  • The fruiting body of Phellinus linteus was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_2$O. The repeated silica gel and ODS column chromatographies of the EtOAc fraction led to isolation of four sterols. From the result of spectral data including NMR, MS and IR, the chemical structures of the sterols were determined as ergosta-7,24(28)-dien-3${\beta}$-ol (episterol, 1), 5${\alpha}$,8${\alpha}$-epidioxyergosta-6,9(11),22-trien-3${\beta}$-ol (dehydrop-eroxyergosterol, 2), 5${\alpha}$,8${\alpha}$-epidioxyergosta-6,22-dien-3${\beta}$-ol (ergoterol peroxide, 3), and $3{\beta}$,$5{\alpha}$-dihydroxy-6${\beta}$-methoxyergosta-7,22-diene (6-O-methylcerevisterol, 4). The ergosterols have been first isolated from this mushroom in this study.

  • This study analyzed Tuber melanosporum, Tuber aestivum,and Tuber magnatum, specifically the content of 𝛽-glucan, amino acids, nucleic acid-related substances, vitamin C, and ergosterol. The 𝛽-glucan content was highest in T. magnatum (20.54%). The free amino acid content was highest in T. aestivum, with the major amino acids being cystine, glutamic acid, and aspartic acid. The total amino acid content was the highest in T. magnatum, the primary amino acids being cystine, glutamic acid, and lysine. The content of nucleotide related compounds was the highest in T. magnatum (5.09 mg/100 g). The highest content of vitamin C (10.15 mg/100 g) and ergosterol (596.91 mg/100 g) was found in T. magnatum, which had the highest useful component content among the three truffles. Additional studies investigating the variation in useful components according to collection time and growth environment are needed.

Isolation and Structure Determination of Metabolites from Cultures of Aspergillus protuberus (Aspergillus protuberus 배양물로부터 대사체 분리 및 구조 결정)

  • Marine-derived microbes have yielded a variety of metabolites so far. In the course of the project to find metabolites from marine microbes, an isolate of Aspergillus protuberus (SF 5767) was selected for chemical investigation. A large scale culture of this strain in PDA media was extracted with an organic solvent and the extract was fractionated by silica gel column chromatography. Repeated reverse phase HPLC of the fractions led to the isolation of three metabolites. Their chemical structures were elucidated as deoxybrevianamide E (1), brevianamide V (2), and ergosterol peroxide (3) on the basis of spectroscopic data including MS, NMR, and UV. To the best of our knowledge, chemical investigation of A. protuberus was conducted for the first time in this study.

  • The determination of the optimum conditions for the conversion of ergosterol to vitamin $D_2$ in shitake mushrooms (Lentinula edodes) was studied using response surface methodology (RSM). The effects of the three main variables ambient temperature ($20-40^{\circ}C$), exposure time (60-180 min), and irradiation intensity ($0.6-1.8W/m^2$) were investigated. According to the RSM ridge analysis, the optimum conditions were as follows: ambient temperature of $34.2^{\circ}C$, exposure time of 175.6 min, and irradiation intensity of $1.41W/m^2$. Under these optimum conditions, the maximum vitamin $D_2$ content of $117.93{\mu}g/g$ in shitake mushrooms was obtained, which agreed fairly well with the predicted value of $122.60{\mu}g/g$.

Screening of Sterol Biosynthesis Inhibitors from Natural Products Using Recombinant Yeast Carrying Human Lanosterol Synthase

  • For the screening of inhibitors of sterol biosynthesis from natural products, a simple and rapid assay method was developed using recombinant yeast carrying human lanosterol synthase, main target of this assay method. Sterol biosynthesis inhibition activity was monitored only by the inhibition of growth of the recombinant yeast. By changing the substrate, this assay method can figure out which step is inhibited in the sterol biosynthesis by the test material. With this assay method total 102 plant samples were screened for their inhibitory activity of sterol biosynthesis. Among plant water extracts screened, 11 plant samples showed inhibitory activity on sterol biosynthesis in ergosterol (-) medium. For selection of the specific inhibitory materials, 11 plant samples were reassayed in ergosterol (+) medium. After all 5 plant samples, Abutilon avicennae Gaertn. (stem), Alnus japonica Steud. (stem), Amaranthus mangostanus L. (aerial part), Philadelphus schrenckii Pupr. (leaf) and Pimpinelia brachycarpa Nakai (aerial part), showed specific inhibitory activity.

Anti-Candida Activity of YH-1715R, a New Triazole Derivative

  • YH-1715R, (2R,3R)-2-(2,4-difluorophenyl)-3-(3-methoxy-1,2,4-isothiazol-3-yl-thio)-1-( 1H-1,2,4-triazol-l-yl)-2-butanol, a new triazole derivative obtained by the structural modification of fluconazole, was found to exhibit potent anti-Candida activity against a wide variety of Candida albicans (C. albicans) (MIC: 0.4-12.5 mg/l). To investigate the mode of action of YH-1715R, its effect on ergosterol biosynthesis in cell-free extracts and whole cells of C. albicans was examined. The inhibitory activity of YH-1715R was approximately ten-fold higher than that of fluconazole. To determine the primary action mechanism of YH-1715R, its inhibitory activity against lanosterol $14\alpha$-demethylase (14$\alpha$-DM), a major target for azole, was measured using gas-liquid chromatography. YH-1715R and fluconazole were found to inhibit 14a-DM with an $IC_{50}$ of 0.015 $\mu$M and 0.01$8\mu$M, respectively, plus the mode of inhibition of YH-1715R and fluconozole was noncompetitive with a $K_i$ of 0.0533$\mu$M and 0.0975$\mu$M.

Effect of Color of Light Emitting Diode on Development of Fruit Body in Hypsizygus marmoreus

  • This study was conducted to identify a suitable color of light for development of the fruit body in Hypsizygus marmoreus. To accomplish this, samples were irradiated with blue (475 nm), green (525 nm), yellow (590 nm), or red (660 nm) light emitting diodes (LEDs) to induce the formation of fruiting bodies after mycelia growth. The diameter and thickness of the pileus and length of stipes in samples subjected to blue LED treatment were similar to those of subjected to fluorescent light (control), and the lengths of the stipes were highest in response to treatment with the red LED and darkness. The commercial yields of plants subjected to blue and green LED treatment were similar to those of the control. In conclusion, cultivation of H. marmoreus coupled with exposure to blue LED is useful for inducing high quality fruit bodies as well as higher levels of ergosterol, DPPH radical scavenging activity, total polyphenol content and reducing power.

The Effects of Glomus etunicatum Innoculation of Robinia pseudoacacia Seedlings on Soil Aggregate Formation in Coal Mine Tailings

  • An investigation was conducted on the effects of arbuscular mycorrhizal fungus, Glomus etunicatum on the formation of soil aggregate and mycorhizal development in the roots of Robinia pseudoacacia seedlings in coal mine tailings and forest soil. G. etunicatum formed mycorrhizas by 35.1 % in coal mine tailings and by 48.9 % in forest soil. Its infection was the typical Arum-type forming inter-cellular hyphae and intra-cellular arbuscules. Ergosterol contents were 3.20 ppm in forest soil and 1.92 ppm in coal mine tailings. The formation of soil aggregate per 50 g pot soil was 19.6 g and 9.5 g in inoculated and noninoculated forest soil and 16.5 g and 11.0 g in inoculated and non-inoculated coal min tailings, respectively. In conclusion, G. etunicatum inoculation increased the formation of soil aggregate both in forest soil and coal mine tailings, but was less effective in the latter.

Monitoring of Ergosterol Biosynthesis Inhibitor (EBI) Pesticide Residues in Commercial Agricultural Products and Risk Assessment (국내 유통 농산물 중 EBI계 농약 모니터링과 위해도 평가)

  • Establishment of simultaneous analysis method and monitoring for individually analyzing residual eight ergosterol biosynthesis inhibitors, EBI (difenoconazole, diniconazole, fenarimol, fenbuconazole, hexaconazole, myclobutanil, nuarimol and paclobutrazol) pesticides in commercial agricultural products, were conducted. The simultaneous analysis method for the pesticides was established using a GC/MS/MS for EBI pesticides. Residual amount of those pesticides were investigated in 989 commercial agricultural products (fifteen kinds of cereal grains, vegetables, beans, nuts, fruits and mushrooms) from seven metropolitan cities and eight provinces. In EBI pesticides analysis, linearity of GC/MS/MS analysis was 0.9974-0.9992, and that of recoveries were 86-135% with relative standard deviations (RSD) <20%. The limit of quantification (LOQ) of the method ranged from 0.5 to 5.0 mg/kg for eight EBI pesticides. According to the monitoring of the EBI pesticides in commercial agricultural products, difenoconazole, fenarimol, hexaconazole showed various residual levels (total frequency of 8/989 detection, 0.8%). Paclobutrazole showed in excess levels of the MRLs (maximum residue limits) for pesticides in one chard sample by the Korea Food Code. As a result of exposure assessment on the detected 8 individual pesticides, all pesticides (difenoconazole, fenarimol, hexaconazole, paclobutrazole) were evaluated as safe level in comparison to toxicologically acceptable daily intake.

Comparison of Growth Characteristics of Tricholoma matsutake Mycelium Among the Types of Air Bubble Bioreactor (공기부양식 생물반응기의 형태별 송이균사의 생장특성 비교)

  • In order to select suitable bioreactor type inhibiting cell stress during submerged culture of Tricholoma matsutake mycelium, the growth characteristics and ergosterol contents were investigated using the external-loop type of air-lift bioreactor (ETAB), balloon type of air bubble bioreactor (BTBB) and column type of air bubble bioreactor (CTBB). Dry weights of the T. matsutake in the BTBB, ETAB and CTBB were 12 g, 11.4 g, and 9.5 g per 1 litter, respectively. BTBB, ETAB and CTBB reached stagnant phases 16, 20, and 24 days after cultivation, respectively, The BTBB was more suitable for liquid culture of T. matsutake mycelium compared to other bioreactors owing to much mycelia product and short culture period. The ergosterol contents produced by the mycelium in the bioreactors were in sequence of BTBB, CTBB, and ETAB at every growth phase. BTBB might affect the mycelium on producing the smallest size of pellets. BTBB and CTBB got the mycelium precipitated and coagulated under operation of bioreactor sparser, whereas ETAB shown no effect of above phenomenon. A renovated bioreactor combined between a balloon shape of BTBB and an external-loop of ETAB was developed to enhance the efficiency of culture technique.
