Development of Quantitative Extraction Method of Amygdalin without Enzymatic Hydrolysis from Tonin(Persicae Semen) by High Performance Liquid Chromatography

  • Tonin(Persicae Semen) is the herb medicine that contains amygdalin as a major ingredient. Amygdalin in water is decomposed into benzaldehyde, HCN, and glucose by emulsin, a hydrolysis enzyme in tonin. A useful and practical method for the optimum extraction condition of amygdalin without enzymatic hydrolysis is required. The extraction yield of amygdalin of natural formula to nin was 0.1 % from crude powders, 1.4% from small pieces, 3.5% from half pieces and 2.4% from whole pieces. The extraction yield of amygdalin of outer shell-eliminated to nin was 0.3% from crude powders, 1.4% from small pieces, and 3.5% from half pieces and whole pieces respectively. The extraction yield of amygdalin was most high when using the size larger than half.

  • This study was conducted to investigate the physicochemical properties of citrus peel extracts with different hot water extraction and enzymatic hydrolysis conditions. Enzymatic hydrolysis was also employed using Viscozyme L and results were compared with that of optimized hot water extract. Hot water extraction was performed under different parameters; the sample to solvent ratio(1:20, 1:15, 1:10), extraction time(2, 4 hrs), extraction temperature(85, $95^{\circ}C$) and enzymatic hydrolysis(0, 1%) and the subsequent extracts were used for determining their physicochemical properties, such as total yield, total phenolics, total flavonoids, and electron donating ability (EDA). With the increase in the sample to solvent ratio and extraction time, total yield, total phenolics, total flavonoids and EDA increased. But extraction temperature did not significantly affect the hot water extract. As hot water extract was hydrolyzed by the enzyme, total yield and active ingredients increased rapidly. In the result of total yield, total phenolics, total flavonoids and EDA, the activity of enzyme-treated extract was higher than those of enzyme-untreated extract. Based upon the overall hot water extraction efficiency, it was found that 20 times volume or 120 min at a time at $95^{\circ}C$ after enzyme treatment was optimal.

  • Hizikia fusiformis hydroysates by five carbohydrases (Viscozyme, Celluclast, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase) were investigated for their extraction efficacy (yield and total total polyphenolic content) and antioxidative activity (DPPH radical and hydrogen peroxide scavenging activity). Termamyl and Ultraflo of the carbohydrases and Flavourzyme and Alcalase of proteases were selected by their high eficacy of extraction and antioxidative activity. Selected enzymes were used to investigate the optimum enzymatic reaction time and dosage (enzyme/substrate ratio) suitable for hydorolysis. Optimum reaction time for the enzymatic hydrolysis was 3 days and optimum dosage of hydrolysis was observed as 5%. Simultaneously, Ultraflo of the two carbohydrases and Alcalse of the two proteases were selected as the most effective enzymes. Combination of Ultraflo and Alcalase under optimum hydrolysis conditions could intensify the extraction efficacy of antioxidative materials form H. fusiformis. The hydrolysate obtained by combining the enzymes was separated into four different molecular weight fractions (<1kD, 1-10 kD, 10-30 kD and >30 kD) and recorded the polyphenolic content distribution and respective antioxidative ability. The fraction <1kD was identified as less effective and those fractions > 1kD indicated comparatively higher antioxidative activities related to their polyphenolic content.

  • A major problem in the enzymatic hydrolysis of lignocellulosic substrates is the very strong bonding of cellulase to lignin and even cellulose in the hydrolysis residues. This phenomenon inhibits recycle of the cellulase which is a major expense of the enzymatic hydrolysis process. In this paper, autohydrolyzed wood was delignified by two-stage with a 0.3% Na OH extraction and oxygen-alkali bleaching and was subjected to enzymatic hydrolysis with cellulase. Also, an improved almost quantitative recycle process of cellulase enzyme was discussed. In enzyme recovery by affinity method. the first recycling showed relatively high hydrolysis rate of 97.4%. Even at the third recycle. hydrolysis rate was 86.7 percents. In the case of cellulase recovery by ultrafiltration method, first 2 recycling treatments resulted very high hydrolysis rate(97.0-97.7%). Even the third recycling showed about 94.2%. Authoydrolysis of oak wood followed by 2-stage delignification with alkali and oxygen-alkali produced a substrate for enzymatic hydrolysis that allowed almost quantitative recycle of cellulase.

Evaluation of Secondary Acid and Enzymatic Hydrolysis of Hemicellulose in Hot Water Pre-Pulping Extract of Mixed Hardwoods

  • Pre-pulping extracts were found to contain a dilute amount of xylo-oligosaccharides and acetic acid as the major components, and many minor components including other organic acids, lignin-derived phenolics, and sugar degradation products. Once separated from the pulp, a secondary hydrolysis step was required to hydrolyze oligomeric hemicellulose sugars into monomeric sugars before fermentation. The following study detailed the extent of hemicellulose recovery by pre-pulping using hot water extraction and characterized the hydrolysis of the extract with respect to comparing acid and enzymatic hydrolysis. The secondaryhydrolysis of hot water extracts made at an H-Factor of 800 was tested for a variety of acid and enzyme loading levels using the sulfuric acid and xylanases. The maximum fermentable sugar yield from acid and enzyme hydrolysis of the extract was 18.7 g/${\ell}$ and 17.7 g/${\ell}$ representing 84.6% and 80.1% of the maximum possible yield, respectively.

  • Steam-exploded woods were delignified by two-stage with a 0.3% NaOH extraction and oxygen-alkali bleaching and were subjected to the enzymatic hydrolysis with cellulase enzyme. Also, an improved almost quantitative recycle process of cellulase enzyme was discussed. In enzyme recovery by affinity method, The first recycling showed relatively high hydrolysis rate of 96.4%. Even at the third recycle, hydrolysis rate was 87.0 percents. In the case of cellulase recovery by ultrafiltration method, first 2 recycling treatments resulted in very high hydrolysis rates, 96.8% and 95.0%, respectively. Even the third recycling showed about 93.6%. Steam-explosion treatment of oak wood followed by 2-stage delignification with alkali and oxygen-alkali produced a excellant substrate for the enzymatic hydrolysis that allowed almost quantitative recycle of cellulase.

Optimization of Enzymatic Pretreatment for the Production of Fermented Ginseng using Leaves, Stems and Roots of Ginseng

  • This study sought to optimize the extraction and enzymatic treatment conditions of Panax ginseng leaves, stems, and roots for the production of fermented ginseng. The optimization enhanced the extraction of total saccharide, a nutrient and growth-activating factor for Lactobacillus bacteria. The hydrolysis of ginseng leaves, stems, and roots was tested with eight enzymes (Pentopan, Promozyme, Celluclast, Ultraflo, Pectinex, Ceremix, Viscozyme, and Tunicase). The enzymatic hydrolysis conditions were statistically optimized by the experimental design. Optimal particle size of ginseng raw material was <0.15 mm, and optimal hydrolysis occurred at a pH of 5.0-5.5, a reaction temperature of 55-$60^{\circ}C$, a Ceremix concentration of 1%, and a reaction time of 2 hr. Ceremix produced the highest dry matter yield and total saccharide extraction. Ginseng leaves were found to be the most suitable raw material for the production of fermented ginseng because they have higher carbohydrate and crude saponin contents than ginseng roots.

보리수 나무 열매로부터 라이코펜 생산을 위한 효소 분해 및 유기용매 추출 복합 공정의 최적화 (Optimization of Combined Process of Enzymatic Hydrolysis and Solvent Extraction for Production of Lycopene from Elaeagnus umbellata)

  • 보리수 나무 열매로부터 효소 분해 및 유기용매 추출의 복합 공정에 의한 라이코펜의 생산 조건을 최적화하였다. 보리수 나무의 열매를 증류수에 현탁하여 Celluclast, Ceremix, AMG, Viscozyme, Promozyme, Pectinex, Tunicase 및 Ultraflo와 같은 가수분해 효소를 사용하여 효소 반응을 수행하였고 효소 반응액을 에틸아세테이트, 아세톤, 클로로포름 및 헥산과 같은 유기 용매로 추출하였다. 가수분해 효소 중 Ceremix 복합 효소를 사용하고, 추출 용매는 클로로포름을 이용하여 추출하였을 때 가장 높은 lycopene 추출률을 나타냈다. 보리수 나무 열매의 효소처리 조건 최적화를 위해 실험계획법을 이용한 결과, 효소 반응의 pH는 5.5로, 반응 온도는 54.4℃로, 효소 첨가 농도는 0.58%로 하였을 때 lycopene 추출률이 가장 높았고 이 때 lycopene의 함량은 22.6 mg/100g을 나타냈다. 이와 같이 효소적 가수분해 공정과 유기 용매 추출 공정을 결합하여 활용하는 복합 공정은 기존 용매추출법에 비해 유효성분인 lycopene의 수율을 2.3배 증가시켰다. 이는 향후 바이오헬스 케어 제품의 기능성 소재로의 개발에 응용할 수 있는 결과라고 사료된다.
