpetococcus pyogenes were erythromycin resistant. Also, the rates of resistance to other macrolides and clindamycin in these clinical isolates were as high as to eryth -romycin. Almost all of the macrolide-resistant isolates were positive for erm of the methylase gene, or mef of the efflux gene.
