Studies on Improvement of Artificial Cultivation and Antioxidative Activity of Poria cocos (복령의 인공 재배법 개선과 항산화활성에 관한 연구)

  • This study was carried out to improve an artificial culture techniques and antioxidative activity of the crude extract isolated from sclerotia of Poria cocos(Fr.) Wolf. In the test of different spawns and inoculation method, the sclerotia formation, number of sclerotia and production yield were excellent in the both sides inoculation method of log spawn, whereas the both side inoculation method of sawdust spawn was poor in sclerotia formation and yield. The optimal spawn and inoculation method for the quality and productivity of P. cocos was in the order of log spawn (both sides inoculation > log spawn(cutting section inoculation) > sclerotia (both sides inoculation) > sawdust spawn (both sides inoculation). The physiological activity substance, crude extract content of P. cocos NIAST 13007 was about 83%. As the concentration of crude extracts increased, the relative viscosity tended to be increased. However, as the concentration of sodium chloride increased, the relative viscosity did not affected. In antioxidative activities, electron donating ability (EDA) of P. cocos was about 10% of butylated hydroxytoluene (BHT). Thiobarbituric acid (TBA) value was similar to that of the vitamin C, however the peroxide value (POV) was lower than those of BHT and vitamin C.

Optimization of Preparation Conditions and Quality Characteristics of Sweet Pumpkin Stock (단호박 스톡 제조조건의 최적화 및 품질 특성)

  • The stock that is the first step for preparation of soups and purees links to the taste of food. Many types of vegetable have been used in stocks, but this study focused on stocks prepared with sweet pumpkin. The stock preparation conditions including the weight of sweet pumpkin, the water volume, and the boiling time at $97^{\circ}C$ were optimized by response surface methodology. The quality characteristics of the resulting stock were investigated. The color, flavor, taste and overall acceptability were dependent parameters. A model equation was proposed with regard to the sweet pumpkin weight, water volume, and boiling time at $97^{\circ}C$. A sweet pumpkin weight of 357.9 to 403.0 g, a water volume of 689.8 to 768.5 mL, and a boiling time of 9.9 to 10.3 min at $97^{\circ}C$ were found to be the optimal stock preparation conditions. The quality characteristics of the sweet pumpkin stock prepared under the optimized conditions were pH 6.64, total acidity 0.18%, soluble solids $2.39\;^{\circ}Brix$, color value (L, 99.07 a, -2.43 b, 11.82), total polyphenol 280.75 mg/L, and electron donating ability 21.32%.

Total Polyphenol Content and Antioxidative Activities of Mistletoe(Viscum album) Extracts by Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 겨우살이(Mistletoe, Viscum album) 추출물의 폴리페놀 함량 및 항산화 활성)

  • This study was performed to investigate antioxidant activities of Mistletoe (MS) extracts by measuring electron donating ability (EDA), super oxide dismutase (SOD)-like activity and total polyphenol content (TPC). The extracts were obtained using supercritical carbon dioxide, hot-water and ethanol extraction. EDAs was the higher in ethanol extract (70.53%) than water (36.73%) or supercritical carbon dioxide extract (15.73% at 400 bar, $30^{\circ}C$) at concentration of 10 mg/mL. However, SOD-like activities and TPCs were the highest in the supercritical carbon dioxide extract. At concentration of 1 mg/mL, supercritical carbon dioxide extract (22.54% at 200 bar, $40^{\circ}C$) was higher SOD activity than water (7.85%) and ethanol extract (6.12%). Regarding TPCs of MS extracts, the content was the highest in supercritical carbon dioxide extract (17.57 wt% catechin equivalent at 200 bar, $50^{\circ}C$) followed by ethanol and water extract. These results suggest that it can be useful to develop functional foods using antioxidative active compounds of MS with high polyphenol contents.

Characteristics of Wine Fermented from Mulberry Juice (오디 착즙액을 이용한 와인발효 특성)

  • We sought to ferment wine from mulberry (Morus alba) juice. The soluble solid content was $9.5{\sim}14.5^{\circ}brix$ on day 6 of fermentation, and gradually fell later; sugar was not further consumed when $3^{\circ}brix$ was attained. Alcohol content rose dramatically on day 6 of fermentation, being 4.5% (v/v) at fermentation temperatures of 16C and 18C, 6.5% (v/v) at 20C, and 8.0% (v/v) at 25C, rising further to 10.5~11.5% (v/v) on day 48, at higher culture temperatures. Citric acid, malic acid, and oxalic acid were present in mulberries. The levels of both citric and oxalic acid fell after fermentation, whereas malic acid concentration increased. All of fructose, glucose, maltose, and sucrose were fermented. Electron-donating ability (EDA) was elevated to over 90% of the control value in mulberry juice diluted to 40% (v/v). SOD-like activities in juice and wine were 80.1% and 72.1% of the control value. Nitrite-scavenging abilities (NSAs) were 86.2% and 85.2% of control in undiluted juice and wine, respectively. Mulberry juice had an activation level higher than that of mulberry wine, but functionality neither rose nor fell after fermentation. Insensory evaluation, the overall wine score was better than average, at 5.00, demonstrating the commercial potential of mulberry wine.

The Antioxidant Activities of the Some Commercial Teas (국내 시판되는 일부 다류 제품의 항산화 효과)

  • The antioxidant activities and their antioxidant compounds of a group of teas obtained in local markets were investigated. A total of 18 teas were tested for their antioxidant activities based on their ability to scavenge ABTS (2,2'-Azino-bis-3-ethylbenzothiazoline-6-sulfonic acid) cation radical and DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical. The former was expressed as mg of ascorbic acid equivalents per 1 tea bag (L-ascorbic acid equivalent antioxidant capacity, AEAC) and the latter was expressed as percentage of electron donating activity (EDA%). A good correlation of AEAC and EDA was observed between the two methods. The concentrations of total polyphenolics and flavonoids in tea extracts were measured by spectrophotometric methods. Total ascorbic acid was determined via the 2,6-dicholoroindophenol titrimetric method. According to the AEAC value and EDA, black tea, brown rice green tea, green tea, herb tea and malva tea showed relatively high antioxidant activities. Polyphenolic compounds were the major naturally occurring antioxidant compounds found in teas and the high concentrations of polyphenolic compounds were observed in black tea, green tea and herb tea. Overall, six teas out of 18 teas tested in the study showed better antioxidant activities and higher amounts of total polyphenolic compounds.

Optimal Extract Methods of Antioxidant Compounds from Coat of Grape Dreg (포도부산물인 과피로부터 항산화 물질 최적 추출방법 확립)

  • Optimal extraction method for antioxidant compounds from coat of grape dreg was established. Extracts were prepared with ethanol solutions containing phosphoric, formic, acetic, HCl, TFA, and citric acids. Antioxidant compounds of grape coat were determined by HPLC analysis and evaluated for antioxidizing effects using in vitro model system. Peonidin-3-glucoside content was the highest in 0.1% HCl-added ethanol extract. The extract prepared from pure ethanol without organic acids showed the highest content of cyanidin-3-glucoside among samples tested. Resveratrol and quercetin contents, the most important antioxidants, were highest in 0,1% HCl-added extract. Electron-donating ability was high in 0.1% acetic acid-added extract. SOD-like activities were 95.08% and 94.39% in 0.1% formic and 0.1% citric acid extracts, respectively. Inhibitory effects on peroxidation of egg yolk lecithin were observed in phosphoric (60.51%), citric (59,27%), and formic acid (56,77%) extracts. Hydrogen radical-scavenging activity was 59.47% in 0,1% HCl extract. Results suggest addition of 0.1% HCl in ethanol solution affords the highest yield in antioxidant compounds and antioxidant activities.

Biological activity of St. John's wort (Hypericum perforatum L.) (St. John's wort(Hypericum perforatum L.)의 생리활성 효과)

  • The physiological activity of St. John's wort extracts were examined. Total phenol contents in the ethanol extracts $(246.0{\pm}10.5\;{\mu}g/ml)$ with St. John's wort leaf was higher than that in water extract $(237.4{\pm}13.2\;{\mu}g/ml)$. The electron donating ability in the water extracts and in the ethanol extracts were 95.0% and 95.2% respectively. Antioxidant protection factor of the ethanol extract was higher than that of the water extract. The water extract from St. John's wort leaves did not show an antimicrobial activity against Helicobacter pylori, but the ethanol extract revealed high antimicrobial activities such as 11 mm of clear zone in $100\;{\mu}g/ml$ of phenol content and 13 mm of clear zone in $150\;{\mu}g/ml$ of phenol content. The hot water extract showed an angiotensin converting enzyme inhibitory activity of 19.2%. The xanthin oxidase inhibitory activity of hot water and ethanol extract were very high, amounting to 84.8% and 100% respectively. The results suggested a possibility for developing the phenol compounds in St. John's wort as anti Helicobacter pylori, anti-oxidant and anti-gout agents.

Study on the Anti-oxidant and Anti-inflammatory Activities of Sarcocarp and Calyx of Persimmon (Cheongdo Bansi) (청도 반시의 과육과 감꼭지(시체: 枾滯)의 항산화 및 항염증 활성 연구)

  • Biological activities of sarcocarp and calyx of persimmon (Cheongdo Bansi) were investigated. The electron donating ability of the calyx extract was 91% at a 500 ${\mu}g/mL$ level, which was higher than that of the sarcocarp extract (40%). The superoxide dismutase (SOD)-like activity of calyx extracts was about 22% at a 1,000 ${\mu}g/mL$ level. During the entire experimental period, there was no cytotoxicity found from sarcocarp and calyx up to 50 ${\mu}g/mL$. Our results indicated that sarcocarp and calyx significantly inhibited NO production and iNOS and COX-2 expression accompanied by an attenuation of interleukin (IL)-$1{\beta}$, IL-6, and tumer necrosis factor (TNF)-${\alpha}$ formation in human keratinocyte. These results suggest that sarcocarp and calyx of persimmon may have significant effect on inflammatory factors and can be used as potential anti-inflammatory agents.

Preliminary Screening of Some Jeju Island Native Plants for Whitening and Antioxidant Activity (제주도 자생식물들에 대한 미백 및 항산화 효능 탐색)

  • In this study, we investigated the whitening and anti-oxidant activity of 37 Jeju island native plants. The active ingredients of the plants were prepared by methanol extraction. Whitening activity of plant extracts was examined from the inhibitory effect of tyrosinase and the inhibition of melanin synthesis of the B16-F1 cell line. Their anti-oxidant activity was measured by electron donating ability of DPPH (1,1-diphenyl-2-picrylhydrazyl) and ROS (reactive oxygen species) scavenging activity in V79-4 lung fibroblast cells using DCF-DA (dichlorofluorescin diacetate). Cytotoxicity of the extracts on cell s based experiments was investigated by MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide) assay. Also, the toxicity test using a rabbit and the human skin patch test were carried out for examining the safety of the extracts which showed the high whitening activity. It is interesting that the extracts of Lespedeza cuneata, Ligustrum lucidum (stem), Morus bombycis (stem) and Prunella vulgaris var. lilacina showed both potent whitening and anti-oxidant activities.

Quality characteristics of frozen brown rice Jeung-pyun dough containing different amounts of acorn flour (도토리가루의 첨가량을 달리하여 제조한 냉동 현미증편반죽의 품질특성)

  • In this study, physicochemical properties of acorn flour was investigated, and characteristics of the Jeung-pyun dough added with the acorn flour at various concentrations were evaluated after storage at -$18^{\circ}C$ for 4 weeks and fermentation. Total polyphenolic content, electron donating ability (EDA) of 0.5%, gelatinization onset temperature (To), gelatinization peak temperature (Tp), and gelatinization conclusion temperature (Tc) of acorn flour were 3,525.12 mg%, 16.71%, $68.41^{\circ}C$, $73.83^{\circ}C$, and $82.96^{\circ}C$, respectively. These all values were increased in the Jeung-pyun dough possibly due to addition of the acorn flour. The yeast count was not affected by the addition levels of acorn flour and the frozen storage period before fermentation. The amount of carbon dioxide gas evolved from Jeung-pyun dough during fermentation was significantly changed with the concentration of acorn flour, but it was negligible. The Jeung-pyun added with 6% acorn flour showed an small increase in the amount of carbon dioxide after frozen storage of 1 wk and fermentation. The pH of the fermented Jeung-pyun samples decreased along with the increasing storage period as well as the increasing acorn flour content, ranging from 4.21 to 5.34. Therefore, the frozen Jeung-pyun dough containing 6~15% of acorn flour and stored for 3 weeks was the most desirable among all the tested samples in this study.
