Electron spin resonance (ESR) spectroscopy was used to detect irradiated meat containing bones (chicken, pork and beef), to investigate the effect of irradiation dose on the ESR signal intensity and to identify the stability of radicals under 9 weeks of storage. Chicken, pork and beef were irradiated with doses 0, 1, 3, 5 and 7 kGy at room temperature using a Co-60 irradiator. Bones were pieced and dried, which were placed in a quartz tube within an Electron paramagnetic resonance (EPR) spectrometer resonator cavity. The irradiated bone presented an asymmetric absorption in shape, different from that of a non-irradiated one. The signal intensity of smaller animals are lower than larger species. Variation was observed between samples of the same species depending on the calcification status of the bone. Moreover different irradiation doses produced different signal areas that make possible to estimate the absorbed dose of treated meat. The ESR signal stability after irradiation was stable in even after a 9 week storage at room temperature.

For the research of the natural marine antioxidant, an antioxidant-producing marine actinomycetes was isolated from sea water in Jeju coastal area. The strain was identified based on 16S rDNA sequencing, the morphology by a method of scanning electron microscopy, physiological and biochemical characteristics and cellular fatty acid analysis. The isolated strain ACT-18 was gram positive, aerobic, non-motile spores. Substrate mycelia are dark green and yellow gray aerial mycelia. The cell size of the strain was $0.5{\sim}1.0\;{\mu}m$. 16S rDNA sequence analysis showed that were Gram-positive bacteria grouped on Streptomyces sp. Results of cellular fatty acid analysis showed that major cellular fatty acids were $C_{15:0}$ anteiso (39.33%), $C_{16:1}$ cis 9 (11.96%), $C_{16:0}$ (13.08%) and $C_{17:0}$ anteiso (10.99%). The antioxidant activity of methanol extract from Streptomyce sp. ACT-18 was evaluated by measuring 1,1-diphenyl- 2-picrylhydrazyl (DPPH), hydroxyl, and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. DPPH radical scavenging activity of SBME (Streptomyces Broth Methanol Extract) A-18 was 46% at 0.1 mg/mL. Hydroxyl radical scavenging activity of SBME A-18 was 63% at 0.1 mg/mL. Alkyl radical scavenging activity of SBME A-18 was 39% at 0.1 mg/mL.

Recently, free radicals such as active oxygen species, nitric oxide, etc are believed to be one of the key substances in physiological and pathological, toxicological phenomena, and oxidative damages, and all organism have defencing system against such as free radicals. Formation and extinction of free radicals may be regulated through bio-redox system, in which various enzymes and compounds should be involved in very complicated manner. Thus, direct and non-invasive measurement of in vivo free radical reactions with living animals must be essential to understand the role of free radicals in pathophysiological phenomena. Electron spin resonance spectroscopy (ESR) is very selective and sensitive technique to detect free radicals, but a conventional ESR spectrometer has large detect in application to living animals, since high frequent microwave is absorbed with water, resulting in generation of high fever in living body. In order to estimate in vivo free radical reactions in living whole animals, we develop in vivo ESR-CT technique using nitroxide radicals as spin probes. Nitroxide radicals and their reduced forms, hydroxylamines, are known to interact with various redox systems. We found that! ! the signal decay due to reduction of nitroxyl radicals is influenced by aging, inspired oxygen concentration, ischemia-referfusion injury, radiation, etc. In the present paper, I will introduce in vivo ESR technique and my laboratory recent results concerning non-invasive evaluation of free radical reactions in living mice.
Electron spin resonance (ESR) spectroscopy was used to investigate the effects of irradiation doses and storage times on three spices. Three imported spices (Brassica juncea, Origanum vulgare, Laurus nobilis) were irradiated with doss of 0, 1, 5 and 10 kGy using a Co-60 irradiator at room temperature. ESR signals were measured from each sample by a Bruker-EPR spectrometer. The gamma-irradiated Brassica and Laurus nobilis exhibited characteristic signals, which were different from those of the non-irradiated ones. Non-irradiated and irradiated Origanum vulgare exhibited single lines, with higher ESR signals in irradiated sample than in non-irradiated sample. The ESR signals increased linearly with increasing irradiation doses $(1{\sim}10kGy)$. A strong positive correlation coefficient $(R^2=0.9939{\sim}0.9993)$ was obtained between the irradiation doses and the corresponding ESR signal intensities. Although the ESR signal intensities decreased with storage time, signals from the three irradiated spices were observed even after 12 weeks of storage at room temperature.
The antioxidative activity of various enzymatic extracts from leaves of Perilla frutescens var. japonica was evaluated by measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. For this study, the leaves were enzymatically hydrolyzed by 8 carbohydrases (Dextrozyme, AMG, Promozyme, Maltogenase, Termamyl, Viscozyme, Celluclast, and BAN) and 9 proteases [Flavourzyme, Neutrase, Protamex, Alcalase, PP-trypsin (trypsin from porcine pancreas), papain, pepsin, $\alpha$-chymotrypsin, and BP-trypsin (trypsin from bovine pancreas)]. The DPPH radical scavenging activities of Promozyme and Alcalase extracts were the highest, and the $IC_{50}$ values were 77.25 and $109.66\;{\mu}g/mL$, respectively. All enzymatic extracts of the leaves scavenged hydroxyl radical, and the $IC_{50}$ values of Celluclast and pepsin extracts which were the highest activity were 243.34 and $241.86\;{\mu}g/mL$, respectively. The BAN and $\alpha$-chymotrypsin extracts showed the highest scavenging activities, and the $IC_{50}$ values were 21.13 and $33.23\;{\mu}g/mL$, respectively. The pepsin extracts from the leaves showed protective effect on $H_2O_2$-induced DNA damage. In addition, the pepsin extracts decreased cell death in PC-12 cells against $H_2O_2$-induced oxidative damage. The findings of the present study suggest that enzymatic extracts of the leaves possess antioxidative activity.
Electron Spin Resonance (ESR) spectroscopy has been used to detect the presence of radiation-induced free radicals in biological samples since the mid 1950s and to irradiate foods containing cellulose, crystalline sugar, and bone. Therefore, we analyzed the ESR spectrum of irradiated infant formula and its ingredients in this study. Samples were irradiated with 2 different radiation sources of $^{60}Co$ gamma rays and electron beams (EBs), and the absorbed doses were 0, 1, 3, 5, and 7 kGy. ESR measurements were performed under normal atmospheric conditions using a JEOL JES-FA100 spectrometer equipped with an X-band bridge. Irradiated infant formula showed anunsymmetrical spectrum ($g_1$=2.0050, $g_2$=2.0006); in contrast, non-irradiated samples showed asymmetrical spectrum. The ingredients of irradiated samples showed a multi-component ESR signal in glucose and lactose and a singlet-type spectrum in milk powder (g=2.0050). $R^2$ of the dose-response curve showed a fine linearity of over 0.95 across the entire sample. We also compared the spectra of identical samples irradiated with $^{60}Co$ gamma rays and EBs, because EBs can be used for food irradiation in foreign countries, although this is not permitted in Korea. However, we could not find any significant differences according to the types of radiation source. Thus, ESR spectroscopy can be used to detect irradiated infant formula and several types of primary ingredients in this formula.
Radiation damage in a single crystal of ammonium sulfate caused by ${\gamma}$-irradiation at room temperature has given rise to several paramagnetic centers. Electron spin resonance (ESR) spectra of crystal are obtained with the X-band EPR spectrometer at room temperature. An intense and isotropic peak of Gaussian shape at g = 2.0036 is assigned to $SO_3^-$, which shows power saturation effects. Angular dependence of spectra is studied for the rotations about three mutually perpendicular axes a, b and c. The g-values are obtained from the relative distances between isotropic peak of $SO_3^-$ and anisotropic peak of the species. Principal $g^-$values and direction cosines were calculated by diagonalizing the 3${\times}$3 matrix whose elements are the $g^-$values for each species. From the analysis of characteristic principal $g^-$values and direction cosines for ammonium sulfate single crystal, anisotropic peaks corresponding to $SO_4^-,\;SO_2^-$ and defect structure corresponding to electron excess type are identified.

Study was carried out with electron spin resonance (ESR) spectroscopy to identify irradiated Korean medicinal herbs. Pueraria thunbergiana Benth., Angelica gigas Nakai, Agaricus blazei Murill and Astragalus membranaceus Bunge were irradiated with doses of 0, 1, 5 and 10 kGy at room temperature using a Co-60 irradiator. The irradiated Korean medicinal herbs exhibited an asymmetric absorption, which was different from the non-irradiated ones. The strength of ESR signals linearly increased by dose-dependent manner (1~10 kGy) and highly positive correlation coefficients ($R^2$=0.9428~0.9942) were obtained between the irradiation doses and the corresponding ESR signal intensities. Detection of the irradiated Korean medicinal herbs was possible even after 6 weeks of storage although the signal intensities of the irradiated samples decreased until 2 weeks at room temperature.
Electron spin resonance (ESR) spectroscopy was used to investigate the effect of irradiation dose on the ESR signal intensity of irradiated crabs and the stability of these radicals under 9 weeks of storage. Swimming and small crabs were irradiated with doses of 0, 1, 3, 5 and 7 kGy using a Co-60 irradiator at ambient temperature. A claw, a walking leg and a cars- pace of the crab pieced and dried were placed in a resonant quart tube within an EPR X-band spectrometer. The irradiated crabs presented an asymmetric absorption in shape at g$_1$=2.002 $\pm$ 0.003 and g$_2$=1.998$\pm$0.005, and were different from the non-irradiated ones. The intensity of the ESR signals was greatest in the claw, intermediate in the carapace and lowest in the walking leg. Samples given low and high doses of irradiation could also be distinguished. The ESR signal after irradiation was stable, even after a 9-week storage.

Dried banana, pineapple and pistachio were irradiated with doses of 0, 0.5, 1, 2 and 5 kGy at room temperature using a Co-60 irradiator. Multiplet ESR signals were observed in irradiated dried banana and pineapple, and singlet ESR signal was observed in irradiated pistachio, while these characteristic signals were not detected in non-irradiated samples. Since the amount of free radicals linearly increased with the applied doses $(0.5{\sim}5\;kGy)$, highly positive correlation coefficients $(R^2=0.9874{\sim}0.9974)$ were obtained between the irradiation doses and the corresponding free radical concentrations. The characteristic ESR signals were observed in irradiated samples even after 40 days of storage at room temperature.
