Plant Regeneration through Somatic Embryogenesis from Cotyledon of Herbaceous Peony (Paeonia lactiflora Pall.) (芍藥(Paeonia lactiflora Pall.)의 子葉組織으로부터 體細胞胚發생을 통한 식물체 獲得)

  • The experiments were carried out to determine the optimum conditions for the direct embrogenesis from the cotyledon derived zygotic embryo culture of Paeonia lactiflora Pall. Different peony tissues derived from zygotic embryos were cultured on MS medium with and without 2,4-D. Somatic embryos were formed from the cotyledons cultured on the medium without 2,4-D. The somatic embryogenesis from cotyledons was promoted in the growth regulator-free MS medium containing 1.65~3.3 g/L $NH_4NO_3$ and 30~40 g/L sucrose. The maximum frequency (80.0%) of somatic embryo formation was obtained from the cotyledons excised from zygotic embryos that cultured on MS medium containing 3.3 g/L $NH_4NO_3$. Epicotyl and roots were elongated from a somatic embryo by adding 0.3 mg/L GA$_3$ in the medium or the cold treatment at 4$^{\circ}C$ more than three weeks at 4$^{\circ}C$.

  • Dynamic parameters of biochemistry including the POD (peroxidase) activity, contents of DNA and RNA, isozymes of POD and ES (esterase) in American send (Panax quinquefolium L.) seed are reported in the present paper. The dynamic changes of POD activity proved that the PAS (physiological afterripening stage) is a stage in which some substances are prepared for seed germination. The POD activity correlated with ER (embryo ratio) significantly DNA content changed little only within 0.0036∼0.013 mg/ml, which did not correlate with ER. RNA content changed from 0.1539 to 1.0313 mg/ml and correlated significantly with RE during all of the embryo afterripening. None of the POD isozyme band was obtained in ESGS (embryo slowly growth stage), but five bands in ERGS (embryo rapidly growth stage) and six bands in PAS. Four bands of ES isozymes were obtained in ESGS, but six bands in ERGS, particularly, the content of ES isozymes increased in PAS. All of these may provide some information for understanding the dormancy mechanisms of American song seed.

  • Effects of media and plant growth regulators on the germination of somatic embryos of Angelica tree(Aralia elata Seem.) was studied for the mass production of Angelica tree through tissue culture. MS medium was found to be the most effective for the germination of somatic embryos(65% germination rate), Among the MS medium, the medium containing 25% less inorganic salts and 1% less sucrose was found to be the most effective. Gelling agent with 0.2-0.3% gelrite promoted the germination of somatic embryo$(65{\sim}70%)$ and caused good growth of shoots and roots. 0.1 mg/l of BA and kinetin treatment caused $65{\sim}70%$ germination rate of somatic embryos and good growth of shoots and roots, and resulted in high percentage of dry matter. 1mg/l or 5 mg/l treatment of putrescine, and 10 mg/l treatment of spermidine caused 90% germination rate of somatic embryos and good growth of plant organs, and inhibited vitrification of regenerated plants.

  • In this experiment, interactions between light and other factors such as chilling, alternating temperature, moisture, content, oxygen, and seed coat which affect germination of Oenothera lamarckiana Ser. seed were investigated to study the physiological effects of light on the germination. Light induced the initial stage of seed germination before radical protrusion by affecting embryo rather than seed coat even under anaerobic condition or low water potential (-18 bars). This light effect on germinability of seed was suppressed by blue light irradiation and the effect was increased with increment of blue light intensity and irradiation time. However, the blue light effect was reversible. Chilling, alternating temperature, softening of seed coat and light showed promotive interaction in the induction of seed germination. Irradiation of filtered light (monochrome), however, reduced chilling effect on germination. Hydrogen-ion concentration and gibberellic acid treatment had no effect on light or dark germination.

Studies on the Physiological Chemistry of Dormancy and Germination in Panax. ginseng Seeds 2. Changes in Abscisic acid content during Stratification of Seeds. (인삼종자의 휴비 및 발아에 대한 생리화학적 연구)

  • Abscisic acid (ABA) content of the seed and endocarp during stratification were analyzed and then examined in relation to the embryo growth and germination. In mature red fruitlet, the ABA content was remarkably higher in sarcocarp than those in both seed ans endocarp. During the stratification before dehiscence, ABA content was gradually decreased in both seed and endocarp. After 90 days(dehiscent percentage; 96%) it came to 90 pmol/ g DW(69% decrease) and to 41 pmol/ g DW (80% decrease) in seed and in endocarp, respectively. The ratio of free from to total ABA content showed constant decrease in seed, but remained at higher level in endocap than in seed. Correlation between the decrease of ABA content and embryo growth showed higher significance in seed than in endocarp. During the stratification after dehiscence, ABA content in seed was gradually decreased at 4$^{\circ}C$ and 15$^{\circ}C$, After 90 days it came to 28 pmol/ g DW (69% decrease) and to 46 pmol/ g DW (49% decrease) at 4$^{\circ}C$ and at 15$^{\circ}C$, respectively. The ratio of free form to total ABA content was gradually increased at 4$^{\circ}C$, but remained almost constant at 15$^{\circ}C$. Correlation between the decrease of ABA content and days to first germination showed positive singificance only at 4$^{\circ}C$, whereas the correlation between the decrease and mean germination percentage per day showed negative significance at 4$^{\circ}C$, but positive significance at 15$^{\circ}C$. The above results indicate the ABA of the seed end endocarp during the stratification before dehiscence seems to be concerned with the immature embryo growth, but that of the seed during the stratification after dehiscence seems to show little effect on the germination capability(degree of breaking physiological dormancy).

A combined pathway of somatic embryogenesis and organogenesis to regenerate radiata pine plants

  • This study describes for the first time in Pinus genus a plant regeneration system via a combined pathway of somatic embryogenesis and organogenesis from immature seeds of radiata pine. Somatic embryos were obtained from embryogenic line 2162 of Pinus radiata D. Don on EDM basal medium containing $60{\mu}M$ ABA and 6% sucrose. The explants used for organogenesis experiments were either freshly collected somatic embryos or somatic embryos germinated for 1 week. Germination medium was half-strength LP medium, supplemented with 0.2% activated charcoal. Different induction periods and BA concentrations were assayed for shoot induction. After induction treatments, explants were elongated on the same medium used for germination stage. Rooting medium was quarter-strength LP medium supplemented with three different auxin treatments: $1.5mg\;L^{-1}$ 1-naphthalene acetic acid (NAA), $1.5mg\;L^{-1}$ indole-3-butyric acid (IBA) and $1mg\;L^{-1}$ IBA with $0.5mg\;L^{-1}$ NAA (MIX). The effect of the photon flux ($120mmol\;m^{-2}\;s^{-1}$ and darkness) in the first week of the explants in the rooting media was also tested. This methodology could offer an alternative to overcome some problems associated with somatic embryogenesis such as the seasonality of embryogenic tissue (ET) initiation or a low embryo production from the ET, a particularly important issue in the case of genetically transformed ETs.

  • It was determined optimal Culture conditions and suitable growth regulators for seed germination, growth of callus, and protoplasts derived from cultured and mesophyll cells in Gremastra appendiculata. Induction of fusion between protoplasts of cultured and mesophyll cells was examined. The best conditions of seed germination and growth of callus were achieved on Hyponex medium contained plant growth regulators(2mg/l 2, 4-D, lmg/l Kinetin). Viability and regeneration of cell wall in protoplasts was determined with fluorescence microscope. Also, fused protoplasts were achieved by using PEG solution between protoplasts of cultured and mesophyll cells.

Pre-Harvest Sprouting Variation of Rice Seeds Located on Each Panicle Position According to Grain Filling Days (벼 등숙일수에 따른 이삭 착생 부위별 종자의 수발아 변이)

  • This research was implemented to investigate pre-harvest sprouting characteristics in two Korean rice cultivars, Shindongjin (SDJ) and Hopum (HP). The panicles of both varieties were sampled after 15 days after heading (DAH) to 60 DAH at intervals of 5 days. As soon as sampled, the panicles were imbibed in water for 14 days at $25^{\circ}C$ to determine the vigor and germination ability of seeds according to location on panicle. To investigate the cause of non-germination of seeds in panicles, tetrazolium test and dormancy breaking were performed. The preharvest sprouting of HP started at 20 DAH while that of SDJ began at 30 DAH. The germination of seeds located in high rank branches within a panicle and the terminal seeds within a branch were earlier and faster and the germination patterns were same in both cultivars. The times at more than 50% of germination in a panicle were 35 DAH (57.0%) in HP and 45 DAH (56.8%) in SDJ. Preharvest sprouting was more than 80% at 50 DAH (82.6% of HP, 92.3% of SDJ) and more than 99% at 60 DAH (99.5%, 99.4%, HP and SDJ). These results indicated that the rate of PHS in a panicle increased with accumulation of the days after heading. The cause of non-germinated seeds at 15-25 DAH in panicle was immature embryo. After 30 DAH, however, when the non-germinated seeds were hulled, they started to germinate due to dormancy breaking, in which the germination percentage was higher in SDJ than HP. In conclusion, the pre-harvest sprouting varied according to days after heading, the seed position on panicle, and the dormancy intensity of varieties.

Somatic Embryogenesis and Plant Regeneration with Embryogenic Tissue Lines in Pinus densiflora (소나무 배발생조직 라인 별 체세포배 유도 및 식물체 재분화)

  • This experiment was conducted to evaluate growth rate among 15 embryogenic tissue lines (ETLs), comparison of maturation on somatic embryos (SEs) with 13 ETLs and efficiency with various concentrations of gelrite on SEs germination in Japanese red pine (Pinus densiflora). In comparison of ETLs growth rate (folds) with 15 lines, the 05-4 line (5.3 folds) showed the highest rate, on the other hand, the lowest one was recorded in the line of 05-37 (1.4 folds). The 13 ETLs were tested for the extent of SEs production. The best production was recorded in the line of 05-4 (39.8/90 mg F.W.). However, most of ETLs, except 5 lines (05-4, 12, 21, 29 and 37), did not produce SEs at all, therefore, big differences in the ability of SEs production existed among the ETLs tested. Effects of various gelrite concentrations for SEs germination with 3 ETLs were also compared. The highest result was obtained from 0.2% gelrite concentration with 05-4 line (47.3%), there was a inclination that the rate of germination was gradually declined over 0.2% gelrite concentration with the 05-4 and 29 lines. respectively. In contrast, in the line of 05-37, no SEs germination occurred on medium with 0.1 or 0.2% gelrite. In conclusion, the growth rate, SEs production and germination frequency were appeared to deeply depended on the ETLs.

Effect of Seed Moisture Content on Seed Storage of Dehisced Ginseng Seeds (종자 수분함량에 따른 개갑 인삼 종자의 저장성 연구)

  • Ginseng (Panax ginseng C.A. Meyer) is a perennial plant and propagates by seeds, and those need after-ripening for germination. To be ready for climate change and to ensure a stable seed supply, the technique for storing seeds in short-term and long-term in large quantities is required. In this study, dehisced ginseng seeds from two locations, batch #1 and batch #2, were stored at -3.5℃ with different moisture content, and after 3, 15, and 27 months of storage, the percentage of radicle emergence and shoot emergence were measured. After 3 months, radicle emergence and shoot emergence were normal only when the seed moisture content was more than 35%, and overall, germination was higher in batch #2 than in batch #1. After 15 months, the partially dehydrated seeds, with a moisture content between 45 to 54%, showed the highest germination rates, and most of the undried seeds were spoiled and failed to germinate. Seeds with moisture content lower than 25% had poor germination, too. The partially dehydrated seeds also succeeded in germination and growth in the soil after 15 months of storage, but deteriorated after one more 1 year, too. In summary, ginseng seeds look like have temperate recalcitrant seed characteristics, and partial dehydration allows extension of seed longevity.
