Effects of Gibberellin Biosynthetic Inhibitors on Oil, Secoisolaresonolodiglucoside, Seed Yield and Endogenous Gibberellin Content in Flax

  • Flax (Linum usitatissimum L.) has been used for the edible oil in Korea. The evaluate the effect of plant growth retardants on flaxseed yield, oil content, and gibberellin level, chloromequat chloride (CMC), paclobutrazol (PBZ), and prohexadione-Ca (PHC) were used as plant growth retardants in this study. These plant growth retardants were foliar-sprayed to flax plant at 55 days after seeding. The concentrations of each plant growth retardant were as followed: CMC 250, 500, 1000 ppm, PBZ 40, 80, 160 ppm and PHC 500, 1000, 2000 ppm. PHC treatment to forming bolls was more stimulated than CMC and PBZ. The highest ripened seed rate was observed in PHC treatment at 2000ppm. The high see yields were obtained in PHC treatment following PBZ and CMC, in turn. Seed yield that significantly increased in PBZ and PHC was found to be increased 12.4 ~ 23.9% as compared to the control. The PHC showed higher flaxseed production and oil yield than that of CMC and PBZ. The results obtained in the present study suggest that higher concentration of plant growth retardant (PHC) increased flaxseed yield and oil content. The optimal concentration of PHC treatment was observed in 2000 ppm. It concludes that the foliar application of PHC 2000 ppm may be useful for the increasing oil and higher seed production in flax plants.

Comparison of Seed Oil Characteristics from Korean Ginseng, Chinese Ginseng (Panax ginseng C.A. Meyer) and American Ginseng (Panax quinquefolium L.)

  • The chemical characteristics of seed oils of Asian ginseng (Panax ginseng C.A. Meyer) at different ages grown in Korea (3, 4 and 5-year old) and China (5-year old), and American ginseng (Panax quinquefoliu L., 5-year old) grown in China were compared. Total fatty acid composition showed a significantly higher oleic acid content in American (87.50%) than in Korean (68.02~69.14%) and Chinese ginseng seed oils (61.19%). At the sn-2 position, the highest oleic acid (81.09%) and lowest linoleic acid (15.77%) were found in American ginseng seed oil. The main triacylglycerol species in ginseng seed oils were triolein (OOO) and 1,2-dioleoyl-3-linoleoyl-glycerol (LOO)/1,3-dioleoyl-2-linoleoyl-glycerol (OLO). In addition, the seed oils possessed an ideal oxidative stability showing 16.55~23.12 hr of induction time by Rancimat test. The results revealed that ginseng seed oil could be developed as a new healthy edible oil, and that the oil's chemical characteristics were strongly associated with the ginseng species and habitats.

  • The Korean origin edible oil sterol part of the rape, sesame and perilla oil can be separated from the other constituents of the non-saponifiable fraction, by the thin layer chromatography on the preparative Plates. The composition of sterols have been determined by gas liquid chromatography and thin layer chromatographic anlysis. Two sterols, ${\beta}$-sitosterol and campesterol were present in all of the oils. And brassicasterol were found in rapeseed oil in addition to the two sterols that were common to all of the oils studied. It was noted that ${\beta}$-sitosterol was the major sterol in the Korean original edible oils. The results showed that contents of sterols were campesterol 24.31%, ${\beta}$-sitosterol 58.90% ana Brassicasterol 11.54%, and $\Delta^7$-sterol 5.25% by method of triangulation and campesterol 26.16%, ${\beta}$-sitosterol 57.50%, brassicasterol 11.70% and ${\Delta}^7$-sterol 4.64% by method of Planimetry of rape seed oil. By sesame seed oil sterol compositions were campesterol 20.35%, stigmasterol 9.15%, ${\beta}$-sitosterol 43.49%, ${\Delta}^7$-sterol 11.25% and others 15.76% by method of triangulation and campesterol 16.79%, stigmasterol 8.69%, ${\beta}$-sitosterol 44.58%, ${\Delta}^7$-sterol 14.28% and others 15.56% by method of planimetry. Campesterol 12.45%, stigmasteriol 5.40%, ${\beta}$-sitosterol 72.32% and ${\Delta}^7$-Sterol 9.83% by method of triangulation-and campesterol 13.00%, stigmasterol 3.76%, ${\beta}$-sitosterol 74.57% and ${\Delta}^7$ sterols 8.67% by method of planimetry of perilla oil. Contents of totalsterol in Korean edible oils were 0.82% by rape, 0.58% by sesame and 0.45% by perilla, respectively.

  • 황협1호는 2000년에 도입유전자원인 KLG50064와 KLG50073을 인공교배하여 얻어진 YK3을 온실과 포장에서 세대 단축을 하면서 선발된 YK3-4B-225계통으로, '04년도에 생산력 검정시험에서 협채소용으로 선발되어 밀양1호의 계통명을 부여하였으며, 이들은 '05년 3월과 8월에 온실에서 재배하여 생육특성과 협채수량을 조사하였고 협채소용으로 유망 시 되어 협채용 강낭콩 신품종 "황협1호"로 선정되었다. 1. 유한신육형으로 화색은 백색, 배축은 녹색이고, 제1본엽은 기형엽이다. 협채수확 시기의 협은 황색이고 협단면은 타원형이며, 협모양은 약간 S자형으로 굽었고, 종피색은 백색이다. 2. 개화일수는 43일 성숙일수는 49일이었고, 경장은 42 cm, 주경절수 8.6개, 분지수 4.8개, 협수는 27개였다. 100립중은 21.7 g이었고, 협채 수확기에 협폭은 넓은 반면 협두께는 얇아 협모양은 타원형이다. 3. 채협 후 협선도 유지기간이 길며, 성분 분석 결과 수분 함량은 87.2%였고, 단백질과 지방은 각각 건조중의 19.7%, 0.6%였다. 당함량 중 Sucrose와 Glucose는 각각 3.5%, 14.0%였고, Vitamin C의 함량은 100 g중 152 mg이었다. 4. 협채수량은 온실재배시 23.55톤/ha로 강낭콩 1호 보다 4% 증수되었다. 5. 황협1호는 시설재배로 전국에서 재배될 수 있으며 결주 방지를 위한 육묘이식 재배와 생육기간 내내 충분한 토양수분 유지가 필요하다.

  • Experiments were carried out in order to compare the natural antioxidants in the acetonitril extracts of raw and baked seeds of Korean sesame and perilla by thin layer and gas chromatography. The sample was dissolved in n-pentane and extracted with acetonitril and the acetonitril extract was separated by thin layer chromatography using silica gel. The spots were detected by spraying with 2, 6 -dichloroquinone -4-chlorimide, phosphomolybdic acid and dimethylamine as chromogenic reagents. Natural antioxidant, such as ${\delta}-tocopherol$ detected in raw and baked sesame and perilla seed oil by TLC and sesamol was detected only in raw and baked sesame seed oil by GC.

Elimination of Saturated Fatty Acids, Toxic Cyclic nonapeptide and Cyanogen Glycoside Components from Flax Seed Oil

  • Flax seed(Linseed, Linum usitatissimum L.) and its oil, a richest source of alpha-linolenic acid(ALA)(${\omega}-3$), contain saturated fatty acids, neurotoxic cyanogen glycosides and immuno-suppressive cyclic-nonapeptides. Present paper describes the development of two chemical processes, Process-A and -B, to remove saturated fatty acids and to destroy cyclic nonapeptides and cyanogen glycosides from flax seed oil. Process-A consists of three major steps, i.e., extraction of fatty acid mixture by alkaline saponification, removal of saturated fatty acid by urea-complexation, and triglyceride reconstruction of unsaturated fatty acid via fatty acyl-chloride activation using oxalyl chloride. Process-B consists of preparation of fatty acid ethyl ester by transesterification, elimination of saturated fatty acid ester by urea-complexation, and reconstruction of triglyceride by interesterification with glycerol-triacetate (triacetin). The destruction of lipophilic cyclic nonapeptide during saponification or transesterification processes could be demonstrated indirectly by the disappearance of antibacterial activity of bacitracin, an analogous cyclic-decapeptide. The cyanogen glycosides were found only in the dregs after hexane extraction, but not in the flax seed oil. The reconstructed triglyceride of flax seed oil, obtained by these two different pathways after elimination of saturated fatty acid and toxic components, showed agreeable properties as edible oil in terms of taste, acid value, iodine and peroxide value, glycerine content, and antioxidant activity.

  • The Korean origin sunflower (Helianthus Annus Linn.) seed of netural lipid were analysed by thinchrography, High performance liquid chromatography, preparative Thinlayer and Gas liquid chromatography. 1) The seed oil triglyceride components were conveniently separated based on their degree of unsaturation by employing the chromatography on silica gel sintered rod impregnated with 12.5% silver nitrate. Sunflower seed oil was composed of triglyceride, especially trilinolein 57. 74% triolein 25.28%, tripalmitin 7 55% ana tristearin 9.43% by a thinctrography. 2) The fatty acid compositions of seed oil have been determined by a high performance liquid chromatographic analysis using a ALC/GPC 244 type from Waters Association (Japan) with ${\mu}$ Bondapak FFAA column. It contained stearic acid 8.59%, oleic acid 27. 19%, palmitic acid 7.50% and linoleic acid 56.72% respectively. 3) The composition of sterols were determined by a preparative Thinlayer and Gas liquid chromatographic analysis. It was noted that sitosterol was the major sterol in the Korean sunflower seed. The results showed that contents of sterols were cholesterol trace, campesterol $13_.^{22\sim}13.9%$, stigmasterol $13.8{\sim}14.1%$, If, sitosterol $58.4{\sim}60.7%$, ${\vartriangle}^7$-stigmastenol $10.2{\sim}10.5%$ and ${\vartriangle}^{7,24(25)}$-stigmastenol $3.6{\sim}3.8%$ by method of planimetry and triangulation.

Characteristics and Germination of Xanthoceras sorbifolia Bunge Seeds Originated from Inner Mongolia and Liaoning, China

  • Seeds of Xanthoceras sorbifolia Bunge were collected from two plantations and two superior trees in Inner Mongolia: and one plantation and one superior tree in Liaoning, China in late August, 2011. Yellowhorn or goldenhorn is an important tree species, from the aspects of source of edible oil and biodiesel and pioneering capacity of degraded and desert land. Characteristics investigated were seed length, width, and weight: weight and volume of 1,000 seeds: and weight and volume of one-liter seeds. The seeds of Qingsonglingxiang No. 1, growing alone in an open space, showed the highest values in seed length (16.08 mm), width (14.48 mm) and weight (1.40 g), while those of Tree No. 160 in Ar Khorqin Banner were the lowest ones: that is, 11.48mm for length, 11.81 mm for width, and 0.73 g for weight, respectively. Traits of seeds varied quite much between trees and among areas; for example, Tree No. 38 and No. 160 produced quite different seeds in several traits, although they are adjacent to each other in the same farm. Weight of 1,000 seeds varied from 718.0 g to 1,010.1 g and volume from 0.76 L to 1.52 L. Weight of one-liter seeds were 522.3 g to 688.2 g, while the number of seeds were 603 to 935. Seeds which were soaked in the water at $4^{\circ}C$ for 2 days showed the highest germination rate (89%) in a 30-day test, which was about 10% to 40% higher than those of non-treatment and dipping treatment at $36^{\circ}C$ followed by keeping under room temperature for 2 days. 81% of seeds in the wet sand at room temperature germinated, while 23% of seeds deprived of seed coat germinated. It is necessary to understand seed traits to select superior clones or provenances for the increased, unfluctuating production of seed.

녹차 종실유의 제조법에 따른 열산화 안정성 비교 (Effects of Extraction Method on the Thermal Oxidative Stability of Seed Oils from Camellia sinensis L.)

  • Camellia sinensis L. (green tea) seed oils were prepared by roasting at $213^{\circ}C$ and pressing (RP), pressing (P), and nhexane extraction (H). The physico-chemical properties of the RP, P, and H samples, including fatty acid composition, color, and sensory characteristics were analyzed. RP, P and H samples were thermally oxidized at $180^{\circ}C$, and oxidative stability was determined by DPPH, CDA, and p-AV at 0, 20, 40, 60, and 80 min. Compared to the P and H samples, RP resulted in significantly higher thermal oxidative stability according to the DPPH, CDA, and p-AV results (p<0.05). The ratio of unsaturated fatty acids to saturated fatty acids among RP, P, and H samples were significantly different (p<0.05). The oleic acid and linoleic acid contents in green tea seed oils were 58 and 23%, respectively. Hunter's color value of lightness (L) for the RP, P, and H samples was not significant. Redness (a) of RP was $3.47{\pm}0.119$ and yellowness (b) of H was $60.10{\pm}2.483$, which were significantly different. Compared to RP samples, H and P samples had the highest color and off-odor values in the sensory evaluation. RP samples showed the highest taste value and were significant overall (p<0.05). The thermal stability of RP extraction was more stable than any other method. Camellia sinensis L. seed oil extracted by RP had better sensory characteristics than other edible oils, including soybean oil, grape seed oil, and extra virgin olive oil.

Supercritical $CO_2$ Extraction of Whole Berry Oil from Sea Buckthorn ($Hippopha\ddot{e}$ rhamnoides var. sp) Fruit

  • The whole berry, pulp, and seed of sea buckthorn fruit were extracted with supercritical $CO_2$ to produce edible oils. The effects of extraction pressure, temperature, and $CO_2$ flow rate on the oil yield and extraction rate were investigated, and the fatty acid composition, tocopherol, and carotenoid contents of the oils were compared. The results showed that the extraction rate was affected by pressure, temperature, and $CO_2$ flow rate and, in general, the yield increased with a rise in any of the 3 variables. Fatty acids in the whole berry and pulp oil were dominated by monounsaturated fatty acids (>64%), followed by saturated fatty acids (about 30%). In contrast, fatty acids in the seed oil consisted mainly of polyunsaturated (>60%) and monounsaturated fatty acids (>24%). The seed oil had a slightly higher content of tocopherols, but a much lower content of carotenoids, compared with the whole berry or pulp oil.
