Dynamic Profile of the Copper Chaperone CopP from Helicobacter Pylori Depending on the Bound Metals

  • Copper is an elemental ion in living organisms. CopP from Helicobacter Pylori (HpCopP) is a copper(I)-binding protein and was suggested as regulator of copper metabolism in vivo. Previously, the metal binding property of HpCopP for Ag(I), Cu(I), and Cu(II) as well as the tertiary structure of HpCopP was shown. In this study, the dynamic profiles of HpCopP depending on metal binding were studied using ${^1H}-^{15}N$ steady-state NOE analysis. The heteroNOE experiment was performed for apo-CopP or metal-bound CopP. The obtained NOE values were analyzed and compared to figure out the effect of metals on the structural flexibility of HpCopP. As a result, Ag(I) and Cu(I) ions improved the rigidity of the structure while Cu(II) ion increased the flexibility of the structure, suggesting the oxidation of the CXXC motif decreases the structural stability of HpCopP.

금속 프탈로시아닌 유도체의 Langumuir-Blodgett 박막 제작에 관한 연구 (Study on Preparation of Metal Phthalocyanine Derivative Langmuir-Blodgett Film)

  • It is well known that the metallo-phthalocyanine(MPcs) are sensitive to toxic gaseous molecules such as $NO_2$ and also chemically and thermally stable. Therefore, lots of MPcs have been studied for the potential chemical sensor for $NO_2$ gas using quartz crystal microbalance or electrical conductivity. In this study, 2,3,9,10,16,17,23,24-[octa-(dodecyloxy)] copper phthalocyanine and 2,3,9,10,16,17,23,24-[octa-(octyloxy)] copper phthalocyanine were synthesized and their possibility of LB film preparation were tested. It was confirmed by using FT-IR, DSC, NMR, UV-Vis absorption spectroscopy and Elemental Analysis that CuPc derivatives were successfully synthesized. From the ${\pi}$-A characteristics and limiting areas of two CuPc derivatives it was found that the preparation of LB films with these CuPc derivatives is possible.

Dimesogenic Compounds with Chiral Tails: Synthesis and Liquid Crystalline Properties of a Homologous Series of a, w-Bis[4-(4'-(S)-( -)-2-methylbutoxycarbonylbiphenyl- 4-oxycarbonyl)phenoxy]alkanes

  • A series of new liquid crystalline dimesogenic compounds with chiral tails was synthesized, and their thermal and liquid crystalline properties were studied. The chain length of the central polymethylene spacers (x) was varied from dimethylene (2) to decamethylene (12). These compounds were characterized by elemental analysis, IR and NMR spectroscopy, differential scanning calorimetry (DSC), and cross-polarizing microscopy. All compounds were found to be enantiotropically liquid crystalline, and the values of melting ($T_m$) and isotropization temperature ($T_i$) as well as enthalpy change (Δ$H_i$) and entropy change for isotropization (Δ$S_i$) decreased in a zig-zag fashion revealing the so-called odd-even effect as x increases. Their mesomorphic properties fall into three categories depending upon x; (a) compounds with x=2 and 4 formed two different mesophases, smectic and cholesteric phases in that order on heating, and vice versa on cooling, (b) compounds with x=3, 7, 8, 10 and 11 reversibly formed only the cholesteric phase, and (c) compounds with x=5, 6, 9 and 12 exhibited only a cholesteric phase on heating, whereas on cooling they formed two different mesophases, cholesteric and smectic phases, sequentially.

  • The addition products of glutathione to ${\beta}$ -nitrostyrene derivatives were synthesized. ${\beta}$ -Nitrostyrene (1a), p-methyl-${\beta}$-nitrostyrene (1b), 3,4,5-trimethoxy-${\beta}$-nitrostyrene (1c), o-, m- and p-chloro-${\beta}$-nitrostyrene (1e, 1f, 1g) and o-, m- and p-methoxy-${\beta}$-nitrostyrene (1h, 1i, 1j) undergo addition reactions with glutathione to form S-(2-nitro-1-phenylethyl)-L-glutathione (5a), S-[2-nitro-1-(p-methyl)phenylethyl]-L-glutatione (5b), S-[2-nitro-1-(3', 4', 5'-trimethoxy)phenylethyl]-L-glutathione (5c), S-[2-nitro-1-(o-chloro)phenylethyl]-L-glutathione (5e), S-[2-nitro-1-(m-choro)phenylethyl]-L-glutathione (5f), S-[2-nitro-1-(p-chloro)phenylethyl]-L-glutathione (5g), S-[2-nitro-x-(o-methoxy)-phenylethyl]-L-glutathion e(5h), S-[2-nitro-x-(m-methoxy)phenylethyl]-L-glutathion e (5i), and S-[2-nitro-1-(p-methoxy)phenylethy])-L-glutathione (5j), respectively. The structure of adducts were identified by UV and IR-spectra, molecular weight measurement, and elemental analysis.

  • The addition reactions of cysteine without blocking amino and carboxyl groups to substituted and unsubstituted ${\beta}$-nitro-styrene derivatives were investigated. ${\beta}$-Nitrostyrene(1a), p-methyl-${\beta}$-nitrostyrene(1b), 3,4,5-trimethoxy-$[\beta}$ -nitrostyrene(1c), $[\varpi}$-3,4-methylenedioxy-${\beta}$ -nitrostyrene(1d), o-, m- and p-chloro-${\beta}$ -nitrostyrene (1e, 1f, 1g) and o-, m- and p-methoxy-${\beta}$-nitrostyrene (1h, 1i, 1j) easily undergo addition reactions with cysteine to form S-(2-nitro-1-phenylethyl)-L-cysteine(3a), S-[2-nitro-1-(p-methyl)phenyl-ethyl]-L-cysteine(3b), S-[2-nitro-1-(3',4',5'-trimethoxy) phenylethyl]-L-cysteine(3c), S-[2-nitro-1-($[\vatpi}$ -3',4'-methylenedioxy)phenylethyl]-L-cysteine(3d), S-[2-nitro-1-(o-chloro)phenylethyl]-L-cysteine(3e), S-[2-nitro-1-(m-chloro)-phenylethyl]-L-cysteine(3f), S-[2-nitro-1-(p-chloro)phenylethyl]-L-cysteine(3g), S-[2-nitro-1-(o-methoxy)phenylethyl]-L-cysteine(3h), S-[2-nitro-1-(m-methoxy)phenylethyl]-L-cysteine(3i) and S-[2-nitro-1-(p-methoxy)phenylethyl]-L-cysteine(3j), respectively. The structure of adducts were confirmed by means of UV-spectrum, IR-spectrum, molecular weight measurement and elemental analysis. The various factors effecting the yield of cysteine adducts to ${\beta}$-nitrostyrene derivatives were also studied.

Synthesis and Characterization of Fe-fullerene/TiO2 Photocatalysts Designed for Degradation of Methylene Blue

  • Fe-fullerene/$TiO_2$ composite photocatalysts were prepared with titanium (IV) n-butoxide (TNB) by a sol-gel method. The samples were characterized by scanning electron microscopy (SEM), Transmission electron microscope (TEM), specific surface area (BET), X-ray diffraction analysis (XRD) and energy dispersive X-ray spectroscopy (EDX). The photocatalytic activities were evaluated by the photocatalytic degradation of methylene blue (MB) solution. XRD patterns of the composites showed that the Fe-fullerene/$TiO_2$ composite contained a typical single and clear anatase phase. The surface properties shown by SEM present a characterization of the texture on Fe-fullerene/$TiO_2$ composites and showed a homogenous composition in the particles for the titanium sources used. The EDX spectra for the elemental identification showed the presence of C and Ti with strong Fe peaks for the Fe-fullerene/$TiO_2$ composite. From the photocatalytic results, the excellent activity of the Fe-fullerene/$TiO_2$ composites for degradation of methylene blue under UV light irradiation could be attributed to both the effects between photocatalytic reaction of the supported $TiO_2$, decomposition of the organometallic reaction by the Fe compound and energy transfer effects such as electron and light of the fullerene.

Hydrogenation of trans-Cinnamaldehyde with Hydrido-Carbonyl Osmium(II) Complexes of Chelating Phosphine Ligands

  • A series of new hydridocarbonyl osmium(Ⅱ) complexes, OsHCl(CO)(PPh3)(L-L)[L-L=Ph2P(CH2)nPPh2 (n=1 (1), 2 (2), 3 (3), cis-Ph2PCH=CHPPh2 (4), and Fe(η5-C5H4PPh2)2 (5)] has been synthesized from OsHCl(CO)(PPh3)3 and chelating diphosphines. These complexes have been characterized by IR, 1H NMR and elemental analysis. The catalytic activities of these complexes both for the transfer hydrogenation of trans-cinnamaldehyde with 2-propanol as the hydrogen donor, and for the selective hydrogenation of trans-cinnamaldehyde with H2, have been examined. Complexes (1)-(5) were shown to have higher selectivities for the transfer hydrogenation of the C=O bond of aldehyde than for the transfer hydrogenation of the C=C bond of aldehyde. The selectivities for the transfer hydrogenation with 2-propanol as well as for the hydrogenation with H2 have been found to decrease in the order 3 > 5 > 2 > 4 > 1. Complex (3) has shown to possess almost 90% of the selectivity to cinnamyl alcohol for transfer hydrogenation. It is also found that there is a correlation between the ν(CO) of each complex and the hydrogenation, of the C=O bond of trans-cinnamaldehyde. Overall, the selectivities with the complexes (1)-(5) are greater for the transfer hydrogenation with 2-propanol than for the hydrogenation with H2.

Synthesis and Electrochemical Studies of Cu(II) and Ni(II) Complexes with Tetradentate Schiff Base Ligands

  • A series of tetradentate Schiff-base ligands; 1,3-bis(salicylideneimino) propane, 1,4-bis(salicylideneimino)butane, and 1,5-bis(salicylideneimino)pentane, and their Cu(Ⅱ) and Ni(Ⅱ) complexes have been synthesized. The properties of ligands and complexes have been characterized by elemental analysis, IR, NMR, UV-Vis spectra, molar conductance, and thermogravimetric anaylsis. The mole ratio of Schiff base to metal at complexes was found to be 1 : 1. All complexes were four-coordinated configuration and non-ionic compound. The electrochemical redox processes of the ligands and their complexes in DMF solution containing 0.1 M TEAP as supporting electrolyte have been investigated by cyclic voltammetry, chronoamperometry, differential pulse voltammetry at glassy carbon electrode, and by controlled potential coulometry at platinum gauze electrode. The redox process of the ligands was highly irreversible, whereas redox process of Cu(Ⅱ) and Ni(Ⅱ) complexes was observed as one electron transfer process of quasi-reversible and diffusion-controlled reaction. Also the electrochemical redox potentials of complexes were affected by chelate ring size of ligands. The diffusion coefficients of Cu(Ⅱ) and Ni(Ⅱ) complexes in DMF solution were determined to be 4.2-6.6×10-6 cm2/sec. Also the exchange rate constants were determined to be 3.6-9.7×10-2 cm/sec.

Synthesis and Characterization of New Intermetallic Compounds $M_3(AsTe_3)_2$ (M=Cr, Fe, Co)

  • The new amorphous intermetallic compounds, M3(AsTe3)2: M=Cr, Co, Fe, were synthesized by the precipitation reaction of the Zintl anion AsTe33- with the divalent transition metal halides in aqueous solution and analyzed by EDS equipped with SEM and PIXE. The empirical formula of the specimens was found to be Fe3.0As1.8Te5.9, Co3.0As2.1Te6.5, and Cr3.0As2.0Te6.9 by the quantitative elemental analysis. The dc specific resistivity of the materials was measured as a function of temperature in the range from 20 to 300 K, in which their resistivity of Cr3(AsTe3)2 was largely dependent on temperature, while those of Co3(AsTe3)2 and Fe3(AsTe3)2 were only slightly dependent on temperature. To characterize the spin glass state of the specimens, the ac and dc magnetic susceptibility were measured and it was found that Co3(AsTe3)2 and Fe3(AsTe3)2 undergo a transition to a spin glass state at 6 K and 38 K, respectively. Magnetization data are reported as both thermal remanent magnetization (TRM) and isothermal remanent magnetization (IRM) as a function of magnetizing field and temperature.

Evaluation of antibacterial activity and cytotoxic effects of green AgNPs against Breast Cancer Cells (MCF 7)

  • The present work reports a facile, rapid and an eco-friendly method for the synthesis of silver nanoparticles using Luffa acutangula (L. acutangula) leaves extract and their antibacterial and cytotoxic effects. The synthesized silver nanoparticles (AgNPs) were characterized by UV-Visible spectroscopy (UV-Vis), Fourier transform infrared spectroscopy (FT-IR) and X-ray diffraction analysis (XRD). Additionally the topography, morphology and the elemental composition of the particles were determined by Scanning Electron Microscopy (SEM) and Energy dispersive spectrophotometric (EDS) technique and the measured particle sizes from SEM micrographs are in the range of 12.5 to 24.5nm. The in-vitro antimicrobial activity of the synthesized nanoparticles was high against gram positive Staphylococcus aureus and moderate against gram negative Escherichia coli and Pseudomonas aeruginosa strains. Further, the cytotoxic effects of synthesized AgNPs were evaluated against Human Breast Cancer (MCF 7) cell line.
