Principle of Emulsion PCR and Its Applications in Biotechnology

  • Emulsion polymerase chain reaction (PCR) is performed on compartmentalized DNA, allowing a large number of PCR reactions to be carried out in parallel. Emulsion PCR has unique advantages in DNA amplification. It can be applied in many molecular biological assays, especially those requiring highly sensitive and specific DNA amplification. This review discusses the principle of emulsion PCR and its applications in biotechnology. Related technologies are also discussed.

Emulsion Mapping in Pork Meat Emulsion Systems with Various Lipid Types and Brown Rice Fiber

  • This study was conducted to evaluate emulsion mapping between emulsion stability and cooking yields, apparent viscosity, and hardness of reduced-fat pork emulsion systems. The reduced-fat emulsion systems were supplemented with different lipid types and brown rice bran fiber (BRF) concentrations. Compared to the control with 30% back fat, lower emulsion stability and higher cooking yield of meat emulsion systems were observed in T1 (30% back fat+1% BRF), T2 (30% back fat+2% BRF), T3 (30% back fat+3% BRF), T4 (30% back fat+6% BRF), and T15 (10% back fat+10% canola oil+2% BRF). Lower emulsion stability and higher apparent viscosity were observed in T1, T2, T3, T4, and T8 (20% back fat+3% BRF) compared to the control. Lower emulsion stability and higher hardness was detected in all treatments compared with the control, except T5 (20% back fat), T10 (10% back fat+10% canola oil+2% BRF), T11 (10% back fat+10% olive oil+2% BRF), T12 (10% back fat+10% grape seed oil+2% BRF), and T13 (10% back fat+10% soybean oil+2% BRF). This approach has been found particularly useful for highlighting differences among the emulsified properties in emulsion meat products. Thus, the results obtained with emulsion mapping are useful in making emulsified meat products of desired quality characteristics, partially replacing pork back fat with a mix of 10% back fat, 10% canola oil and 2% BRF was most similar to the control with 30% pork back fat.

에멀젼 반전법으로 제조된 쿼세틴을 함유하는 나노에멀젼에 대한 에탄올의 영향 (The Effects of Ethanol on Nano-emulsions Containing Quercetin Prepared by Emulsion Inversion Point Method)

  • 본 연구에서는 고농도의 쿼세틴을 함유하는 안정한 나노에멀젼의 조성을 찾고 POE (30) hydrogenated castor oil (HCO-30)/오일/쿼세틴/에탄올/물로 이루어진 나노에멀젼에 대한 에탄올의 영향을 연구하였다. 나노에멀젼은 저에너지법인 emulsion inversion point (EIP) 법과 고에너지법인 균질기(homogenizer)를 병합하여 제조하였다. 나노에멀젼에 대한 에탄올과 다른 성분들의 영향을 평가하기 위해 입자 크기, 에멀젼의 형태, 그리고 입자 분포와 같은 물리적 특성을 조사하였다. 나노에멀젼에 대한 쿼세틴의 최적 농도는 0.2 %였다. 계면활성제인 POE (30) hydogenated castor oil (HCO-30) 농도에 따른 나노에멀젼의 입자 크기를 보면, 2.00 %부터는 에멀젼의 입자 크기는 300 nm 이하였으며 HCO-30을 4.75 % 함유하는 나노에멀젼이 가장 안정하고 입자 크기도 172.40 nm로 나타났다. 마지막으로 에탄올 4.00 %를 함유한 나노에멀젼의 입자 크기는 128.15 nm이고 입도 분포 또한 좁게 나타났다. 나노에멀젼의 불안정화 과정은 Ostwald ripening에 의한 것으로 보여진다. 나노에멀젼에 대한 에탄올의 영향을 연구함으로써 소량의 에탄올을 이용하여 약물의 봉입률을 증가시킬 수 있으며, 안정한 나노에멀젼을 만듦으로써 스킨 로션, 에센스 및 향수와 같은 화장품과 제약 등에 응용될 수 있을 것으로 사료된다.

Removal study of As (V), Pb (II), and Cd (II) metal ions from aqueous solution by emulsion liquid membrane

  • Emulsion Liquid Membrane (ELM) is a prominent technique for the separation of heavy metal ions from wastewater due to the fast extraction and is a single-stage operation of stripping-extraction. The selection of the components (Surfactant and Carrier) of ELM is a very significant step for its preparation. In the ELM technique, the primary water- in-oil (W/O) emulsion is emulsified in water to produce water-in-oil-in-water (W/O/W) emulsion. The water in oil emulsion was prepared by mixing the membrane phase and internal phase. To prepare the membrane phase, the extractant D2EHPA (di-2-ethylhexylphosphoric acid) was used as a mobile carrier, Span-80 as a surfactant, and Paraffin as a diluent. Moreover, the internal (receiving) phase was prepared by dissolving sulphuric acid in water. Di-(2- ethylhexyl) phosphoric acid such as surfactant concentration, carrier concentration, sulphuric acid concentration in the receiving (internal) phase, agitation time (emulsion phase and feed phase), the volume ratio of the membrane phase to the receiving phase, the volume ratio of the external feed phase to the primary water-in-oil emulsion and pH of feed were studied on the percentage extraction of metal ions at 20℃. The results show that it is possible to remove 78% for As(V), 98% for Cd(II), and 99% for Pb(II). 화장품용 유화 제조기술 최근동향 (Recent Emulsion Technology in Cosmetics)

  • Emulsions are mixture of immiscible liquids in which one is dispersed in all over the other. They have been applied to many applications including cosmetics, foods, drug delivery system (DDS), fine chemicals, and chemical separations. Especially, emulsion technology is one of the most useful technique to formulate cosmetics such as eye cream, foundation, and foam cleansing. In general, the emulsions can be generated by mechanical agitation of two immiscible fluids. However, the emulsions obtained by conventional method have limited in stability, monodispersity, and complicate process. We describe here preparation techniques of representative cosmetic emulsions such as liposome, liquid crystal emulsion, nanoemulsion, multiple emulsion, and pickering emulsion. Furthermore, various factors which can control the physical properties of each cosmetic emulsions are briefly discussed.

달걀 노른자위 레시틴의 첨가가 물/카놀라 기름 에멀션의 지방질 산화에 미치는 영향 (Effect of Addition of Egg Yolk Lecithin on the Lipid Oxidation of a Water/canola Oil Emulsion)

  • 350 ppm 농도로 물/카놀라 기름 에멀션에 첨가된 달걀 노른자 위 레시틴은 에멀션 기름의 철 이온에 의한 자동산화와 클로로필에 의한 감광산화에 유의한 영향을 나타내지 않았다. 에멀션의 산화 중 달걀 노른자위 레시틴으로부터 유래한 인지방질은 분해되었으며 감광산화에 비해 자동산화에서 분해 속도가 높았다. 그러나 PC와 PE 사이의 분해 속도는 두 종류의 산화에서 모두 유의한 차이를 보이지 않았다(p>0.05).

Preparation and Characterization of Tributyrin Sub-micron Emulsion as Carrier for Paclitaxel

  • Paclitaxel is a well known anticancer agent and has been a pharmaceutical challenge because of its extremely poor water-solubility and susceptibility to the p-glycoprotein (p-gp)-mediated efflux in multi-drug resistant (MDR) cancer cells. Tributyrin (TB), a triglyceride with relatively short fatty acid chains, was chosen as solubilizing vehicle for paclitaxel based on the solubility study (26.6 mg/mL). Tributyrin (10%) o/w emulsion containing paclitaxel (5%), egg phosphatidylcholine (5%) and pegylated phospholipid (0.5%) was prepared by high pressure homogenization to obtain submicron-sized emulsion. The mean particle size of the resultant TB emulsion was 395.5 nm. Paclitaxel in TB emulsion showed higher anticancer activity against human breast cancer cell line, MCF-7, than free form delivered in DMSO solution. On the other hand, its anticancer activity was significantly reduced in MCF-7/ADR, a MDR variant cancer cell line of MCF-7, and recovered by the presence of verapamil, suggesting of the susceptibility to the p-gp mediated efflux even though paclitaxel was encapsulated into emulsion. The TB emulsion showed great potential as a promising vehicle for water-insoluble anticancer agent, paclitaxel.

  • Soft segment가 각각 ester 및 ether형 polyol로된 2종의 polyurethane(PU) ionomer를 emulsion 블렌딩하여, 블렌드의 분산점도, 및 emulsion cast film의 접촉각, 기계적 특성을 조성의 함수로 측정하였다. 블렌드의 기계물성은 혼합법칙으로부터 심한 scattering이나, negative deviation을 보였으며, 이는 ester 및 ether형 polyol간의 상용성의 결여에 기인하는 것으로 보았다. 접촉각 측정 결과로부터 유화입경이 작은 PPG PU 입자가 주로 도막표면층을 형성하는 것을 확인할 수 있었다.

  • In this study, two biodegradable polymer(PLA, PBS) emulsions were treated on agricultural packaging paper such as fruit bagging paper. Water-repellency, mechanical properties, and SEM image with thermal aging were measured on the emulsion treated fruit bagging paper. Biodegradable polymers(such as poly lactic acid, poly butylene succinate) emulsion treated fruit bagging paper had higher water-repellency and strength than other water-repellent(such as acrylic repellent, linseed oil and paraffin wax) treated fruit bagging paper. According to FE-SEM results of polymer emulsion coated fruit bagging paper, the colloidal particles of emulsion after thermal treatment (looks like being) were adhered to the fibers. Thus, using biodegradable polymer emulsion is expected to protect a fruit for a long time.

Physico-chemical Properties of Chicken Meat Emulsion Systems with Dietary Fiber Extracted from Makgeolli Lees

  • Makgeolli lees is a jigaemi by product produced by makgeolli brew processing. Jigaemi has high fiber content and therefore can potentially be used in the development of foods rich in dietary fiber. The effects of makgeolli lees fibers on the composition and physico-chemical properties of chicken emulsion systems were studied. The moisture and ash contents, yellowness, and viscosity of chicken meat emulsion systems with makgeolli lees fiber were all higher than those of control. Moreover, chicken batters supplemented with makgeolli lees fiber were characterized by lower cooking loss and better emulsion stability. Chicken emulsion systems with makgeolli lees fiber also had improved emulsion stability and emulsion viscosity, and the best results were obtained with meat batter containing 2% makgeolli lees fiber.
