쌀을 주식으로 하는 우리나라의 여건상 연간 추정치로 싸라기 약 12만톤, 미강 약 49만톤, 왕겨 약 79만톤의 벼 도정 부산물이 발생하고 있다. 본 연구에서는 벼 도정 부산물 중 비식량 자원인 왕겨를 대상으로 고효율 효소 당화를 위한 바이오매스 전처리 방법을 탐색하였다. 왕겨 원시료의 초기 조성은 셀룰로스 34.5%, 헤미셀룰로스 20.5%, 리그닌 25.3%, 회분 14.6%로 나타났는데, 억새 등 초본계 바이오매스와 비교하여 특이하게 높은 성분은 회분으로 이는 벼에 대한 규산질 비료의 시용에서 기인한 것이다. 바이오매스 전처리에 많이 사용되는 암모니아, 희황산 용매와 규산염에 침식성을 가지는 가성소다 용매를 이용하여 각 용매별 단독 및 알칼리-산 복합 처리 하였을 때 효소 가수분해 효율, 고상시료 성분변화 등을 상호 비교하였다. 예비실험을 통하여 암모니아 처리조건은 15%(w/w) $150^{\circ}C$ 20분, 가성소다 처리조건은 1.5%(w/w) $150^{\circ}C$ 20분, 희황산 처리조건은 1.0%(w/w) $150^{\circ}C$ 10분으로 설정하였다. 암모니아 단독, 희황산 단독, 암모니아-희황산 복합 처리 시료의 효소 가수분해 효율은 각각 37.8%, 39.1%, 42.8%로 약 40%선에서 큰 차이가 없었다. 반면 가성소다 단독, 가성소다-희황산 복합 처리시료의 효소 가수분해 효율은 각각 62.7%, 82.8%로 나타나 앞선 3가지 처리방법 대비 50%, 100%에 가까운 효소 가수분해 효율 향상을 보였다. 이 때 전처리 고상시료의 성분 변화를 살펴보면 회분 함량에서 큰 차이를 보였는데 암모니아 단독, 가성소다 단독, 희황산 단독, 암모니아-희황산 복합, 가성소다-희황산 복합 처리에서 각각 47.8%, 77.1%, 43.5%, 55.8%, 94.7%의 회분 성분 기각률(rejection rate)을 나타냈다. 이는 왕겨 효소 가수분해 효율의 최대 저해요인이 회분임을 추정할 수 있다. 왕겨 전처리 알칼리 용매는 암모니아보다 가성소다가 더 효과적이었고 희황산 복합 처리시 그 효과가 크게 상승하였다. 따라서 규산염(회분) 함량이 높은 바이오매스는 고온 고압 조건에서 가성소다 용액으로 처리한 후 그 고상분을 희황산 용액으로 복합 처리하는 시스템이 효소 당화 증진에 매우 유리함을 확인하였다.

Enzyme-aided bleaching of softwood and hardwood kraft pulps by a xylanase preparation from an alkalophilic fungus Cephalospotium sp. RYM-202 was studied. Maximal solubilization of Pulp xylan was obtained at 5$0^{\circ}C$ in both kraft pulps. The optimum pH of the enzyme for the hydrolysis of pulp xylan was 8.0 and more than 90% of the maximal activity was detected at 9.0. The positive effects of xylanase pretreatment on bleachability of softwood and hardwood kraft pulps were observed. The kappa number of softwood and hardwood kraft pulps was decreased by 3.7 and 2.0 units, respectively. The pulp fibre integrity was not significantly affected by xylanase pretreatment when the physical properties of handsheets made from xylanase-treated pulps were compared with those of handsheets from untreated pulps. These results indicate that the alkaline xylanase of Cephalospotium sp. RYM-202 is well suitable for application in enzymatic prebleaching of softwood and hardwood kraft pulps under the alkaline conditions.

Despite the easing of social distancing, demand for non-face-to-face services continues to rise. Recently, the EU is pursuing a comprehensive plastic use reduction by expanding the scope of plastic use regulations for packaging plastics according to the New Cyclical Economy Action Plan(NCEAP). In response to this trend, the packaging industry is moving away from conventional non-degradable/petroleum-based plastics and conducting research on packaging materials using biodegradable plastics such as PLA(Poly Lactic Acid), PBAT(Poly Butylene Adipate-co-butylene Terephthalate). On the other hand, ginkgo leaves occur in large quantities in Korea and act as a cause of slip accidents and flooding. In this study, a method to utilize ginkgo biloba leaf as a new alternative biomass resource was proposed by producing lactic acid through pretreatment, enzymatic saccharification, and fermentation processes. For the efficiency of lactic acid production, a comparative analysis of lignin content from before and after browning was performed. In addition, the degree of glucan extraction was evaluated by applying a pretreatment method using three catalysts: hot water, sulfuric acid, and sodium hydroxide. It is difficult to expect high production of lactic acid with single process. Therefore, an integrated process operation using both the pretreated hydrolyzate and the residual solid enzymatic saccharification solution must necessarily be applied.

A two-step process was investigated for pretreatment and fractionation of rice straw. The two-step fractionation process involves first, soaking rice straw in aqueous ammonia (SAA) in a batch reactor to recover lignin-rich hydrolysate. This is followed by a second-step treatment in a fixed-bed flow-through column reactor to recover xylo-oligomer-rich hydrolysate. The remaining glucan-rich solid cake is then subjected to an enzymatic process. In the first variant, SAA treatment in the first step dissolves lignin at moderate temperature (60 and $80^{\circ}C$), while in the second step, hot-water treatment is used for xylan removal at higher temperatures ($150{\sim}210^{\circ}C$). Under optimal conditions ($190^{\circ}C$ reaction temperature, 30 min reaction time, 5.0 ml/min flow rate, and 2.3 MPa reaction pressure), the SAA-hot-water fractionation removed 79.2% of the lignin and 63.4% of the xylan. In the second variant, SAA was followed by treatment with dilute sulfuric acid. With this process, optimal treatment conditions for effective fractionation of xylo-oligomer were found to be $80^{\circ}C$, 12 h reaction time, solid-to-liquid ratio of 1:12 in the first step; and 5.0 ml $H_2SO_4/min$, $170^{\circ}C$, and 2.3 MPa in the second step. After this two-step fractionation process, 85.4% lignin removal and 78.9% xylan removal (26.8% xylan recovery) were achieved. Use of the optimized second variant of the two-step fractionation process (SAA and $H_2SO_4$) resulted in enhanced enzymatic digestibility of the treated solid (99% glucan digestibility) with 15 FPU (filter paper unit) of CTec2 (cellulase)/g-glucan of enzyme loading, which was higher than 92% in the two-step fractionation process (SAA and hot-water).

Alakline peroxide pretreatment for the delignification of poplar wood was performed. sinceit is a simple and efficent method for enhancing the enzymatic digestibility of wood residues. Approximately one-half of their lignin and most of the hemicellulose present in poplar wood were removed when the wood sawdust was reacted at 25$^{\circ}C$ for 100 hrs in an alkaline solution (pH 11.5) of 1% peroxide. The rate of decomposition as well as the saccharification efficiency were enhanced up to 350% and 260% respectively in comparision with those of the controll. This enhancement is comparable with that pretreated with 1% sodium hydroxide and 20% peracetic acid successively. The advantages of alkaline peroxide as delignifying agents against other chemicals were also discussed.

Exposure of cells to ultraviolet B (UVB) radiation can induce production of free radicals and reactive oxygen species (ROS), which damage cellular components. In addition, these agents can stimulate the expression of matrix metalloproteinase (MMP) and decrease collagen synthesis in human skin cells. In this study, we examined the anti-photoaging effects of extracts of Tetraselmis suecica (W-TS). W-TS showed the strongest scavenging activity against 2,2-difenyl-1-picrylhydrazyl (DPPH) and peroxyl radicals, followed by superoxide anions from the xanthine/xanthine oxidase system. We observed that the levels of both intracellular ROS and lipid peroxidation significantly increased in UVB-irradiated human skin fibroblast cells. Furthermore, the activities of enzymatic antioxidants (e.g., superoxide dismutase) and the levels of non-enzymatic antioxidants (e.g., glutathione) significantly decreased in cells. However, W-TS pretreatment, at the maximum tested concentration, significantly decreased intracellular ROS and malondialdehyde (MDA) levels, and increased superoxide dismutase and glutathione levels in the cells. At this same concentration, W-TS did not show cytotoxicity. Type 1 procollagen and MMP-1 released were quantified using RT-PCR techniques. The results showed that W-TS protected type 1 procollagen against UVB-induced depletion in fibroblast cells in a dose-dependent manner via inhibition of UVB-induced MMP-1. Taken together, the results of the study suggest that W-TS effectively inhibits UVB-induced photoaging in skin fibroblasts by its strong anti-oxidant ability.
This study evaluated the microbial conversion of coconut oil waste, a major agro-residue in tropical countries, into single cell oil (SCO) feedstock for biodiesel production. Copra cake was used as a low-cost renewable substrate without any prior chemical or enzymatic pretreatment for submerged growth of an oleaginous tropical mangrove fungus, Aspergillus terreus IBB M1. The SCO extracted from fermented biomass was converted into fatty acid methyl esters (FAMEs) by transesterification and evaluated on the basis of fatty acid profiles and key fuel properties for biodiesel. The fungus produced a biomass (8.2 g/l) yielding 257 mg/g copra cake SCO with ~98% FAMEs. The FAMEs were mainly composed of saturated methyl esters (61.2%) of medium-chain fatty acids (C12-C18) with methyl oleate (C18:1; 16.57%) and methyl linoleate (C18:2; 19.97%) making up the unsaturated content. A higher content of both saturated FAMEs and methyl oleate along with the absence of polyunsaturated FAMEs with ≥4 double bonds is expected to impart good fuel quality. This was evident from the predicted and experimentally determined key fuel properties of FAMEs (density, kinematic viscosity, iodine value, acid number, cetane number), which were in accordance with the international (ASTM D6751, EN 14214) and national (IS 15607) biodiesel standards, suggesting their suitability as a biodiesel fuel. The low cost, renewable nature, and easy availability of copra cake, its conversion into SCO without any thermochemical pretreatment, and pelleted fungal growth facilitating easier downstream processing by simple filtration make this process cost effective and environmentally favorable.

This work was conducted to evaluate the effect of dilute sodium hydroxide (NaOH) on barley straw at boiling temperature and fractionation of its biomass components into lignin, hemicellulose, and reducing sugars. To this end, various concentrations of NaOH (0.5% to 2%) were applied for pretreatment of barley straw at $105^{\circ}C$ for 10 min. Scanning electron microscopy (SEM), atomic force microscopy (AFM), and Fourier transform infrared (FTIR) spectroscopy studies revealed that 2% NaOH-pretreated barley straw exposed cellulose fibers on which surface granules were abolished due to comprehensive removal of lignin and hemicellulose. The X-ray diffractometer (XRD) result showed that the crystalline index was increased with increased concentration of NaOH and found a maximum 71.5% for 2% NaOH-pretreated sample. The maximum removal of lignin and hemicellulose was 84.8% and 79.5% from 2% NaOH-pretreated liquor, respectively. Reducing sugar yield was 86.5% from 2% NaOH-pretreated sample using an enzyme dose containing 20 FPU of cellulase, 40 IU of ${\beta}$-glucosidase, and 4 FXU of xylanase/g substrate. The results of this study suggest that it is possible to produce the bioethanol precursor from barley straw using 2% NaOH at boiling temperature.
Lignocellulose ($2^{nd}$ generation) is difficult to hydrolyze due to the presence of lignin and the technology developed for cellulose fermentation to ethanol is not yet economically viable. However, recent advances in the extremely new field of biotechnology for the ethanol production are making it possible to use of agriculture residuals and nonedible crops biomass, e.q., rice straw and miscanthus sinensis, because of their several superior aspects as agriculture residual and nonedible crops biomass; low lignin, high contents of carbohydrates. In this article, as the basic study of AP(Ammonia Percolation), the properties and the optium conditions of process were established, and then the overall efficiency of AP was investigated. The important independent variables for AP process were selected as ammonia concentration, reaction temperature, and reaction time. The percolation condition for maximizing the content of cellulose, the enzymatic digestibility, and the lignin removal was optimized using RSM(Response Surface Methodology). The determined optimum condition is ammonia concentration; 11.27%, reaction temperature; $157.75^{\circ}C$, and reaction time; 10.01 min. The satisfying results were obtained under this optimized condition, that is, the results are as follows: cellulose content(relative); 39.98%, lignin content(relative); 8.01%, and enzymatic digestibility; 85.89%.
The purpose of this study was to investigated the effects of Laminaria japonica fucoidan extract (LJFE) through the enzymatic regulation against the hepatotoxicity-inducing carbon tetrachloride $(CCl_4)$ in LJFE and $CCl_4-treated$ rats. LJFE of 100mg/kg concentration was intraperitoneally administered into rats at dose of 1.5m11kg for 14 days. On the day 15, 3.3ml/kg of $CCl_4$ dissolved in olive oil (1:1) was injected 12 hours before anesthetization. We examined the levels of glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT) in serum of rats, superoxide dismutase(SOD) in mitochondrial fraction, and catalase(CAT), glutathione peroxidase(GPx) in liver homogenate. $CCl_4-treatment$ markedly increased the levels of GOT and GPT, and significantly decreased those of SOD, CAT and GPx. But LIFE pretreatment decreased the levels of GOT and GPT, by 40% and 64%, respectively and increased those of SOD, CAT and GPx, by 114%, 36.1% and 55.9%, respectively These results showed the LIFE had the enzymatic regulatory effects against the hepatotoxicity-inducing $CCl_4$ in the preventive way.
