Influence of Dietary Supplementation of Condensed Tannins through Leaf Meal Mixture on Intake, Nutrient Utilization and Performance of Haemonchus contortus Infected Sheep

  • The study assessed the effect of dietary supplementation of leaf meal mixture (LMM) containing condensed tannins (CT) on feed intake, nutrient utilization and performance of sheep infected with Haemonchus contortus. Eighteen adult sheep of similar age and body weight ($25.03{\pm}1.52$) were included in this study and out of these, 12 sheep were infected with single dose of infective third stage larvae of H. contortus at 2,000 larvae per sheep. The experimental sheep were allocated in three different groups' i.e. negative control (NC; no infection), control (C; H. contortus infected) and treatment (T; H. contortus infected+CT at 1.5% of the DM through LMM) and the experiment was conducted for a period of 90 d. The intake of dry matter (DM), organic matter (OM) and digestibility of DM, OM, neutral detergent fibre (NDF) and acid detergent fibre (ADF) were comparable among three animal groups. However, digestibility of crude protein (CP) and ether extract (EE) were significantly (p<0.05) higher in NC group as compared to both C and T groups. Nitrogen (N) retention (g/d or % of N intake) was significantly (p = 0.038) lower in C group as compared to T and NC groups. Daily intake (g/kg $W^{0.75}$) of digestible crude protein (DCP), digestible organic matter (DOM) and total digestible nutrient (TDN) did not differ significantly (p<0.05) in the three groups. Haemoglobin (Hb) and packed cell volume (PCV) were significantly (p<0.001) higher in treatment group as compared to control. The level of Hb and PCV reduced (p<0.001) after 30 days of experimental feeding. CT significantly (p<0.001) reduced serum urea in T group as compared to NC and C groups. Serum proteins differed significantly (p<0.01) among the three groups. The activity of serum enzymes AST, ALT, ALP and LDH were also statistically non significant (p<0.05) among treatments. The weight of abomasal lymph nodes (ALN) in T group was higher (p<0.05) than in C group. Treatment group had lower (p<0.05) total worms and fecal egg count compared to control group. It may be concluded that dietary supplementation of CT through LMM significantly improved the N retention, and inhibited the different developmental stages of Haemonchus contortus in experimental sheep.

Separation of Highly Purified Antimicrobial Lysozyme Using Ultrafiltration and Characteristics of Membrane Fouling (한외여과 공정을 이용한 고순도 향균 Lysozyme 의 분리 및 막 침착 특성)

  • The value of lysozyme as a natural food preservative is continuously increased due to its unique antimicrobial activity. To determine the optimum separation concentration among the various hen egg white protein (HEWP) concentrations (0.25, 0.5, 1.0, w/v), protein concentrations, lysozyme concentrations, specific activities (SA), and purification factors of prefiltered solution (PFS) and PM30 permeate solution (PMS) were compared. The purity of lysozyme separated at each step was analyzed and confirmed by gel permeation chromatography and electrophoresis. The fouling deposits on membrane were observed by SEM. The non-enzymatic proteins were removed over 99% by ultrafiltration (UF). The increased feed concentration did not contribute to the increase of SA. SA of PMS was 18 to 31 times higher than that of PFS. The optimum feed concentration was decided as 0.25% based on SA and purification factor. The non-enzymatic region of gel chromatogram was proved to be ovalbumin. The thickness of deposit on the UF membrane was approximately $0.9{\mu}m$ and removed by cleaning with 0.1 N NaOH. Therefore, UF using PM30 membrane was very effective to separate the antimicrobial lysozyme from various HEWPs.

Comparison of Growth and Body Composition in olive flounder Larvae(Paralichthys olivaceus) Fed Domestic Experimental and Imported Commercial Microparticulated Diets (국내 실험 미립자 사료와 수입 미립자 사료를 공급한 넙치(Paralichthys olivaceus) 자.치어의 성장과 체조성 비교)

  • This experiment was conducted for compare domestic experimental microparticulated diets with imported commercial microparticulated diets in olive flounder larvae, Paralichthys olivaceus (Temminck et Schlegel). Fish larve were fed four microparticulated diets from 8th day after hatching. Four diets were two commercial microparticulated diets D and H, and experimental microparticulated diets K1 and K2 formulated each with different protein sources (diet K1 ; squid meal, blood meal, yeast extract, chlorella powder, olive flounder muscle, Lys, and Met ; diet K2 ; whole egg protein, krill meal, short-necked clam meal, squid muscle, live yeast, yeast extract, and casein). There were no significant differences on body weight, body length and survival rates among four diet treatments up to the 40th day after hatching. At the 83th day after hatching, fish fed diet D had a significantly higher survival rate than that of fish fed diet K2, whereas there was no significant difference between to diet H and K1. Fish fed diet D had a significantly higher body weight than these of fish fed diaet K1 and K2, whereas there was no significant difference between fish fed diet D and H. There was no significant difference on EPA and DHA of body fatty acid composition among four diet treatments up to the 83th day after hatching. These results show that nutritionally well-ballanced domestic microparticulated diets for olive flouner lavae can be developed.

Gene expression changes in silkworm embryogenesis for prediction of hatching time

  • The silkworm's dormancy and embryonic development are accomplished through the interaction of various genes. Analysis of the expression of several interacting genes can predict the embryonic stage of silkworms. In this study, we analyzed the changes in the expression level of genes at each stage during the embryonic development of dormant silkworm eggs and selected genes that can predict the hatching time. Jam123 and Jam124 silkworms were collected after egg laying, and the silkworm eggs were preserved using a double refrigeration method and expression analysis was performed for 23 genes during embryogenesis. There were 5 genes showing significant changes during embryogenesis: UDP-glucuronosyltransferases (BmUGTs), heat shock protein hsp20.8 (BmHsp20.8), Cytochromes b5-like proteins (BmCytb5), Krüppel homolog 1 (BmKr-h1), and cuticular protein RR-1 motif 41 (BmCpr41). As a result of quantitative comparison of the expression levels of these 5 genes through real-time PCR, the BmUGTs gene showed a difference between Jam123 and Jam124, making it difficult to see it as an indicator for predicting hatching time. However, the BmHsp20.8 gene had a common expression decreased at the imminent hatching stage. In addition, it was confirmed that the expression level of the BmCytb5 gene decreased to the lowest level at the time of imminent hatching, and the expression of the BmKr-h gene was made only at the time of imminent hatching. The expression of the last BmCpr41 gene can be confirmed only at the time of imminent hatching, and it was confirmed that it shows a rapid increase right before hatching. Taken together, these results suggest that expression analysis of BmHsp20.8, BmCytb5, BmKr-h1, and BmCpr41 genes can determine the stage of embryogenesis, predict hatching time, which facilitate better management of silkworm eggs.

Inhibition of Glycation End Products Formation and Antioxidant Activities of Ilex paraguariensis: comparative study of fruit and leaves extracts

  • Objectives: Ilex paraguariensis (Aquifoleaceae) is cultivated to produce "yerba mate". Due to its nutritional, energizing, hypoglycemic and antioxidant effects, it is used in the elaboration of food, pharmaceuticals, and cosmetics. The oxidative stress related to protein glycation and production of advanced glycation end products (AGEs) leads to the development of several diseases. The objective of this work was to compare the antioxidant and anti-AGEs activity of a decoction of fruits (F) and leaves (L). Methods: The antioxidant activity was assayed by the DPPH assay and the inhibition of egg yolk lipid peroxidation (ILP), and anti-AGEs activity, through the inhibition of the formation of fructosamine (IF), β-amyloid (Iβ), protein carbonylation (IC) and AGEs (IA). Polyphenols were quantified by HPLC. Results: Maximum response ± SEM: For F 0.01 ㎍/mL: IF = 42 ± 4%, IC = 17 ± 2% and for 10 ㎍/mL: IA = 38 ± 4%, Iβ = 67 ± 7%. For L 0.1 ㎍/mL: IF = 35 ± 2%, IC = 19 ± 2% and for 100 ㎍/mL: IA = 26 ± 3%, Iβ = 63.04 ± 2%. The DPPH IC50 = 134.8 ± 14 ㎍/mL for F and 34.67 ± 3 ㎍/mL for L. The ILP IC50 = 512.86 ± 50 ㎍/mL for F and 154.8 ± 15 ㎍/mL for L. By HPLC L presented the highest amounts of flavonoids and caffeoylquinic acids. F and L showed strong anti-AGEs activity, affecting the early stages of glycation at low concentrations and the late stages of glycation at high concentrations. The highest activity for both F and L was seen in the IF and Iβ. F presented the highest anti-AGEs potency. L presented the highest antioxidant potency, which was related to the highest content of polyphenols. Conclusion: The fruits of I. paraguariensis could be a source of antioxidant and anti-AGEs compounds to be used with medicinal purposes or as functional food.

Histological Change of Uterus Endometrium and Expression of the Eggshell-related Genes according to Hen Age (닭의 산란연령에 따른 자궁내막조직의 변화 및 난각 관련 유전자의 발현양상)

  • The eggshell is an intricate and highly ordered structure composed of multiple layers and a calcified matrix. The eggshell is formed at the uterine segment of the chicken oviduct. In this study, histological changes in the uterine endometrium and the expression of the eggshell-related genes were investigated according to hen age. We analyzed the expression of eggshell protein-related genes, such as OCX-32, OCX-36, OC-17, OC-116, and eggshell-ion-related genes, such as CABL-1, SPP1, SCNN1G, ATP2A2, CA2, and CALM1. In chicken uterine endometrium, histological deformation, fibrosis, atrophy and elimination of micro-villi were found with increasing hen age. The concentration of blood-ion components did not significantly change with age. The amount of telomeric DNA in uterine endometrial cells decreased with increasing hen age. The expression of most of the eggshell-related genes changed significantly with increasing hen age. The expression of some ovo-proteins, which play a role in eggshell formation, increased with increasing hen age; however, there were no significant correlations among eggshell protein genes. Eggshell ion-related genes, such as ATP2A2, SCNN1G, CA2, and CALM1, were closely related to each other. The OCX-32 and OCX-36 genes were closely related to some of the eggshell ion genes. Eggshell protein-related genes, such as the OCX-32, OCX-36 genes and ion-related genes such as CALB-1, ATP2A2, SCNN1G, CA2, CALM1, affected eggshell formation, mutually or independently. This study shows that, uterine although endometrial cell damage occurs with increasing hen age, normal eggshells can be formed in old hens. This suggests that eggshell protein-and eggshell ion-related genes also control the homeostasis of eggshell formation.

STUDIES ON THE CYSTINE COMPONENT IN THE SERICULTURAL PROTEINS OF BOMBYX MORI L. (가잠사단백질의 각과정에서의 Cystine 성분에 대한 연구)

  • The purpose of this treatise is to prove the presence of cystine in silk fiber through wide sampling throughout all the sericultural processes of Bombyx mori.; also to show that disulfide cross linkages exist in the silk fiber. The conclusions reached were as follows: 1. Crystalline cystine was obtained from silk fibroin using Folin's Method. 2. Analytical data showing the cystine content of silk fiber and its related materials were obtained using Sullvan's Method as follows: Material Percent Cystine A. Mulberry leaf protein 0.175 B. Silkworm egg 0.33 C. Silkworm Body, matured, fat extracted, without silk gland 0.41 D. Silk gland, matured 1.23 E. Silkworm feces none F. Silkworm pupa, fat extracted 0.30 G. Silkworm moth, fat extracted 0.60 H. Raw Silk 0.22 I. Fibroin 0.175 J. Sericin 0.30 3. The presence of cystine in the silkworm was substantiated the existence of 0.175 % methionine in mulberry leaves and 0.12% methionine in the silk gland. 4. Part of the sulfhydryl compounds in the silk gland is believed to transfer to serine and methionine, with the former being secreted into the liquid silk finally as silk fiber and the latter used for nutritive purposes in the growing of silk gland tissue. 5. The cystine content is variable by mulberry species, silkworm species, sex, breeding process, and other culturing environments. 6. Hybrid silkworms require more nutritive amino acids for effective growth than the original parents, and secrete less of them as silk fiber. 7. From such an observation, the amino acid composition of silk fiber is believed to be fairly flexible. Cystine if included in the amorphous part of the fiber, especially in sericin. 8. The result from enriching the silkworm diet with pure cystine or wool cystine did not result in any advantage, therefore it is believed that the natural cystine and methionine contents in the mulberry leafaregoodenoughforsilkwormnutrition. 9. The disulfide cross linkage in silk fiber was verified by using the Harris Method. Contraction took place following the treatment of the fiber with various salts and acids. Comparisons were made with wool fiber. 10. During these experiments, the fibrious structure of silk fiber and the net-globular liquid form were photographed microscopically. It is believed that the globules of liquid silk are net-formed by the inter attraction of the OH ion of the globular peptide and the H ion of water as shown by the hair cracking behavior of the film. The net-globular protein precipitation from the mulberry protein solution showed that mulberry is a proper diet for the formation of fibrous protein in the silk fiber. 11. The significance of the presence of cystine in silk fiber as emphasized in this paper should result in modification of the general conception that cystine is absent from this fiber. Studies on the Activated Sludge of Food Industries for Animal Feed - Part2. Nutritive Value of Brewery's Activated Sludge - (맥주공장(麥酒工場) Sludge의 사료적(飼料的) 가치(價値)에 관하여 - 제2보(第二報) 식품공장 활성오니(活性汚泥)의 사료화에 관한 연구 -)

  • This experiment was carried out to investigate the nutritive value of brewery's activated sludge on the performance and nutrients utilization of egg type chicken of babcock fed the different levels of sludge. The chemical composition, content of amino acids and mineral in brewery's activated sludge were also analyzed. 3,6,9 and 12% of brewery's activated sludge were supplemented with basal ration as a substituted ingredient to soybean oil meal in experimental ration. The results obtained were as follow: 1. Chemical composition analysis 1) Brewery's activated sludge had 42.50% of crude protein on the air dried basis, and had 15,69% of crude ash, and had 2,060 kcal of metabolizable energy per kg of sludge. 2) Total amino acid content of brewery's activated sludge was 42.50% and 99% crude protein of brewery's activated sludge was a true amino acid, and brewery's activated sludge contained especially more methionine and threonine that those of soybean oil meal. 3) In case of mineral content of brewery's activated sludge, phosphorus, magnesium, copper and iron were plentifully included. However, calcium content in brewery's activated sludge was very low. 2. Feeding trial 1) Body gain of chicken fed the different levels of sludge was decreased in proportion to increasing level of sludge was decreased in proportion to increasing level of sludge. However, no statistical differences were found out between treatments. 2) Diet intake of chicken fed the different levels of sludge was significantly (p<0.05) increased as the supplementation level of sludge in ration increased. 3) Feed conversion of chicken fed the different levels of sludge was high in proportion to increasing level of sludge in ration. However, there were no significant differences between treatments. 3. Digestion trial 1) Utilization of dry matter of chicken fed the different levels of sludge was decreased as the level of sludge in ration increased. However, no statistical differences were found out between treatments. 2) Utilization of crude protein of chicken fed the different levels of sludge was significantly (p<0.01) increased as the level of sludge was higher. Utilization of crude protein of control treatment and of sludge 3% treatment was higher than that of other treatments. 3) Utilization of crude ash of chicken fed the different levels of sludge was significantly (p<0.05) decreased in proportion to increasing level of sludge in ration. 4) Utilization of NFE of chicken fed the different levels of sludge was slightly decreased in proportion to increasing level of sludge in ration. However, no statistical differences were found out between treatments. Therefore according to this experiment, it may be concluded that brewery's activated sludge can be supplemented with chicken ration by $6{\sim}9%$.

Nutritional Adequacy of Target Pattern in Dietary Guidance System - Dietary Reference Intakes for Koreans 2015 - (식사구성안 권장식사패턴의 영양 적정성 평가 - 2015 한국인 영양소 섭취기준 -)

  • Nutritional adequacy of Target patterns in Dietary Guidance System were evaluated. Study subjects were 18 Target patterns designed for 2 children groups, 4 adolescent and 4 adult male and female groups in Dietary Reference Intakes for Koreans 2015. Nutrients examined were 4 macro-nutrients(protein, fat, carbohydrate, and dietary fiber), 5 minerals(Ca, P, Fe, Na, and K), and 5 vitamins(vitamin A, thiamin, riboflavin, niacin, and vitamin C). Energy and nutrients contents were calculated based upon assigned multiples of single serving in Target pattern and Representative Food Composition Table by Excel program. Among the 18 Target patterns, only 5 were mean adequacy ratio(MAR) of 1.0, fulfilled the expectations of Dietary Guidance System. Protein and Na contents were more than dietary reference intakes(DRI) in all 18 Target patterns. Dietary fiber, Ca, and K were deficient nutrient in 15, 10, and 9 Target patterns, respectively. The units of vitamin A were not same in Target pattern and DRI, retinol equivalent(RE) in Target pattern and retinol activity equivalent(RAE) in DRI. When calculate RE content of vitamin A in Target pattern to RAE content, 15 Target patterns did not reach to DRI of vitamin A. The Target pattern of elderly women showed the lowest mean adequacy ratio(MAR), 7 nutrients were less than DRI, need to be revised. Average energy contribution ratio of protein, fat, and carbohydrate were 16.04%, 24.35%, and 60.91%, respectively, fulfilled the adequate acceptable macro-nutrient distribution range(AMDR). To improve the nutritional inadequacy of dietary fiber, Ca, K, and vitamin A in Target pattern, effective mean may be the increasing vegetable group assignment. Also decreasing the meat fish egg legume group assignment in some Target pattern may be the way of preventing of protein overconsumption. The energy in adequate amount for condiment use also need to be considered.

  • Fertilized egg, by successive cell divisions, differentiates into different tissues and organs with various structures and functions. Different cells and tissues contain different proteins, products of selective gene expression. Not all the genes in any genomes are equally active, temporal and spatial gene expression being the general rule. Present paper attempts to review the tanscriptional mechanisms or the initiations of transcription from several angles. In some of the organisms the genes in the process of transcription or the genes in the inactive state can be seen under the light microscope. Some bands of Drosophila polytene chromosomes may exhibit a swollen or puff appearance under certain conditions. A puff, unfolded or decondensed form of chromomere, represents sets of intense transcriptional activity or RNA synthesis. The heterochromatic X chromosome whose genes remain inactive in the female mammals can be visualized as a dark staining structure called Barr body, Configuration of chromatin differs between transcribed and nontranscribed chromatin. Modification to the chromatin facilitates RNA synthesis. The movement of large polymerase molecule along the DNA would probably be facilitated if some modifications of the chromatin configuration is effected. Methylation of cytosines in CG sequences is associated with inactive genes. Methylation can play a role in determination of mammalian cells during embryogenesis. Demethylation is necessary for the gene to be expressed during development A histone modification that is also known to be correlated with transcriptional capacity of chromatin is acetylation of the lysine residues of the core histones. Chromatin containing a high level of histone acetylation is very sensitive to DNase 1. For the transcription to occur TBP must first bind to the TATA box. Another TF, TF IIB, then binds to the promoter-TBP complex, facilitating the access of RNA polymerase to the transcription initiation site. As recently as eight years ago researchers assumed that histones were irrelevant to the regulation of gene expression. Histones combine with the DNA to form nucleosome of the chromatin. Histones are vital participant in gene regulation. Histone and basal factors compete for access to TATA box. When DNA is exposed to basal factors before histones are introduced, the basal factors assemble on TATA boxes preventing the access of histones, allowing transcription to occur, for transcription to begin, activator protein at the upstream activation sequence or enhancer must interact with the tail of histone H4 at TATA box and cause the histone role particle to dissociate from the TATA box leading to partial breakup of the histone core particle and allowing the basal factors to bind to the TATA box. New concept of genomic flux in contrast to the old concept of static genome has been developed based on the powerful new molecular techniques. Genomic changes such as repetitive DNAs and transposable elements, it is assumed but not yet proved, may affect some of the developmental patterns that characterize particular cells, tissues, organs, and organisms. In the last decade or so remarkable achievement have been made in the researches of the structures and functions of TFs and the specific target sequences located in promoters or enhancers where these TFs bind. TFs have independent domains that bind DNA and that activate transcription. DNA binding domain of TFs serves to bring the protein into the right location. There are many types of DNA binding domains. Common types of motifs can be found that are responsible for binding to DNA. The motifs are usually quite short and comprise only a small part of the protein structure. Steroid receptors have domains for hormone binding, DNA binding, and activating transcription. The zinc finger motif comprises a DNA binding domain. Leucine zipper consist of a stretch of amino acids with a leucine residue in every seventh position Two proteins form a dimer because they interact by means of leucine zippers on similar α-helical domain. This positions their DNA binding basic domains for interaction with the two halves of a DNA sequence with dyad symmetry of TGACTCA, ACTGAGT.
