  • We investigated simultaneous removal of heavy metals such as Cr, Ni, and Zn by adsorption onto powdered activated carbon (PAC) and PAC modified with sodium diethyldithiocarbamate (PAC-SDDC). Modification of PAC was confirmed by Fourier transform infrared spectroscopy and Scanning electron microscopy and energy dispersive X-ray spectroscopy. Both PAC and PAC-SDDC reached adsorption equilibrium within 48 h, and the adsorption kinetics followed a pseudo-second order reaction kinetics. The removal of metals was enhanced with increasing both adsorbent dosage and followed the descending order of Cr > Ni > Zn for PAC and Cr > Zn > Ni for PAC-SDDC, respectively. Adsorption kinetics followed pseudo-second order kinetics. Adsorption kinetic results were well fitted by the Freundlich isotherm except for Cr adsorption onto PAC. The optimum pH for heavy metal adsorption onto PAC was 5, whereas that for PAC-SDDC ranged from 7 to 9, indicating that modification of PAC with SDDC significantly enhanced heavy metal adsorption, especially under neutral and alkaline pH conditions. Our results imply that SDDC modified PAC can be applied to effectively remove heavy metals especially Cr in plating wastewaters without adjusting pH from alkaline to neutral.

  • Phospholipase A$_2$(PLA$_2$) catalyzes the hydrolysis of the sn-2 position of membrane glycerophospholipids to liberate arachidonic acid(AA), a precursor of eicosanoids including prostaglandins(PGs) and leukotrienes (LTs). The same reaction also produces lyso-phospholipids. So far, at least 19 enzymes that possess PLA2 activity have been identified, consists of low-molecular-weight, Ca$\^$2+/-requiring, secretory enzymes that have been implicated in a number of biological processes, such as modification of eicosanoid generation, inflammation, host defense, and atherosclerosis. • The purpose of Environmental Impact Statement (EIS) is an integral component of decisions on proposed plans, projects, and actions. Adapted from the National Environmental Policy Act(NEPA, 1969) environmental impact statement in Korea has aimed to analyse and prepare the mitigation in the process of big scale development and major industrial planning. During the past 19 years EIS has played the central role for the environmental issues in the process of economic development. Nevertheless EIS act needs some modification in the system as well as in the contents of evaluation process. EIS should be served as decision-making process by assessing environmental issues in the beginning stage of proposed actions. Other consideration was suggested to make EIS system better used for environment conservation.

  • Effects of acid ($HNO_3$ and HCl) modification on the adsorption properties of Pb(II) and Cu(II) onto bamboo-based activated carbon (BAC) were investigated through a series of batch experiments. The carbon content increased and oxygen content decreased with acid treatment. $HNO_3$ induced carboxylic acids and hydroxyl functional groups while HCl added no functional group onto BAC. The pseudo-second order model better described the kinetics of Pb(II) and Cu(II) adsorption onto experimented adsorbents, indicating that the rate-limiting step of the heavy metal sorption is chemical sorption involving valency forces through sharing or exchange of electrons between the adsorbate and the adsorbent. The equilibrium sorption data followed both Langmuir and Freundlich isotherm models. The adsorption capacities of BAC were affected by the surface functional groups added by acid modification. The adsorption capacities were enhanced up to 36.0% and 27.3% for Pb(II) and Cu(II), respectively by the $HNO_3$ modification, however, negligibly affected by HCl.

  • Consequence-Based Engineering (CBE) is a new paradigm proposed by the Mid-America Earthquake Center (MAE) to guide evaluation and rehabilitation of building structures and networks in areas of low probability - high consequence earthquakes such as the central region of the U.S. The principal objective of CBE is to minimize consequences by prescribing appropriate intervention procedures for a broad range of structures and systems, in consultation with key decision makers. One possible intervention option for rehabilitating unreinforced masonry (URM) buildings, widely used for essential facilities in Mid-America, is passive energy dissipation (PED). After the CBE process is described, its application in the rehabilitation of vulnerable URM building construction in Mid-America is illustrated through the use of PED devices attached to flexible timber floor diaphragms. It is shown that PED's can be applied to URM buildings in situations where floor diaphragm flexibility can be controlled to reduce both out-of-plane and in-plane wall responses and damage. Reductions as high as 48% in roof displacement and acceleration can be achieved as demonstrated in studies reported below.

  • Starch serves not only as an energy source for plants, animals, and humans but also as an environmentally friendly alternative for fossil fuels. Progress in understanding of starch biosynthesis, and the isolation of many genes involved in this process have enabled the genetic modification of crops in a rational manner to produce novel starches with improved functionality. Starch is composed of two glucose polymers, amylose and amylopectin. The amylose and amylopectin ratio in starch affects its physical and physicochemical properties. Alteration in starch structure can be achieved by modifying genes encoding the enzymes responsible for starch biosynthesis and starch hydrolysis. Here, we describe recent findings concerning the starch modification in sweetpotato. Sweetpotato [Ipomoea batatas (L.) Lam] ranks seventh in annual production among food crops in the world as an important starch source. To develop transgenic sweetpotato plants with modifying starch composition, we constructed transformation vectors overexpressing granule bound starch synthase I and inhibiting amylopectin synthesis genes such as starch branching enzyme and isoamylase under the control of 35S promoter, respectively. Transformation of sweetpotato (cv. Yulmi) is in progress.

  • The purpose of this study is to promote utilization of paper mill sludge as an adsorbent for stabilizing heavy metals in contaminated water by measuring the adsorption capacity of paper mill sludge for cadmium and arsenic. To measure adsorption capacity of paper mill sludge, sorption isotherm experiments were analyzed by Langmuir and Freundlich isotherm models. Also, two methods of chemical modifications were applied to improve the adsorption capacities of paper-mill-sludge: the first method used sodium hydroxide (NaOH), called PMS-1, and the second method used the NaOH and tartaric acid ($C_4H_6O_6$) together, called PMS-2. For Cd adsorption, PMS-1 presented the increase of reactivity while PMS-2 presented the decline of reactivity compared to that of untreated paper-mill-sludge. In case of As adsorption, both PMS-1 and PMS-2 showed the decrease of adsorption capacities. This is because zeta-potential of paper mill sludge was changed to more negative values during chemical modification process due to the hydroxyl group in NaOH and the carboxyl group in $C_4H_6O_6$, respectively. Therefore, we may conclude that the chemical treatment process increases adsorption capacity of paper mill sludge for cation heavy metals such as Cd but not for As.

  • Background: The roots of Panax ginseng contain two types of tetracyclic triterpenoid saponins, namely, protopanaxadiol (PPD)-type saponins and protopanaxatiol (PPT)-type saponins. In P. ginseng, the protopanaxadiol 6-hydroxylase (PPT synthase) enzyme catalyses protopanaxatriol (PPT) production from protopanaxadiol (PPD). In this study, we constructed homozygous mutant lines of ginseng by CRISPR/Cas9-mediated mutagenesis of the PPT synthase gene and obtained the mutant ginseng root lines having complete depletion of the PPT-type ginsenosides. Methods: Two sgRNAs (single guide RNAs) were designed for target mutations in the exon sequences of the two PPT synthase genes (both PPTa and PPTg sequences) with the CRISPR/Cas9 system. Transgenic ginseng roots were generated through Agrobacterium-mediated transformation. The mutant lines were screened by ginsenoside analysis and DNA sequencing. Result: Ginsenoside analysis revealed the complete depletion of PPT-type ginsenosides in three putative mutant lines (Cr4, Cr7, and Cr14). The reduction of PPT-type ginsenosides in mutant lines led to increased accumulation of PPD-type ginsenosides. The gene editing in the selected mutant lines was confirmed by targeted deep sequencing. Conclusion: We have established the genome editing protocol by CRISPR/Cas9 system in P. ginseng and demonstrated the mutated roots producing only PPD-type ginsenosides by depleting PPT-type ginsenosides. Because the pharmacological activity of PPD-group ginsenosides is significantly different from that of PPT-group ginsenosides, the new type of ginseng mutant producing only PPD-group ginsenosides may have new pharmacological characteristics compared to wild-type ginseng. This is the first report to generate target-induced mutations for the modification of saponin biosynthesis in Panax species using CRISPR-Cas9 system.
