A Study on Ways to Improve Catalog Enriched Content Services in Domestic Public Libraries (국내 공공도서관의 목록 보강콘텐츠 서비스 개선방안에 관한 연구)

  • The purpose of this study is to derive implications through a comparison of the current status of catalog enriched content services provision in U.S. public libraries and domestic public libraries. In addition, we are seeking ways to improve the catalog enriched content services for domestic public libraries in the future. From early September to mid-October 2023, specific books were searched on public library websites in the U.S. and Korea, and the functions of the enriched content services shown in the search results were compared. The results are as follows: First, domestic public library enriched content services require a separate company to develop and provide an enriched content services solution. Second, the enriched content services platform must discover domestic information sources that can be utilized in the areas of book-centered, book recommendation, and community engagement. Third, it is necessary to develop enriched content using public data such as the Library Information Naru. Fourth, each integrated library must that data generated from local community engagement services can be utilized as an enriced content service.

A Study on the Enriched Contents of OPACs of Public Libraries in Korea (공공도서관 온라인 목록의 보강된 콘텐츠에 관한 연구)

  • The purpose of this study is to analyze the present state of the enriched contents of OPACs of public libraries in Korea. Examined are the OPACs of 125 public libraries which hold Jeon Du-hwan Hoegorok, and 260 public libraries which hold Banil Jongjokjuui retrieved from KOLIS-NET of the National Library of Korea from Sept. through Oct. 2019. The enriched contents of these controversial books are mostly provided by Naver, Daum, and Aladin Internet Bookstore, and their so-called summaries, reviews, etc. are actually only excerpts supplied by publishers. They are given only as a link to external sources or the whole or partial display in a bibliographic record. Since these enriched contents contain only publishers' promotional statements, but not any objective and professional review, they are not much useful for library users. Therefore, more active participation of librarians in creating or evaluating the enriched contents are proposed, especially for such controversial books.

Enhancing the performance of a long-life modified CANDLE fast reactor by using an enriched 208Pb as coolant

  • The investigation of the utilization of enriched 208Pb as a coolant to enhance the performance of a long-life fast reactor with a Modified CANDLE (Constant Axial shape of Neutron flux, nuclide densities, and power shape During Life of Energy production) burnup scheme has performed. The analyzes were performed on a reactor with thermal power of 800 MegaWatt Thermal (MWTh) with a refueling process every 15 years. Uranium Nitride (enriched 15N), 208Pb, and High-Cr martensitic steel HT-9 were employed as fuel, coolant, and cladding materials, respectively. One of the Pb-nat isotopes, 208Pb, has the smallest neutron capture cross-section (0.23 mb) among other liquid metal coolants. Furthermore, the neutron-producing cross-section (n, 2n) of 208Pb is larger than sodium (Na). On the other hand, the inelastic scattering energy threshold of 208Pb is the highest among Na, natPb, and Bi. The small inelastic scattering cross-section of 208Pb can harden the neutron energy spectrum. Therefore, 208Pb is a better neutron multiplier than any other liquid metal coolant. The excess neutrons cause more production than consumption of 239Pu. Hence, it can reduce the initial fuel loading of the reactor. The selective photoreaction process was developing to obtain enriched 208Pb. The neutronic was calculated using SRAC and JENDL 4.0 as a nuclear data library. We obtained that the modified CANDLE reactor with enriched 208Pb as coolant and reflector has the highest k-eff among all reactors. Meanwhile, the natPb cooled reactor has the lowest k-eff. Thus, the utilization of the enriched 208Pb as the coolant can reduce reactor initial fuel loading. Moreover, the enriched 208Pb-cooled reactor has the smallest power peaking factor among all reactors. Therefore, the enriched 208Pb can enhance the performance of a long-life Modified CANDLE fast reactor.

Characterization of an alkaline esterase from an enriched metagenomic library derived from an oil-spill area

  • A novel esterase gene (est7S) was cloned from an enriched metagenomic library derived from an oil-spill area. The gene encoded a protein of 505 amino acids, and the molecular mass of the Est7S was estimated to be 54,512 Da with no signal peptide. Est7S showed the highest identity of 40% to an esterase from a sludge metagenome compared to the characterized enzymes with their properties, although it showed 99% identity to a carboxylesterase in the genome sequence of Alcanivorax borkumensis SK2. Est7S had catalytic triad residues, Ser183, Glu312, and His420, and the GESAG motif in most family VII lipolytic enzymes. Est7S was purified from the crude extract of clone SM7 using Sephacryl S-200 HR and HiTrap Q column chromatographies. The purified Est7S was optimally active at $50^{\circ}C$ and pH 10.0. Est7S showed a high specific activity of 366.7 U/mg protein. It preferred short length esters, particularly p-nitrophenyl acetate, efficiently hydrolyzed R- and S-enantiomers of methyl-3-hydroxy-2-methylpropionate, and glyceryl tributyrate. These properties of Est7S may provide potential merits in biotechnological applications such as detergent and paper processing under alkaline conditions.

  • The search for relevant and actionable information is a key to achieving clinical and research goals in biomedicine. Biomedical information exists in different forms: as text and illustrations in journal articles and other documents, in images stored in databases, and as patients' cases in electronic health records. This paper presents ways to move beyond conventional text-based searching of these resources, by combining text and visual features in search queries and document representation. A combination of techniques and tools from the fields of natural language processing, information retrieval, and content-based image retrieval allows the development of building blocks for advanced information services. Such services enable searching by textual as well as visual queries, and retrieving documents enriched by relevant images, charts, and other illustrations from the journal literature, patient records and image databases.

Identification of New Microsatellite Markers in Panax ginseng

  • Microsatellites, also called simple sequence repeats (SSR), are very useful molecular genetic markers commonly used in crop breeding, species identification and linkage analysis. In the present study, we constructed a microsatellite-enriched genomic library of Panax ginseng, and identified 251 novel microsatellite sequences. Tri-nt repeat units were the most abundant (46.6%), followed by di-nt repeats (35.5%). The $(AG)_n$ motif was most common (23.1%), followed by the $(AAC)_n$ motif (22.3%). From the genotyping of 94 microsatellites using marker-specific primer sets, we identified 11 intraspecific polymorphic markers as well as 14 possible interspecific polymorphic markers differing between P. ginseng and P. quinquefolius. The exact allele structures of the polymorphic markers were determined and the alleles were named. This study represents the first report of the bulk isolation of microsatellites by screening a microsatellite-enriched genomic library in P. ginseng. The microsatellite markers could be useful for linkage analysis, genetic breeding and authentication of Panax species.

Development of Gene Based STS Markers in Wheat

  • The objective of this study is to develop the gene based sequence tagged site (STS) markers in wheat. The euchromatin enriched genomic library was constructed and the STS primer sets were designed using gene based DNA sequence. The euchromatin enriched genomic (EEG) DNA library in wheat was constructed using the $Mcr$A and $Mcr$BC system in $DH5{\alpha}$ cell. The 2,166 EEG colonies have been constructed by methylated DNA exclusion. Among the colonies, 606 colonies with the size between 400 and 1200 bp of PCR products were selected for sequencing. In order to develop the gene based STS primers, blast analysis comparing between wheat genetic information and rice genome sequence was employed. The 227 STS primers mainly matched on $Triticum$ $aestivum$ (hexaploid), $Triticum$ $turgidum$ (tetraploid), $Aegilops$ (diploid), and other plants. The polymorphisms were detected in PCR products after digestion with restriction enzymes. The eight STS markers that showed 32 polymorphisms in twelve wheat genotypes were developed using 227 STS primers. The STS primers analysis will be useful for generation of informative molecular markers in wheat. Development of gene based STS marker is to identify the genetic function through cloning of target gene and find the new allele of target trait.

A Case Study on the Next Generation Library Catalogs (차세대 도서관 목록 사례의 고찰)

  • The purpose of this study is to investigate the major features of Next Generation Library Catalogs. 'Next Generation Melvyl Pilot' of University of California Library System and 'SearchWorks' of Stanford University Library are examined. While the former is developed, based on OCLC WorldCat Local, the latter is based on the Blacklight, an Open Source Catalog Software. Both commonly provide the features, including enriched contents, facet navigation, keyword searching, relevancy ranking of search results, and user contribution, etc., but some functions vary in scopes and contents. Also, it seems that both are in process of development rather than complete implementations.

Distribution and Frequency of SSR Motifs in the Chrysanthemum SSR-enriched Library through 454 Pyrosequencing Technology (국화 SSR-enriched library에서 SSR 반복염기의 분포 및 빈도)

  • Chrysanthemums, often called mums or chrysanths, belong to the genus Chrysanthemum, which includes about 30 species of perennial flowering plants in the family Asteraceae. We extracted DNA from Dendranthema grandiflorum ('Smileball') to construct a simple sequence repeat (SSR)-enriched library, using a modified biotin-streptavidin capture method. GS FLX (Genome Sequencer FLX System which provides the flexibility to perform the broad range of applications) sequencing (at the 1/8 run specification) resulted in 18.83 mega base pairs (Mbp) with an average read length of 280.06 bp. Sequence analyses of all SSR-containing clones revealed a predominance of di-nucleotide motifs (16,375, 61.5%) followed by tri-nucleotide motifs (6,616, 24.8%), tetra-nucleotide motifs (1,674, 6.3%), penta-nucleotide motifs (1,283, 4.8%), and hexa-nucleotide motifs (693, 2.6%). Among the di-nucleotide motifs, the AC/CA class was the most frequently identified (93.5% of all di-nucleotide types), followed by the GA/AG class (6.1%), the AT/TA class (0.4%), and the CG/GC class (0.03%). When we analyzed the distribution of different repeat motifs and their respective numbers of repeats, regardless of the motif class, of 100 SSR markers, we found a higher number of di-nucleotide motifs with 70 to 80 repeats; we also found two di-nucleotide motifs with 83 and 89 repeats, respectively, but their product lengths were within optimum size (297 and 300 bp). In future work, we will screen for polymorphisms of possible primer pairs. The results will provide a useful tool for assessing molecular diversity and investigating the population structure among and within Chrysanthemum species.

Construction of a Large Synthetic Human scFv Library with Six Diversified CDRs and High Functional Diversity

  • Antibody phage display provides a powerful and efficient tool for the discovery and development of monoclonal antibodies for therapeutic and other applications. Antibody clones from synthetic libraries with optimized design features have several distinct advantages that include high stability, high levels of expression, and ease of downstream optimization and engineering. In this study, a fully synthetic human scFv library with six diversified CDRs was constructed by polymerase chain reaction assembly of overlapping oligonucleotides. In order to maximize the functional diversity of the library, a ${\beta}$-lactamase selection strategy was employed in which the assembled scFv gene repertoire was fused to the 5'-end of the ${\beta}$-lactamase gene, and in-frame scFv clones were enriched by carbenicillin selection. A final library with an estimated total diversity of $7.6{\times}10^9$, greater than 70% functional diversity, and diversification of all six CDRs was obtained after insertion of fully randomized CDR-H3 sequences into this proofread repertoire. The performance of the library was validated using a number of target antigens, against which multiple unique scFv sequences with dissociation constants in the nanomolar range were isolated.
