추출온도 및 pH가 김에서 추출한 Crude Porphyran의 화학특성에 미치는 영향 (Effect of Extraction Temperature and pH on the Chemical Properties of Crude Porphyran Extracted from Porphyra yezoensis)

  • 추출용매비 및 추출시간을 각각 50배 (w/v), 3시간으로 일정하게 하고 추출온도 ($60\~100^{\circ}C$)와 pH($2.0\~13.5$)를 달리하면서 crude porphyran을 제조하여 수율과 화학조성 을 조사하였다. Crude porphyran의 화학조성은 추출온도와 pH에 따라 크게 달라져서 산성영역에서 추출한 crude porphyran은 중성영역에서 추출한 것에 비해 수율 및 황산기 함량은 더 많으면서 단백질을 더 적게 함유하나 분자량도 작아지는 것으로 나타났고, 알칼리 영역에서 추출하는 경우는 황산기가 떨어져 나가고 단백질의 혼입량이 많아지는 것으로 나타났다. 따라서 실험목적에 따라 추출조건이 달라져야 함을 알 수 있었는데 수율의 증가 및 저분자화를 위해서는 pH $3\~4$의 조건이, 그리고 시료내 존재하는 porphyran을 분자량의 변화 없이 추출하기 위해서는 중성 영역에서 추출하는 것이 적절할 것으로 사료된다.

Influence of "Historical Effects" on the Rheological Properties of a Polyacrylonitrile Copolymer Solution

  • Polyacrylonitrile (PAN) copolymers of different molecular weights were synthesized by a suspension polymerization and precipitation polymerization method. The rheology behaviors of the synthesized PAN copolymers were investigated in relation to their molecular weight, solid content and melting temperature. The influence of "historical effects" on the spinning solution of PAN was studied by analyzing the laws of viscosity considering the diversification time and temperature. The viscosity disciplines of each spinning solution conformed well to the rheological universal laws in a comparison of the suspension polymerization product with that of precipitation polymerization. Viscosity changes in the swelling process of dissolution were gentler in the suspension polymerization product; a small amount of water will quickly debase the solution viscosity, and high-speed mixing can greatly shorten the time required by the spinning solution to reach the final viscosity.

Effects of Sodium Fluoride on the Water Transport in Leaves of Barley and Rice under Salt Stress in the Light

  • The kinetics of the loss of leaf fresh weight during incubation of barley and rice leaves in 9% or 15% NaCl solutions were biphasic, indicating the existence of a controlling mechanism for water transport. The first rapid phases reached their plateaus within 1 and 2 h in the case of rice and barley leaves, respectively. When barley leaves were fed with sodium fluoride, an inhibitor of phosphatase inhibitor, through their epicotyls for 3 h in darkness, prior to the treatment of NaCl, the biphasic pattern shown during NaCl treatment was disappeared resulting in linear decreases in the relative fresh weights. The results suggest that NaF accelerates salt-induced water efflux from plant cells, possibly by inhibiting the protection mechanism that may act in NaF-untreated leaves. The linear water loss can be explained in terms of phosphorylation of aquaporin by blocking its dephosphorylation in the presence of the phosphatase inhibitor to keep aquaporin in a phosphorylated form. However, the effect of NaF shown in barley leaves were not observed in rice. These results suggest that the regulation of water transport depends on plant species, and the mechanism for the controlling water transport in rice is different from that of barley.

Influence of Temperature, Oxygen, m-Chlorophenylhydrazone Cerulenin, and Quinacrine on the Production of Extracellular Proteases in Bacillus cereus

  • Bacillus cereus KCTC 3674 excretes at least two kinds of extracellular proteases into the growth medium. Two major bands of the protease activity with molecular weights of approximately 100 and 38 kDa were obtained after gelatin-SDS-PAGE. The protease with a molecular weight of 38kDa was identified as an extracellular neutral (metallo-) protease. The neutral protease was quite thermostabile but labile to alkaline pH. On the contrary, the 100-kDa protease was thermolabile but stable to alkaline pH. The production of 38-kDa neutral protease was strongly affected by temperature, oxygen, carbonylcyanied m-chlorophenylhydrazone(CCCP) that was defined as a protonophofre, and cerulenin which inhibited lipid synthesis and caused changes in the membrane composition. On the other hand, the production of the 100-kDa protease was strongly affected by only temperature and cerulenin. Quinacrine (0.2 mM), which inhibits the penicillinase-releasing proteases of Bacillus licheniformis, had no effect, whatsoever, on the production of extracellular proteases in B.cereus KCTC 3674.

양식넙치로 부터의 Lysozyme 정제와 어류병원성 세균에 대한 정균작용 (Purification and antibacterial effect of lysozyme from flounder, Paralichthys olivaceus)

  • 넙치의 혈청, 체표점액 및 각 조직으로부터 lysozyme의 분리와 정제를 Chitin-coated Cellulose column 크로마토그라피를 사용하여 분리하였다. 15% SDS-PAGE의 전기 영동상을 조사한 결과 넙치의 신장, 유문수, 아가미 점액 및 혈청으로부터 분리된 lysozyme은 동일한 분자량(약 14000)을 나타내었다. 넙치의 방어기작으로서 lysozyme의 역할을 밝히기 위해 그람양성 세균 2주, 그람음성 세균 5주에 대하여 넙치 신장 유래 lysozyme의 정균작용을 조사한 결과 그람 양성 2주 그람 음성 세균 2주에 대하여 정균작용을 나타내었다. 이것은 넙치 유래의 lysozyme은 그람 양성 세균뿐만 아니라 그람 음성 세균에 대해서는 생체 방어 작용을 수행하는 것으로 고려된다.

Isolation and Characterization of Two Korean Mistletoe Lectins

  • Two isolectins (KML-IIU and the KML-IIL) were individually isolated from the previously reported Korean mistletoe lectin, KML-C, by using an immunoaffinity column. Molecular weights of the KML-IIU and the KML-IIL were 64 kDa and 60 kDa respectively. Both of the lectins were composed of heterogeneous A and B subunits linked with a disulfide bond, and showed the same carbohydrate-binding specificities for Gal and GalNAc. However, they are different not only in biophysical properties (glycosylation and amino acid compositions) but also bioactivities (cell killing and cytokine induction). The KML-IIL showed 17-145 times stronger in cytotoxicities to various human and mouse cancer cell lines than the KML-IIU. The KML-IIL also induced TNF-$\alpha$ secretion from mouse peritoneal macrophages 4.5 times better than the KML-IIU. The results demonstrated isolectins in Korean mistletoe were varied in bioactivities and the KML-IIL may be developed as an anti-cancer agent.

Cashew Nut Oil: Extraction, Chromatographic and Rheological Characterisation.

  • Oil was extracted from cashew nuts. The physicochemical parameters of the oil were determined. A chromatographic assay of the oil was carried out using Gas Chromatography-Mass Spectrometry. Seventeen compounds were detected: Phenol, Phenol 2-methyl-, Cyclohexene 4, 4-dimethyl-, m-Fluoro-2-diazoacetophenone 4-dimethyl-, Tetradecanoic acid, Phenol 4-octyl-, n-Hexadecanoic acid. Others are 9, 12-Octadecadienoic acid (Z, Z) - methyl ester, Hexadecanoic acid methyl ester, Methyl stearate, Dodecanoic acid methyl ester, 9, 12, 15-Octadecatrienoic acid methyl ester, 9, 12, 15-Octadecatrienoic acid (Z, Z, Z)-, Oleic acid, Octadecanoic acid, Tetracosanoic acid and 9-Octadecenoic acid methyl ester. Among the components are omega three and omega six essential free fatty acids. The rheological profiling and flow properties of cashew nut oil were determined using a Programmable Rheometer. Cashew nut oil exhibits slight dilatant behaviour at the low end of shear rate. The long chain and high molecular weight of its constituents controlled its rheology. Long-chained 9-Octadecenoic acid methyl ester, 9, 12-Octadecadienoic acid (Z, Z) - methyl ester, Tetracosanoic acid and methyl stearate, coupled with their high molecular weights are responsible for the shear thickening effect observed. Two models, Carreau-Yasuda and Ostwald-de Waele Power Law were employed to fit the rheological data. The Carreau-Yasuda model followed well the data.

Dispersion polymerization of styrene and Methylmethacrylate using macromonomers as a reactive stabilizer

  • The novel linear- (V-LUM) and cross-type macromonomers (C-VUM) of vinyl-terminated bifunctional polyurethane were synthesized and applied to the dispersion polymerization of styrene and MMA in ethanol. The existence of the vinyl terminal groups and the grafted macromonomer with styrene and PMMA was verified using 1H NMR and 13C NMR. Monodisperse polystyrene (PS) microspheres were successfully obtained above 15 wt % of macromonomer relative to styrene. The macromonomer can efficiently stabilize higher surface area of the particles compared to a conventional stabilizer, PVP. The grafting ratio of the PS calculated from 1H NMR linearly increased up to 0.048 with 20 wt % of the macromonomer and the high molecular weights (501,300 g/mol) of PS with increased glass transition and enhanced thermal stability were obtained. Furthermore, the stable and monodisperse PMMA microspheres having a weight-average diameter of $5.09{\mu}m$ and a good uniformity of 1.01 were obtained with 20 wt% L-VUM. The molecular weight increased, but the size of the PMMA particles decreased with the macromonomer concentration due to the increased stabilizing effect. The molecular weight of the PMMA was approximately two fold higher than that by a conventional PVP. The L-VUM acts as a reactive stabilizer, which gives polyurethane-grafted PS or PMMA with a high molecular weight. In addition, the XPS result showed that the C-PS (PS using the C-VUM) was anchored with a larger amount of PEG than that of the L-PS (PS using the L-VUM) on the particle surface. Thus, the reaction and stabilizing mechanism of the macromonomers for the formation of PS particles is proposed.

키토산 분자량에 따른 Rhodamine 6G-Sodium Dodecyl Sulfate계의 응집효과에 관한 연구 (Effect of Molecular Weight of Chitosan on Flocculation of Suspended Solids in Rhodamine 6G-Sodium Dodecyl Sulfate System)

  • 산업폐기물인 꽃게의 껍질로부터 chitin을 탈아세틸화시켜 분자량이 서로 다른 chitosan을 제조하였다. Rhodamine 6G(Rh 6G)-sodium dodecyl sulfate(SDS)계 및 Rh 6G-chitosan계들의 흡수 및 형과 spectra를 조사하였고, chitosan의 분자량 및 pH에 따른 Rh 6G-SDS-chitosan계의 응집효과에 대해 조사하였다. Rh 6G-SDS계의 흡광도나 형광세기는 S/D(SDS 농도/Rh 6G 농도)=32 이하에서 감소하다가 그 이상에서 다시 증가하였다. 부유물질(SS) 제거율이나 투광도로부터 S/D=32에서 chitosan을 첨가한 Rh 6G-SDS-chitosan계의 응집 성능이 Rh 6G-SDS계에 비해 훨씬 우수함을 알 수 있었다. 그리고 chitosan의 농도나 분자량이 크면 클수록, S/D 첨가 범위는 32에서 100까지 확대되는 것을 알 수 있었다. 부유물질의 제거율은 chitosan의 분자량이 클수록 pH 2∼9에서 우수한 성능을 가지는 반면, pH 12 이상에서 부유물질의 제거율은 현저히 저하되는 것을 알 수 있었다.

사과의 Hemicellulose가 Ca-Pectate Gel형성에 미치는 영향 (Effect of Apple Hemicellulose on the Ca-Pectate Gel Formation)

  • Fuji 사과의 hemicellulose가 사과 Ca-pectate gel 형성에 미치는 영향을 조사하기 위하여 hemicellulose를 $HF_1$(1N KOH 가용성), $HF_2$(2N KOH 가용성), $HF_3$(3N KOH 가용성), $HF_4$(4N KOH 가용성)으로 분획한 후 다시 gel여과 정제하여 사과 Ca-pectate gel 형성에 미치는 영향을 조사하였다. Fuji hemicellulose는 KOH의 농도가 1N에서 4N로 증가됨에 따라 hexose peak은 점차 저분자쪽과 고분자쪽의 용출이 증가되었으며 pentose peak는 특히 고분자쪽의 용출이 증가되었다. Gel여과에 의하여 분획된 hemicellulose는 분자량 1만${\sim}$143만에 이르는 $8{\sim}10$개의 peak로 구분되었으며 사과 Ca-pectate gel 형성시에 첨가시킨 결과 고분자보다 저분자쪽이, pentose보다 hexose와 uronic acid가 많이 함유된 쪽에서 gel의 hardness, adhesiveness 및 gumminess 가 증가되었다.
