Single-strand DNA Binding of Actinomycin D with a Chromophore 2-Amino to 2-Hydroxyl Substitution

  • A modified actinomycin D was prepared with a hydroxyl group that replaced the amino group at the chromophore 2-position, a substitution known to strongly reduce affinity for double-stranded DNA. Interactions of the modified drug on single-stranded DNAs of the defined sequence were investigated. Competition assays showed that 2-hydroxyactinomycin D has low affinity for two oligonucleotides that have high affinities ($K_a\;=\;5-10{\times}10^6\;M^{-1}$ oligomer) for 7-aminoactinomycin D and actinomycin D. Primer extension inhibition assays performed on several single-stranded DNA templates totaling around 1000 nt in length detected a single high affinity site for 2-hydroxyactinomycin D, while many high affinity binding sites of unmodified actinomycin D were found on the same templates. The sequence selectivity of 2-hydroxyactinomycin D binding is unusually high and approximates the selectivity of restriction endonucleases. Binding appears to require a complex structure, including residues well removed from the polymerase pause site.

  • The sequential sampling inspection method is an extension of the double-sampling and multiple-sampling methods and its theory is based on the sequential probability ratio test(SPRT). In this paper, the characteristics of SPRT for testing the propotion of defectives are approximated by using the estimated excess over the boundaries. The use of the estimated excess shows good performances in estimating the operating characteristic function and the average sample number of SPRT compared to the method by neglecting the excess. It also makes it possible to determine the boundary values which satisfy the desired error probabilities.

  • Electrification of existing railroad as well as extension of double track, need a large amount of electric energy. Especially, increase in energy consumption of high cost causes much problems in domestic economy. It is neccessary to save energy for the crisis of exhaust of fossil energy. About $20{\~}25\%$ of regenerated energy of an electric railcar is lost on down slope run or on braking. In order to save energy in electric railway system, Therefor, application of energy regeneration system is proposed and introduced in the present paper.

  • In this paper, a cyclic mechanical model is presented to simulate the behaviour of top and seat with web angle beam-to-column connections. The introduced mechanical model is compared with Eurocode 3 Annex J, its extension, and with experimental data. To have a better insight regarding the actual response of the joints, available results of the experiments, carried out on full-scale top and seat angle joints under monotonic and cyclic loading, are first considered. Subsequently, a finite element model of the test setup is developed. The application of the proposed model, its comparisons with the experimental curves and with the Eurocode 3 Annex J and with its modification, clearly show the excellent quality of the model proposed.

  • In this paper we present a method of determining the unknown degree of any 2D discrete linear shift-invariant system which is characterized only by the coefficients of the double power series of a transfer function, i.e. a 2D impulse response array. Our method is based on a 2D extension of Berlekamp-Massey algorithm for synthesis of linear feedback shift registers, and it gives a novel approach to identification and approximation of 2D linear systems, which can be distinguished in its simplicity and potential of applicability from the other 2D Levinson-type algorithms. Furthermore, we can solve problems of 2D Pade approximation and 2D system reduction on a reasonable assumption in the context of 2D linear systems theory.

  • Autophagy is an evolutionally conserved cytoplasmic degradation system in which varieties of materials are sequestered by a double membrane structure, autophagosome, and delivered to the lysosomes for the degradation. Due to the wide varieties of targets, autophagic activity is essential for cellular homeostasis. Recent genetic evidence indicates that autophagy has a crucial role in the regulation of animal lifespan. Basal level of autophagic activity is elevated in many longevity paradigms and the activity is required for lifespan extension. In most cases, genes involved in autophagy and lysosomal function are induced by several transcription factors including HLH-30/TFEB, PHA-4/FOXA and MML-1/Mondo in long-lived animals. Pharmacological treatments have been shown to extend lifespan through activation of autophagy, indicating autophagy could be a potential and promising target to modulate animal lifespan. Here we summarize recent progress regarding the role of autophagy in lifespan regulation.

  • This study investigated the length-tension and velocity-force relations of the torso erectors. A myoelectric based approach was used wherein a dynamic biomechanical model incorporating active and passive tissue charactreistics provided music kinematic estimates during controlled sagittal plan extension motions. A double linear optimization formulation from the literatured provided muscle tension estimates. The data supported a linear length-tension relation toward full flexion for both the erector spinae and latissimus muscles. Velocity trends agreed with that predicted by Hill's exponential relation. The results have implications for muscle tension estimation in biomechanical torso modeling, and suggest a possible low back pain injury mechanism.

  • A communication network protocol among programable devices built-i manufacturing field, MAP (Manufacturing Automation Protocol), has to provide the job transfer service carrying out a lot of jobs distributively, and the MMS( manufacturing message specification) defines the above application layer protocol. In this paper user software required in job transfer was implemented. So as to provide each service with compatibility and extension, each module was designed according to the functions. A method is selected in double-assigning their domain and program applicable even in complex process, which loads and proceeds several programs sequentially. In order to confirm the logical validity of the designed protocol, local applying test is accomplished for the application layer of response station. Modelling the job flow process, each serice module is verified with the I/O primitives.

  • 80-nm self-aligned n-and p-channel I-MOS devices were demonstrated by using a novel fabrication method featuring double sidewall spacer, elevated drain structure and RTA process. The fabricated devices showed a normal transistor operation with extremely small subthreshold swing less than 12.2 mV/dec at room temperature. The n- and p-channel I-MOS devices had an ON/OFF current of 394.1/0.3 ${\mu}A$ and 355.4/8.9 ${\mu}A$ per ${\mu}m$, respectively. We also investigated some critical issues in device design such as the junction depth of the source extension region and the substrate doping concentration.
