The Relationship among Social Support, Work-Family Conflict and Work-Family Enrichment of Married Nurses (기혼간호사의 사회적 지지와 일-가정 갈등, 일-가정 향상과의 관계)

  • Purpose: This study is to identify the relationship among married nurses' social support, work-family conflict and work-family enrichment and to examine the influence of social support on work-family conflict and work-family enrichment. Methods: Data were collected from married nurses working at three hospitals with more than 100 beds in J Province and analyzed by descriptive statistics, t-test, ANOVA, Mann-Whitney U test, Kruskal-Wallis H test, Pearson's correlation analysis and hierarchical regression analysis using SPSS 21 program. Findings: In the correlation analysis only social support and work-family enrichment showed positive correlation. As a result of hierarchical multiple regression analysis, social support still acted as a significant influence factor on work-family conflict and work-family enrichment even in the state of considering the effect of control variables. Conclusion: In order to lower work-family conflict and to enhance work-family enrichment of married nurses, it is necessary to introduce and implement welfare policies for work-family reconciliation at the workplace level and at home level it is necessary to provide concrete measures so that married nurses can find the value of life as workers while house working and nurturing.

Acidophilic Bacterial Communities of Soil and Enrichment Cultures from Two Abandoned Mine Sites of the Korean Peninsula

  • Bacterial diversity based on the denaturing gradient gel electrophoresis (DGGE) analysis of PCR-amplified 16S rRNA gene sequences was determined for soil samples from two abandoned mine sites and the corresponding enrichment cultures using soil sample as key inoculum. Sequencing analysis of DGGE bands obtained from both the soil samples matched mostly with sequences of uncultured and newly described organisms, or organisms recently associated with the acid mine drainage environment. However, the enrichment of soil samples in ferrous sulfate and elemental sulfur media yielded sequences that were consistent with well-known iron- and sulfur-oxidizing acidophilic bacteria. Analysis of enrichment cultures of soil samples from Dalsung mine revealed abundant ${\gamma}$-$Proteobacteria$, whereas that of Gubong mine sample displayed acidophilic groups of ${\gamma}$-$Proteobacteria$, ${\alpha}$-$Proteobacteria$, $Actinobacteria$ and $Firmicutes$. Chemical elemental analysis of the mine samples indicated that the Dalsung site contained more iron and sulfate along with other toxic components as compared with those of the Gubong site. Biogeochemistry was believed to be the primary control on the acidophilic bacterial group in the enrichment samples.

  • Gene set enrichment analysis (GSEA) is a computational method to identify statistically significant gene sets showing significant differences between two groups of microarray expression profiles and simultaneously uncover their biological meanings in an elegant way by employing gene annotation databases, such as Cytogenetic Band, KEGG pathways, gene ontology, and etc. For the gone set enrichment analysis, all the genes in a given dataset are first ordered by the signal-to-noise ratio between the groups and then further analyses are proceeded. Despite of its impressive results in several previous studies, however, gene ranking by the signal-to-noise ratio makes it difficult to consider highly up-regulated genes and highly down-regulated genes at the same time as the candidates of significant genes, which possibly reflect certain situations incurred in metabolic and signaling pathways. To deal with this problem, in this article, we investigate the gene set enrichment analysis method with Fisher criterion for gene ranking and also evaluate its effects in Leukemia related pathway analyses.

Identification of key genes and functional enrichment analysis of liver fibrosis in nonalcoholic fatty liver disease through weighted gene co-expression network analysis

  • Nonalcoholic fatty liver disease (NAFLD) is a common type of chronic liver disease, with severity levels ranging from nonalcoholic fatty liver to nonalcoholic steatohepatitis (NASH). The extent of liver fibrosis indicates the severity of NASH and the risk of liver cancer. However, the mechanism underlying NASH development, which is important for early screening and intervention, remains unclear. Weighted gene co-expression network analysis (WGCNA) is a useful method for identifying hub genes and screening specific targets for diseases. In this study, we utilized an mRNA dataset of the liver tissues of patients with NASH and conducted WGCNA for various stages of liver fibrosis. Subsequently, we employed two additional mRNA datasets for validation purposes. Gene set enrichment analysis (GSEA) was conducted to analyze gene function enrichment. Through WGCNA and subsequent analyses, complemented by validation using two additional datasets, we identified five genes (BICC1, C7, EFEMP1, LUM, and STMN2) as hub genes. GSEA analysis indicated that gene sets associated with liver metabolism and cholesterol homeostasis were uniformly downregulated. BICC1, C7, EFEMP1, LUM, and STMN2 were identified as hub genes of NASH, and were all related to liver metabolism, NAFLD, NASH, and related diseases. These hub genes might serve as potential targets for the early screening and treatment of NASH.

A Multidimensional System for Phosphopeptide Analysis Using TiO2 Enrichment and Ion-exchange Chromatography with Mass Spectrometry

  • Although offline enrichment of phosphorylated peptides is widely used, enrichment for phosphopeptides using $TiO_2$ is often performed manually, which is labor-intensive and can lead to irreproducible results. To address the problems associated with offline enrichment and to improve the effectiveness of phosphopeptide detection, we developed an automated online enrichment system for phosphopeptide analysis. A standard protein mixture comprising BSA, fetuin, crystalline, ${\alpha}$-casein and ${\beta}$-casein, and ovalbumin was assessed using our new system. Our multidimensional system has four main parts: a sample pump, a 20-mm $TiO_2$-based column, a weak anion-exchange, and a strong cation-exchange (2:1 WAX:SCX) separation column with LC/MS. Phosphorylated peptides were successfully detected using the $TiO_2$-based online system with little interference from nonphosphorylated peptides. Our results confirmed that our online enrichment system is a simple and efficient method for detecting phosphorylated peptides.

NGSEA: Network-Based Gene Set Enrichment Analysis for Interpreting Gene Expression Phenotypes with Functional Gene Sets

  • Gene set enrichment analysis (GSEA) is a popular tool to identify underlying biological processes in clinical samples using their gene expression phenotypes. GSEA measures the enrichment of annotated gene sets that represent biological processes for differentially expressed genes (DEGs) in clinical samples. GSEA may be suboptimal for functional gene sets; however, because DEGs from the expression dataset may not be functional genes per se but dysregulated genes perturbed by bona fide functional genes. To overcome this shortcoming, we developed network-based GSEA (NGSEA), which measures the enrichment score of functional gene sets using the expression difference of not only individual genes but also their neighbors in the functional network. We found that NGSEA outperformed GSEA in identifying pathway gene sets for matched gene expression phenotypes. We also observed that NGSEA substantially improved the ability to retrieve known anti-cancer drugs from patient-derived gene expression data using drug-target gene sets compared with another method, Connectivity Map. We also repurposed FDA-approved drugs using NGSEA and experimentally validated budesonide as a chemical with anti-cancer effects for colorectal cancer. We, therefore, expect that NGSEA will facilitate both pathway interpretation of gene expression phenotypes and anti-cancer drug repositioning. NGSEA is freely available at www.inetbio.org/ngsea.

A Comprehensive Review of Recent Advances in the Enrichment and Mass Spectrometric Analysis of Glycoproteins and Glycopeptides in Complex Biological Matrices

  • Protein glycosylation, a highly significant and ubiquitous post-translational modification (PTM) in eukaryotic cells, has attracted considerable research interest due to its pivotal role in a wide array of essential biological processes. Conducting a comprehensive analysis of glycoproteins is imperative for understanding glycoprotein bio-functions and identifying glycosylated biomarkers. However, the complexity and heterogeneity of glycan structures, coupled with the low abundance and poor ionization efficiencies of glycopeptides have all contributed to making the analysis and subsequent identification of glycans and glycopeptides much more challenging than any other biopolymers. Nevertheless, the significant advancements in enrichment techniques, chromatographic separation, and mass spectrometric methodologies represent promising avenues for mitigating these challenges. Numerous substrates and multifunctional materials are being designed for glycopeptide enrichment, proving valuable in glycomics and glycoproteomics. Mass spectrometry (MS) is pivotal for probing protein glycosylation, offering sensitivity and structural insight into glycopeptides and glycans. Additionally, enhanced MS-based glycopeptide characterization employs various separation techniques like liquid chromatography, capillary electrophoresis, and ion mobility. In this review, we highlight recent advances in enrichment methods and MS-based separation techniques for analyzing different types of protein glycosylation. This review also discusses various approaches employed for glycan release that facilitate the investigation of the glycosylation sites of the identified glycoproteins. Furthermore, numerous bioinformatics tools aiding in accurately characterizing glycan and glycopeptides are covered.

혐기성 PCE 탈염소화 관련 미생물 군집 특성

  • Tetrachloroethylene(PCE) dechlorination was investigated in an anaerobic enrichment culture from landfill soil. Anaerobic PCE dechlorinating microorganisms could convert 150mg/L of PCE via trichloroethylene(TCE) to cir-1,2-dichloroethylene(CDCE) within 2 days at the optimum temperature of 30 to 35$^{\circ}C$. The enrichment culture could dechlorinate TCE but did not degrade other chlorinated aliphatic compounds, such as cDCE, trans-1,2-dichloroethylene, 1,1-dichloroethylene, 1,1-dichloroethane, 1,2-dichloro- ethane, and 1,1,1-trichloroethane during 5 days incubation. Several isolates from the enrichment culture did not show dechlorinating activity of PCE. Microbial analysis of the dechlorinating enrichment culture by using Polymerase chain reaction-Denaturing gradient gel electrophoresis (PCR-DGGE) method showed that at least three microorganisms were related to the anaerobic PCE dechlorination in the enrichment

The influences of work-family conciliation strategies on work-family conflict and enrichment in dual-earner couples who live in Daegu with children (대구광역시에 거주하는 맞벌이부부의 일-가족조정전략이 일-가족 갈등과 일-가족 향상에 미치는 영향)

  • The purpose of this study was to investigate the influences of work-family conciliation strategies on work-family conflict and enrichment in dual-earner couples who live in Daegu with children. The subjects of this study were 176 dual-earner couples, who have under 16 youngest children and work over 15 hours each week. The research tool was questionnaires. For data analysis, factor analysis, Cronbach ${\alpha}$, paired t-test and multiple regression were performed. The main results of this study were as following. First, there were significant differences in management and planning skill strategy, work-family conflict and enrichment according to gender. Second, positive attitudes strategy toward multiple responsibilities, wives use, had an influence on work${\rightarrow}$family conflict in wives. Also, partner coping, management and planning skill, and positive attitudes strategy, wives use, and management and planning skill strategy, husbands use, had an influence on family${\rightarrow}$work conflict in wives. GoBean: a Java GUI application for visual exploration of GO term enrichments

  • We have developed a biologist-friendly, Java GUI application (GoBean) for GO term enrichment analysis. It was designed to be a comprehensive and flexible GUI tool for GO term enrichment analysis, combining the merits of other programs and incorporating extensive graphic exploration of enrichment results. An intuitive user interface with multiple panels allows for extensive visual scrutiny of analysis results. The program includes many essential and useful features, such as enrichment analysis algorithms, multiple test correction methods, and versatile filtering of enriched GO terms for more focused analyses. A unique graphic interface reflecting the GO tree structure was devised to facilitate comparisons of multiple GO analysis results, which can provide valuable insights for biological interpretation. Additional features to enhance user convenience include built in ID conversion, evidence code-based gene-GO association filtering, set operations of gene lists and enriched GO terms, and user -provided data files.
