miR-485 Acts as a Tumor Suppressor by Inhibiting Cell Growth and Migration in Breast Carcinoma T47D Cells

  • MicroRNAs (miRNAs) are small, non-coding RNAs (18-25 nucleotides) that post-transcriptionally modulate gene expression by negatively regulating the stability or translational efficiency of their target mRNAs. In this context, the present study aimed to evaluate the in vitro effects of miR-485 mimics in breast carcinoma T47D cells. Forty-eight hours after T47D cells were transfected with miR-485 mimics, an MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay was utilized to determine the effects on cell viability. Colony formation and cell migration assays were adopted to determine whether miR-485 affects the proliferation rates and cell migration of breast carcinoma T47D cells. Our results showed that ectopic expression of miR-485 resulted in a significant decrease in cell growth, cell colony formation, and cell migration. These findings suggest that miR-485 might play an important role in breast cancer by suppressing cell proliferation and migration.

Ectopic Migration of an Adult Heartworm in a Dog with Dirofilariasis

  • A 3-yr-old female mongrel dog was referred to the Veterinary Teaching Hospital of Chungnam National University in the Republic of Korea. An adult heartworm, Dirofilaria immitis, was found in the abdominal cavity of the dog during spaying. Dirofilariasis in this dog was also diagnosed by modified Knott's test, ELISA test, and PCR analysis. The present case is the first report on the migration of an adult dog heartworm to the abdominal cavity of a dog in the Republic of Korea.

RNA Binding Protein Rbms1 Enables Neuronal Differentiation and Radial Migration during Neocortical Development by Binding and Stabilizing the RNA Message for Efr3a

  • Various RNA-binding proteins (RBPs) are key components in RNA metabolism and contribute to several neurodevelopmental disorders. To date, only a few of such RBPs have been characterized for their roles in neocortex development. Here, we show that the RBP, Rbms1, is required for radial migration, polarization and differentiation of neuronal progenitors to neurons in the neocortex development. Rbms1 expression is highest in the early development in the developing cortex, with its expression gradually diminishing from embryonic day 13.5 (E13.5) to postnatal day 0 (P0). From in utero electroporation (IUE) experiments when Rbms1 levels are knocked down in neuronal progenitors, their transition from multipolar to bipolar state is delayed and this is accompanied by a delay in radial migration of these cells. Reduced Rbms1 levels in vivo also reduces differentiation as evidenced by a decrease in levels of several differentiation markers, meanwhile having no significant effects on proliferation and cell cycle rates of these cells. As an RNA binding protein, we profiled the RNA binders of Rbms1 by a cross-linked-RIP sequencing assay, followed by quantitative real-time polymerase chain reaction verification and showed that Rbms1 binds and stabilizes the mRNA for Efr3a, a signaling adapter protein. We also demonstrate that ectopic Efr3a can recover the cells from the migration defects due to loss of Rbms1, both in vivo and in vitro migration assays with cultured cells. These imply that one of the functions of Rbms1 involves the stabilization of Efr3a RNA message, required for migration and maturation of neuronal progenitors in radial migration in the developing neocortex.

이소갑상선 19예의 신티그라피 소견 (Scintigraphic Findings of Nineteen Cases of Ectopic Thyroid)

  • 저자들은 이소갑상선이 의심되어 갑상선신티그라피로 확진된 19명의 환자를 대상으로 갑상선 스캔과 갑상선 기능검사의 결과 및 임상증상등을 조사하여 다음과 같은 결과를 얻었다. 1) 이소갑상선의 성별분포는 여자 16명, 남자3명으로 여자가 남자의 5.3배였고, 30세 이전인 경우가 97%였다. 2) 이소갑상선이 발견된 위치는 설근부 10예, 설하부1예, 전 후두부 5예 그리고 Combined form이 3예였다. 3) 갑상선기능검사를 시행한 16예에서 갑상선기능저하증이 7예, 준임상적 갑상선기능저하증이 4예 그리고 갑상선기능 정상이 5예였다. 4) 설갑상선의 경우는 갑상선기능저하증이 90%에서 나타났으나, 이보다 아래로 내려온 경우에는 33%로 갑상선 기능이상은 이소갑상선의 위치에 따라 유의한 차이가 있었다(p<0.05). 5) 갑상선의 주된 임상증상은 설근부의 인후이물감, 연하곤란이었고, 이보다 아래에 위치한 경우는 전경부의 종괴가 주된 임상증상이었다. 6) 갑상선기능저하증으로 진단된 7예와 준임상적 갑상선 기능저하증 4예중 3예에서는 갑상선호르몬을 투여하였고 갑상선 기능이 정상인 2예에서 이소갑상선 제거술이 시행되었으며 그 나머지는 추적관찰하였다. 7) 설근부나 전경부에 종괴가 있을 경우에는 반드시 갑상선스캔을 시행하여 이소갑상선의 가능성을 배제하여야 한다. 또한 이소갑상선의 기능이 정상인 환자에서도 갑상선기능저하증으로 진행될 수 있으므로 추적관찰이 필요하다.

Knockdown of Circ_0000144 Suppresses Cell Proliferation, Migration and Invasion in Gastric Cancer Via Sponging MiR-217

  • Previous studies have uncovered the role of circ_0000144 in various tumors. Here, we investigated the function and mechanism of circ_0000144 in gastric cancer (GC) progression. The expression of circ_0000144 in GC tissues and cells was detected through quantitative real-time polymerase chain reaction (qRT-PCR) method. Gain- and loss-of-function experiments including colony formation, wound healing and transwell assays were performed to examine the role of circ_0000144 in GC cells. Furthermore, western blot was conducted to determine the expressions of epithelial mesenchymal transition (EMT)-related proteins. The interaction between circ_0000144 and miR-217 was analyzed by bioinformatic analysis and luciferase reporter assays. The circ_0000144 expression was obviously upregulated in GC tissues and cells. Silencing of circ_0000144 inhibited cell proliferation, migration and invasion of GC cells, but ectopic expression of circ_0000144 showed the opposite results. Moreover, circ_0000144 sponged miR-217, and rescue assays revealed that silencing miR-217 expression reversed the inhibitory effect of circ_0000144 knockdown on the progress of GC. Our findings reveal that circ_0000144 inhibition suppresses GC cell proliferation, migration and invasion via absorbing miR-217, providing a new biomarker and potential therapeutic target for treatment of GC.

  • 1세된 남아에서 세열낭종을 의심한 좌측 경부 종양이 수술을 시행한 결과 고형종양이였고, 조직검사상 이소성 흉선으로 판명되었다. 술후 초음파상 흉선은 정상적으로 있었고, 현재 환자의 면역기능은 아무 이상없이 정상으로 자라고 있다.

인간 신장암 Caki세포에서 Par-4에 의한 MMP-2 활성 저해를 통한 세포 이동 조절 (Par-4 Modulates Cell Migration through Inhibition of MMP-2 Activity in Human Renal Carcinoma Caki Cells)

  • Par-4는 다양한 세포사멸 자극에 세포 사멸을 조절하고, 종양 억제기능을 가지고 있다. 그러나, Par-4에 의한 암세포의 이동 및 침윤에 대한 연구는 수행되지 않았다. 본 연구에서 Par-4단백질의 과발현이 인간 신장암 Caki세포에서 MMP-2의 활성화를 억제하지만 MMP-9 활성에는 영향을 주지 않았다. Par-4에 의한 MMP-2의 활성 억제는 leucine zipper domain이 결실된 Par-4 에서는 확인되지 않았다. Par-4 siRNA를 이용한 knock-down 실험에서 PMA 처리 시 세포이동 및 침윤 증가함을 확인하였다. Par-4의 과발현과 knock-dwon에서 MMP-2 mRNA 발현의 변화를 확인 할 수 없었다. 이 점은 Par-4 매개의 MMP-2 활성 억제는 전사 후 조절을 통하여 야기됨을 추측 할 수 있다.

STC2 is upregulated in hepatocellular carcinoma and promotes cell proliferation and migration in vitro

  • The human glycoprotein, stanniocalcin 2 (STC2) plays multiple roles in several tumor types, however, its function and clinical significance in hepatocellular carcinoma (HCC) remain unclear. In this study, we detected STC2 expression by quantitative real-time PCR and found STC2 was upregulated in HCC tissues, correlated with tumor size and multiplicity of HCC. Ectopic expression of STC2 markedly promoted HCC cell proliferation and colony formation, while silencing of endogenous STC2 resulted in a reduced cell growth by cell cycle delay in G0/G1 phase. Western blot analysis demonstrated that STC2 could regulate the expression of cyclin D1 and activate extracellular signal-regulated kinase 1/2 (ERK1/2) in a dominant-positive manner. Transwell chamber assay also indicated altered patterns of STC2 expression had an important effect on cell migration. Our findings suggest that STC2 functions as a potential oncoprotein in the development and progression of HCC as well as a promising molecular target for HCC therapy.

유주 비장 1례 (A Case of Wandering Spleen)

  • Wandering spleen is a rare condition characterized by migration of the spleen from its normal position due to laxity or absence of the supporting splenic ligaments. Ultrasonography shows the absence of the spleen in left upper quadrant (LUQ) and the presentation in the lower abdomen of a homognous mass whose sonographic appearance is consistent with that of a spleen. Contrast-enhanced CT reveals the absence of the spleen in LUQ, a soft tissue mass resembling spleen in the lower abdomen, and the splenic pedicle of whirled appearance. We present a case of wandering spleen, which could be diagnosed with US and CT by the ectopic location of spleen and the whirling pattern of splenic pedicle.

  • Permanent canine are very important teeth in point of function as well as esthetic. Therefore impaction of canines is a frequently encountered clinical problems. The incidence of impaction ranges between 1-3%. The cause of canine impaction can be result of ectopic position of tooth germ, defect of dental follicle, early loss or delayed exfoliation primary canine, space problem, supernumarary teeth, cyst, odontoma, Impaction frequently involves further complication such as root resorption of adjacent teeth, cyst formation and migration of the neighboring teeth. Various treatment modalities include extraction of primary canine, surgical opening and orthodontic traction, autotransplantation, surgical extraction of impacted canine.
