Effects of Albizziae Cortex Extracts on the Elastase Activity and DPPH and NO Scavenging Activities

  • Elastic fibers are found in the skin, lungs, arteries, veins and other structures. Elastases destroy the elastic fibers and cause the emphysema and pulmonary hypertension. Oxidative stress is needed for these pathologic changes. Accordingly, present study was designed to investigate the effect of Albizziae Cortex extracts (ACE) on elastase activity and anti-oxidative effects of ACE. The in vitro inhibitory effects on elastase and di(phenyl)-(2,4,6-trinitrophenyl)iminoazanium (DPPH) and nitric oxide (NO) free radical scavenging activities of ACE were measured. The elastase activity was significantly inhibited by ACE. DPPH and NO free radicals were significantly scavenged as well. ACE showed the elastase-inhibiting effects and anti-oxidative activities in vitro. These results suggest that ACE may have potential roles in the treatment of pulmonary emphysema and pulmonary hypertension.

  • This study was carried to establish a simple isolation and purification method of ginsenoside $F_1$ from leaves of Panax ginseng and was to evaluate the inhibitory effect of purified ginsenoside $F_1$ on the activities of elastase and tyrosinase. The content of ginsenoside $F_1$ was 90-fold higher in leaves than in root of ginseng. Ginsenoside $F_1$ was isolated from EtOAc fraction between EtOAc and alkalized water of 80% EtOH extract after remove of hydrophobic components. The 50% inhibitory concentration ($IC_{50}$) of ginsenoside $F_1$ on elastase activity and tyrosinase activity was 1.07 mM and 1.81 mM, respectively. Especially, inhibitory effect of ginsenoside $F_1$ on tyrosinase activity was higher than that of arbutin ($IC_{50}$; 2.20 mM). These results indicate that ginsenoside $F_1$ have a potential for industrial cosmetic materials.

비타민나무 추출물 및 착즙 분말의 항산화 활성 및 Nitric Oxide 생성과 Elastase에 대한 저해활성 (Antioxidative and Inhibitory Activities of Extract and Juice Powder from Hippophae rhamnoides L. against Nitric Oxide and Elastase Production)

  • Background: The flowering plant Hippophae rhamnoides L. has been used for many studies on fruit or leaf extracts. This study was conducted to investigate the development of a new cosmetic material from H. rhamnoides fruits and leaves that have by antioxidant, anti-inflammatory and wrinkle improvement activities. Methods and Results: The antioxidant abilities of H. rhamnoides extracts, including of a water-soluble fruit powder (FW), a fatsoluble fruit powder (FF), a supercritical extract of fruit by-product (BS), and a mixture of leaf and fruit (MIX), were investigated in vitro. A DPPH radical assay for antioxidant activity was performed for these fractions alongside assay to evaluate the total phenolic and flavonoid content (TPC and TFC). As expected, the MIX had the highest DPPH radical scavenging activity ($RC_{50}=10.27{\mu}g/m{\ell}$), and the TPC and TFC also were highest in MIX ($225.7mg{\cdot}GAE/g$, and $25.18mg{\cdot}QE/g$, respectively). Nitric oxide (NO) production in LPS-induced RAW264.7 cells was estimated and the results indicated an over 75% decrease of NO production in FF and MIX. In other assays, the highest elastase inhibitory activity was found in FW. Conclusions: These results revealed that H. rhamnoides extracts have a high potential for antioxidant, anti-inflammatory and antiwrinkle activities. H. rhamnoides products are suggested to be applied as the functional materials of cosmetic ingredients.

황기(黃芪)의 elastase 활성과 DPPH, NO 소거능에 미치는 영향 (Effects of Astragali Radix Extracts on the Elastase Activity and DPPH and NO Scavenging Activities)

  • Objectives : Elastic fibers are found in the skin, lungs, arteries, veins and other structures. The defects of elastic matrix aggravate hypertension which is associated with alteration in the great arteries, arteries, and arterioles. The elastase inhibitors were undergoing in clinical studies about emphysema and pulmonary hypertension. This study was designed to investigate the effect of Astragali Radix extracts (AR) on elastase activity and anti-oxidative effects. Methods : The elastase inhibitory activity and DPPH (1,1-diphenyl-2-picrylhydrazyl) and NO free radical scavenging activities of AR were measured. Results : The elastase activity was significantly inhibited by AR. The significant DPPH and NO free radical scavenging activities were observed in AR as well. Conclusion : AR showed the anti-elastase effects and anti-oxidative activities in vitro. These results suggest that AR may be a possible drug for the treatment of pulmonary emphysema and pulmonary hypertension.

산거울 추출물의 Elastase 활성 저해 및 Matrix Metalloproteinase-1 발현 억제 효과 (Inhibitory Effects of Carex humilis Extract on Elastase Activity and Matrix Metalloproteinase-1 Expression)

  • 본 연구에서는 산거울 추출물의 항주름 활성을 연구하기 위하여 항산화, 엘라스타제 활성 억제, Matrix metalloproteinase-1 (MMP-1) mRNA 발현 및 단백질의 생성 억제 관련 실험을 진행하였다. 산거울의 뿌리부분을 95 % 에탄올로 추출하고 유기 용매로 분획하여 각 추출물 분획물에 대한 항산화 활성과 엘라스타제 억제 효능을 측정한 결과, EtOAc 분획이 각각 SC50=4.89 ${\mu}g/mL$, IC50=23.5 ${\mu}g/mL$ 로 가장 우수한 결과를 나타내었다. 따라서 활성이 가장 좋은 EtOAc 분획물로 부터 실리카겔 컬럼 크로마토그래피를 이용하여 활성물질을 분리하고, 분리된 물질이 ${\alpha}$-viniferin임을 분광학적 분석결과로 확인하였다. RT-PCR을 이용한 MMP-1 mRNA 발현능 평가에서 EtOAc 분획물과 ${\alpha}$-viniferin은 각각 50 ~ 60 % (10 ~ 100 ${\mu}g/mL$), 약 60 ~ 75 % (0.5 ~ 2 ${\mu}g/mL$)의 저해효과를 나타내었으며, Western-blot을 이용한 MMP-1 단백질 생성을 평가한 결과에서 역시 EtOAc 분획물과 ${\alpha}$-viniferin은 같은 농도에서 각각 50 ~ 65 %와 55 ~ 65 % 저해 효과를 나타내었다. 결론적으로, ${\alpha}$-viniferin을 함유한 산거울의 EtOAc 분획층의 주름 개선용 기능성 화장품 개발을 위한 소재로 개발 가능성을 확인하였다.

대황(大黃)의 Elastase 활성 억제와 Tyrosinase 억제연구 (The effect of Elastase inhibition and Tyrosinase by Rheum undulatum Stokes extracts)

  • Objective : Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search new materials from the traditional herbal medicines which inhibit the aging process of skin in vitro. Methods : herbal medicines(80% ethanol or water extracts) were screened for their inhibitory activities against elastase. Results : Rheum undulatum (final concentrstion 1 mg/ml) appeared over 30% of inhibition of elastase activity. so we are investigated anti wrinkle effects of Rheum undulatum look through MMP-1 inhibition activity. also Extracts of Rheum undulatum showed higher anti tyrosinase activity than arbutin (final concentration 1 mg/ml). Conclusion : These results suggest that herbal medicines could be strong potential sources of inhibition of anti-aging and whitening effects for the skin.

솔순 열수 추출물의 생리활성 (Physiological Activities of Hot Water Extract from Pine Bud (Pinus densiflora))

  • 본 연구에서는 솔순의 기능성을 규명하기 위해 열수 추출물을 대상으로 여러 생리활성과 아질산염 소거작용에 대하여 분석하였다. 우선, 솔순의 항산화 활성을 측정하기 위하여 DPPH, SOD 유사활성, XO 저해활성을 측정하였다. 그결과 DPPH법을 통해 측정한 솔순의 항산화력은 2 mg/mL에서 71.4%의 radical 소거능을 나타내었으며, SOD 유사활성은 2 mg/mL 농도에서 85.4%로 비교적 높은 활성을 나타내었다. Xanthine oxidase 저해활성 측정 실험에서는 2 mg/mL에서 70.9%의 높은 저해활성을 나타내었다. 솔순 열수추출물의 아질산염 소거능 측정 실험에서는 pH 1.2와 3.0에서 53~59%, pH 6.0에서 40%의 소거능을 나타내었다. 항고 혈압 측정실험에서는 시판되는 항고혈압제 captopril과 비교분석 하였는데, 0.1 mg/mL에서 93.0%였고 같은 농도의 솔순 열수 추출물은 14.6%의 낮은 저해율을 나타냈다. 하지만, 1 mg/mL에서 44.7%의 저해율을 나타내어 솔순 열수 추출물의 비교적 양호한 저해활성을 보였다. 혈당강하능의 지표로 $\alpha$-glucosidase 활성 억제능을 분석한 결과 솔순 열수 추출물 3 mg/mL의 농도에서 43.3%의 저해활성을 나타냈으며, 주름 예방 효과의 지표로 elastase 활성 억제능을 측정한 결과 10 mg/mL의 농도에서 59.6%의 저해능을 보였다. 이상의 결과들은 솔순 열수 추출물의 우수한 생리활성을 증명하고 있는 것으로 천연의 건강 및 미용 소재로써 그 활용도가 높을 것으로 판단된다.

Development of an Agar Diffusion Method to Measure Elastase Inhibition Activity Using Elastin-Congo Red

  • The pancreatic and neutrophil elastases are associated with several illnesses including lung and vascular diseases, various cancers, and pancreatitis. The development of a potent and specific inhibitor to the elastases could lead to new therapies. In this study, an agar diffusion method was modified to include a substrate-dye conjugate (Elastin-Congo red) as a substrate of elastase and an indicator of elastase inhibitory activity. The Elastin-Congo red agar plates consisted of 0.1 % Elastin-Congo red and 2.5% agar. The elastase and elastase inhibitors were simultaneously loaded into wells, ultimately resulting in halo formations in which the halo diameter decreased as the concentration of elastase inhibitor increased. The concentration of elastase inhibitor in the samples, therefore, was inversely proportional to the halo diameters. This simplified method provided an excellent correlation with the standard microplate technique, which uses a chromogenic substrate. The concentration of elastase inhibitor obtained from the culture supernatant of a recombinant elastase inhibitor produced by the yeast Pichia pastoris was easily determined. This study has established a simple modified and inexpensive agar diffusion method that is potentially useful for the identification, quantification, and screening of new elastase inhibitors.

EFFICACY AND BIOLOGICAL ACTIVITIES OF A NEW ANTI-AGING AGENT OBTAINED FROM ARECA CATECHU

  • Inhibitory effects of the new material obtained from Areca catechu seed (CC-516) according to a special process, and its applicability to the skin as a cosmetic raw material in terms of its efficacy were presented. Areca catechu extract out of 150 medicinal plants, exhibited high inhibitory effect on the porcine pancreatic elastase ($IC_{50}$ : $40.8{\mu}$g/ml). It also had an inhibitory effect on the human leukocyte elastase ($IC_{50}$ : 48.1$\mu$g/ml), hyaluronidase ($IC_{50}$ : $416{\mu}$g/ml), antioxidative activity ($IC_{50}$ : $45.4\mu$g/ml) and free radical scavenging activity ($SC_{50}$ : $10.2{\mu}$g/ml). The cream contained 3% of CC-516 improved skin hydration above 16.5%. Especially, the skin elasticity increases more than 35% and skin wrinkles decreased more than 23%. The CC-516 was designed to be utilized in cosmetology. The cream containing 3% of this product has not only protecting effect on the skin mechanical properties provided by the collagen and the elastin in the derm but also restructuring effect of scarring or aging tissue.

흑마늘 추출물을 첨가한 화장품의 기능성 평가: Tyrosinase와 Elastase 저해 활성 및 산화 스트레스에 미치는 영향 중심으로 (Physiological Effects of Formulations Added with Black Garlic Extract on Skin Care: Oxidative Stress, Tyrosinase and Elastase Activities)

  • 본 연구는 흑마늘을 함유한 화장품 시료에 대해 in vitro에서 tyrosinase 및 elastase 저해 활성과 hairless 마우스를 이용해 in vivo에서의 산화스트레스에 대한 항산화 효능을 평가하였다. 그 결과 흑마늘 화장품은 대조 화장품에 비해 유의적으로 낮은 피부 투과율을 나타내었으며(p<0.05, p<0.01) 흑마늘 화장품은 대조 화장품에 비해 유의적으로 높은 tyrosinase(p<0.05, p<0.01, p<0.001)와 elasetase(p<0.01) 활성 억제능을 나타내었다. 또한 UVB로 증가된 피부조직내 지질 과산화물과 과산화수소의 생성에 대해 대조 화장품에 비해 흑마늘 화장품은 더 높은 억제 경향을 나타내었다. 따라서 이러한 본 연구의 결과는 흑마늘이 기능성화장품 소재로의 가능성이 큼을 시사한다.
