Water Uptake, Cotyledon Damage after Imbibition and Hypocotyl Elongation in Soybean with Different Seed Size and Color (콩 종실크기 및 종피색에 따른 침종후 수분흡수특성, 자엽손상 및 배축 신장력의 차이)

  • The experiment was conducted to determine if seed size and seed coat color of soybean might be effective in water uptake and cotyledon damage after imbibition, and hypocotyl elongation. Eight soybean cultivars were separated into two classes of large and small seed based on seed weight, and each class included two cultivars with yellow and black seed color, respectively. Small seed size group was superior in water uptake by seed for 24 hour in imbibition at $25^{\circ}C$ , but its differences decreased as soaking time increased. Small seed cultivars germinated faster and had better germination rate than large ones. However, cultivars with black seed coat showed more slow water uptake at initial time and faster germination than yellow seed, but in 24 hour after imbibition, cultivars with black seed coat had higher water uptake rate than yellow seeds. Small seed cultivar group showed no cotyledon damage in imbibition for 24 hour while large seed cultivars were damaged 78% of cotyledon, and black seed showed low cotyledon damage compared to yellow seed. Hypocotyl length was shorter in large seed rather than in small seed, but hypocotyl thickness in large seed was more thick than in small seed. In correlation coefficients, seed coat rate, embryo rate exhibited significantly negative association with seed weight, and the correlation of seed weight with water uptake in 3 hour after soaking was significantly negative, but in 24 hour showed positive correlation.

Construction of Tomato yellow leaf curl virus Clones for Resistance Assessment in Tomato Plants (토마토 작물의 TYLCV 저항성 평가에 이용할 수 있는 감염성 클론 개발)

  • Five isolates of Tomato yellow leaf curl virus (TYLCV) collected from various regions of Korea were amplified using PCR and determined the sequences of full-length genome, respectively. The PCR-amplified DNA of each TYLCV isolate was introduced into a binary vector to construct infectious clone containing 1.9 copies of the corresponding viral genome. Various cultivars and breeding lines of tomato were inoculated with Agrobacterium tumefaciens harboring infectious clone of each TYLCV isolate to assess resistance against TYLCV. Susceptible cultivar 'Super-sunread' revealed typical yellowing and narrowing of the upper leaves. In contrast, breeding linesTY12, GC9, GC171, and GC173, which contained the TY-1 and/or TY-3 genes that confer resistance against TYLCV in nature, were completely symptomless, suggesting that the lines were resistant to challenging TYLCV isolates. Symptoms of TYLCV in susceptible tomato cultivars are significantly different from those of TYLCV in the resistant tomato cultivars at 30 days after agroinfiltration. Although genomic DNAs of TYLCV were detected from the breeding lines TY12, GC9, GC171, and GC173 using real-time PCR analysis with specific primers, levels of TYLCV DNA accumulation in the resistant breeding lines were much lower than those of TYLCV DNA accumulation in susceptible tomato cultivars. Similar symptom severity and levels of TYLCV DNA accumulation were observed from TYLCV infections mediated by Bemisia tabaci in the resistant and susceptible tomato cultivars. Concentration of agrobacterium did not affect the response of tomato cultivars against TYLCV inoculation. Taken together, these results suggest that TYLCV inoculation via agroinfiltration is as effective as inoculation through Bemisia tabaci and is useful for breeding programs of TYLCV-resistant tomato.

Effects of Antihypertensive Drugs on Renal Function and Glomerular Morphology in Chronic Renal Failure Rats (만성신부전 백서에서 항고혈압제의 종류에 따른 신부전의 진행과 사구체의 형태학적 변화)

  • Purpose: Hypertension accelerates the progression of chronic renal disease, whether it results from, or causes, the renal disease. Therefore, the control of hypertension is one of the important factors that retard the rate of renal deterioration. We compared the effects of different antihypertensive agents on renal function and glomerular morphology In subtotal nephrectomized rats. Materials and methods: After induction of chronic renal failure with 5/6 nephrectomy, the rats were divided into three groups; control group (Group C), enalapril group (Group E), and nicardipine group (Group N). Systolic blood pressure was measured by tail cuff method every 4 weeks until 12 weeks after nephrectomy. At 12 weeks after nephrectomy, all rats were placed in metabolic cages for 24 hour urine collections to measure urinary protein and creatinine excretion. After urine collection and blood sampling for serum creatinine, all rats were sacrificed. The renal tissue was processed for morphometric study with light microscope and electron microscope. Results: 1. The blood pressure of Group C increased progressively, but both enalapril and nicardipine prevented the development of hypertension, and the two drugs were equally effective in maintaining normal blood pressure throughout the study. 2. Twenty-four hour urinary protein excretion was lower in Group E compared to Group C and Group N 3. Mesangial expansion score in both treated groups were significantly lower than the control group. Mean glomerular volume in Group E was significantly reduced compared to Group C and Group N. There was no significant difference in mean glomerular volume between Group C and Group N. 4. There was no significant difference in podocyte structural changes, estimated by filtration slit length density, among control, enalapril and nicardipine treated groups. Conclusion: Control of hypertension with enalapril or nicardipine afforded considerable protection from mesangial expansion in the rat remnant kidney model. But protein excretion and glomerular growth were significantly reduced in Group E compared to Group N. There was no significant difference in podocyte structural changes among the 3 groups.

  • Rock bridge in rock masses can be considered as one of several types of opening-mode fractures, and also it has been known to have a great influence on the stability of structures in rock mats. In the beginning of researching a rock bridge it used to be studied only in characteristics of its behavior, as considering resistance of material itself. However the distribution pattern of rock bridges, which can affect the stability of rock structures, is currently researched with a fracture mechanical approach in numerical studies. For investigating the effect of rock bridges on the development pattern of hydraulic fractures, the author analyzed numerically the stress state transition in rock bridges and their phenomena with a different pattern of the rock bridge distributons. From the numerical studies, a two-crack configuration could be defined to be representative of the most critical conditions for rock bridges, only when cracks are systematic and same in their length and angle. Moreover, coalescence stresses and onset of propagation stresses could be known to increase with decreasing s/L ratio or increasing d/L ratio. The effect of pre-existing crack on hydraulic fracturing was studied also in numerical models. Different to the simple hydraulic fracturing modeling in which the fractures propagated exactly parallel to the maximum remote stress, the hydraulic fractures with pre-existing cracks dial not propagate parallel to the maximum remote stress direction. These are representative of the tendency to change the hydraulic fractures direction because of the existence of pre-existing crack. Therefore s/L, d/L ratios will be identical as a function effective on hydraulic fractures propagation, that is, the $K_{I}$ vague increase with decreasing s/L ratio or increasing d/L ratio and its magnification from onset to propagation increases with decreasing s/L ratio. The scanline is a commonly used method to estimate the fracture distribution on outcrops. The data obtained from the scanline method can be applied to the evaluation of stress field in rock mass.s.

Studies on the Indigenous Vesicular-Arbuscular Mycorrhizal Fungi(VAMF) in Horticultural Crops Grown Under Greenhouse -III.   • Effects of the indigenous Vesicular-arbuscular mycurrhizal fungi(VAMF) on early growth response of greenhouse grown crops were experimented. This study was done to evaluate the benefit of indigenous VAMF inoculation on the early growth and the subsequent growth after transplanting of some crops such as cucumber, tomato, hot pepper, eggplant, and melon. Leaf area, shoot dry weight, and plant length of mycorrhizal greenhouse crops showed the tendency of significant or no significant increase over control plants receiving no inoculation. The levels of VA mycorrhizal colonization were increased with plant growth, and infection rates of horticultural crop except hot pepper around one week after transplanting were decreased, while that of 8 weeks after emergence of mycorrhizal seedlings were increased again and infected by around 50% at harvesting time. In spore densities in the rhizosphere soil of craps experimented, the number of spore ranged from $72.7{\pm}26.3$ to $100{\pm}10.3g^1$ on dried soil basis and high density showed in both cucumber and tomato. Total nitrogen contents in shoots were lower in the mycorrhizal plants than non-mycorrhizal one, whereas P uptake in mycorrhizal hot pepper and tomato were highly ramarkable. The K contents in the shoots of mycorrhizal cucumber and eggplent were highly enhanced. Inoculation of the indigenous VAMF enhanced shoot Ca and Mg in both tomoto and melon. The contents of Fe, Zn, Mn and Cu in shoots of mycorrhizal crops were higher than non-mycorrhizal plants and vice versa in case of eggplent. Inoculation of the indigenous VAMF to horticultural crops were effective for alleviation of transplanting shock, and pretransplanting infection improved subsequent growth by reducing the time required for establishment of a functional mycorrhizal symbiosis following transplanting.

The Effect of Low-Intensity Pulsed Ultrasound on Fracture Healing in the Rabbit Model (토끼모델에서 저강도 맥동초음파가 골절치유에 미치는 영향)

  • The purpose of this research was to determine the effects on the healing of fibular fractures in rabbits of low-intensity pulsed ultrasound (50 $mW/cm^2$ and 500 $mW/cm^2$) applied for periods of 4, 14 and 24 days following fibular osteotomy. Thirty-six male Japanese white rabbits were randomly divided into three groups of twelve for three treatment protocols: (1) ultrasound treatment at intensities of 50 $mW/cm^2$ and 500 $mW/cm^2$ until the 4th day following fibular osteotomy, (2) ultrasound treatment at intensities of 50 $mW/cm^2$ and 500 $mW/cm^2$ until the 14th day following fibular osteotomy, and (3) ultrasound treatment at intensities of 50 $mW/cm^2$ and 500 $mW/cm^2$ until the 24th day following fibular osteotomy. The low-intensity pulsed ultrasound was applied to only one fibula of each rabbit (these served as the experimental group). The other fibula of each rabbit served as the control group. The selection of which fibula was to be treated was made randomly. The animals were sacrificed on the 4th, 14th and 24th day after the start of ultrasound treatments. Percent of trabecular bone area and fibular radiography were carried out to compare the degree of fibular bone healing. A microscope was also used to determine any histologic changes. For statistical differences in radiological changes due to length of treatment period (4, 14 and 24 days respectively), the Wilcoxon signed-ranks test was used to compare the experimental and control groups. For statistical differences in fracture healing due to differences in ultrasound intensity, radiological studies were compared using the Mann-Whitney Test. And, to compute percentage differences in areas of trabecular bone, Two-way analysis of variance (ultrasound intensity x each group) was used. Experiment results were as follows: 1. In animals sacrificed on the 4th day, no difference was found in the radiological studies of the fibulae in the experimental and control groups (p>.05). However, experimental groups showed more rapid bone repair than control group. 2. Both radiographic and percent of trabecular bone area studies showed significant differences in rabbits sacrificed after 14 days. Fracture healing was significantly increased in the experimental group (p<.05) 3. In the animals sacrificed on the 24th day, histologic study showed rapid bone repair but fibular radiologic studies did not show statistical differences between the two groups (p>.05). 4. On the 14th day, bone union on radiograph was significantly more rapid in the treatment group with pulsed ultrasound of 50 $mW/cm^2$ than the group with 500 $mW/cm^2$ (p<.05). Histologic studies showed that both the 14 and 24 days groups had more rapid bone repair in animals treated with 50 $mW/cm^2$ ultrasound intensity than those treated with 500 $mW/cm^2$ intensity. In conclusion, it has been shown that the low-intensity pulsed ultrasound has a positive effect on bone fracture healing in the early stage and the range of pulse ultrasound from 50 $mW/cm^2$ to 500 $mW/cm^2$ is effective for fracture healing. Further study is needed to investigate the influence of pulsed ultrasound on delayed union and non-union in bone fractures and also for the clinical use of low-intensity pulsed ultrasound for bone healing in humans.

  • Recently, consumer needs are diversifying as information technologies are evolving rapidly. A lot of IT devices such as smart phones and tablet PCs are launching following the trend of information technology. While IT devices focused on the technical advance and improvement a few years ago, the situation is changed now. There is no difference in functional aspects, so companies are trying to differentiate IT devices in terms of appearance design. Consumers also consider design as being a more important factor in the decision-making of smart phones. Smart phones have become a fashion items, revealing consumers' own characteristics and personality. As the design and appearance of the smartphone become important things, it is necessary to examine consumer values from the design and appearance of IT devices. Furthermore, it is crucial to clarify the mechanisms of consumers' design evaluation and develop the design evaluation model based on the mechanism. Since the influence of design gets continuously strong, various and many studies related to design were carried out. These studies can classify three main streams. The first stream focuses on the role of design from the perspective of marketing and communication. The second one is the studies to find out an effective and appealing design from the perspective of industrial design. The last one is to examine the consumer values created by a product design, which means consumers' perception or feeling when they look and feel it. These numerous studies somewhat have dealt with consumer values, but they do not include product attributes, or do not cover the whole process and mechanism from product attributes to consumer values. In this study, we try to develop the holistic design evaluation model based on consumer values based on three-step approach from product attributes, perceived attributes, to consumer values. Product attributes means the real and physical characteristics each smart phone has. They consist of bezel, length, width, thickness, weight and curvature. Perceived attributes are derived from consumers' perception on product attributes. We consider perceived size of device, perceived size of display, perceived thickness, perceived weight, perceived bezel (top - bottom / left - right side), perceived curvature of edge, perceived curvature of back side, gap of each part, perceived gloss and perceived screen ratio. They are factorized into six clusters named as 'Size,' 'Slimness,' 'No-Frame,' 'Roundness,' 'Screen Ratio,' and 'Looseness.' We conducted qualitative research to find out consumer values, which are categorized into two: look and feel values. We identified the values named as 'Silhouette,' 'Neatness,' 'Attractiveness,' 'Polishing,' 'Innovativeness,' 'Professionalism,' 'Intellectualness,' 'Individuality,' and 'Distinctiveness' in terms of look values. Also, we identifies 'Stability,' 'Comfortableness,' 'Grip,' 'Solidity,' 'Non-fragility,' and 'Smoothness' in terms of feel values. They are factorized into five key values: 'Sleek Value,' 'Professional Value,' 'Unique Value,' 'Comfortable Value,' and 'Solid Value.' Finally, we developed the holistic design evaluation model by analyzing each relationship from product attributes, perceived attributes, to consumer values. This study has several theoretical and practical contributions. First, we found consumer values in terms of design evaluation and implicit chain relationship from the objective and physical characteristics to the subjective and mental evaluation. That is, the model explains the mechanism of design evaluation in consumer minds. Second, we suggest a general design evaluation process from product attributes, perceived attributes to consumer values. It is an adaptable methodology not only smart phone but also other IT products. Practically, this model can support the decision-making when companies initiative new product development. It can help product designers focus on their capacities with limited resources. Moreover, if its model combined with machine learning collecting consumers' purchasing data, most preferred values, sales data, etc., it will be able to evolve intelligent design decision support system.

Improvement of the Seed Production Method of the Pen Shell -The Occurrence of Larvae and the Early Growth of the Spat- (키조개 채묘의 개발연구 -부유유생의 출현과 부착치패의 초기성장-)

  • In order to set up a predictive model for effective spat collection of pen shell, Atrina (Servatrina) pectinata, the survival rate and time required at each developmental stage of drifting larvae were surveyed during the period from June 8 to October 16 in 1986 at the Yongku inlet Chilchon Is., Chinhae Bay, the southern part of Korea. And also the experiments of spat collection were carried out In Yongku inlet during the period from July 6 to November 23 in 1987 and In Yoja Bay during the period from July 9 in 1987 to February 15 in 1588. The advent of D-shape larvae ca. $135\times144um $ long had three peaks in that area: August 1, 12 and 25. Umbo shape larvae ca. $300\times317um$ and full grown larvae ca. $455\times450um$ long also sowed three peaks: August 9, August 22 and September 4 for the former, and August 23, September 3 and September 16 for the latter. Nine to ten days was required for D-shape larvae to develop to umbo shape larvae. The instantaneous survival rate was 0.94 with a total survival rate if $54\%$ at this intermorphological stage. The required time of umbo to full grown larvae varied from twelve to fourteen days with a instantaneous survival rate of 0.88 and total survival rate of $19\%$. Twenty-two to twenty-three days was required for each group of the D-shape larvae to reach a fullgrown stage, and their total survival rate was $10\%$ during this developmental period. The number of the spat attached to the spat collector is 0.16 inds. per $m^2$ vertical spat collector in Yongku inlet and 0.48 inds. per $m^2$ horizontal spat collector in Yoja Bay. The average shell length of spat attached was $0.51({\pm}0.15)\;mm$ on September $27,\;38.52({\pm}6.98)\;mm$ on November 21 in 1987 and $49.00({\pm}10.77)mm$ on February 15 in 1988.

Response of Yield and Nitrogen Use Efficiency for Garlic on Different Types and Rates of Organic Fertilizer (유기질 비료의 종류 및 시비량이 마늘의 수량과 질소이용효율에 미치는 영향)

  • Organic fertilization (OF) was an effective management strategy to improve crop yield and soil fertility. Determination of optimum application rate has been received great interest as a major research field. In this study, we evaluated optimum application level of three different organic fertilizers; mixed expeller compost (MEC), mixed organic fertilizer (MOF) and organic complex fertilizer (OCF), for garlic cultivation. Treatments consisted of MEC, MOF, OCF (50, 100, 150% of standard nitrogen input), inorganic fertilizer(NPK, N-P2O5-K2O : 250-78-128 kg ha-1) and no-fertilization (control). Overall, the yields of garlic under MEC, MOF and OCF 100% (5,337, 5,617 and 5,276 kg ha-1) were higher than under control (4,496 kg ha-1). The 100% of three OFs treatements showed the highest yield and bulb diameter, similar with NPK, while leaf length and bulb height were not significantly different among all treatments. The 150% of three OFs rather decreased yield. The highest nitrogen use efficiency (38.4 and 38.0%) was observed in MOF 100% and OCF100%, which was the most similar with that in NPK (38.8%). As OF application rate increased, pH was decreased, but soil NO3-N and NH4-N were increased. There was no significant difference in soil organic matter (OM), av. P2O5 and Ex. cation values. From these results, 100% application of OFs could be recommended as a suitable input level for garlic cultivation, regardless of organic fertilizer types. Further study might be required to evaluate long-term OF application effect on soil health and crop productivity.
