$\small{D}$-Phenylglycine aminotransferase ($\small{D}$-PhgAT) from Pseudomonas stutzeri ST-201 is useful for enzymatic synthesis of enantiomerically pure $\small{D}$-phenylglycine. However, its low protein solubility prevents its application at high substrate concentration. With an aim to increase the protein solubility, the N-terminus of $\small{D}$-PhgAT was genetically fused with short peptides ($A_1$${\alpha}$-helix, $A_2$${\alpha}$-helix, and ALAL, which is a hybrid of $A_1$ and $A_2$) from a ferredoxin enzyme of a halophilic archaeon, Halobacterium salinarum. The fused enzymes $A_1$-$\small{D}$-PhgAT, $A_2$-$\small{D}$-PhgAT, and ALAL-$\small{D}$-PhgAT displayed a reduced pI and increased in solubility by 6.1-, 5.3-, and 8.1- fold in TEMP (pH 7.6) storage, respectively, and 5-, 4.5-, and 5.9-fold in CAPSO (pH 9.5) reaction buffers, respectively, compared with the wild-type enzyme (WT-$\small{D}$-PhgAT). In addition, all the fused $\small{D}$-PhgAT displayed higher enzymatic reaction rates than the WT-DPhgAT at all concentrations of L-glutamate monosodium salt used. The highest rate, $23.82{\pm}1.47$ mM/h, was that obtained from having ALAL-$\small{D}$-PhgAT reacted with 1,500 mM of the substrate. Moreover, the halophilic fusion significantly increased the tolerance of $\small{D}$-PhgAT in the presence of NaCl and KCl, being slightly in favor of KCl, where under the same condition at 3.5 M NaCl or KCl all halophilic-fused variants showed higher activity than WT-$\small{D}$-PhgAT.

Recently, International Atomic Energy Agency and major leading countries in radioactive waste management tend to subdivide the categories of radioactive waste based upon risk-graded approach. In this context, the category of very low level waste has been newly introduced, or optimized management options for this kind of waste have been pursued in many countries. The application of engineered surface landfill type facilities dedicated to dispose of very low level waste has been gradually expanded, and it was analyzed that their design concept of isolation has been much advanced than those of the old fashioned surface trench-type disposal facilities for low and intermediate level waste, which were usually constructed in 1960's. In addition, the management options for very low level waste in major leading countries are varied depending upon and interfaced with the affecting factors such as: national framework for clearance, legal and practical availability of low and intermediate level waste repository and/or non-nuclear waste landfill, public acceptance toward alternative waste management options, and so forth. In this regard, it was concluded that optimized long-term management options for very low level waste in Korea should be also established in a timely manner through comprehensive review and discussions, in preparation of decommissioning of large nuclear facilities in the future, and be implemented in a systematic manner under the framework of national policy and management plan for radioactive waste management.

Biodiesel produced using microorganisms, which are recognized as the third-generation biomass, is among the various known renewable energy sources that can replace fossil fuels used in conventional transportation. Scenedesmus obliquus has been identified as an excellent species for biodiesel production, as it grows faster and can accumulate up to 40-50 percent of the dry cell weight. Enhancing production using S. obliquus requires measuring the cell mass for controlling the cultivation process. In the current study, S. obliquus was cultured for 75 days, and growth changes of the microalgae were measured by absorbance, microscopic imaging, and Raman spectroscopy. Between days 60 to 75 of culture, the change in absorbance was observed to be less than 3%, whereas the number of microalgae observed microscopically was more than three times higher. Moreover, the Raman spectroscopy results showed three strong peak values of β-carotene at 997 cm-1, 1148 cm-1, and 1515 cm-1, with peak values of β-carotene showing greater than 3-fold increase during the culture period. Therefore, we predict that application of Raman spectroscopy will help in identifying the growth elements and growth degree in microalgae culture during increased biomass production.

The NAUT (Non-contact Air coupled Ultrasonic Testing) technique is one of the ultrasonic testing methods that enables non-contact ultrasonic testing by compensating for the energy loss caused by the difference in acoustic impedance of air with an ultrasonic pulser receiver, PRE-AMP and high-sensitivity transducer. As the NAUT is performed in a state of steady ultrasonic transmission and reception, testing can be performed on materials of high or low temperatures or specimens with a rough surface or narrow part, which could not have been tested using the conventional contact-type testing technique. For this study, the internal defects of spot weld, which are often applied to auto parts, and CFRP parts, were tested to determine if it is practical to make the NAUT technique commercial. As the spot welded part had a high ultrasonic transmissivity, the result was shown as red. On the other hand, the part with an internal defect had a layer of air and low transmissivity, which was shown as blue. In addition, depending on the PRF (Pulse Repetition Frequency), an important factor that determines the measurement speed, the color sharpness showed differences. With the images obtained from CFRP specimens or an imaging device, it was possible to identify the shape, size and position of the internal defect within a short period of time. In this paper, it was confirmed in the above-described experiment that both internal defect detection and image processing of the defect could be possible using the NAUT technique. Moreover, it was possible to apply NAUT to the detection of internal defects in the spot welded parts or in CFRP parts, and commercialize its practical application to various fields.
As an ozone contactor, we newly adopted HJLR (High-performance Jet Loop Reactor) for the decolorization of Reactive black 5 and the mineralization of oxalic acid, which has been applied exclusively in biological wastewater treatments and well-known for high oxygen transfer characteristics. The ozonation efficiency for organic removals and ozone utilization depending on the mass transfer rate were compared to those of Stirred bubble column reactor, which was controlled by varing energy input in the HJLR and Stirred bubble column reactor. The results were as follows; first, the decolorization rate of Reactive black 5 in the HJLR reactor was nearly proportional to the increasing $k_La$. When the $k_La$ was increased by 25 % from $13.0hr^{-1}$ to $16.4hr^{-1}$, 30 % of the k' (apparent reaction rate constant) was increased from 0.1966 to $0.2665min^{-1}$ (Stirred bubble column; from 0.1790 to $0.2564min^{-1}$). Ozone transfer was found to be a rate-determining step in decolorizing Reactive black 5, which was supported by that no residual ozone was detected in all of the experiments. Second, the mineralization of oxalic acid was not always proportional to the increasing $k_La$ in the RJLR reactor. The rate-determining step for this reaction was OH(OH radical) production with ozone transfer, because residual ozone was always detected during the ozonation of oxalic acid in contrast with Reactive black 5. This result indicates that the increase of $k_La$ in the HJLR reactor is beneficial only when there are in ozone transfer limited regions. In addition, regardless of $k_La$, the mineralization of oxalic acid was nearly accomplished within 60 minutes. It was interpreted as that the longer staying of residual ozone by whirling liquid in the HJLR reactor contributed to an high ozone utilization(83-94%), producing more OR radicals.
Rising sea levels along the coast from global warming causes the increase of wave energy along the coast. This rise in sea levels results in relatively deep water levels, which would incur the loss of sand that had not occurred in the past from erosion in coastal areas. Generally, it has been challenging to protect against coastal erosion, and the slope, cross-sectional shape, and materials are selected for the site conditions depending on the change in external forces. However, the application of counter measures based on insufficient understanding of the phenomenon is causing various damage, indicating the need for technological development and converging technologies to improve credibility. In this study, we developed eco-friendly permeable biopolymer concrete blocks to control the coastal erosion by using the Bio-Coast, an effective porous structure that mitigates the destructive erosion caused by the rising sea levels. The hexagonal design of Bio-Coast was derived from the honeycomb, columnar joints, and clover, which are durable and stable structures in nature, and the design was changed to apply bumps on the Bio-Coast filling in the form of a clover to reduce wave overtopping and run-up. Applying the field condition of beaches on the east coast of Korea, the block weight and size were decided and the prototype blocks were manufactured and are ready for field placement. In particular, it is intended to protect coastal areas from destructive erosion by natural and artificial external forces, and to extend the design to river,s lakes, and natural walking trails, to improve the efficiency of quality control and process control through the use of blocks.
In reality, it is a green revolution of the entire agricultural matrix in Korea that integrated pest control plays an important role in the possible breakthrough in rice self-sufficiency. In paddy agroecosystem as man-modified environment, rice is newly established every year by transplantation under diverse water regimes which affect a microclimate. Standing water benefits rice by regulating the microclimate, but it favors the multiplication of certain pets through the amelioration of the microclimate. Further, the introduction of high yielding varieties with the changing of cultural practices results in changing occurrence pattern of certain pests. In general, japonica type varieties lack genes resistant to most of the important pests and insect-borne virus diseases, whereas indica type possesses more genes conferring varietal resistance. Thus, this differences among indica type, form the background of different approaches to pest management. The changes in rice cultivation such as double cropping, growing high-yielding varieties requiring heavy fertilization, earlier transplanting, intensvie-spacing transplanting, and intensive pesticide use as a consequence of the adoption of improves rice production technology, have intensified the pest problems rather than reduced them. The cultivation of resistant varieties are highly effective to the pest, their long term stability is threathened because of the development of new biotypes which can detroy these varieties. So far, three biotypes of N. lugens are reported in Korea. Since each resistant variety is expected to maintain several years the sequential release of another new variety with a different gene at intervals is practised as a gene rotation program. Another approach, breeding multilines that have more than two genes for resistance in a variety are successfully demonstrated. The average annual rice losses during the last 15 years of 1977-’91 are 9.3% due to insect pests without chemical control undertaken, wehreas there is a average 2.4% despite farmers’insecticide application at the same period. In other words, the average annual losses are prvented by 6.9% when chemical control is properly employed. However, the continuous use of a same group of insecticides is followed by the development of pest resistance. Resistant development of C. suppressalis, L. striatellus and N. cincticeps is observed to organophosphorous insecticides by the mid-1960s, and to carbamates by the early 1970s in various parts of the country. Thus, it is apparent that a scheduled chemical control for rice production systems becomes uneconomical and that a reduction in energy input without impairing the rice yield, is necessarily improved through the implementation of integrated pest management systems. Nationwide pest forecasting system conducted by the government organization is a unique network of investigation for purpose of making pest control timely in terms of economic thresholds. A wise plant protection is expected to establish pest management systems in appropriate integration of resistant varieties, biological agents, cultural practices and other measures in harmony with minimizing use of chemical applications as a last weapon relying on economic thresholds.
Drying process is very important for commercialization of velvet antlers. So far, artificial drying has mostly depended on experience of farmers resulting in low efficiency of drying, deterioration during drying, and contamination by dust and bacteria. Various drying techniques for high-quality production have currently been commercialized in some developed countries, and one of them is a drying technique using microwave. In this study, application of the technique using an electronic (microwave) oven for drying of velvet antler was investigated. The results of this study are as follows. 1. It was found that the drying period was significantly affected by cornification of velvet antlers. Accordingly, it can be assumed that the velvet antlers should partly be sliced to save drying period and required drying energy. 2. It was also found that the drying speed was changeable according to tempering period and even with same drying period. The tempering period should be controlled according to processing rate. 3. The results indicated that a repeated drying of 2-min drying and 3-min tempering is the most effective when a microwave oven was used. 4. The results also showed that the drying technique using microwave of electronic oven can be effectively used for drying of velvet antlers. 5. Only drying and tempering periods were examined in this study, however, ingredient change of deer antler after drying is necessary to be investigated in the future.

Many machineries and equipments are being changing to various and complicated by development of recent technology and arrival of convergence age in distant future. These various and complicate equipments need more precise outcomes and low-power consumption sensors to get close and exact results. In this paper, we proposed fault tolerance and feedback theorem for sensor network and MEMS circuit which has a benefit of energy efficiency through wireless sensor network. The system is provided with independent sensor communication if possible as unused action, using idle condition of system and is proposed the least number of circuits. These technologies compared system efficiency after examining product of each Moving Distance by developed sensor which gives effects to execution of system witch is reduced things like control of management side and requirement for hardware, time, and interaction problems. This system is designed for practical application; however, it can be applied to a normal life and production environment such as "Ubiquitous City", "Factory Automata ion Process", and "Real-time Operating System", etc.
The transient and rapid expression system of a foreign protein in planta is a very useful technique in biotechnology application. We have investigated optimum condition of Agrobacterium-infiltration technique in which expression level of foreign proteins were maximized without detrimental effects on plants using GFP and Cucumber mosaic virus 2b protein, which is known as an enhancer of gene expression and a suppressor of post-transcriptional gene silencing(PTGS). The optimum expression level of both RNA and protein of GFP with minimum leaf impairment was obtained at $OD_{600}$=0.2 of Agrobactrium inocula. The steady-state levels of GFP RNA and protein generally peaked at 3 and 7 days post-infiltration(dpi), respectively. In the presence of 2b, both the magnitude and duration of GFP expression was highly increased and we could detect GFP level until 17 dpi. On the other hands, the 2b-mediated higher accumulation of foreign proteins resulted in the repression of normal leaf growth, possibly due to the limitation of supply of energy or materials required for growth maintenance. Using this Agrobacterium-infiltration system with 2b and GFP, we tested a hypothesis for the threshold model of PTGS initiation. Four GFP transgenic lines of N. benthamiana, which shows different expression level of GFP were tested to determine the threshold level for PTGS initiation. Agrobacterium-infiltration of GFP into those GFP-transgenic plants resulted in the co-silencing of the transgenic GFP. It was found that very low concentration of Agrobacterium with GFP and GFP+2b($OD_{600}$=0.002-0.02) which could not phenotypically induce an additive GFP expression, was enough to trigger PTGS pathway in all GFP transgenic plants. This strongly indicates that each GFP-transgenic plant should be expressing the transgenic GFP at its own pre-determined level and there was no buffer zone of additive GFP-expression to the threshold. In other words, the PTGS seems to be immediately activated as a self-defensive mechanism if an internal balance of gene expression is broken.
