  • Opisthorchis viverrini and Clonorchis sinensis are parasites known to be carcinogenic and causative agents of cholangiocarcinoma in Asia. The standard method for diagnosis for those parasite infections is stool examination to detect parasite eggs. However, the method has low sensitivity, and eggs of O. viverrini and C. sinensis are difficult to distinguish from each other and from those of some other trematodes. Here, we report a multiplex real-time PCR coupled with high resolution melting (HRM) analysis for the differentiation of O. viverrini and C. sinensis eggs in fecal samples. Using 2 pairs of species-specific primers, DNA sequences from a portion of the mitochondrial NADH dehydrogenase subunit 2 (nad 2) gene, were amplified to generate 209 and 165 bp products for O. viverrini and C. sinensis, respectively. The distinct characteristics of HRM patterns were analyzed, and the melting temperatures peaked at $82.4{\pm}0.09^{\circ}C$ and $85.9{\pm}0.08^{\circ}C$ for O. viverrini and C. sinensis, respectively. This technique was able to detect as few as 1 egg of O. viverrini and 2 eggs of C. sinensis in a 150 mg fecal sample, which is equivalent to 7 and 14 eggs per gram of feces, respectively. The method is species-specific, rapid, simple, and does not require fluorescent probes or post-PCR processing for discrimination of eggs of the 2 species. It offers a new tool for differentiation and detection of Asian liver fluke infections in stool specimens.

  • A 43-yr-old patient visited our department with intermittent chest pain he had suffered with for the past 2 months. Chest CT showed an egg shaped mass in the left chest wall. Local resection was performed for distinguishing the mass from a neurogenic tumor. The diagnosis was periosteal Ewing sarcoma of the rib. Since there was no evidence of metastasis based on the PET-CT, a 2nd operation was done with wide resection and thoracoplasty. The patient was then treated with combined chemotherapy. There has been no local recurrence for the fast 1 year. The patient's age and tumor origin were distinct from the usual characteristics of Ewing's sarcoma. Periosteal Ewing's sarcoma of the rib has rarely been reported. We report here on a case of periosteal Ewing's sarcoma of the rib along with a review of the relevant medical literature.

  • The seasonal fluctuation of the large 28-spotted lady beetle, Henosepilachna vigintioctomaculata (Motschulsky), was observed in potato fields of Gangneung region, Gangwondo Province. The effects of their leaf consumption over a 24 hour period and developmental periods by different temperatures were investigated, and then the developmental threshold (DT) and effective cumulative temperature (ET) of each stage were calculated. Various developmental stages of H. vigintioctomaculata were captured in the fields from May to September, with three peaks in late May, late June, and late August. Leaf consumption by larval stages increased with the development of instar. The 4th instar larvae consumed potato leaves much more than other stages. Egg hatchability and emergence rate of H. vigintioctomaculata were lower at $10^{\circ}C$ and $30^{\circ}C$, but higher at around $20{\sim}23^{\circ}C$. The developmental periods of each stage became shorter as temperature increased; 90 days at $10^{\circ}C$, 40 days at $20^{\circ}C$ and 25 days at $25^{\circ}C$. Base on developmental velocity to temperature, calculated DT and ET were $7.3^{\circ}C$ and 83.

  • Sustained-releasing praziquantel (SRP) tablet was designed for single dose treatment regimen of clonorchiasis. A previous pre-clinical study confirmed its sustained-releasing characteristics and a better cure rate than conventional praziquantel (PZQ). In this clinical study, the pharmacokinetics of this SRP tablet were investigated in human volunteers (phase 1; 12 volunteers), and its curative efficacy was examined in clonorchiasis patients (phase 2; 20 volunteers). In the phase 1 clinical study, blood concentrations of both tablets showed wide individual variation. The $AUC_{last}$ of SRP was $497.9{\pm}519.0ng{\cdot}hr/ml\;(mean{\pm}SD)$ and PZQ of $628.6{\pm}695.5\;ng{\cdot}hr/ml$, and the $AUC_{inf}$ of SRP was $776.0{\pm}538.5\;ng{\cdot}hr/ml$ and of PZQ $658.6{\pm}709.9\;ng{\cdot}hr/ml$. $C_{max}$ values of SRP and PZQ were $90.7{\pm}82.2ng/ml\;and\;214.9{\pm}251.9\;ng/ml$, and $T_{max}$ values were $3.42{\pm}1.43\;hr\;and\;1.96{\pm}1.23\;hr$, respectively. SRP tablets showed similar AUC values, but lower $C_{max}$ and longer $T_{max}$ values than PZQ. In the phase 2 study, SRP at 30 mg/kg (single dose) achieved a 60% cure rate and a 95.5% egg reduction rate. The cure rate of a single dose SRP was unsatisfactory compared with that of the conventional PZQ dose, but much better than that achieved by a single dose PZQ.

  • Mushroom sciarid fly Lycoriella ingenua (Dufour), syn. L. mali (Fitch) is one of the most common fly pests affecting the mushroom cultivation in Korea. This study was carried out to investigate the development at different temperatures and damages of L. ingenua in A. bisporus mushroom cultivation. Rearing of mushroom flies were carried out on mycelial culture in Petri dishes. The development of L. ingenua from egg to adult at the temperature of 16, 20, 24, $28^{\circ}C$ were 35.2, 25.8, 23.5, and 22.2 days, respectively. Adult flies invade mushroom farms and oviposit in freshly spawned compost. Damages are mainly caused by the larvae of 1-7 mm, which feed on growing mycelium and also developing stipes and caps of mushrooms. Adult flies spoil the appearance of the fruiting bodies, spread various fungal and bacterial disease, and transmit mites. The damages by mushroom flies increased without pest control causing yield loss of up to 27%.

  • The objective of this study was to evaluate the characteristics of Korean Native Cattle sperm frozen-thawed with L-cysteine and/or catalase. The semen from bulls was collected by the artificial vagina method, and Triladyl containing 20% egg-yolk and/or L-cysteine (L), catalase (C) and L-cysteine + catalase was added to the diluted semen for cryopreservation. The results showed that sperm viability was significantly higher in the L-cysteine + catalase ($69.49{\pm}3.16%$) group than in the control ($60.5{\pm}3.94%$) group (p<0.05). Acrosome damage was significantly lower in the L-cysteine ($17.12{\pm}1.08%$) group than in the control ($21.46{\pm}1.14%$), catalase ($20.54{\pm}0.76%$), and L-cysteine + catalase ($19.29{\pm}0.65%$) groups (p<0.05). In addition, the level of intact mitochondria in the spermatozoa was significantly higher in the L-cysteine ($58.65{\pm}1.39%$) group than in the control ($50.63{\pm}2.37%$) group (p<0.05). The hydrogen peroxide level in the frozen-thawed sperm was significantly lower in the L-cysteine ($3.74{\pm}1.66%$), catalase ($4.65{\pm}1.87%$), and L-cysteine + catalase ($8.11{\pm}2.15%$) groups than in the control ($13.22{\pm}1.6%$) group (p<0.05). The glutathione level was significantly higher in the L-cysteine ($1.33{\pm}0.03%$) group than in the control ($1.08{\pm}0.06%$), catalase ($1.05{\pm}0.02%$) and L-cysteine + catalase ($1.11{\pm}0.03%$) groups (p<0.05). In conclusion, L-cysteine and catalase could protect the membrane of Korean Native Cattle sperm from damage during sperm cryopreservation. Especially, L-cysteine was more effective for keeping acrosomes and mitochondria intactness during sperm cryopreservation.

  • EPN-resistant diamondback moth were tested the developmental characteristics, longevity, fecundity and synergistic effect with synergists when treated with the bistrifluron, an inhibitor of chitin synthesis. Inhibition rate of egg hatch was below at the recommended concentration (50 ppm) to the EPN-resistant strain than EPN-non selected strain, however, mortality within 24hr after hatching was showed as 50-60%. They did not show the difference between two strains. Mortality of larval instars were showed effective to two strains with no difference. Inhibition rate of emergence did not show any effect when treated with pupae. Moreover, they did not affect to the longevity and fecundity. When bistrifluron was treated at 50 ppm to the adult, however, longevity and fecundity was decreased with no difference between two strains. Bistrifluron to the EPN-resistant strain showed synergistic effect as high as 137.7 and 73.1 folds with synergist such as PBO (microsomal oxidase inhibitor) and polyoxin B (chitin synthetase inhibitor), respectively.

  • This study was carried out to measure and analyze the vibration levels and modes produced by a hopper type feeder used at laying hen farms, to prevent the vibration resonance and to reduce the vibration of the hopper type feeder (6-tier). The most vibration in a layer house were produced by hopper type feeder as shown the previous study. According to the measurement results, the ratio of transmissibility was as high as 100-638% for natural frequency of 170 Hz or less. And vibration simulations were taken by the results of vibration mode analysis and the weakest points to vibrations were determined accordingly. Then measurement and analysis were taken for those points. The quantities of vibration were 4.6354-4.9118 g($g=9.81^m/s^2$) by axis. In this study, it was found that hopper type feeder generated vibration as much as to influence the ratio of laying eggs of layer. And by the analysis of vibration on the weakest points, design methods and equipment were suggested for isolating/reducing of vibration by each axis.

  • Mites were collected from organic cultivations of greenhouse cucumbers and identified as Tyrophagus neiswanderi (Acari: caridae). T. neiswanderi (length, $490.1{\mu}m;$ width, $288.1{\mu}m$) is a very small, milky-white, and egg-shaped mite, and it mainly causes damage to the leaves, flowers, and fruits of cucumber plants. In the early growing season of cucumbers, the shoots of seedlings became pale and yellow because of T. neiswanderi, and eventually shrinkage or bud-failing was observed in the plants. In the middle of the growing season, T. neiswanderi caused white spots on the leaves and flowers of the plants, and the spots gradually became holes. T. neiswanderi also caused severe damage to young fruits by feeding on the rinds of the fruits, inducing malformations and lowering the economic value.

  • This study was designed to evaluate the effect of L-cysteine on sperm characteristics and oocyte cleavage in vitro in Korean native cattle. For this study, the freezing of diluted semen were added with Triladyl containing 20% egg-yolk and/or 0, 5, 10 and 20 mM L-cysteine before cryopreservation. The viability in frozen-thawed sperm were estimated by SYBR14/PI double stain, acrosome damage with FITC-PNA, mitochondria intact with Rhodamin123 and hydrogen peroxide($H_2O_2$) level with carboxy-DCFDA by flow-cytometry. The developmental capacity was also assessed with cleavage rates in oocytes fertilized in vitro by frozen-thawed sperm. In results, the sperm viability was significantly increased in 10 mM and 20 mM concentrations of L-cysteine than other groups (p<0.05). In addition, acrosome damage was significantly decreased in 10 mM and 20 mM concentrations of L-cysteine than other groups (p<0.05). The mitochondria intact was also significantly increased in 10 mM and 20 mM concentrations of L-cysteine than other groups (p<0.05). On the other hand, the cleavage rates were significantly increased in 0 mM, 5 mM and 10 mM groups than 20 mM concentration of L-cysteine (p<0.05). The oocyte degeneration of oocytes were significantly decreased in 0 mM, 5 mM and 10 mM groups than in 20 mM L-cysteine group (P<0.05). However, there are no significantly differences among the L-cysteine treatment groups. We suggest that concentration of 10 mM L-cysteine have beneficial impact for sperm cryopreserved in Korean native cattle. This result also could be recommended for artificial insemination program if supported by an improvement in the fertility results and required further study.
