Evaluation of Ventilation Systems in an Enclosed Nursery Pig House (무창자돈사의 환기시스템 정립 및 환기효율 평가)

  • An experiment was conducted to establish the most suitable ventilation system for the enclosed nursery pig house in Korea, comparing four different ventilation systems ; i) air enters through perforated ceiling and exhausts through chimney (NA), ii) air enters through perforated ceiling and exhausts through side walls (NB), iii) air enters through perforated ducts and exhausts through side walls (NC) and iv) air enters through perforated ducts and exhausts through chimney(ND). The experiment was carried out during winter and summer separately. The experimental pigs were weaned at fourteen days old in winter (December-February) and at twenty one days old in summer (June-August). The main results of the experiment are as follows : A preliminary experiment showed that in the NC system during summer, air can reach all the pig rooms in the house and the air flow rates of the upper, middle (1.2 m height of the room) and low (at the height of pig stature) parts of the room were measured at 7.0-8.08, over 0.5 and over 0.2 m/s, respectively, which flow rates were much higher(p$<$0.05) than those in other system. At the minimum ventilation efficiency during winter, air flow rates of upper, middle and low parts of the room equipped with the NC system were detected at over 1, less than 0.5 and around 0.07 m/s, respectively. It is concluded that the separated ventilation system air-entering through ducts is the most suitable for the ventilation system of the enclosed nursery pig house and the exhausting system through side walls is more efficient for ventilation than the system through roof. Furthermore, to sustain proper temperature and reduce energy waste as well as heat consumption, a future research should be carried out to develop the environmental control system in relation to developing a heat regulator.

Analysis of Wind Vorticity and Divergence in the High-latitude Lower Thermosphere: Dependence on the Interplanetary Magnetic Field (IMF) (고위도 하부 열권 바람의 소용돌이도와 발산 분석: 행성간 자기장(IMF)에 대한 의존도)

  • To better understand the physical processes that control the high-latitude lower thermospheric dynamics, we analyze the divergence and vorticity of the high-latitude neutral wind field in the lower thermosphere during the southern summertime for different IMF conditions. For this study the National Center for Atmospheric Research Thermosphere-Ionosphere Electrodynamics General Circulation Model (NCAR-TIEG CM) is used. The analysis of the large-scale vorticity and divergence provides basic understanding flow configurations to help elucidate the momentum sources that ulti-mately determine the total wind field in the lower polar thermosphere and provides insight into the relative strengths of the different sources of momentum responsible for driving winds. The mean neutral wind pattern in the high-latitude lower thermosphere is dominated by rotational flow, imparted primarily through the ion drag force, rather than by divergent flow, imparted primarily through Joule and solar heating. The difference vorticity, obtained by subtracting values with zero IMF from those with non-zero IMF, in the high-latitude lower thermosphere is much larger than the difference divergence for all IMF conditions, indicating that a larger response of the thermospheric wind system to enhancement in the momentum input generating the rotational motion with elevated IMF than the corresponding energy input generating the divergent motion. the difference vorticity in the high-latitude lower thermosphere depends on the direction of the IMF. The difference vorticity for negative and positive $B_y$ shows positive and negative, respectively, at higher magnetic latitudes than $-70^{\circ}$. For negative $B_z$, the difference vorticities have positive in the dusk sector and negative in the dawn sector. The difference vorticities for positive $B_z$ have opposite sign. Negative IMF $B_z$ has a stronger effect on the vorticity than does positive $B_z$.

Growth kinetics and chlorine resistance of heterotrophic bacteria isolated from young biofilms formed on a model drinking water distribution system (모델 상수관망에 형성된 초기 생물막에서 분리한 종속영양세균의 생장 동역학 및 염소 내성)

  • The present work quantified the growth of young biofilm in a model distribution system that was fed with chlorinated drinking water at a hydraulic retention time of 2 h. Bacterial biofilms grew on the surface of polyvinyl chloride (PVC) slides at a specific growth rate of $0.14{\pm}0.09day^{-1}$ for total bacteria and $0.16{\pm}0.08day^{-1}$ for heterotrophic bacteria, reaching $3.1{\times}10^4cells/cm^2$ and $6.6{\times}10^3CFU/cm^2$ after 10 days, respectively. The specific growth rates of biofilm-forming bacteria were found to be much higher than those of bulk-phase bacteria, suggesting that biofilm bacteria account for a major part of the bacterial production in this model system. Biofilm isolates exhibited characteristic kinetic properties, as determined by ${\mu}_{max}$ and $K_S$ values using the Monod model, in a defined growth medium containing various amounts of acetate. The lowest ${\mu}_{max}$ value was observed in bacterial species belonging to the genus Methylobacterium, and their slow growth seemed to confer high resistance to chlorine treatment (0.5 mg/L for 10 min). $K_S$ values (inversely related to substrate affinity) of Sphingomonas were two orders of magnitude lower for acetate carbon than those of other isolates. The Sphingomonas isolates may have obligate-oligotrophic characteristics, since the lower $K_S$ values allow them to thrive under nutrient-deficient conditions. These results provide a better understanding and control of multi-species bacterial biofilms that develop within days in a drinking water distribution system.

Influence of Daytime Temperature on the Time Required for Fruit Harvest and Yield of Hot Pepper (주간온도가 고추의 수확 소요일수 및 수량에 미치는 영향)

  • Due to climate change, the occurrence of abnormal weather conditions has become more frequent, causing damage to vegetable crops grown in Korea. Hot pepper, Chinese cabbage and radish, the three most popular vegetables in Korea, are produced more in the field than in the greenhouse. It has been a trend that the time for field transplanting of seedlings is getting earlier and earlier as the spring temperatures keep rising. Seedlings transplanted too early in the spring take a longer time to resume the normal growth, because they are exposed to suboptimal temperature conditions. This study examined the influence of air temperature during seedling growth on the time required to reach the first fruit maturity and yield of hot pepper. Seedling plants of 'Super Manita' hot pepper was grown in temperatures $2.5^{\circ}C$ and $5.0^{\circ}C$ lower than the optimum temperature (determined by the average of temperatures for the past 5 years). Seedlings were transplanted into round plastic containers (30-cm diam., 45-cm height) and were placed in growth chambers in which the ambient temperature was controlled under natural sunlight. The growth of seedlings under lowered temperatures was reduced compared to the control. The mineral (K, Mg, P, N) concentrations in the leaf tissues were higher when plants were grown with the ambient temperature $2.5^{\circ}C$ lower than the optimum, regardless of changes in other growth parameters. Tissue calcium (Ca) concentration was the highest in the plants grown with optimum temperature. The carbohydrate to nitrogen (C/N) ratio, which was the highest (18.3) in the plants grown with optimum temperature, decreased concomitantly as the ambient temperature was lowered $2.5^{\circ}C$ and $5.0^{\circ}C$. The yield of the early harvested fruits was also reduced as the ambient temperature became lower. The first fruit harvest date for the plants grown with optimum temperature (June 27) was 13 days and 40 days, respectively, earlier than that in plants grown with $2.5^{\circ}C$ (July 10) and $5.0^{\circ}C$ (Aug 6) lower ambient temperatures. The fruit yield per plant for the optimum temperature (724 g) was the greatest compared to those grown with $2.5^{\circ}C$ (446 g) and $5.0^{\circ}C$ (236 g) lower temperatures. The result of this study suggests that the growers should be cautioned not to transplant their hot pepper seedlings too early into the field, since it may delay the time of first harvest eventually reducing total fruit yield.

Clinical Significance of the Ghrelin Concentrations in Plasma and Tumor Tissue from Patients with Gastric Cancer (위암 환자의 혈장 및 종양 조직에서 측정된 그렐린 농도의 임상적 의의)

  • Purpose: Ghrelin, produced primarily in the gastrointestinal tract, including the stomach, has been reported to reflect nutritional status and to control homeostasis by influencing food intake and adiposity. The purpose of this study is to evaluate nutritional status, as well as plasma and gastric tissue ghrelin levels, in patients with gastric cancer who underwent a gastrectomy. Materials and Methods: Eighty patients were analyzed by the degree of weight loss $(weight\;loss{\geq}5%\;or\;<5%)$ and the extent of gastrectomy (subtotal or total gastrectomy). Blood samples were collected from all patients preoperatively and postoperatively especially at seven days. Gastric tissues, including tumor and normal tissues, were obtained from the resected stomach. levels of plasma and tissue ghrelin were measured with a commercial ELISA kit. Results: There were no significant differences in the clinical characteristics and ghrelin levels of plasma, gastric tumor tissue and normal tissue by the degree of weight loss. The ghrelin levels in plasma and tumor tissue showed no correlations with each other while the ghrelin level in tumor tissue was significantly lower than that in normal tissue. The degree of cellular differentiation also had an association with ghrelin production. A gastrectomy proved to decrease significantly plasma ghrelin levels, body mass index, and biochemical markers, regardless of the extent of gastric resection. Conclusion: These results show that gastric cancer affects the production of ghrelin in the gastric mucosa and that ghrelin is mainly produced in stomach even though it could be partially covered by endogenous ghrelin from other organs following a gastrectomy. However, we should further investigate which other factors have an impact on energy consumption, ghrelin secretion, and changes in ghrelin levels after a gastrectomy.

Current status of sweetpotato genomics research (고구마 유전체 연구현황 및 전망)

  • Sweetpotato [Ipomoea batatas (L.) Lam] grows well in harsh environmental conditions, and is cultivated as one of the top seven food crops in the world. Recently, sweetpotato is drawing interest from people as a healthy food because it is high in dietary fiber, vitamins, carotenoids and overall nutrition value. However, few studies have been conducted on sweetpotato genome sequencing in spite of its importance. This review is aimed at increasing the efficiency of sweetpotato genome sequencing research as well as establishing a base for gene utilization in order to control useful traits. Recently, animal and plant genome sequencing projects increased significantly. However, sweetpotato genome sequencing has not been performed due to polyploidy and heterogeneity problems in its genome. Meanwhile research on its transcriptome has been conducted actively. Recently, a draft of the diploid sweetpotato genome was reported in 2015 by Japanese researchers. In addition, the Korea-China-Japan Trilateral Research Association of Sweetpotato (TRAS) has conducted research on gene map construction and genome sequencing of the hexaploid sweetpotato Xushu 18 since 2014. The Bill & Melinda Gates Foundation launched the 'sweetpotato genomic sequencing to develop genomic tools for Sub-Sahara Africa breeding program'. The chloroplast genome sequence acquired during sweetpotato genome sequencing is used in evolutionary analyses. In this review, the trend of research in the sweetpotato genome sequencing was analyzed. Research trend analysis like this will provide researchers working toward sweetpotato productivity and nutrient improvement with information on the status of sweetpotato genome research. This will contribute to solving world food, energy and environmental problems.

A Study on Serum Leptin Values by Elisa Method in Children (ELISA법으로 측정한 소아 혈중 LEPTIN 치에 관한 연구)

  • Purpose: Leptin is an adipocyte-derived blood-borne satiety factor that acts on its cognate leptin receptor in the hypothalamus, thereby regulating food intake and energy expenditure. We measured the leptin concentrations in serum of normal and obese children with human leptin ELISA kit, unlike previous study with leptin RIA kit and investigated the relationship between leptin concentrations and body mass index, gender, and age. Methods: We measured serum concentrations of leptin in 67 children who were visited to the Department of Pediatrics, Chungnam National University Hospital during the period of 5 months from February, 1999 to June, 1999. Height, weight, obesity index, and body mass index were measured in 67 subjects. Leptin values in serum were measured by sandwich ELISA method. Data analysis was done according to the obesity, body mass index, gender and age. Results: The mean concentration of leptin was $7.69{\pm}8.83\;ng/ml$ in normal children group and $36.34{\pm}18.57\;ng/ml$ in obese group. Serum leptin concentrations were significant correlation with the body mass index (p<0.01). Serum leptin concentration was significant higher in the group of over 10 years of age (p<0.01). Leptin levels showed no significant difference by gender. Conclusion: Serum leptin levels were significantly higher in obesity group than in control one, and they were correlated with body mass index and age. Measurements of leptin value by sandwich ELISA method are very useful and easily applicable to determine obesity.

Physicochemical Properties of Jadeite Powder and Its Application to Cosmetic Formulations (경옥가루의 물리화학적 특성 및 화장품 제형 응용 연구)

  • In this study, physicochemical properties of the natural jadeite powder were investigated and also the wash-off pack and liquid foundation containing the jadeite powder were prepared. In addition, each of these formulations was evaluated by various functional effects, sensory evaluation, stability and skin safety. In the wash-off pack, the far-infrared ray emissivity and radiation energy values increased as a function of the jadeite powder content. At a 3% jadeite powder content, the skin temperature increased by about $0.5^{\circ}C$ when the jadeite powder-containing formulation was applied to the skin. Besides, the chroma of the liquid foundation containing the jadeite powder more clearly expressed the original color of the skin. Moisture content measurements of the wash-off pack and liquid foundation containing the jadeite powder showed the highest moisture uptake of 5.0% and 63.0%, respectively. In sensorial test, the wash-off pack formulations containing the jadeite powder demonstrated improved affinities toward a skin, adherency, and moistness and combatted itching. The liquid foundation containing jadeite powder showed also improved affinities except for the coverage when compared to control formulations. Furthermore, the stability evaluation for 8 weeks revealed neither discoloration nor separation phenomenon for the formulations containing the jadeite powder. Moreover, the pH was found to be stable up to 8 weeks and the viscosity up to 4 weeks. Skin safety assessments showed that all formulations containing the jadeite powder were non-irritating. These results suggest that the jadeite powder as an inorganic pigment may serve as a new multi-functional cosmetic ingredient with stability and safety.

Study on the Whitening Efficacy and Skin Barrier by Lysosome-related Organelle Extract (LOE) from Egg White (난백(Egg White)에서 추출한 리소좀 추출물(LOE)의 미백 효능 및 피부장벽에 관한 연구)

  • Lysosomes are cellular organelles involved in energy metabolism and intracellular digestion in eukaryotic cells, including protease, nuclease, glycosidase, lipase, and phosphatase. Our previous studies have confirmed that egg white lysosomes had melanin decolorization and reduction activity. However, there have been few studies on skin barrier and skin regeneration as well as inhibition of melanin production by egg white lysosomes on B16F10 melanocyte cell line. In this study, we attempted to identify the effect of lysosome-related organelle extract (LOE) extracted from egg white on the melanin content change and skin barrier enhancement in cells. First, cytotoxicity evaluation was performed on B16F10 melanocyte cell line to confirm the whitening efficacy of LOE. Cytotoxicity by LOE was not observed at 20 mg/mL concentration, but cytotoxicity was observed at 40 mg/mL, and the maximum concentration value was set to 20 mg/mL in all subsequent experiments. LOE samples of 5, 10, 20 mg/mL inhibited melanin production by 61.5 ± 4.0%, 61.4 ± 7.3%, 58.3 ± 8.3%, respectivly, compared to α-MSH, a negative control in melanin contents assay. MITF mRNA expression was reduced by about 39.7 ± 3.2% compared to the α-MSH treatment group. TEER assay using HaCaT showed that LOE increased TEER resistance in a dose-dependent manner, indicating that LOE is involved in strengthening the skin barrier. LOE also increased the TEER resistance under TNF-α treatment. Skin barrier was normally restored by LOE even under the condition of inflammation. LOE had a positive effect on cell division and cell migration promotion, confirmed by the observing the effect of promoting cell migration by LOE through cell migration assay. Taken together, we expect that LOE can be developed as a cosmetic material to enhance has effects on skin regeneration and skin barrier strengthening as well as whitening function if enzyme stabilization and formulation technology are combined.

Hydrogeochemical Evolution Related to High Fluoride Concentrations in Deep Bedrock Groundwaters, Korea (국내 심부 암반지하수에서의 고농도 불소 산출과 관련된 수리지구화학 진화)

  • To understand the geologic and hydrogeochemical controls on the occurrence of high fluoride concentrations in bedrock groundwaters of South Korea, we examined a total of 367 hydrochemistry data obtained from deep groundwater wells (avg. depth=600 m) that were drilled fur exploitation of hot springs. The fluoride concentrations were generally very high (avg. 5.65mg/L) and exceeded the Drinking Water Standard (1.5 mg/L) in $72\%$ of the samples. A significant geologic control of fluoride concentrations was observed: the highest concentrations occur in the areas of granitoids and granitic gneiss, while the lowest concentrations in the areas of volcanic and sedimentary rocks. In relation to the hydrochemical facies, alkaline $Na-HCO_3$ type waters had remarkably higher F concentrations than circum-neutral to slightly alkaline $Ca-HCO_3$ type waters. The prolonged water-rock interaction occurring during the deep circulation of groundwater in the areas of granitoids and granitic gneiss is considered most important for the generation of high F concentrations. Under such condition, fluoride-rich groundwaters are likely formed through hydrogeochemical processes consisting of the removal of Ca from groundwater via calcite precipitation and/or cation exchange and the successive dissolution of plagioclase and F-bearing hydroxyl minerals (esp. biotite). Thus, groundwaters with high pH and very high Na/Ca ratio within granitoids and granitic gneiss are likely most vulnerable to the water supply problem related to enriched fluorine.
