Development of an Enzyme-linked Immunosorbent Assay Using Vitellin for Vitellogenin Measurement in the Pale Chub, Zacco platypus

  • Objectives Fish vitellogenin (VTG) is produced in the female liver during oogenesis through the estradiol cycle and produced in the male liver by endocrine disrupting chemicals (EDCs) such as alkylphenols. In this study, we propose that the VTG concentration in the pale chub could be detected using monoclonal antibodies and polyclonal antibodies against vitellin (Vn) in a VTG enzyme-linked immunosorbent assay (ELISA) system. Methods Monoclonal antibodies and polyclonal antibodies were produced using the Vn extracted from the matured ovum of the ovary. The VTG was extracted from the plasma of the male pale chub. The Vn and VTG were confirmed by measuring the molecular weight of their proteins using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and the specificity of the antibodies was checked through western blotting methods. The assay system was validated with respect to optimal assay concentrations, specificity, recovery, and intra- and inter-assay variations. Results The Vn consisted of two protein bands with apparent molecular weights of 64 and 37 kDa. The SDS-PAGE indicated protein weights of 146 and 77 kDa in the VTG. The assay range was 15.6 ng/mL to 2,000 ng/mL, and the value of the intra- and inter-assay variations were within 10.0% and 14.7%, respectively. The recovery rate was $99.5{\pm}5.5%$. Conclusions A sandwich ELISA was developed that could be used to qualify the VTG of pale chub in screening for EDCs. Pale chub is an ideal species for observing estrogen activity in the environment because of its extensive habitat and extensive food chain. The ELISA developed here would be more favorable than those for other species for determining the effect of long-term food chain accumulation of EDCs in aquatic environments.

The Effect of Coagulant and Molecular Weight on the Wet Spinnability of Regenerated Silk Fibroin solution

  • The regenerated silk fibroin with various molecular weights (MW) was prepared by different dissolution condition and the effect of coagulant on the wet spinnability of the various MW silk fibroin solutions dissolved in formic acid was investigated by the observation of wet spun filament in coagulant and the measurement of maximum draw ratio. The observation on the wet spun filament in coagulation bath revealed that good fibers without bead were formed in a high MW and a very high MW silk fibroin samples. In contrast, beads were observed in the silk fibroin sample with medium MW. The maximum draw ratio of wet spun silk fibroin filament decreased with MW reduction. The decrease of maximum draw ratio in isopropanol, acetone, DMF and THF was remarkably higher than that in methanol and ethanol, indicating that the coagulant type strongly influenced the wet spinnability. The two simple evaluation methods used in this study showed complementary information for wet spinnability: (a) The observation of filament in coagulant was effective to check a continuous fiber formation and a bead formation, and (b) the maximum draw ratio measurement was useful to examine the post drawing ability related to molecular orientation.

  • The effects of additives on the Zn electrodeposition in chloride-based electrolyte were investigated using circulation cell with three electrodes system. The cathodic polarization increased with the addition of polyethylenglycol (hereafter PEG) in electrolyte. This was attributed to the adsorption of the additives on the electrode and the inhibition of migration of metal ion. The PEG, however, did not have any noticeable effect on the properties of plating solutions at the concentration used. The effect of PEG on the electrocrystallization was related to its molecular weight. With the increase of molecular weight, the cathodic polarization increased, while the surface roughness was improved with the decrease of brightness. Especially, the PEG mixed with different molecular weights was the most effective. The orientation and the type of the deposited grains were changed and refined by PEG, which resulted in the modification of deposited surface roughness and brightness.

  • The stress relaxation of poly(methyl acrylate)-poly(acrylonitrile) copolymer samples was carried out in air, distilled water, pH 3, 7 and 11 solutions at various temperatures using a tensile tester equipped with a solvent chamber. The relaxation spectra of poly(methyl acrylate)-poly(acrylonitrile) copolymers were obtained by applying the experimental stress relaxation curves to the equation of relaxation spectrum derived from the Ree-Eyring and Maxwell model. The determination of relaxation spectra was performed from computer calculation using a Laplace transform method. It was observed that the relaxation spectra of these samples are directly related to the distribution of molecular weights and self-diffusions of flow segments.

Isolation from Gloydius blomhoffii siniticus Venom of a Fibrin(ogen)olytic Enzyme Consisting of Two Heterogenous Polypeptides

  • Objective: This study was undertaken to isolate a fibrin(ogen)olytic enzyme from the snake venom of Gloydius blomhoffii siniticus and to investigate the enzymatic characteristics and hemorrhagic activity of the isolated enzyme as a potential pharmacopuncture agent. Methods: The fibrinolytic enzyme was isolated by using chromatography, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and fibrin plate assay. The characteristics of the enzyme were determined by using fibrin plate assay, protein hydrolysis analysis, and hemorrhage assay. Its amino acid composition was determined. Results: The fibrin(ogen)olytic enzyme with the molecular weight of 27 kDa (FE-27kDa) isolated from G. b. siniticus venom consisted of two heterogenous disulfide bond-linked polypeptides with the molecular weights of 15 kDa and 18 kDa. When more than $20{\mu}g$ of FE-27kDa was applied on the fibrin plate, fibrinolysis zone was formed as indicating its fibrinolytic activity. The fibrinolytic activity was inhibited completely by phenylmethanesulfonylfluoride (PMSF) and ethylenediaminetetraacetic acid (EDTA) and partially by thiothreitol and cysteine. Metal ions such as $Hg^{2+}$ and $Fe^{2+}$ inhibited the fibrinolytic activity completely, but $Mn^{2+}$ did not. FE-27kDa preferentially hydrolyzed ${\alpha}$-chain of fibrinogen and slowly hydrolyzed ${\beta}$-chain, but did not hydrolyze ${\gamma}$-chain. High-molecular-weight polypeptides of gelatin were hydrolyzed partially into polypeptides with molecular weights of more than 45 kDa. A dosage of more than $10{\mu}g$ of FE-27kDa per mouse was required to induce hemorrhage beneath the skin. Conclusion: FE-27kDa was a serine proteinase consisting of two heterogeneous polypeptides, hydrolyzed fibrin, fibrinogen, and gelatin, and caused hemorrhage beneath the skin of mouse. This study suggests that the potential of FE-27kDa as pharmacopuncture agent should be limited due to low fibrinolytic activity and a possible side effect of hemorrhage.

Effect of Gelling Agent Molecular Weight on Self-Discharge Behavior for Zinc-Air Batteries (아연-공기 전지용 전해질의 Gelling Agent 분자량에 따른 자가 방전 억제 효과)

  • A zinc-air battery is one of most promising advanced batteries due to its high specific energy density, low cost, and environmental friendliness. However, zinc anodes in zinc-air batteries lead to several issues including self-discharge, corrosion reaction, and hydrogen evolution reaction (HER). In this paper, viscosity of electrolyte has been controlled to suppress the corrosion reaction, HER, and self-discharge behavior. Various viscosity average molecular weights of poly(acrylic acid) (PAA) are adopted to prepare the electrolyte. The evaporation of electrolytes is proportional to the increase in molecular weight. In addition, enhanced self-discharge behavior is obtained when the gelling agent with high molecular weight is used. In addition, the zinc-air cell assembled with lower viscosity average molecular weight of PAA (Mv ~ 450,000) delivers 510.85 mAh/g and 489.30 mAh/g of discharge capacity without storage and with 6 hr storage, respectively. Also, highest capacity retention (95.78 %) is obtained among studied materials.

  • Monomer casting nylons were synthesized by casting anionic polymerization of ${\varepsilon}$-caprolactam. Polymerization rates, molecular weights of the products and the conversions were determined while varying the content of catalysts in the range of 0.2~0.6 mol% and 0.1~1.0 mol% for initiator. The polymerization rates were enhanced as the ratio of catalysts to initiator increased. The maximum molecular weight was observed when the ratio of catalysts to initiator was 0.8, and as the ratio increased the molecular weight decreased. On the other hand, when the ratio of catalysts to initiator was below 0.8, the conversions and the molecular weights were abruptly diminished due to the termination of growing chains.

Honey Bee Venom (Apis mellifera) Contains Anticoagulation Factors and Increases the Blood-clotting Time

  • Objectives: Bee venom (BV) is a complex mixture of proteins and contains proteins such as phospholipase and melittin, which have an effect on blood clotting and blood clots. The mechanism of action of honey bee venom (HBV, Apis mellifera) on human plasma proteins and its anti-thrombotic effect were studied. The purpose of this study was to investigate the anti-coagulation effect of BV and its effects on blood coagulation and purification. Methods: Crude venom obtained from Apis mellifera was selected. The anti-coagulation factor of the crude venom from this species was purified by using gel filtration chromatography (sephadex G-50), and the molecular weights of the anti-coagulants in this venom estimated by using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Blood samples were obtained from 10 rabbits, and the prothrombin time (PT) and the partial thromboplastin time (PTT) tests were conducted. The approximate lethal dose (LD) values of BV were determined. Results: Crude BV increased the blood clotting time. For BV concentrations from 1 to 4 mg/mL, clotting was not observed even at more than 300 seconds, standard deviations $(SDs)={\pm}0.71$; however, clotting was observed in the control group 13.8 s, $SDs={\pm}0.52$. Thus, BV can be considered as containing anti-coagulation factors. Crude BV is composed 4 protein bands with molecular weights of 3, 15, 20 and 41 kilodalton (kDa), respectively. The $LD_{50}$ of the crude BV was found to be $177.8{\mu}g/mouse$. Conclusion: BV contains anti-coagulation factors. The fraction extracted from the Iranian bees contains proteins that are similar to anti-coagulation proteins, such as phospholipase $A_2(PLA_2)$ and melittin, and that can increase the blood clotting times in vitro.

Effect of PVA Polymerization on Synthesis of YAG:Ce3+ Phosphor Powders Prepared by a Solid-liquid Hybrid Route (PVA 중합도가 고상-액상 혼합 방식에 의한 YAG:Ce3+ 형광체 분말 합성에 미치는 영향)

  • YAG:$Ce^{3+}$ phosphor powders were synthesized using $Al(OH)_3$ seeds by means of a PVA-polymer-solution route. Various types of PVA with different molecular weights (different polymerization) were used. All dried precursor gels were calcined at $500^{\circ}C$ and then heated at $1500^{\circ}C$ in a mix of nitrogen and hydrogen gases. The final powders were characterized via XRD, SEM, PSA, PL, and PKG analyses. The phosphor properties and morphologies of the synthesized powders were dependent on the PVA type. As the molecular weight of the PVA was increased, the particle size gradually decreased with agglomeration, and the luminous intensity of the phosphor increased. However, the phosphor powder prepared from the PVA exhibiting very high molecular weight, showed a 531 nm (blue) shift from the 541 nm (yellow) wavelength of the YAG:$Ce^{3+}$ phosphor. Finally, the synthesized YAG:$Ce^{3+}$ phosphor powder prepared from the PVA with 89,000 - 98,000 molecular weight showed phosphor properties similar to those of a commercial phosphor powder, but without a post-treatment process.

  • Hydrophilic reactive hot-melt polyurethane adhesive(HRHA) using a hydrophilic polyol having different molecular weight and NCO index was synthesized. This HRHA was synthesized using Polyethylene glycol(PEG) as a hydrophilic polyol, Polypropylene glycol(PPG) and Polycaprolactone diol(PCL) as hydrophobic polyols, and Methylene diphenyl diisocyanate(MDI) as an isocyanate. The changes in IR spectrum, viscosity and thermal properties of HRHA with different PEG molecular weights and NCO index were investigated, and the tensile strength and elongation of the HRHA casting film and the peel strength, moisture permeability and water pressure of the HRHA coated fabric were confirmed. In this experiment, as the molecular weight of PEG and NCO index increased, the adhesive strength, tensile strength, elongation and moisture permeability was increased but viscosity and Tg was decreased.
