The purpose of this research presents a method to position and makes the structure for eco-corridors reasonably with collectable analysing results of various effects shown in mammals' road-kill at 429 points. Target animals of this research are Leopard cat, Siberian weasel, Raccoon dog, Korean hare, Eurasian red squirrel, Siberian chipmunk and Water deer. The results derived from the empirical analysis on the contents above are followed. First, according to the results as for Leopard cat road kill analysis, which is designated as Endangered Species Class II, the eco-corridor might be located at near village having stead food in order to decrease the frequencies of road-kill, because its road kill points were mainly collected at 4 lane hilly road with mountain-road-farm area geological type of. Second, because Siberian weasel's road kill was detected at 2 lane hilly road with mountain-road-stream geological type, the eco-corridor might be located at near a mill to decrease road-kill frequencies. Third, the road-kill frequency of Eurasian red squirrel can be reduced when the eco-corridor is located at the area across coniferous tree near 4 lane west sea freeway with mountain-road-mountain. Fourth, the road-kill of Raccoon dog can be reduced when the eco-corridor is located at 4 lane mountain road or hilly road with the geological type having farm land-road-mountain(stream). Fifth, Korean hare's road-kill can be reduced when the eco-corridor is located at grass land across ridge line of mountain, because wild rabbit road kill was happened at 4 lane mountain road or 2 lane mountain road(mountain-road-mountain). Sixth, As for Siberian chipmunk, the eco-corridor might be located at the side slope of mountain road at 2 lane mountain road under the speed of 60km/h with mountain-road-mountain. Seventh, For Water deer, the eco-corridor might be located at 4 lane hilly road with mountain-road-farm land. As for Common otter, Amur hedgehog, Yellow-throated marten, Weasel, it is difficult to specify the proper site of eco-corridor due to the lack of data. Eco-corridors for carnivores might be well located at 4 lane hilly road or 2 lane hilly road with mountain-road-farm land, and the track for herbivores might be well located as a overhead bridge on mountain-road-mountain type across mountains. In order to position eco-corridors for wildlife properly, we have to research animal's behavior with ecological background, and to consider the local uniqueness and regularly collect the empirical road-kill data in long term 3 to 5 year, which can be the foundation for the more suitable place of wild life eco-corridors.
The cloning of canids was succeeded in 2005, several years after the birth of Dolly the sheep and also after the cloning of numerous other laboratory and farm animal species. The delay of successful somatic cell nuclear transfer (SCNT)was due to the unique reproductive characteristics of the female dogin comparison to other domestic mammals, such as ovulation of immature canine oocyte and a requirement of 25 days for the completion of meiosis within the oviduct (Holst & Phemister, 1971). When the technology for the recovery of in vivo matured oocyte was established, the application of cloning also became possible and cloned dog offspring were obtained. This report summarizes the progress of technical procedures that are required for cloning canids and the application of this technique. The first cloned dog, Snuppy, was achieved using an in vivo-matured oocyte which was enucleated and transferred with an adult skin cell of male Afghan hound. After establishment of a criterion of well-matured oocyte for the improvement of SCNT efficiency, we obtained three cloned female Afghan hound and a toy poodle cloned from 14 year-old aged Poodle using SCNT through this factor. To date, cloned dogs appeared to be normal and those that have reached puberty have been confirmed to be fertile. Through application of canine SCNT technique, first, we demonstrated that SNCT is useful for conserving the breed of endangered animal from extinction through cloning of endangered gray wolves using inter-species SCNT and keeping the pure pedigree through the cloning of Sapsaree, a Korean natural monument. Secondly, we showed possibility of human disease model cloned dog and transgenic cloned dog production through cloning of red fluorescent protein expressing dog. Finally, SCNT can be used for the propagation of valuable genotypes for making elite seed stock and pet dog. In summary, dog cloning is a reproducible technique that offers the opportunity to preserve valuable genetics and a potential step towards the production of gene targeted transgenic cloned dogs for the study of human diseases.
The techniques of IVM, IVF and IVC of canine oocytes may provide useful information for gamete salvage programs and the conservation of endangered canidae. This investigation has been made to determine the efficiency of in vitro maturation (IVM) as a basic experiment to study the development of canine oocytes after in vitro fertilization (IVF). The rate of oocytes developing to the Mil stage was higher in the hormone treated group (10 IU/ml hCG+eCG, 14.7%, p<0.05) than in the control group (0 IU/ml hCG+eCG, 10.0%). The monospermy and pronuclear rates of canine oocytes were investigated after caffeine treatment on IVF. Canine oocytes were fertilized in the Fert-TALP medium supplemented with 0, 10, 20 or 30 mM caffeine (Fert I, Fert II, Fert III or Fert IV, respectively). The highest pronuclear formation rate was obtained in the Fert I for 24 h IVF (6.7%, 6/89). Therefore, it is believed that unlike in other mammals, caffeine in canine IVF does not increase the efficiency of fertilization rate, and is not an important factor.

Efficient management of road-kill data is difficult at national scale when there are many organizations that are in charge of different road types. Here, we described the first case to integrate road-kill data through Korea Road-kill Observation System (KROS) by the Korean government. The system was launched in June 2018 to approximately 3,000 road menders. During 15 months, 5,812 road-kill observations were registered on KROS including mammals, birds, reptiles and amphibians. Of them, about 86 % was occupied by five species (Hydropotes inermis, Felis catus, Capreolus pygargus, Nyctereutes procyonoides, and Canis lupus familiaris) listed in number of occurrences. The observed road-kill frequency rapidly increased until April 2019 and peaked on May 2019. However, as the system is just starting, the results from KROS cannot be treated as the exact representation of road-kill trend in the country. Although the efficient method to manage national road-kill statistic is arranged, still there are some limitations to overcome to make the system stable.
National parks are designated for the purpose of maintenance, conservation and utilization of different habitats. Therefore, it is necessary to select habitats of a high value as a protected area in order to balance conservation and development. However, the existing national park boundary adjustment and new designation criteria only focused on the endangered species and protected area, without proper evaluation of the habitat value of actual species. Therefore, this study aims to quantitatively evaluate habitat function in terms of biodiversity and habitat value, so that it can be referred to for the designation and boundary adjustment of national parks. We assessed species diversity and habitat values for each of the habitat types, for mammals only, as they are able to choose preferred habitats. In order to evaluate biodiversity, we used Maxent to derive species richness map and used InVEST's Habitat quality model to evaluate habitat value. As a result of evaluation, species richness was high in the national park boundary area. Also, even if the same edge is adjacent to the development area depending on the land cover, the species richness is low. Compared with Wolaksan and Sobaeksan National Park, the species richness and habitat value of the northern area, which is connected with other forests, were higher than those of the southern area where roads were developed. Therefore, it is expected that the use of the result of this study for the national park boundary adjustment and management will enhance the function of the national park as a habitat.
Background: Research on the reproductive physiology of Water and Sika deer, an endemic in Korea, still needs to be completed. This study analyzed the ovarian development and morphological characteristics of wild Water deer and Sika deer. Methods: Water deer and Sika deer ovaries were collected from the Korean Peninsula and Russia-Korean Peninsula border during the estrus and pregnancy seasons, respectively. And, morphological and physiological analysis and immunohistochemistry were conducted to confirm the detection of Ca2+ and assess the morphological changes in the ovaries. Results: The results of morphological analysis of ovaries during pregnancy and estrus, the development of the corpus luteum and follicles of Water deer showed similar patterns to other mammals. In contrast, the corpus luteum of Sika deer differed in tissue morphology and composition from Water deer. Ca2+ related to tissue metabolism was detected in the theca cells zone of Water deer on the estrus and was highly detected in the luteum cells zone during pregnancy. The hormone receptor protein expression patterns were generally higher in the ovaries of Water deer on the estrus and the pregnancy than in Sika deer. The expression of LH receptor was relatively low in the lutein cell zone, unlikely that of Water deer. The expression of VEGF was also different from Water deer, and the response in Sika deer was relatively very low compared to Water deer in expressing all proteins-related development. Conclusions: Therefore, the results of the study were shown that the composition of the corpus luteum of Sika deer is not clear compared to Water deer, and there are many differences in the functional and morphological formation of the corpus luteum.
삼천포천은 제방을 따라 제내지에 인공구조물이 설치되어 있으며, 하상구조가 인간활동으로 교란되어 있다. 본 연구결과 부착조류와 저서성 무척추동물의 생물다양성이 여타 하천에 비해 낮은 것으로 나타났다. 따라서 생태하천 복원 시 제방의 제외지와 제내지를 동시에 친환경적으로 복원을 고려함으로써 생물다양성 증대를 도모하고 하상 복원 등을 통해 회유성 어류들이 하천생태계로 유입되어 생태계의 안정성 및 건강성을 회복해야 할 필요가 있다(Palmer et al., 2014). 이를 위해서는 생태하천 복원시 사용되는 재료는 무생물적 재료를 지양하고 과거부터 주로 서식하는 자생식물 등을 주재료로 사용하고 하도 구조는 하상의 현재 유로형상 (flow channel pattern)을 유지하도록 유도해야 한다. 유로 연장이 필요한 경우 직강화 등에 의해 축소되어서는 안되며 축소된 유로는 복원을 권장한다. 분절된 지류는 본류와 연결시키고 연력이 곤란한 웅덩이는 대상하천에 맞게 소택형 습지를 형성하도록 유도한다. 또한 생태통로(Eco-corridor) 확보 및 추이대 조성을 통해 생태환경 기능을 개선할 필요가 있으며 제내지의 폐천 부지는 하천구역으로 편입하여 생태습지 등으로 이용을 적극 고려해야 한다(Choi and Lee, 2019). 생태적 복원을 위해 주차장, 체육시설 및 공원 등은 배제해 자연성을 유지하되 주민의 활용도를 고려하는 것도 중요하다. 이를 통하여 삼천포천의 자연성 회복을 통한 생물다양성 증진 및 주민에게 다양한 문화 휴식 공간 제공 등의 생태계 서비스 능력을 향상을 기대한다(Bennett et al., 2009).
본 연구에서는 습지보호지역 보전계획을 달성할 수 있도록 습지보호지역의 생태계 보전관리 도구로서 시민과학연구 방법론을 제시하고 2022년부터 주민역량강화 사업이 추진되고 있는 습지보호지역 3개소(광주광역시 장록, 경남 고성 마동호, 고창 인천강하구 습지보호지역)를 대상으로 적용성을 검토하였다. 본 연구에서 제안한 시민과학연구 방법은 습지보호지역 보전계획에서 제시하고 있는 보전·이용·관리 목표에 근거하여 습지보호지역의 이해당사자와 그들의 주요 관심사 및 정보수요 파악 후, 각 주체별 정보 수요를 충족할 수 있는 성과물을 도출할 수 있는 연구 활동을 수행하고 시민과학자를 포함한 이해당사자에게 연구 성과를 환류 및 확산하는 절차이다. 본 연구에서는 해당 방법론을 적용하여 3개 습지보호지역에서 생태계 모니터링(생태계교란 식물 등 식물 모니터링, 육상곤충 및 포유류 흔적 조사, 신규 습지 발굴)을 실시하고 식물 16종, 육상곤충 43종, 멸종위기야생생물 I급 수달, II급 삵을 포함한 포유류 5종의 서식현황 정보를 취득하였다. 시민과학자들이 수집한 모니터링 정보를 활용하여 생태계교란 식물 분포지도 제작 후 소관 환경청 및 지자체에 관리 근거자료로 제공하여 시민과학연구의 정책 활용성을 제고하였다. 향후 본 연구에서 제안한 시민과학연구 방법론이 습지보호지역의 보전관리 도구로 정착하기 위해서는 시민과학연구 자료 축적, 정책수요에 부합하는 연구 성과의 도출과 함께 성과물의 정책 활용성과 사회·경제적 파급효과에 대한 검토가 면밀히 이루어져야 할 것이다.
도토리거위벌레는 큰턱을 이용하여 도토리에 구멍을 뚫고 가지를 자르는 독특한 행동을 한다. 이러한 행동을 이해하기 위해 다양한 현미경을 이용하여 구기의 해부학적 특징을 관찰하였다. 실체현미경을 이용한 연구에서는 도토리거위벌레 수컷의 평균 내전건과 외전건의 두께는 각각 32.87 ㎛, 53.83 ㎛이었고, 암컷의 평균 내전건과 외전건의 두께는 각각 40.81 ㎛, 75.55 ㎛이었다. 내전건과 외전건의 유의한 두께 차이로 인해 큰턱을 닫는 힘보다 벌리는 힘이 더 강하다는 것을 유추할 수 있다. 광학현미경을 이용한 관찰에서는 주둥이 안쪽에 위치한 내전건과 외전건의 너비 차이와 큰턱 쪽에서는 원형으로 위치하며 두부로 갈수록 납작해져 두부 외피 안쪽에 근육으로 연결된다는 것을 확인할 수 있었고, 내전건의 위치와 모양의 변화에 대한 관찰을 수행하였다. 전자현미경을 통한 연구에서는 외전건이 내전건보다 더 굵으며 이로 인해 큰턱을 안쪽으로 당기는 힘보다 바깥쪽으로 밀어내는 힘이 더 크다는 것을 확인하였다. 관찰 결과를 바탕으로, 도토리거위벌레, 밤바구미, 왕거위벌레의 큰턱의 형태와 이빨의 위치 그리고 큰턱의 움직임에 대해 비교 분석하였고, 도토리거위벌레가 보여주는 특징적인 행동은 해부학적/형태적 특징과 연관되어 있음을 확인할 수 있었다.
본 연구는 탐진강 하구역 생물군집의 군집 동태 및 분포 조건을 통해 기수지역 생태계 관리의 기초정보를 제공하기 위해 각 분류군별 전문가들이 정밀 조사한 결과를 분석하였다. 식생을 포함한 총 11개 생물 분류군에 대한 생물종의 다양성, 풍부성, 상관관계 및 생태적 특성을 분석하였다. 분석 결과 식생은 하천의 물리적 환경과 염도 등 서식처 환경조건에 따라 7개 상관형에 18개 단위식생의 분포가 확인 되었다. 생물종 다양성은 멸종위기야생생물 9분류군을 포함해 총 1,125개 분류군이 확인되었다. 각 분류군별 종조성과 분포 특성은 넓은 구간에 걸쳐 형성되는 자연성 높은 기수역의 하구역 특성을 잘 반영하고 있었다. 특히, 하천을 중심으로 유속과 염도, 토성 등 기반환경 차이에 따른 종다양성과 그 분포가 뚜렷하게 구분되었다. 일부구간에 분포하는 물막이보, 인공제방과 같은 인위적 교란요소에도 불구하고 현재의 환경조건 하에서 생물종의 다양성과 그 분포는 전반적으로 자연성 높은 것으로 평가되었다. 그러나 물막이보, 간척, 주기적인 제방관리, 경작지 확장 등에 의한 야생생물의 서식처 소실과 축소, 생태계 연결성 단절 등은 지역 생태계의 다양성과 건강성을 위협하는 주요 요인으로 지목되었다. 탐진강 하구역의 생물 분포 현황과 그 특성을 규명하는 최초의 정밀조사로서 탐진강을 비롯한 하구역에 대한 각종 개발압력 및 훼손 위기에 대응 할 수 있는 기초자료로 활용될 수 있을 것으로 기대한다.
