Effect of Glucosylceramides and Sterols Isolated from Agaricus Blazei Extract on Improvement of Skin Cell (신령버섯에서 분리된 Glucosylceramide 및 Sterol의 피부 세포 개선 효과)

  • Agaricus blazei Murill (Almond mushroom) has many beneficial effects, such as anti-cancer, immuneenhancement, and anti-obesity. Also, its skin benefits have been reported for antioxidant, anti-inflammatory, and whitening. In order to elucidate these effects, many studies have been conducted. In this study, we reconfirmed the skin efficacy of the extract of the mushrooms mushrooms. The Agaricus blazei extract showed inhibition of melanin synthesis, enhancement of collagen synthesis, and up-regulation of gene expression (hyaluronan syntahase-2, 3 and aquaporin-3) at 100 ㎍/mL. and identified the ingredients from the extract. We further investigated them to find an applicability as cosmetic ingredients. The ingredients were confirmed comparison of their spectroscopic data with literature values. They were identified as being ergosterol (1), 5-dihydroergosterol (2), cerevisterol (3), cerebroside B (4), cerebroside D (5), adenosine (6), and benzoic acid (7). Among these compounds, we evaluated skin efficacy for two cerebrosides and three ergosterol derivatives that have not been reported its efficacy. As a result, 5-dihydroergosterol (2) inhibited melanogenesis in B16F10 and promoted collagen biosynthesis in human dermal fibroblast. In addition, cerevisterol (3), cerebroside B (4), and cerebroside D (5) inhibited NO production in RAW 264.7 cell. In particular, cerebroside D (5) increased the expression of hyaluronan synthase-2 and aquaporin-3 genes in HaCaT. These results suggest that Agaricus blazei extract and isolated compounds can be used as cosmetic ingredients.

Effect of UV-B irradiation time on storage characteristics of the fruiting body of Heuktari (UV-B조사 시간에 따른 흑타리 자실체의 저장 특성)

  • This experiment was conducted to investigate the changes in freshness parameters and determine the optimal UV-B irradiation conditions for each storage period for Heuktari, an oyster mushroom cultivar, grown under LED lightNo significant changes were observed in the CO2 and O2 content until day 12 of storage; however, after day 15, the CO2/O2 ratios were higher in the UV-treated groups than in the untreated groups. No color difference was observed between the UV-treated and untreated groups. In all UV-treated groups, hardness decreased over time; however, the group irradiated for 15 minutes showed higher hardness values than those of the untreated group. The UV-irradiated group showed higher ergosterol content than did the untreated group, and scanning electron microscopy revealed that mushroom cell wall destruction increased with increased UV treatment time. Overall, these findings confirmed that UV-B irradiation increased the ergosterol content of Heuktari but decreased its freshness and negatively impacted other storage characteristics.

  • Attempts were made to investigate the sterol components of Boletus aerus Bull. which grows wildly in Korea. Its carpophores were collected in Gyeong-Gi Province and extracted with chloroform and methanol. A sterol was isolated and identified as ergosterol by T.L.C., G.L.C. and chemical tests.

  • The carpophores of Lentinus lepideus Fr. which grows wildly in Korea were collected in Gyeong Gi Province to examine the chemical components of the fungus. They were extracted with a mixture of chloroform and methanol. From the extract four spots were detected by TLC. One of these spots was identified as ergosterol by TLC, GLC and chemical tests.

  • Choneophora trispora 단독 혹은 혼합배양하여 Carotene과 Vitamin A의 생성을 연구하였다. 1) $\beta$-Carotene 생성량은 chonephora trispora의 (+)주보다 (-)주가 좋았고 단독배양보다 혼합배양이 좋았다. 2) $\beta$-cionone은 $\beta$-carotene 생합성을 촉진하였다. 3) FeCl$_3$은 균생육과 $\beta$-carotene 생합성을 저해하였다. 4) 균체추출물은 ergosterol, carotene은 확인할 수 있으나 vitamin A는 확인 할수 없었다.

  • Regeneration of protoplast was effective by preincubating spore suspension containing 30$\mu g$/ml of 2-DG for 4 hours, and CBE medium containing casamino acid, bovine serum albumin, ergosterol and myoinositol was found to be more efficient than any other regeneration medium tested in this experiment. The regeneration frequency was about 30%. Optimal conditions for conidial protoplast fusion were obtained by treatment of protoplasts with 10 mM $CaCl_2$ and 30% polyethylene glycol 4000 (pH 7.5) as fusogenic agent at $37^{\circ}C$ for 10 minutes. The fusion frequency was $8.2\times 10^{-4}$. The higher productivity of enzyme of fusant FWN-56 was achived: 2.3-fold for CMCase, 1.5-fold for avicelase, 1.8-fold for $\beta$-glucosidase and 2.5-fold for xylanase compared to that obtained in two parental strains. The genetic stability of fusant after maintenance on minimal medium for more than 4 weeks was high because segregant rate was below 1%. The conidial DNA content of fusant was 1.4-1.6 times higher than that of the parental strains, The nucleus size of fusants were also higher than that of each parental strains.

  • Compositions of fatty acid and sterol of Hirneola auricula-Judge and Gyrophora esculenta produced is Korea were analyzed by gas liquid chromatographic(GLC) and infra red(IR) spectro-photometric techniques. As results, H. auricula showed linoleic acid 33.73, palmitic acid 15.52, stearic acid 5.03, oleic acid 16.03, linolenic acid 17.80, and unknown acid 11.89 % respectively, in their composition, while G. esculenta linoleic acid 46.35, palmitic acid 31.71, oleic acid 16.82, unknown acid 5.12 %, and trace of stearic and linolenic acids, respectively. Sterols were separated by thin layer chromatographic technique from both samples and identified by IR analysis. Two sterols, sitosterol and ergosterol, were present in both samples.

  • Three stigmastane-type sterols and one ergostane-type sterol were isolated from the ethyl acetate soluble fraction of the aerial parts of Artemisia princeps Pampanini (Sajuarissuk). From the results of physico-chemical data including NMR, MS and IR, the chemical structures of the compounds were determined as $stigmasta-5,22-dien-3,{\beta}-ol (stigmasterol, 1),stigmast-5-en-3{\beta}-ol({\beta}-sitosterol,2), 5{\beta},8{\beta}-epidioxy-5{\beta},8{\beta}-ergosta-6,22-dien-3{\beta}-ol(ergosterol peroxide, 3),\;and\;{\beta}-sitosterol\;3-O-{\beta}D-glucopyranoside(daucosterol,4)$.

Influence of Hexaconazole on Biochemical Constituents of Groundnut (땅콩의 생화학성분에 미치는 Hexaconazole의 영향)

  • In this study, hexaconazole 5% SC, an ergosterol biosynthesis inhibitor, was tested on groundnut with its recommended ($500\;mL\;ha^{-1}$) and higher ($2,000\;mL\;ha^{-1}$) concentrations under greenhouse conditions in India. Its influence on biochemical constituents of groundnut plants was assessed apart from its disease management potential against late leaf spot caused by Phaeoisariopsis personata (Berk and Curt). Likewise, leaf samples were collected from hexaconazole 5% SC-sprayed plants at different time intervals. Thereafter, their analyses showed considerable differences in the plant constituents, such as chlorophyll, soluble protein, and total phenol contents and the activity of nitrate reductase enzyme. The induction activity of defense-related enzyme, peroxidase, was also analyzed. However, no difference was observed in the isozymic pattern. Moreover, the ground kernels collected from treated plants also showed no difference in the estimated carbohydrate and other constituents.

The Conditions Affecting Ethanol Tolerance of Yeast strains in Alcohol Ferwlentation - Study on the Aeration and Lipid Addition (알콜발효에서 효모의 에탄올 내성 조건-통기와 lipid첨가에 대한 연구)

  • The alcohol fermentation was carried out to study the effect of aeration and unsaturated fatty acids added on the ethanol tolerance of Saccharomyces cerevisiae STV89 and Kluyveromyces fragilis CBS397. The cell growth rate and ethanol production rate was stimulated by aeration and the cell mass production and ethanol production were also substantially improved. With respect to strains, the maximum specific growth rate and overall ethanol productivity of K. fragilis under aerated condition were 6.4 fold and 4.4 fold higher than those of strictly anaerobic condition, although those of S. cerevisiae were increased 1.7 times and 2.3 times by aeration. The addition of ergosterol, linoleic acid and oleic acid also improved the cell growth and ethanol production of S. cerevisiae and K. fragilis. Thus it was found that oxygen and unsaturated fatty acids added played a decisive role on the increase of ethanol tolerance of yeast strains.
