Effects of Protein Functionality on Myofibril Protein-Saccharide Graft Reaction

  • The myofibril protein (MP) isolate-saccharide graft reactions was prepared using the Maillard reaction with saccharides. The effects of various saccharides on protein functionality and quality of the Maillard reaction were investigated and compared with those of MP. The grafting degree of the MP isolate-saccharide graft reaction was significantly higher in the reducing sugar-treated groups (lactose, glucose, fructose, and palatinose). The browning intensity of the MP isolate-saccharide graft reaction with fructose, sucrose, and erythitol was higher than that observed in the control reaction (p<0.05). MP that reacted with reducing sugars (glucose, fructose, palatinose, and lactose) had fainter bands than MP that reacted with non-reducing sugars (sucrose, erythitol, trehalose, sorbitol, and xylitol). MPs conjugated with glucose exhibited higher protein solubility. The palatinose and lactose treatments were maximum in water binding capacity, though no significant difference in oil binding capacity among the saccharide treatments was observed. The emulsion stability of the MP isolate-saccharide graft reaction with palatinose and erythitol was higher than that of the control reaction. Therefore, reducing sugars have good protein functionality in the MP isolate-saccharides graft reaction.

당류 및 유화제 첨가농도에 따른 찹쌀떡 저장 중 물성 변화 (Changes in the Texture Properties of Glutinous Rice Cake under Various Concentrations of Saccharides and Emulsifiers during Their Storage at 4℃)

  • 찹쌀떡의 경화 억제 및 저장성 개선을 위하여 대규모 떡 생산업체에서 실제 사용되고 있는 당류 및 유화제를 이용하여 저장 중 물성 변화를 조사하고자 하였다. 이때 유화제 및 당류의 첨가농도는 떡 생산업체에서 사용빈도가 높은 농도를 중심으로 설정하였다. 즉, 찹쌀떡에 첨가한 유화제는 에스피로 0.1, 0.5, 1%로 첨가하였고, 당류로는 설탕을 5, 10, 15%, 저당을 5, 10, 20%, 맥아당을 10, 15, 20%로 각각 첨가하여 찹쌀떡을 제조한 뒤 $4^{\circ}C$에 저장하면서 물성을 측정하였다. 그 결과, 에스피 첨가농도는 0.5%일때, 설탕 첨가농도는 15% 일때, 저당 첨가농도는 20%일때, 맥아당 첨가농도는 10% 일때 저장기간에 따른 물성 변화가 완만하게 일어나고 찹쌀떡의 경화 억제에 효과적인 것으로 나타났다.

Effects of natural mono- and di-saccharide as alternative sweeteners on inflammatory bowel disease: a narrative review

  • Objectives: The incidence of inflammatory bowel disease (IBD) is increasing globally, and excessive added sugar consumption has been identified as one of the contributing factors. In the context of IBD, it is essential to explore functional sweeteners that can improve metabolic health and minimize the risk of IBD-related symptoms. This review article aims to shed light on the effects of natural mono- and di-saccharides as alternative sweeteners, specifically focusing on potential benefits for IBD. Methods: A comprehensive literature review was performed using PubMed and Google Scholar databases with articles published after the year 2000. The search terms 'IBD', 'added sugar', 'sweeteners', 'mono-saccharide', and 'di-saccharide' were combined to retrieve relevant articles. A total of 21 manuscripts, aligning with the objectives of the study, were selected. Papers focusing on artificial or high-intensity sweeteners were excluded to ensure relevant literature selection. Results: Multiple studies have emphasized the association between the high consumption of added sugars such as simple sugars and the increased risk of developing IBD. This is suggested to be attributed to the induction of pro-inflammatory cytokine productions and dysbiosis of the gut microbiota. Consequently, there is a growing demand for safe and functional sweeteners, in particular mono- and di-saccharides, that can serve as alternatives for IBD patients. Those functional sweeteners regulate inflammation, oxidative stress, and Intestinal barrier protection, and restore microbiome profiles in various IBD models including cells, animals, and humans. Conclusions: Understanding these mechanisms resolves the link between how sugar consumption and IBD, and highlights the beneficial effects of natural alternative sweeteners on IBD when they were administered by itself or as a replacement for simple sugar. Further, exploration of this relationship leads us to recognize the necessity of natural alternative sweeteners in dietary planning. This knowledge could potentially lead to more effective dietary strategies for individuals with IBD.

Effects of Mono- and Polysaccharides on In Vitro Fertility of Boar Spermatozoa

  • This study was conducted to examine the effect of several saccharides on the induction of capacitation and acrosome reaction (AR) and to examine the effects of mono and polysaccharides on the penetration activity of boar spermatozoa. Spermatozoa were inseminated in medium with fucose, galactose and mannose as monosaccharide, and fucoicIan. galactan and marman as polysaccharide. The penetration rates were significantly (p<0.05) lower in medium with galactose (40.6%), mannose (38.1%), fucose (41.6%) and fucoidan (36.6%) compared with control (56.7%). The rates of AR were increased (40.7 to 59.8%) by the preincubation periods prolonged from 0 to 4 hr (p<0.05). Similar tendencies were observed in AR when spermatozoa were treated with monosaccharides, but not significantly differ among the groups treated with different time of preincubation with some exception of galactose. When spermatozoa were treated with polysaccharides, the rates of AR were significantly (p<0.05) increased by preincubation time prolonged from 0 to 4 hr with an exception of fucoidan. In conclusion, the present study suggests that penetration rate of spermatozoa is higher in presence of polysaccharides than monosaccharides. Also, it may resume that the comparing to control, the all saccharides (L-fucose, D-galactose, D-mannose, fucoidan. galactan and mannan)-treated groups slightly increase the AR pattern as preincubation time prolonged.

EFFECTS OF FRUCTO-OLIGOSACCHARIDES ON MILK-YIELD AND MILK-COMPONENTS OF DAIRY COWS

  • Fructo-oligosaccharides are found in many plants, such as onion, burdock and wheat. They are not well hydrolyzed by digestive enzymes in animals, but are peculiarly assimilated by Bifidobacterium and some useful bacteria. In our previous experiment (Kobayashi et al., 1987) it was suggested that they were effective in decreasing energy loss in the metabolism of dairy calves. In the present study, the effects of fructo-oligosaccharides on body weight, milk-yield and milk-components (fat, protein and solids-not-fat) were investigated in dairy cows. Lactating cows were fed a standard diet containing fructoligo saccharides at 18.70g, 9.35g and 0.0g (control) per 100kg body weight, day for three weeks. Neither treatments significantly affected any of the parameters examined. The fructo-oligosaccharides were assumed to be hydrolyzed by rumen microorganisms and hardly to affect the bacterium florae in the intestines of the lactating cows.

Effect of Additives on the Fermentation Quality and Residual Mono- and Disaccharides Compositions of Forage Oats (Avena sativa L.) and Italian Ryegrass (Lolium multiflorum Lam.) Silages

  • This study aimed to evaluate the effects of silage additives on the fermentation qualities and residual mono- and disaccharides composition of silages. Forage Oats (Avena sativa L.) and Italian Ryegrass (Lolium multiflorum Lam.) were ensiled with glucose, sorbic acid and pre-fermented juice of epiphytic lactic acid bacteria (FJLB) treatments for 30 days. In both species grass silages, although the respective controls had higher contents of butyric acid (20.86, 33.45g $kg^{-1}$ DM) and ammonia-N/total nitrogen (100.07, 114.91 g $kg^{-1}$) as compared with other treated silages in forage oats and Italian ryegrass, the fermentation was clearly dominated by lactic acid bacteria. This was well indicated by the low pH value (4.27, 4.38), and high lactic acid/acetic acid (6.53, 5.58) and lactic acid content (61.67, 46.85 g $kg^{-1}$ DM). Glucose addition increased significantly (p<0.05) lactic acid/acetic acid, and significantly (p<0.05) decreased the values of pH and ammonia-N/total nitrogen, and the contents of butyric acid and volatile fatty acids as compared with control, however, there was a slightly but significantly (p<0.05) higher butyric acid and lower residual mono- and di-saccharides as compared with sorbic acid and FJLB additions. Sorbic acid addition showed the lowest ethanol, acetic acid and ammonia-N/total nitrogen, and highest contents of residual fructose, total mono- and di-saccharides and dry matter as well as high lactic acid/acetic acid and lactic acid content. FJLB addition had the lowest pH value and the highest lactic acid content, the most intensive lactic acid fermentation occurring in FJLB treated silages. This resulted in the faster accumulation of lactic acid and faster pH reduction. Sorbic acid and FJLB additions depressed clostridia or other undesirable bacterial fermentation, thus this decreased the water-soluble carbohydrates loss and saved the fermentable substrate for lactic acid fermentation.

배양액 제조에 사용된 당의 종류와 농도 및 배양 온도가 Tea Fungus발효 홍차의 pH 변화와 Total Acids생성에 미치는 영향 (Effect of Saccharides and Incubation Temperature on pH and Total Acidity of Fermented Black Tea with Tea Fungus)

  • 항암, 시력 회복, 변비 , 당뇨병 , 신장병 , 불면증 등에 치료 호과가 있어 건강 음료로 이용되고 있는 tea fungus 발효 홍차를 당의 종류(자당, 포도당, 과당, 옥수수물엿), 농도(10-60% w/v), 그리고 배양 온도별($5-50^{\circ}C$)로 제조하여 발효액의 pH변화와 산생산성을 보았다. 자당과 포도당을 첨가하여 제조한 홍차 용액 중에서 산생성 경향이 서로 유사하고, 자당과 옥수수물엿을 사용한 홍차 용액에서 산생성 경향이 서로 유사한 것으로 보아 자당보다 과당의 이용성이 높은것으로 추정된다. 배양액 표면의 막 형성과 생육, 그리고 생성을 위한 최적 온도는 $30^{\circ}C$였다. 20%이상의 자당과 과당을 사용한 홍차 용액을 $30^{\circ}C$에서 14일간 배양하였을 때 0.1-0.2%의 낮은 산 생성을 보였으며, 10%의 자당과 과당을 사용한 홍차 용액에서 14일간 배양시킨 발효 홍차의 total acidity는 각각 1.8% w/v(자당)과 0.68% w/v(과당)가 되었다. 발효 홍차의 pH는 배양 2일 후부터 당의 증류, 농도, 배양 온도에 크게 영향을 받지 않고 모든 배양액에서 2.74-3.56으로 떨어져 배양 14일까지 같은 수준의 pH를 유지하였다.

Fermentation Quality of Italian Ryegrass (Lolium multiflorum Lam.) Silages Treated with Encapsulated-glucose, Glucose, Sorbic Acid and Pre-fermented Juices

  • This experiment was carried out to evaluate the effects of adding encapsulated-glucose, glucose, sorbic acid or prefermented juice of epiphytic lactic acid bacteria (FJLB) on the fermentation quality and residual mono- and disaccharide composition of Italian ryegrass (Lolium multiflorum Lam) silages. The additive treatments were as follows: (1) control (no addition), (2) encapsulated-glucose addition at 0.5% for glucose, (3) glucose addition at 1%, (4) sorbic acid addition at 0.1%, (5) FJLB addition at a theoretical application rate of $2.67{\times}10^5$ CFU (colony forming unit) $g^{-1}$, on a fresh weight basis of Italian ryegrass. Although control and encapsulated-glucose treatments had higher contents of butyric acid (33.45, 21.50 g $kg^{-1}$ DM) and ammonia-N/Total nitrogen (114.91, 87.01 g $kg^{-1}$) as compared with the other treated silages, the fermentation in all silages was clearly dominated by lactic acid. This was well indicated by the low pH (4.38-3.59), and high lactic acid/acetic acid (4.39-22.97) and lactic acid content (46.85-121.76 g $kg^{-1}$ DM). Encapsulated-0.5% glucose and glucose addition increased lactic acid/acetic acid, and significantly (p<0.05) decreased ammonia-N/total nitrogen, and the contents of butyric acid and total volatile fatty acids (VFAs) as compared with the control. However, there were higher butyric acid and lower residual mono-and di-saccharides on the two treatments as compared with sorbic acid and FJLB addition, and their utilization efficiency of water soluble carbohydrates (WSC) was lower than that of both sorbic acid and FJLB additions. Sorbic acid addition showed the lowest content of ethanol and ammonia-N/total nitrogen, and the highest content of residual fructose and total mono-and disaccharides as well as the higher lactic acid/acetic acid value. Sorbic acid addition decreased the loss of mono-and disaccharides, and inhibited the activity of clostridial and other undesirable bacteria, and greatly increased the utilization efficiency of fermentable substrates by epiphytic LAB. FJLB addition had the lowest pH value and the highest lactic acid content among all additive treatments, with the most intensive lactic acid fermentation occurring in FJLB treated silage. This resulted in the faster accumulation of lactic acid and faster pH reduction. Sorbic acid and FJLB addition depressed clostridia or other undesirable bacterial fermentation which decreased the WSC loss and saved the fermentable substrate for lactic acid fermentation.

호상 요구르트의 점도에 미치는 균주의 영향 (The Effect of Starter Culture on Viscosity of Stirred Yogurt)

  • This study was to review recent reports in effects of various starter cultures on the viscosity in stirred yogurt. The rheological properties of yogurt have received considerable attention in the literature. Most yogurts are typically made by mixed cultures of Lactobacillus bulgaricus and Streptococcus thermophilus. The viscosity of yogurt made by mixed cultures was much higher than that of yogurt by single cultures. Since texture of stirred yogurt is the result of both acid aggregation of casein micelles and production of exopoly-saccharides, it is suggested that yogurt be made by the exopolysaccharide-producing cultures in order to increase viscosity, Both types of exopolysaccharides are capsule and loose slime(ropy). But it is desirable to use encapsulated nonropy strains. And Bifidobacteria affects adversely to the viscosity of yogurt. Therefore, starter cultures which have an effect on yogurt viscosity have been widely demonstrated. This review is the search for the development of viscosity in stirred yogurt.

Immobilization of Styrene-acrylamide Co-polymer on Either Silica Particles or Inner Surface of Silica Capillary for the Separation of D-Glucose Anomers

  • Styrene-acrylamide co-polymer was immobilized on porous partially sub-$2{\mu}m$ silica monolith particles and inner surface of fused silica capillary ($50{\mu}m$ ID and 28 cm length) to result in ${\mu}LC$ and CEC stationary phases, respectively, for separation of anomeric D-glucose derivatives. Reversed addition-fragmentation transfer (RAFT) polymerization was incorporated to induce surface polymerization. Acrylamide was employed to incorporate amide-functionality in the stationary phase. The resultant ${\mu}LC$ and CEC stationary phases were able to separate isomers of D-glucose derivatives with high selectivity and efficiency. The mobile phase of 75/25 (v/v) acetonitrile (ACN)/water with 0.1% TFA, was used for HPLC with a packed column (1 mm ID, 300 mm length). The effects of pH and ACN composition on anomeric separation of D-glucose in CEC have been examined. A mobile phase of 85/15 (v/v) ACN/30 mM sodium acetate pH 6.7 was found the optimized mobile phase for CEC. The CEC stationary phase also gave good separation of other saccharides such as maltotriose and Dextran 1500 (MW~1500) with good separation efficiency (number of theoretical plates ~300,000/m).
