In Vivo/In Vitro Properties of Novel Antioxidant Peptide from Pinctada fucata

  • Due to the potential of antioxidants to scavenge free radicals in human body, it is important to be able to prepare antioxidant peptides that meet the industrial requirements for cosmetics and food. Here, we determined in vivo/in vitro activities of antioxidant peptide from P. fucata (PFAOP) prepared by bio-fermentation method. The antioxidant property test results showed the DPPH, hydroxyl, superoxide radical-scavenging, and cellular antioxidant activity. EC50 values of PFAOPs were 0.018 ± 0.005, 0.126 ± 0.008, 0.168 ± 0.005, and 0.105 ± 0.005 mg/ml, respectively, exhibiting higher antioxidant activities than glutathione (p < 0.05). Moreover, anti-proliferation and cytotoxicity activity results illustrated PFAOP has a potent anti-proliferative activity against HepG2, Caco-2, and MCF-7 carcinoma cells with no cytotoxicity. Moreover, the protocols we developed in this work demonstrated several excellent advantages in PFAOP preparation compared to enzymatic hydrolysis or chemical synthesis methods and provide a theoretical foundation for higher-value application of marine-derived functional peptides.

Three Protein Kinases from the Etiolated Oat Seedlings Phosphorylate Oat Phytochrome A In Vitro

  • Phosphorylation of phytochrome may play important functional roles to control plant photomorphogenesis. Many attempts have failed to identify the protein kinase that phosphorylates phytochrome in vivo. It has been reported that a polycation-stimulated protein kinase activity was associated with the purified phytochrome. However, it is not known if the kinase activity is an intrinsic property of phytochrome or whether it comes from a contaminant of the purified phytochrome. In the present study, three protein kinases that phosphorylate phytochrome have been identified from etiolated oat seedlings. A polycationstimulated protein kinase that had very similar enzymatic properties with that associated with the purified phytochrome was identified in the cytosolic extract. It phosphorylated several contaminant proteins in the kinase preparation as well as phytochrome and had a broad substrate specificity. A CK II-type protein kinase phosphorylated phytochrome and the exogenously added casein. It is likely that this kinase may not be a feasible candidate for the kinase phosphorylating phytochrome in vivo since the content of the kinase seemed to well exceed the content of phytochrome in the etiolated oat seedlings. Another protein kinase that had unique enzymatic properties phosphorylated phytochrome very specifically and seemed to be present in a small quantity in the etiohlted seedlings. It is expected that one of three kinases may be responsible for the phytochrome phosphorylation in vivo.

한국산 겨자중 Myrosinase의 정제 및 효소학적 특성 (Purification and Enzymatic Properties of Myrosinase in Korean Mustard Seed(Brassica juncea))

  • 한국산 겨자에서 myrosinase를 분리 및 정제하고, 이들의 효소학적 특성을 조사한 결과는 다음과 같다. 겨자 myrosinase를 DEAE-cellulose, Concanavalin A-Sepharose 및 FPLC Soporose 6 칼럼을 이용하여 분리 및 정제하였을 때 적어도 3개의 이성효소가 존재하는 것으로 나타났으며, Myrosinase II-2의 최종 비활성도는 57.lunits/mg, 정제도는 약 248배였다. SDS-PAGE상에서 myrosinase(II-2)의 단일밴드를 확인한 결과, 그 분자량은 약 67KD로 추정 되었다. 최적 pH는 phosphate 및 Tris-HCI 완충액에서 7.0이 가장 활성이 높았고, 그 효소는 pH 7.0에서 안정하였으며, 최적 활성을 나타내는 온도는 $37^{\circ}C$ 부근이었고, $40^{\circ}C$ 이상에서는 비교적 불안정하게 나타났다. Ascorbic acid의 영향은 1mM에서 가장 안정하였으며, 그 이상의 농도에서는 변화가 없었다. 망간과 마그네슘 및 나트륨은 효소활성을 촉진시키는 것으로 나타났으며 구리, 수은 및 철 이온은 약간 저해하였다. Ascorbic acid analogue 중 dehydroascorbic acid는 효소 활성을 저해하였으며, 나머지 것들은 거의 영향을 미치지 않았다. 2-Mer-captoethanol과 dithiothreitol과 같은 환원제는 효소활성을 억제하였으나 이들과 ascorbic acid를 함께 첨가 하였을 때는 활성이 다소 증가하였다.

Oligomeric Structures Determine the Biochemical Characteristics of Human Nucleoside Diphosphate Kinases

  • Major human Nucleoside diphosphate kinases (NDPKs) exist as hetero-oligomers, consisting of NDPK-A and NDPK-B, rather than homo-oligomer. To investigate their biological function depending on the oligomeric structure in vivo, we characterized the biochemical properties of cellular NDPK. Cellular NDPKs, which are made up of a unique combination of isoforms, were purified from human erythrocyte and placenta. We found that cellular NDPK and recombinant isoforms NDPKs have their own distinct biochemical properties in autophosphorylation, stability toward heat or urea, and DNA binding. Cellular NDPK was found to have unique characteristics rather than the expected additive properties of recombinant isoforms. The mutations in the dimeric interface of NDPK-B (R34G, N69H or K135L) caused defective DNA binding and simultaneously reduced the enzymatic stability These results suggest that the oligomeric interaction could play a major role in the stability of catalytic domain and might be related to the regulation of various cellular functions of NDPK.

Characterization of Polysaccharides Obtained from Purslane (Portulaca olerace L.) Using Different Solvents and Enzymes

  • Physiochemical properties, such as yield and molecular weight distribution of polysaccharide fractions, of polysaccharides in the enzymatic hydrolysates of purslane were investigated and characterized. A higher amount of micro nutrients, such as potassium (9,413 mg/100 g), phosphorus acid (539 mg/100 g), leucine, alanine, lysine, valine, glycine, and isoleucine, was present in whole purslane. The yield of water soluble polysaccharides (WSP) was 0.29, 7.01, and 7.94% when extracted using room temperature water (RTW), hot-water (HW), and hot temperature/high pressure-water (HTPW), respectively, indicating that HW or HTPW extraction may be effective to obtain WSP from purslane. The average ratio of L-arabinose:D-galactose in the WSP was 37:49, 34:37, and 27:29, when extracted using RTW, HW, and HTPW, respectively. These results indicate that water was a suitable extraction solvent for preparation of the arabinogalactan component of whole purslane. A higher yield and total carbohydrate content was obtained by using Viscozyme L instead of Pectinex 5XL during extraction of the WSP, which indicates that enzymatic treatment of purslane may be an effective method to control the Mw of polysaccharides. Finally, it was confirmed that Viscozyme L is a suitable enzyme for the hydrolysis and separation of polysaccharides obtained from purslane.

쌀코지가 도루묵(Arctoscopus japonicus) 식해의 발효특성에 미치는 영향 (Effects of Rice Koji on the Fermentation Characteristics of the Sik-hae from Sailfin Sandfish Arctoscopus japonicus)

  • This study was conducted with a view toward enhancing the industrial utilization of a male sailfin sandfish Arctoscopus japonicus based on its application in the preparation of Sik-hae, a Korean traditional fermented fishery product prepared using hard-boiled rice and enzymatic malt sprouts. As an alternative to rice and malt sprouts, we evaluated the utility of rice Koji for Sik-hae fermentation by investigating the physicochemical changes that occur during fermentation. The sailfin sand fish Sik-hae fermented with 10% or 20% rice Koji was found to show similar fermentation characteristics to the control fermentation with 20% hard-boiled rice and 4% malt sprouts, with respect to acid production, texture profile, and the softening of bones during the entire fermentation period. In particular, compared with the control, the addition of 10% rice Koji reduced the dehydration of Sik-hae and increased the content of free amino acids, which contributed to preventing a marked reduction in pH during fermentation. Accordingly, 10% rice Koji is proposed as a viable alternative enzymatic agent for the preparation of Sik-hae, which can contribute enhancing the desired properties of this traditional food product.

  • 어류 중 수산가공공장에서 대량으로 사용되고 있는 민태(hoki, Johnius belengeri)의 부산물인 frame으로부터 단백질을 효율적으로 회수하고자, 참치 내장 유래 단백질 분해효소를 이용하여 가수분해시키고, 여기서 얻어진 가수분해물의 기능성을 개선할 목적으로 한외여과막을 이용하여 가수분해물에 함유되어 있는 펩티드를 분자량에 따라 분획한 후 각각의 가분해물에 대하여 기능성을 검토하였다. 참치 유문수 유래 단백질 분해효소 (tuna pyloric caeca crude enzyme: TPCCE)를 이용한 경우, 민태 frame 단백질을 가수분해하기 위한 최적 조건은 pH 10.0, 반응온도 $50^{\circ}C$ 및 기질농도 0.4%, 기질 대 효소비 100 : 1(w/w), 반응시간 12시간이었다. 최적 가수분해 조건 하에서 민태 frame 단백질을 TPCCE로 가수분해하여 생성된 가수분해물의 수율은 약 77%(건조중량 기준)였다. 가수분해물을 4종류의 한외여과막(MWCO 30, 10, 5 및 1 K)을 이용하여 분자량의 크기에 따라 4종류로 분획하여 기능성을 검토한 결과, 용해도는 분자량이 낮은 1 K 가수분해물이, 유화성과 포말성은 분자량인 높은 30 K와 10 K가수분해물이 우수하였다.

(${\beta}$-Cyclodextrin 유도체의 합성 (Syntheses of ${\beta}$-Cyclodextrin derivatives)

  • Cyclodextrin are obtained from starch by enzymatic degradation. The three best characterize forms are ${\alpha}$, ${\beta}$, ${\gamma}$ cyclodextrin consisting of 6, 7, and 8 D-glucose units, respectively. Each of the glucose units are in the rigid C1 chair conformation and are linked by ${\alpha}$ 1,4 bonds. This geometry gives the cyclodextrin the shape of a hollow truncated cone with the wider side formed by the secondary 2- and 3-hydroxy groups and the narrower side by the primary 6-hydroxy group. The most characteristics property of the cyclodextrin is their ability to form inclusion complexes with a wide range of guest moleculars. We syntheses per-6-substituted ${\beta}$-cyclodextrin derivatives and investigate structures, spectrospcopic properties. The substituted materials are piperidine, piperazin, morphorine. The synthetic compound showed a good solubility than natural ${\beta}$ cyclodextrin in organic solvents such as methylene chloride, methanol, ethanol, etc.

Uniqueness of Microbial Cutinases in Hydrolysis of p-Nitrophenyl Esters

  • Using fungal (Fusarium solani f. pisi) and bacterial (Pseudomonas mendocina) cutinases, the initial hydrolysis rate of p-nitrophenyl esters was systematically estimated for a wide range of enzyme and substrate concentrations using a 96-well microplate reader. Both cutinases exhibited a high substrate specificity; i.e. a high hydrolytic activity on p-nitrophenyl butyrate (PNB), yet extremely low activity on p-nitrophenyl palmitate (PNP). When compared to the hydrolysis of PNB and PNP by other hydrolases [lipases and esterases derived from different microbial sources, such as bacteria (Pseudomonas cepacia, Psedomonas furescens, Baciilus stearothermophilus), molds (Aspeillus niger, mucor miehei), and yeasts (Candida rugosa, Candida cylindracea)], the above substrate specificity would seem to be a unique characteristic of cutinases. Secondly, the hydrolytic activity of the cutinases on PNB appeared much faster than that of the other hydrolytic enzymes mentioned above. Furthermore, the current study proved that even when the cutinases were mixed with large amounts of other hydrolases (lipases or esterases), the Initial hydrolysis rate of PNB was determined only by the cutinase concentration for each PNB concentration. This property of cutinase activity would seem to result from a higher accessibility to the substrate PNB, compared with the other hydrolytic enzymes. Accordingly, these distinct properties of cutinases may be very useful in the rapid and easy isolation of various natural cutinases with different microbial sources, each of which may provide a novel industrial application with a specific enzymatic function.

Galactomannan 이용에 관한 연구 : Affinity Chromatography에 의한 지렁이 유래 /alpha-Galactosidase의 정제 및 응용법 (Purification and Application of Earthworm /alpha-Galactosidase by Affinity Chromatography)

  • An $\alpha$-D-galactosidase ($\alpha$-D-galactoside galactohydrolase, EC 3. 2. 1. 22) from earthworm was purified by affinity chromatography using N-$\varepsilon$-aminocaproyl-$\alpha$-D-galactopyranosylamine coupled to sepharose and its properties were examined. The specific activity of the purified enzyme, tested with p-nitrophenyl-$\alpha$-D-galactopyranoside as substrate, was 314 units/mg protein, representing an 122-fold purification of the original crude extract. The final preparation obtained from by Sephadex G-25 chromatography showed a single band on SDS-polyacrylamide gel electrophoresis. The molecular weight was determined to be 48,000 by SDS-polyacrylamide gel electrophoresis. The purified galactosidase was showed maximum activity at pH 4.5 and 4$0^{\circ}C$, and was stable in the pH and temperature ranges from 4.0 to 5.5 and 30 to 5$0^{\circ}C$, respectively. The enzyme activity was inhibited by Zn2+, Hg2+ and Co2+. When the purified $\alpha$-galactosidase treated to guar gum for 6 hour, gel-promoting property was increased. It was clear that enzymatic elimination of galactose from guar gum by purified $\alpha$-galactosidase would lead to a significant increase in gelation ability.
