  • The concept of “precision agriculture” or “site-specific farming” is usually confined to the fields of soil science, crop science and agronomy. However, because plants grow in soil, animals eat plants, and humans eat animal products, it could be argued (perhaps with some poetic licence) that the fields of feed quality, animal nutrition and animal production should also be considered in this context. NIR spectroscopy has proved over the last 20 years that it can provide a firm foundation for quality measurement across all of these fields, and with the continuing developments in instrumentation, computer capacity and software, is now a major cog in the wheel of precision agriculture. There have been a few giant leaps and a lot of small steps in the impact of NIR on the animal world. These have not been confined to the amazing advances in hardware and software, although would not have occurred without them. Rapid testing of forages, grains and mixed feeds by NIR for nutritional value to livestock is now commonplace in commercial laboratories world-wide. This would never have been possible without the pioneering work done by the USDA NIR Forage Research Network in the 1980's, following the landmark paper of Norris et al. in 1976. The advent of calibration transfer between instruments, algorithms which utilize huge databases for calibration and prediction, and the ability to directly scan whole grains and fresh forages can also be considered as major steps, if not leaps. More adventurous NIR applications have emerged in animal nutrition, with emphasis on estimating the functional properties of feeds, such as in vivo digestibility, voluntary intake, protein degradability and in vitro assays to simulate starch digestion. The potential to monitor the diets of grazing animals by using faecal NIR spectra is also now being realized. NIR measurements on animal carcasses and even live animals have also been attempted, with varying degrees of success, The use of discriminant analysis in these fields is proving a useful tool. The latest giant leap is likely to be the advent of relatively low-cost, portable and ultra-fast diode array NIR instruments, which can be used “on-site” and also be fitted to forage or grain harvesters. The fodder and livestock industries are no longer satisfied with what we once thought was revolutionary: a 2-3 day laboratory turnaround for fred quality testing. This means that the instrument needs to be taken to the samples rather than vice versa. Considerable research is underway in this area, but the challenge of calibration transfer and maintenance of instrument networks of this type remains. The animal world is currently facing its biggest challenges ever; animal welfare, alleged effects of animal products on human health, environmental and economic issues are difficult enough, but the current calamities of BSE and foot and mouth disease are “the last straw” NIR will not of course solve all these problems, but is already proving useful in some of these areas and will continue to do so.

Improvement of Postharvest Fruit Quality in 'Formosa' Plums (Prunus salicina) after Treatment with 1-methylcyclopropene during Storage

  • Plum is a climacteric fruit and softening is a serious problem for storage and transportation. Thus $1{\mu}L{\cdot}L^{-1}$ of 1-methylcyclopropene (1-MCP) was applied to plums to prolong their shelf life and maintain quality. Japanese plums (Prunus salicina cv. Formosa) were stored at $20^{\circ}C$ and $0^{\circ}C$ for 14 days and 46 days respectively, with or without 1-MCP treatment. Fruits were treated with $1{\mu}L{\cdot}L^{-1}$ 1-MCP at $10^{\circ}C$ for 24 h. Ethylene production and respiration rate were strongly inhibited in 1-MCP-treated fruits at $20^{\circ}C$. It was also observed that there was less ethanol and acetaldehyde evaporation in 1-MCP-treated fruits stored at $20^{\circ}C$ compared to those in control fruits not treated with 1-MCP. Fruit qualities, such as firmness, titratable acidity (TA), skin color, and decay, changed more slowly in 1-MCP-treated fruits stored at $20^{\circ}C$ than in untreated fruits. There were no differences in the ethylene production or respiration rate between the groups of fruits stored at $0^{\circ}C$ throughout the experiment. Chilling injury was also inhibited by the application of 1-MCP during storage at $0^{\circ}C$. When the fruits stored at $0^{\circ}C$ with or without 1-MCP were transferred to $20^{\circ}C$ after 25 days, the differences in ethylene production and respiration rate, firmness, TA, TSS, and acetaldehyde and ethanol evaporation between the initial (after being stored at $0^{\circ}C$ for 25 days) and the final measurements (after being stored at $0^{\circ}C$ for 25 days and then transferred to $20^{\circ}C$ for three days) were lower in 1-MCP treated fruits than in non-treated fruits. The postharvest application of 1-MCP in Formosa plums showed positive effects at both $0^{\circ}C$ and $20^{\circ}C$ storage conditions with regard to quality, such as low ethylene production and low respiration rates, firmness, TA, ethanol, and acetaldehyde evaporation, chilling injury, and decay.

Inhibitory Activity of Garlic Fermented by Pediococcus pentosaceus KACC 91419 against Antibiotic-resistant Pathogens

  • The aim of this study was to screen lactic acid bacteria for the fermentation of garlic and to assess the increase in inhibitory activity of garlic fermented against antibiotic-resistant pathogens for use as an animal feed supplement. We screened 45 strains of lactobacillus for the fermentation of garlic. Of these strains, 23 showed similar growth rates with or without allicin. Cultures of the 23 strains were mixed with an equivalent amount of garlic juice and incubated overnight at $37^{\circ}C$. The three strains with the lowest pH values were Lactobacillus paracasei KCTC 3169, L5 strain, and L. reuteri SW. Garlic juice fermented by the L5 strain more strongly inhibited antibiotic-resistant pathogenic bacteria than L. paracasei KCTC 3169, L. reuteri SW, or garlic juice itself. By examining carbohydrate utilization, morphologic properties and 16S rRNA gene sequences, we identified the L5 strain as Pediococcus pentosaceus and deposited it in the name of P. pentosaceus KACC 91419 into the Korea Agricultural Culture Collection. To identify the antimicrobial compound from the garlic filtrate fermented by P. pentosaceus KACC 91419, we fractionated P. pentosaceus KACC 91419 culture on a C18 column and checked the antimicrobial activity of fractions A6 to A10. Only fraction A9 showed inhibitory activity on Staphylococcus aureus. Comparing the mass spectra of the fractions with and without antimicrobial activity, we observed a single dominant product ion (m/z 157.99) from the fraction showing antimicrobial activity. Its molecular mass (157.99) was 2 atomic mass units less than that of allicin (162.02). This suggests that allicin might be converted to its derivative, which has antimicrobial activity, during fermentation by P. pentosaceus KACC 91419.

중국 길림성 백성지역 흑개토의 이화학성 및 인산 흡착 특성 (Physical-chemical Properties and Phosphorus Adsorption Characteristics of Soils in Baicheng, China)

  • 농경지로의 개발 가능성이 높은 중국 길림성 백성지역의 주요한 토양인 흑개토의 이화학성 및 인산 흡착특성을 조사하였다. 토지 이용방식 또는 재배작물이 서로 다른 중국 길림성 백성지역 4곳과 중국 용정에 위치한 연변 대학교 농과대학 부속 시험장 1곳에서 1993년 8월에 토양을 채취하여 실험을 수행하였다, 중국 길림성 백성지역 토양의 $_PH$는 미경작지 10.2, 경작지는 $7.3{\sim}7.6$으로 비교적 높았다. 양이온 치환용량은 20 cmol(+) $kg^{-1}$ 이상이었으며, 치환성 양이온 중 Na는 특히 표토에 많이 존재하였으며, Ca의 함량이 매우 높아 Ca 포화율이 100% 이상을 나타내었다. 토양 포화 침출액의 주요 양이온은 Na이었으며, ECe와 SAR을 기준으로 백성 미경작지 토양은 염류-나트륨성 토양, 경작지 토양은 나트륨성 또는 일반 토양에 해당하였다. 백성지역 토양의 유효인산 함량은 10mg P $kg^{-1}$ 미만으로 아주 낮았으며, 최대 인산흡착량은 $ $ mg P $kg^{-1}$이었다. 길림성 백성지역 토양의 경우 유효인산 함량이 매우 낮아 토양 비옥도 중진을 위한 인산질 비료의 시용과 함께 토양 투수성의 개선 등과 같은 적절한 토양 관리가 필요하며, 용정지역 토양의 경우에는 인산흡착 특성을 고려하여 인의 과잉 축적을 방지하기 위한 인산질 비료의 적정 시용을 권장하여야 한다.

화학비료, 가축분뇨 및 퇴비의 질소동위원소비 (Nitrogen Isotope Compositions of Synthetic Fertilizer, Raw Livestock Manure Slurry, and Composted Livestock Manure)

  • 화학비료, 가축분뇨 및 퇴비 등 주요 질소원의 질소동위원소비 (${\delta}^{15}N$) 차이를 조사하기 위해 각각 8, 4, 37점의 시료를 채취하여 ${\delta}^{15}N$을 분석하였다. 평균 ${\delta}^{15}N$ 값은 화학비료가 $-1.5{\pm}0.5$‰ (범위: -3.9~+0.5‰‰), 가축분뇨가 $+6.3{\pm}0.4$‰ (+5.3~+7.2‰), 가축분퇴비가 $+16.0{\pm}0.4$‰ (+9.3~+20.9‰)였다. 화학비료가 타 질소원에 비해 ${\delta}^{15}N$ 값이 낮은 것은 화학비료 제조시 이용하는 질소원인 대기 $N_2$${\delta}^{15}N$ 값 (0‰)을 반영하기 때문이다. 반면, 가축분에 비해 퇴비의 ${\delta}^{15}N$ 값이 높은 것은 퇴비화 과정 중 일어나는 질소손실 (특히, 암모니아 휘산)과 관련된 질소동위원소분할효과 ($^{14}N$의 손실속도>$^{15}N$의 손실속도)에 의한 퇴비 중 $^{15}N$ 농축에 의한 결과로 판단된다. 따라서, 본 연구는 ${\delta}^{15}N$ 분석을 통해 현재 우리나라 농업 시스템에서 가장 널리 이용되고 있는 두 가지 질소원 (화학비료와 퇴비)을 구분할 수 있음을 보여준다.

Multiple Genes Related to Muscle Identified through a Joint Analysis of a Two-stage Genome-wide Association Study for Racing Performance of 1,156 Thoroughbreds

  • Thoroughbred, a relatively recent horse breed, is best known for its use in horse racing. Although myostatin (MSTN) variants have been reported to be highly associated with horse racing performance, the trait is more likely to be polygenic in nature. The purpose of this study was to identify genetic variants strongly associated with racing performance by using estimated breeding value (EBV) for race time as a phenotype. We conducted a two-stage genome-wide association study to search for genetic variants associated with the EBV. In the first stage of genome-wide association study, a relatively large number of markers (~54,000 single-nucleotide polymorphisms, SNPs) were evaluated in a small number of samples (240 horses). In the second stage, a relatively small number of markers identified to have large effects (170 SNPs) were evaluated in a much larger number of samples (1,156 horses). We also validated the SNPs related to MSTN known to have large effects on racing performance and found significant associations in the stage two analysis, but not in stage one. We identified 28 significant SNPs related to 17 genes. Among these, six genes have a function related to myogenesis and five genes are involved in muscle maintenance. To our knowledge, these genes are newly reported for the genetic association with racing performance of Thoroughbreds. It complements a recent horse genome-wide association studies of racing performance that identified other SNPs and genes as the most significant variants. These results will help to expand our knowledge of the polygenic nature of racing performance in Thoroughbreds.

보검선인장과 저단선인장의 열매와 줄기 영양성분 조성 (Nutrient Composition in the Parts of Opuntia ficus-indica var. saboten and Opuntia humifusa)

  • This study was conducted to analyze the nutrient compositions of stem and fruit of Opuntia ficus-indica var. saboten (OF) and Opuntia humifusa (OH). The results of the two-way analysis of variance test indicated that the parts, varieties and parts*varieties had influence on nutrient content except crude protein, soluble dietary fiber, Fe, Se, I, vitamin E, niacin and vitamin C. Moisture and crude ash content was higher in stem than in fruit, while crude lipid and insoluble dietary content was higher in fruit than in stem. Mineral content revealed that K (1,313.67 mg% in OF, 1,351.38 mg% in OH) was the highest in fruit and Ca (5,146.29 mg% in OF, 1,388.19 mg% in OH) was the highest in stem. Vitamin C was the most abundant vitamin in the fruit of OF (199.98 mg%) and OH (187.12 mg%). Polyunsaturated fatty acid was the highest among fatty acids (66.9~70.1%), with higher content in the stem (753.89 mg%) than fruit (578.01 mg%) in OF, while higher in the fruit (1,093.63 mg%) than stem (475.07 mg%) in OH. Moisture, crude protein, Mg, Se and riboflavin was higher in OF than OH; whereas, crude lipid, insoluble dietary fiber, total dietary fiber and monounsaturated fatty acid were higher in OH than OF. These results indicated that OF and OH could be a good food source for Ca, K and Vitamin C. Also, different nutrient content by parts and varieties, can be helpful in choice of parts or varieties for consumer purposes.

어병 세균에 대한 소나무과 잎 정유의 항세균 효과 (Antibacterial Activity of Essential Oils from Pinaceae Leaves Against Fish Pathogens)

  • 어병 세균은 어류 양식업의 경제적 피해 뿐만 아니라, 인수공통감염원으로 알려진 전염성 병원균이다. 어병 세균을 제어하기 위해 지속적인 항생제의 사용은 부작용이 수반되기 때문에, 천연 유래 소재의 개발이 요구된다. 본 연구에서는 항생제대체제 개발을 위해 항세균효과가 우수한 침엽수 정유를 발굴하고자 하였다. 소나무과에 속하는 전나무 (Abies holophylla), 곰솔 (Pinus thunbergii), 섬잣나무 (Pinus parviflora), 솔송 (Tsuga sieboldii), 리기테다소나무 (Pinus rigitaeda)의 잎에서 hydro-distillation법을 이용하여 정유를 추출하였으며, 추출된 정유는 어병 세균인 Edwardsiella tarda, Photobacterium damselae, Streptococcus parauberis, Lactococcus garivieae에 대하여 항균력을 평가하였다. 그 결과, 전나무와 곰솔 잎 정유가 그람 음성 세균인 E. tarda와 P. damselae에 대하여 선택적으로 강한 항균력을 나타냈다. GC-MS 분석 결과, 전나무 잎 정유의 주요 성분은 (-)-bornyl acetate (29.45%), D-limonene (20.47%), camphene (11.73%)이고, 곰솔 잎 정유의 주요 성분은 α-pinene (59.81%)으로 각각 확인되었다. 또한, 미량으로 존재하지만 정유와 동일한 효능을 나타내는 유효 성분으로 oxygenated monoterpenes인 neryl acetate, (-)-borneol, (-)-carveol의 세가지 화합물을 구명하였다. 따라서 그람 음성어병세균의 생장억제효과가 우수한 전나무와 곰솔 잎 정유는 사료 첨가제, 수산용 의약품 등 생물학적 제제로 활용 될 수 있을 것으로 사료된다.

일계성 딸기 과실의 부위에 따른 바이오 활성 화합물 함량 비교 (Comparison of Bioactive Compounds Contents in Different Fruit Tissues of June-bearing Strawberry Cultivars)

  • '매향', '설향', '금향', '아키히메'와 '레드펄' 딸기 과실 부위에서 환원당, 유기산, 아스코르빈산, 안토시아닌 및 엘라직산의 함량을 비교하였다. 환원당, 유기산, 아스코르빈산 및 안토시아닌 함량은 딸기 표피에서 다른 부위의 함량 보다 유의하게 높았으나 엘라직산의 함량은 딸기 수과에서 높았다. '매향' 딸기의 표피 및 과육에서 아스코르빈산 함량은 각각 69.2와 $42.2mg{\cdot}100g^{-1}$ FW로 다른 품종에 비해 높았다. 그리고 '금향' 딸기의 표피 및 수과에서 안토시아닌 함량은 각각 $74.0mg{\cdot}100g^{-1}$ FW과 $36.7mg{\cdot}100g^{-1}$ DW으로 타 품종에 비해 유의하게 높았으며 '설향' 딸기 수과에서 엘라직산의 함량은 $215.5mg{\cdot}100g^{-1}$ DW로 타 품종에 비해 유의하게 높았다. 딸기 과실의 부위에 따라 바이오 활성 화합물들의 함량은 크게 차이가 있었으며 '매향', '설향' 및 '금향' 딸기 품종은 바이오 활성 화합물들의 함량이 높아 소비자들에게는 고품질 딸기로 인식될 수 있어 국내 육성 품종 딸기의 보급이 확산될 것으로 기대된다. 그리고 본 기초적 결과들은 딸기 가공 분야 및 영양학 연구 분야에도 유용하게 활용될 것으로 판단된다.

Changes in Nutrient Levels of Aqueous Extracts from Radish (Raphanus sativus L.) Root during Liquefaction by Heat and Non-heat Processing

  • The amount of cellular components including soluble sugars, amino acids, organic acids and glucosinolates (GLS) was investigated during radish root processing to develop a radish beverage. The radish root was divided into two parts, white and green tissue, and processed separately by extracting the juice from the fresh tissue and from the boiled tissue to compare differences in the components content among the preparations. The overall palatability of both the fresh and boiled extracts from the green part of the radish was higher than that of the same extracts from the white part. The sweetness of extract by boiling increased and its pungency decreased, thereby the palatability increased by being compared to the fresh radish extract. The sweetness was affected by sucrose not by glucose or fructose of monosaccharides by showing different sucrose contents according to treatment comparing palatability. Malic acid was identified as primary organic acid, and the content was higher in both the fresh and boiled extracts from the white part than in the extracts from the green part of the radish. The fresh extract from the green part of the radish contained more essential amino acids, such as threonine and valine, and more hydrophilic amino acids including glutamic acid, aspartic acid, and arginine than those of the fresh extract from the white part, suggesting the green fresh part is more palatable than the white fresh part. The main sulfur compound was ethylthiocyanate in radish, and others were butyl isothiocyanate, dimethyl-disulfide, and 4-methylthio-3-butylisothiocyanate. The four GLS were detected much more in the fresh green and fresh white parts of the radish because they evaporated during boiling. The contents of the four sulfur compounds were higher in the white fresh part than in the green fresh part, which is likely the reason the pungency was higher and the palatability was lower in the white fresh part than in the green fresh part of the radish. The ascorbic acid content was higher in the fresh extract compared to the boiled extracts from both the green and white parts. Taken together, these findings indicate that fresh radish extract is superior to obtain in terms of retaining desirable nutritional and functional components for health.
