Resources Use Characteristics of Higher Fungi in Byeonsanbando National Park (변산반도 국립공원 고등균류의 자원이용적 특성)

  • According to the survey on higher fungi from 2009 to 2011 and also in 2015 in Byeonsanbando National Park, a total of 2 division, 6 class, 18 orders, 61 families, 157 genera and 323 species were observed. In case of Agaricales, there were 23 families, 67 genera and 153 species; Boletales, there were 6 families, 27 genera and 45 species; Russulales, there were 3 family, 4 genera and 40 species; Polyporales, there were 6 family, 21 genera, 28 species. Thus, most of them belonged to the following 4 orders: Agaricales, Russulales, Boletales and Polyporales. Dominant species belonged to Boletaceae (37 species), Russulaceae (36 species), Agaricaceae (28 species) and Amamtaceae (25 species). For the habitat environment, the ectomycorrhizal mushrooms were 40.2% (poisonous mushrooms, 46 species; edible & medicinal mushrooms, 51 species; unknown edible & poisonous mushrooms, 26 species), litter decomposing and wood rotting fungi 35.3%(poisonous mushrooms, 10 species; edible & medicinal mushrooms, 52 species; unknown edible & poisonous mushrooms, 46species), grounding Fungi 22.3%(poisonous mushrooms, 8 species; edible & medicinal mushrooms, 31 species; unknown edible & poisonous mushrooms, 29 species). Monthly, most of poisonous mushrooms, edible & medicinal mushrooms and unknown edible & poisonous mushrooms were found in July and August. In terms of altitude, the most species were observed at 1~99m and the populations dropped by a significant level at an altitude of 200m or higher. It seemed that the most diversified poisonous mushrooms, edible & medicinal mushrooms and unknown edible & poisonous mushrooms occurred at climate conditions with a mean air temperature at $24.0{\sim}25.9^{\circ}C$, the highest air temperature at $28.0{\sim}29.9^{\circ}C$, the lowest air temperature at $20.0{\sim}21.9^{\circ}C$, a relative humidity at 77.0~79.9% and a rainfall of 300.0~499.9mm.

  • Acetoin (AC) is a volatile platform compound with various potential industrial applications. AC contains two stereoisomeric forms: (3S)-AC and (3R)-AC. Optically pure AC is an important potential intermediate and widely used as a precursor to synthesize novel optically active materials. In this study, chiral (3R)-AC production from meso-2,3-butanediol (meso-2,3-BD) was obtained using recombinant Escherichia coli cells co-expressing meso-2,3-butanediol dehydrogenase (meso-2,3-BDH), NADH oxidase (NOX), and hemoglobin protein (VHB) from Serratia sp. T241, Lactobacillus brevis, and Vitreoscilla, respectively. The new biocatalyst of E. coli/pET-mbdh-nox-vgb was developed and the bioconversion conditions were optimized. Under the optimal conditions, 86.74 g/l of (3R)-AC with the productivity of 3.61 g/l/h and the stereoisomeric purity of 97.89% was achieved from 93.73 g/l meso-2,3-BD using the whole-cell biocatalyst. The yield and productivity were new records for (3R)-AC production. The results exhibit the industrial potential for (3R)-AC production via whole-cell biocatalysis.

Identification and Distribution of the Pathogenic Microorganisms Isolated from Edible Ice in North Area of Daegu, Korea (대구시 북구지역의 식용얼음에서 세균 분포 및 동정)

  • The definition of edible ice is frozen water for the use of food manufacturing, processing, or cooking, as well as for the direct eating. It has been reported that in the process of ice manufacturing and its selling, edible ice is contaminated with some microorganisms, which causes food poisoning and gastroenteritis. It was shown that besides in the edible ice, germ growth caused by various reasons occurred in the mineral water, tap water, water filtering system, and water purifier. With public awareness, in order to examine the sanitary conditions of edible ice in the Northern area of Daegu metropolitan city, 15 places were randomly selected. As a result, 14 places were found to be contaminated with microorganisms. After incubating on the Brain Heart Infusion (BHI) agar plate, 80% of Gram-negative bacilli, 17% of Gram-positive cocci, and 3% of Gram-negative cocci were cultured. Enterobacter cloacae, Chryseomonas luteola, Pantoea spp., Klebsiella pneumoniae, Acinetobacter baumannii, Acinetobacter calcoaceticus or Providencia rettgeri were detected. Gram-positive cocci cultured in BHI agar plate from 5 specimens were identified as Staphylococcus aureus or Staphylococcus xylosus, which is well known bacteria causing strong food poisoning. This present paper raises questions on the importance and awareness of sanitary conditions of edible ice and the identification of pathogenic microorganisms living in the edible ice in relation to their distribution. The examination of sanitary conditions of edible ice in other areas in Daegu seems to be also needed to find out if there are similar cases.

Classification of the Edible Plants on the Market in Busan (시장에 유통되는 식용식물의 분류)

  • The edible plants sold at the market in Busan were classified into 8 divisions, 11 classes, 46 orders, 67 families, 156 genera, 183 species and 45 varieties, and a total of 228 kinds of plants were included in them. Among the 228 kinds of edible plants, Angiospermae were the highest number of 202 kinds (88.60%), and next came Fungi of 9 kinds (3.95%), Phaeophyta of 5 kinds (2.19%), Rhodophyta of 4 kinds (1.75%), Pteridophyta of 3 kinds (1.32%) Gymnospermae of 2 kinds (0.88%), Chlorophyta of 2 kinds (0.88%), and Lichenes of 1 kind (0.44%) in order In the taxon of family, Cruciferae, Rosaceae, Leguminoceae, Cucurbitaceae, Umbelliferae, Compositae, Liliaceae, and Gramineae include many species. In the taxon of genus, Brassica(14 species), Prunus(9), Lactuca(9), Allium(7), Citrus(5), Cucumis(4), Cichorium(4) in order contain many species. Among tile edible plants, Chamjuknamu(Cedrela sinensis) is wrongly named Gajuknamu(Ailanthns altissima). And there are others which are called in a wrong way: Endive is wrongly called Chicory, Saeneutaribeoseot, Saesongi and Padeudeknamul(Japanese hornwort), Chamnamul. Gyul and Milgam, Geumgam and Gamgyul, Banana and Pacho, Paprika and Pimang(Pimento) are the names of the same plants. The number of the kinds of edible plants is different in each season. Fall has the most edible plants, and it is followed by spring, summer and winter. The edible parts of plants which are sold at the market are leaves, young sprouts, fruits, grains, roots, stems and petioles.

Food Sources of Thiamin, Riboflavin and Niacin Based on Food Composition Table and National Annual Food Supply Data in Food Balance Sheet (우리 나라 식품수급표 자료을 분석한 티아민, 리보플라빈, 나이아시신의 주요 급원식품)

  • The purpose of the study was to find out the rich sources of vitamin B-complexes in Korea. Thirty kinds of vitamin B-complex (thiamin, riboflavin, and niacin) rich foods were selected based on vitamin content in 100g edible portion, single serving size, 100kcal energy content, and 1999 national annual food supply data. The results were summarized as follows: \circled1 Most of the thiamin rich foods selected were fishes: half of the foods selected was fishes when based on the contents in single serving size. On the other hand, about half of the foods selected were vegetables when based on the thiamin contents in 100kcal energy content. A relatively large number of food groups were included such as fishes, vegetables, beans, grains, meats, etc. when based on the contents in 100g edible portion, and national food supply data. \circled2 Most of the riboflavin rich foods selected were fishes and shellfishes, and vegetables: half of the foods selected were fishes and shellfishes when based on the content in 100g edible portion; two-thirds of the foods selected were vegetables when based on the content in single serving size; half of the foods selected were vegetables when based on the content in 100kcal. \circled3 The most important niacin sources are fishes: Most than 60% of the foods selected were fishes when based o the 100g edible portion, single serving size, and 100kcal energy content. Laver is very important source of all 3 vitamin B-complexes in Korea when based on the content in 100g edible portion, 100kcal energy content, and national annual food supply but not in single serving size. On the other hand, rice is rice is sources of all 3 vitamin B-complexes when based on the national annual food supply, but not in single serving size. On the other hand, rice sources of all 3 vitamin B-complexes when based on the national annual food supply, but is not included in 30 rich foods selected based on the content in 100g edible portion, single serving size, and 100kcal energy content. Mushrooms are also very important food sources of thiamin, riboflavin, and niacin when based on the vitamin content in 100g edible portion, single serving size, and 100kcal energy content but not included in 10 rich food selected based on the national food supply data.

Study on the Genetic Diversity and Biological Characteristics of Wild Agaricus bisporus Strains from China

  • 90 wild Agaricus strains from China, including 44 Agaricus bisporus strains identified preliminarily by isozyme electrophoresis, were studied by the techniques of SRAP and ISSR. 18 special SRAP bands and 12 special ISSR bands were analyzed, the strains were clustered and a demdrogram was obtained. The results showed that the strains were divided into 2 groups, wild A. bisporus group and the other Agaricus group. It is similar to the result of isozyme electrophoresis. 41 wild A. bisporus strains from Sichuan and Tibet were divided into 4 groups based on their growing places, suggesting the regionally difference of the strains to be quite obvious. Some white wild A. bisporus strains from Xinjiang and Tibet had special patterns, resulting in lower coefficient values with other wild A. bisporus strains. The biological characteristics of three wild A. bisporus strains were analyzed, and the results showed: 1. The wild strains grew slowly on PDA medium with weak appressed mycelia, and grew normally in kernel or fermented cottonseed shell substrate. 2. They grew faster than control strain As2796 under lower temperature of $16^{\circ}C$, and higher temperature of $32^{\circ}C$, with optimum growing temperature of $20-24^{\circ}C$, which was $4^{\circ}C$ lower than that of control strain. 3. In the cultivation with manure compost via twice fermentation, the mycelia grew normally in compost and quite slowly in casing soil, and the fruitbodies occurred less and late with easily opening and low production. 4. The fruitbody was off-white with flat and scaled cap, long stipe and dark gill. The bisporus basidia occupied 70-80% and trisporus basidia 20-30% of the total basidia. 5. Heterokaryotic monospore isolates could fruit in cultivation, and the homokaryotic isolates could cross with those derived from overseas wild A.bisporus strains. 6. The electrophoresis phenotype of isozymes such as esterase etc. belonged to high production type (H type). 7. The RAPD patterns made much difference from those of high production, good quality or hybrid strains, which indicated that the wild strains produce a new kind of RAPD type.

Characteristics of Mineral Mg Dissolving Sensor in Edible Water using GMR-SV Device (거대자기저항 스핀밸브 소자를 이용한 음용수 미네랄 Mg 용해센서 특성 연구)

  • The measurement dissolution sensor system using GMR-SV device with magnetic sensitivity of 0.8 %/Oe and Mg-film thick of 200 nm and Mg-foil thick of 50 mm was fabricated and characterized. During the water dissolving process of Mg-film and Mg-foil, the subtle variation of magnetic field by the decrease of current in solenoid was detected by the GMR-SV sensor. The variations of Mg bubble number and ORP as a function of time for three different kinds of edible, tap, and distilled water, are measured and compared. A After 45 min, the speed of fast dissolving Mg was shown the order of edible > tap > DI water. The variation of output magnetoresistance as a function of dissolved time of Mg-film and Mg-foil for edible water, which is composed of mineral content of $0.8{\sim}5.4\;mg/l$ was investigated. The response times for the dissolution in edible water were 5 min and 20 min, respectively. From the measurement of dissolving time and speed for Mg-film and Mg-foil using GMR-SV device, the mineral Mg sensor system in edible water can be possible to develop.

Drying characteristics and physical properties of medicinal and edible mushrooms (약용버섯과 식용버섯의 건조방법에 따른 품질특성)

  • In this study, the drying characteristics of medicinal (Ganoderma lucidum, Phellinus linteus) and edible (Pleurotus eryngii, Lentinus edodes) mushrooms were investigated. Their shrinkage ratios, Hunter's color values, and rehydration ratios were evaluated. Mushrooms were cut with rectangular (1 cm) and square ($1{\times}1cm$) shapes, and dried in the room temperature and at $50^{\circ}C$ and $70^{\circ}C$ using hot-air dryer. Initial moisture contents of edible mushrooms were higher than those of medicinal mushrooms, whereas final moisture contents were vice versa. Drying rate was the highest when drying at $70^{\circ}C$ (p<0.05). Moreover the drying rate of square slices was higher than that of rectangular slices (p<0.05). The shrinkage ratios of both medicinal mushrooms and edible mushrooms were the least when drying at $50^{\circ}C$ and $70^{\circ}C$, respectively (p<0.05). The changes of color values were less in the medicinal mushrooms dried in the room temperature than in the hot-air dried medicinal mushrooms (p<0.05). However, in case of edible mushrooms, the changes of color values were less in the hot-air dried mushrooms (p<0.05). Rehydration ratios of medicinal mushrooms dried at $50^{\circ}C$ was the highest (p<0.05). In contrast, rehydration ratios of edible mushrooms was the highest when drying in the room temperature (p<0.05). Thus, the hot-air drying at $50^{\circ}C$ would be suggested as the efficient drying method for both medicinal mushrooms and edible mushrooms.

Analysis of the Genome Sequence of Strain GiC-126 of Gloeostereum incarnatum with Genetic Linkage Map

  • Gloeostereum incarnatum has edible and medicinal value and was first cultivated and domesticated in China. We sequenced the G. incarnatum monokaryotic strain GiC-126 on an Illumina HiSeq X Ten system and obtained a 34.52-Mb genome assembly sequence that encoded 16,895 predicted genes. We combined the GiC-126 genome with the published genome of G. incarnatum strain CCMJ2665 to construct a genetic linkage map (GiC-126 genome) that had 10 linkage groups (LGs), and the 15 assembly sequences of CCMJ2665 were integrated into 8 LGs. We identified 1912 simple sequence repeat (SSR) loci and detected 700 genes containing 768 SSRs in the genome; 65 and 100 of them were annotated with gene ontology (GO) terms and KEGG pathways, respectively. Carbohydrate-active enzymes (CAZymes) were identified in 20 fungal genomes and annotated; among them, 144 CAZymes were annotated in the GiC-126 genome. The A mating-type locus (MAT-A) of G. incarnatum was located on scaffold885 at 38.9 cM of LG1 and was flanked by two homeodomain (HD1) genes, mip and beta-fg. Fourteen segregation distortion markers were detected in the genetic linkage map, all of which were skewed toward the parent GiC-126. They formed three segregation distortion regions (SDR1-SDR3), and 22 predictive genes were found in scaffold1920 where three segregation distortion markers were located in SDR1. In this study, we corrected and updated the genomic information of G. incarnatum. Our results will provide a theoretical basis for fine gene mapping, functional gene cloning, and genetic breeding the follow-up of G. incarnatum.
