Analysis of the Reaction Steps in the Bioconversion of D,L-ATC to L-Cysteine

  • The reaction steps involved in the bioconversion of a chemically synthesized precursor, $D,L-2-amino-{\Delta}^2-thiazoline-4-carboxylic$ acid (D,L-ATC), to L-cysteine and the properties of the involved enzymes were investigated. It was found that the conversion consisted of two steps, i. e., D,L-ATC to S-carbamyl-L-cysteine (S-C-L-cysteine) and S-C-L-cysteine to L-cysteine, and the S-C-L-cysteine was an intermediate between them. While the enzymes involved in the reactions were induced by the addition of D,L-ATC as an inducer, S-C-L-cysteine induced only the enzyme involved in the latter step. The conversion of S-C-L-cysteine to L-cysteine could be also carried out in the presence of hydroxylamine and its rate was much faster than that by the corresponding enzyme. On the other hand, L-cysteine (or L-cystine) was decomposed to evolve $H_2S$ by the enzyme considered to be a kind of desulfhydrase. However, hydroxylamine was a perfect inhibitor for this enzyme.

  • Veratryl alcohol which has been reported as an inducer for lignin peroxidase showed different effects on the enzyme biosynthesis in Phanerochaete chrysosporium depending on the addition time. Enzyme expression was optimally induced by adding veratryl alcohol when the carbon source began to be depleted. Hydrogen peroxide, to some extent, stimulated production of lignin peroxidase, but beyond a certain concentration, inactivated lignin peroxidase. Tween 80 induced the formation of small pellets, which were resistant to the deactivation by shear stress. Lignin peroxidase production was increased twice compared with that of the control by adopting all the optimal factors in the culture system.

Large Increase in Leuconostoc citreum KM20 Dextransucrase Activity Achieved by Changing the Strain/Inducer Combination in an E. coli Expression System

  • A recombinant putative dextransucrase (DexT) was produced from Leuconostoc citreum KM20 as a 160 kDa protein, but its productivity was very low (264 U/l). For optimization, we examined enzyme activity in 7 Escherichia coli strains with inducer molecules such as lactose or IPTG. E. coli BL21-CodonPlus(DE3)-RIL exhibited the highest enzyme activity with lactose. Finally, DexT activity was remarkably increased by 12-fold under the optimized culture conditions of a cell density to start induction ($OD_{600}$) of 0.95, a lactose concentration of 7.5 mM, and an induction temperature of $17^{\circ}C$. These results may effectively apply to the heterologous expression of other large DexT genes.

  • 목재부후균으로 부터 락케이스 효소의 생산 및 유도를 위하여 여러가지 유도약품(inducer)을 사용하였다. 이들 가운데 ferulic acid, pentachlorophenol 및 2,5-xylidine이 매우 높은 락케이스 활성을 나타나게 하였으며, 거의 동일한 유도효과를 보여주었다. 이들 약품 이외에도 sinapic acid, syringic acid 및 coffeic acid 등도 높은 락케이스 활성을 주었는데, 산의 형태가 알데히드류보다도 높은 유도효과를 나타냈다. 그리고 실험한 48개 균주 가운데 38개 균주가 락케이스를 생산하였으며, 이 가운데 32균주가 ferulic acid에 의해 강한 효소유도 활성을 보였다. 이러한 결과는 지금까지 락케이스 효소의 검출이 어려웠던 Abortiporus biennis 및 Gleophyllum odoratum에서도 높은 락케이스 효소의 유도를 가능하게 하였다. 아울러 가장 높은 효소활성을 나타낸 균주로서는 Cerrena unicolor 였으며, 그 락케이스 효소활성이 무처리 및 inducer 첨가시 각각 40,000 및 60,000 nkat/l 정도였다.

Production of Laccase by Trametes sp. CJ-105 (Trametes sp. CJ-105에 의한 Laccase 생산)

  • For Trametes sp. CJ-105, a kind of white-rot fungi which was collected from the mountain of Korea and was proven to be effective in decolorizing a wide range of structurally different synthetic dyes, the optimum conditions for mycelial growth and laccase(E.C. 1.10.3.2) production were investigated. Among various carbon sources, glucose showed the highest potential for the mycelial growth and laccase production, the optimum concentration being 2% glucose. For the nitrogen source, asparagine was good for the mycelial growth, while ammonium tartrate for laccase production(optimum concentration: 0.04%). The addition of thiamine and biotin increased both th emycelial growth and laccase production. When 2,5-xylidine was added as an inducer after the first day of culture, the production of alccase was seven-times higher than that in the absence of the inducer. The optimum pH and temperature conditions for laccase production by Trametes sp. CJ-105 were pH 5.0 and $25^{\circ}C$, respectively. In the 5L fermentation, the production of laccase reached a maximum of 340U/ml at the time when the ammonium ion was being rapidly depleted.

  • From Aspergillus nidulans FGSC 168, mutant strains (TCD27, NCN14, and ACN3) were obtained by NTG and 4-NQO treatments. These four mutant strains were unable to grow on medium supplemented with CMC as a sole carbon source and showed lower levels of CMCase activity in comparison with FGSC168. Some characteristics of these four mutant strains in CMCase production were examined. It was appeared that the permeases for CMCase inducer in all the four mutants were not affected. Two mutants, TCD27 and NCN2, might have some defects in regulatory gene for CMCase production. Analyses of enzyme components of CMCase showed that ACN14 might by defective in structural gene for CMCase.

Biotransformation of Protopanaxadiol-Type Ginsenosides in Korean Ginseng Extract into Food-Available Compound K by an Extracellular Enzyme from Aspergillus niger

  • Compound K (C-K) is one of the most pharmaceutically effective ginsenosides, but it is absent in natural ginseng. However, C-K can be obtained through the hydrolysis of protopanaxadiol-type ginsenosides (PPDGs) in natural ginseng. The aim of this study was to obtain the high concentration of food-available C-K using PPDGs in Korean ginseng extract by an extracellular enzyme from Aspergillus niger KACC 46495. A. niger was cultivated in the culture medium containing the inducer carboxymethyl cellulose (CMC) for 6 days. The extracellular enzyme extracted from A. niger was prepared from the culture broth by filtration, ammonium sulfate, and dialysis. The extracellular enzyme was used for C-K production using PPDGs. The glycoside-hydrolyzing pathways for converting PPDGs into C-K by the extracellular enzyme were Rb1 → Rd → F2 → C-K, Rb2 → Rd or compound O → F2 or compound Y → C-K, and Rc → Rd or compound Mc1 → F2 or compound Mc → C-K. The extracellular enzyme from A. niger at 8.0 mg/ml, which was obtained by the induction of CMC during the cultivation, converted 6.0 mg/ml (5.6 mM) PPDGs in Korean ginseng extract into 2.8 mg/ml (4.5 mM) food-available C-K in 9 h, with a productivity of 313 mg/l/h and a molar conversion of 80%. To the best of our knowledge, the productivity and concentration of C-K of the extracellular enzyme are the highest among those by crude enzymes from wild-type microorganisms.

Screening for Korean Vegetables with Anticarcinogenic Enzyme Inducing Activity Using Cell Culture System

  • There is extensive evidence suggesting the protective role of fruits and vegetables against chemically induced carcinogenesis. We have tested the ability of a representative range of Korean vegetables to act as blocking agents against neoplastic initiation by determining the induction level of quinone reductase , an anticarcinogenci marker enzyme, in hepalclc 7 cells exposed to vegetable extracts. Among thirty vegetables tested, Arcitum lappa(Burdock), Brassica juncea (Mustard leaf), Pteridium aguilinum (Bracken) and Chrysanthemum cornoratium(Crown daisy) caused a significant induction of quinone teductase activity with a limited increase in arylhdrocarbon hydroxylase activity. Combination of crown daisy with burdock had synergistic effect on quinone reductase induction. Quinone reductase-inducing activity was found mostly in hesane and ehtylactate fractions of MeOH extract of crown daisy while it ws not quinone reductase activity in liver, kideny, lung, and small intestine, confirming the presence of potent QR inducer (s) in crown daisy. These sata suggest that some vegetables including crown daisy induced QR merits further investigation as a potential cancer preventive agent in human.

Screening of Wood-Rot Fungi Based on RBBR Decolorization and Its Laccase Activity (RBBR 탈색능을 이용한 목재부후균의 선발 및 이들 균의 Laccase 효소활성)

  • This study was to screen white-rot fungi possesing strong lignin degrading enzymes, glucose-1 oxidase (GOD), laccase (LAC) and Mn-peroxidase (MnP), based on their decolorization activity of Remazol Brilliant Blue R (RBBR). In the midst of 20 tested fungi, 9 isolates were shown 4 kinds of activities such as RBBR decolorization, GOD, LAC and MnP. Relatively high active strains were identified as Phlebia radiata, Trametes versicolor, Abortiporus biennis, Gleophyllum odoratum and Cerrena unicolor. In particular, T. versicolor, G. odoratum, and C. unicolor, which have high activities of LAC, were used to confirm the optimal temperature and pH and to evaluate the effect of inducer, 2,5-xylidine on their LAC activity. The optimum temperatures for mycelial growth were $28^{\circ}C$ for T. versicolor and G. odoratum, and $25^{\circ}C$ for C. unicolor. The optimum pH for mycelial growth was 5.5. Three strains showed the increase of LAC enzyme activity by the addition of 2,5-xylidine. T. versicolor had the highest LAC activity of $22,700nkat/{\ell}$, corresponding to 11.3 times, G. odoratum $15,400nkat/{\ell}$, 9 times and C. unicolor $17,330nkat/{\ell}$, 5.5 times higher than those of the control.

Studies on Microbial Penicillin Amidase ( I ) Optimization of the Enzyme Production from Escherichia coli (미생물 페니실린 아미다제에 관한 연구 (I) E. coli로부터 효소생산 조건의 최적화)

  • To maximize the production of penicillin amidase from Estherichia coli (ATCC 9637), the media composition and several factors affecting the engyme production during fermentation were studied. The optimal media composition was found to be; 3.5% tryptone, 1.5% monosodium glutamate and 0.5% yeast extract. The addition of 0.15% phenylacetic acid as an enzyme inducer at the initial stage of cultivation increased the engyme productivity about 5 fold. It was found that the engyme activity reached maximum within 16hr of cultivation. The maximum production of the enzyme obtained was about 102.5 units/l broth under the optimized condition. The enzyme production was markedly increased by the optimization as compared with those previously reported.
