Modified LOGIT(MLOGIT) Transformation: Prediction of $IC_{50}$ Value from Two Arbitrary Concentration Data

  • A LOGIT transformation is a method to estimate IC50 values with two arbitrary concentration data when complete dose response curves(DRCs) are not available. We propose a modified LOGIT transformation (MLOGIT) which predicts IC50 values more accurately than the conventional LOGIT method.

Influence of Level of Dietary Inorganic and Organic Copper and Energy Level on the Performance and Nutrient Utilization of Broiler Chickens

  • An experiment was conducted to determine the influence of dietary inorganic (copper sulfate) and organic (copper proteinate) forms of copper and energy level on performance and nutrient utilization of broiler chickens. Two hundred day-old commercial Vencobb broiler chicks were purchased and randomly distributed to 20 cages of 10 birds each. These replicates were randomly assigned to one of five treatments in a (($2{\times}2$)+1) factorial arrangement. These two factors were sources of Cu ($CuSO_{4}$ vs. Cuproteinate) and dose of Cu supplements (200 mg and 400 mg/kg dietary dry matter) and the control (no supplemental Cu). After the starter period (up to 3 weeks), from d 22 onwards another factor i.e. energy at two levels (2,900 vs. 2,920 kcal/kg diet) was introduced with the previous factorial arrangements by subdividing each replicate into two equal parts, for two energy levels, without disturbing the dose and source of Cu supplement. Cu-salt supplementation linearly increased (p<0.01) live weight (LW), live weight gain (LWG) and feed conversion ratio (FCR) at 3 weeks, whereas cumulative feed intake (CFI) was unaffected (p>0.05). LWG and FCR were higher (p<0.01) in Cu-proteinate supplemented birds compared to $CuSO_{4}$ supplementation. A linear dose response (p<0.01) of Cu was found for the performance of broiler chickens. Birds having a higher energy level in the finisher stage increased (p<0.01) LWG and FCR. Cumulative feed intake was similar (p>0.05) across the groups up to the 5th week. Cu-proteinate increased performance of broiler chickens compared to $CuSO_{4}$. Dose of supplemental Cu-salt irrespective of source showed a linear response (p<0.01) for performance. Supplementation of Cu-proteinate increased metabolizability of DM (p<0.01), NFE (p<0.05), total carbohydrate (p<0.01) and OM (p<0.01) at the starter period. Increased dose of Cu-salt linearly increased (p<0.01) metabolizability of DM, CP, CF, NFE and OM. Higher energy level in the diet improved DM (p<0.05), EE (p<0.01), NFE (p = 0.01), total carbohydrate (p<0.01) and OM (p<0.01) metabolizability. Cu-proteinate supplementation showed better nutrient utilization compared to CuSO4. Dose of Cu linearly increased DM, CP, EE, NFE, total carbohydrate and OM metabolizability. CF metabolizability was unaffected (p>0.05) among the treatments. In conclusion, dietary supplementation of Cu-salt more than the requirement may improve performance and nutrient utilization in broiler chickens even with a high energy finisher diet. Cu-proteinate showed better performance and nutrient utilization compared to $CuSO_{4}$.

고용량 방사선 조사 후 골육종 세포주(Saos-2)의 아포프토시스 발생 (Induction of Apoptosis in Human Osteosarcoma Cell Lines(Saos-2) by Single Fraction High Dose Irradiation)

  • A single fraction of 50 Gy extracorporeal irradiation, as a modality of limb-sparing operation, has been used to achieve tumor necrosis in osteosarcoma. Although this modality of radiation therapy preserving the mobility of a joint is commonly practiced, the precise knowledge on the radiobiological response of osteosarcoma cell has remained to be elucidated. We therefore observed whether a single high dose irradiation caused apoptosis in osteosarcoma cells and whether the commitment to apoptosis was associated with cell kinetics. We also investigated radiation dose response along the time course for development of apoptosis following single high dose irradiation. The morphologic change in apoptosis was observed by fluorescence with Hoechst 33258 and the degree and the fraction of cells by flow cytometry. Irradiation of osteosarcoma cells with 10, 30 and 50 Gy resulted in chromatin condensation and apoptotic body formation. The degree of apoptosis in osteosarcoma cells was $29.5{\pm}3.56%$, $39.9{\pm}4.83%$ at 24 and 48 hours after 10 Gy irradiation ; $41.1{\pm}3.93%$, $66.9{\pm}5.21%$ at 24 and 48 hours after 30 Gy irradiation ; and $48.0{\pm}3.69%$, $75.6{\pm}4.65%$ at 24 and 48 hours after 50 Gy irradiation. The fraction of cells in cell-cycle kinetic was $39.2{\pm}4.3%$ in G2/M, $22.1{\pm}4.65%$ in G1 at 24 hours after 10 Gy irradiation ; $51.0{\pm}4.3%$ in G2/M, $20.4{\pm}4.7%$ in G1 at 48 hours after 10 Gy irradiation ; $40.3{\pm}3.9%$ in G2/M, $26.1{\pm}4.7%$ in G1 at 24 hours after 30 Gy irradiation ; $59.2{\pm}3.9%$ in G2/M, $5.9{\pm}5.1%$ in G1 at 48 hours after 30 Gy irradiation ; and $44.3{\pm}4.2%$ in G2/M, $21.1{\pm}3.5%$ in G1 at 24 hours after 50 Gy irradiation. The fraction of cells at 48 hours after 50 Gy irradiation could not be observed because of irradiation induced cell death of most of cells. All values for irradiated cells showed accumulation in G2/M phase and reduction in G1 phase, irrespective of irradiation dose. The results suggest that a single fraction of high dose irradiation with 50 Gy results in accumulation of cells at G2/M phase, leading to apoptosis.

Efficacy Analysis of Simplified Intensity-modulated Radiotherapy with High or Conventional Dose and Concurrent Chemotherapy for Patients with Neck and Upper Thoracic Esophageal Carcinoma

  • For patients with neck and upper thoracic esophageal carcinoma, it is difficult to control lymph node metastases with conventional dose therapy. In this study, we assessed the feasibility of simplified intensity-modulated radiotherapy (sIMRT) and concurrent chemotherapy for 44 patients and boosted high-dose to metastatic lymph nodes. Three radiation treatment volumes were defined: PGTVnd, with which 68.1Gy was delivered in high dose group (hsIMRT group), and 60Gy in the conventional dose group (csIMRT group); PTV1, featuring 63.9Gy in the hsIMRT group and 60Gy in the csIMRT group; PTV2, with 54Gy given to both groups. The sIMRT plan included 5 equi-angular coplanar beams. All patients received the cisplatin and 5-FU regimen concurrently with radiotherapy. The treatment was completed within six weeks and one case with grade three acute bronchitis was observed in hsIMRT group. For esophageal lesions, 80% complete response (CR) and 20% partial response (PR) rates were found in the hsIMRT group, and 79.2% CR, with 20.8% PR, in the csIMRT group; for lymph node lesions, 75% CR and 25% PR rates were observed in the hsIMRT group, with 45.8% and 37.5% respectively in the csIMRT group (P<0.05). The differences in 1-, 2- and 3-year relapse-free survival rates were all statistically significant (P<0.05). The major toxicity observed in both groups was Grade I~II leucopenia. sIMRT can generate a desirable dose distribution in treatment of neck and upper thoracic esophageal carcinoma with a better short-term efficacy. Boosted high dosing to metastatic lymph nodes can increase the relapse-free survival rate.

6 MV 광자선과 6 MeV 전자선 하에서 TLD 기판 위에 얹힌 금속 박막의 효과 (The Effects of Metal Plate loaded on TLD chip in 6 MV Photon and 6 MeV Electron Beams)

  • 본 실험은 의료용 가속기로부터 나오는 6MV 광자선과 6 MeV 전자선을 고체 팬텀위의 LiF 열형광 선량계 (TLD-l00)에 쪼여서 수행하였다. TLD-l00의 방사선 반응감도를 증가시키기 위해 TLD-l00 기판 (표면적 3.2 $\times$ 3.2 $\textrm{mm}^2$) 위에 같은 면적의 금속박막 (주석 혹은 금)을 얹어서 실험하였다. SSD l00cm, 방사선장의 크기 10$\times$10 $\textrm{cm}^2$의 조건 하에서 표면 흡수선량을 측정하였다. 측정결과 각 금속들로 인하여 TLD-l00 의 신호강도가 증강된 것이 관측되었다. 그리고 표면 흡수선량이 방사선량에 따라서 매우 선형적인 값을 가지는 것으로 나타났다 .6 MV 광자선의 경우 1 mm 의 금속박막을 TLD-l00 에 얹은 결과 표면 흡수선량이 각 각 14%, 56% 증가되었다 .6MeV의 전자선의 경우에는 금박막은 TL 반응감도가 13% 증가되었으나 주석의 경우에는 전혀 변화가 없었다. 금속박막을 얹은 TLD-l00의 방사선량 반응감도는 금속박막의 전자 입자밀도에 따라 증가하는 것으로 관측되었다. 이것은 TLD-l00보다 큰 전자밀도를 가진 부가물질(금속박막 )로부터 TLD-l00으로 산란전자가 유입되는 데 기인하는 것으로 보인다. 이 결과로부터 금속박막을 얹은 TL 선량계가 치료광자선용 증폭 선량계로서의 역할을 할 수 있을 것임을 시사한다. 즉 금속박막으로 인해서 TLD-l00 의 방사선 량 반응감도가 증가되었으므로 높은 감도의 보다 작은 TL 선량계의 개발이 가능하게 되었다.

Analytical Consideration of Surface Dose and Kerma for Megavoltage Photon Beams in Clinical Radiation Therapy

  • Background: In radiation therapy, estimation of surface doses is clinically important. This study aimed to obtain an analytical relationship to determine the skin surface dose, kerma and the depth of maximum dose, with energies of 6 and 18 megavoltage (MV). Materials and Methods: To obtain the dose on the surface of skin, using the relationship between dose and kerma and solving differential equations governing the two quantities, a general relationship of dose changes relative to the depth was obtained. By dosimetry all the standard square fields of $5cm{\times}5cm$ to $40cm{\times}40cm$, an equation similar to response to differential equations of the dose and kerma were fitted on the measurements for any field size and energy. Applying two conditions: a) equality of the area under dose distribution and kerma changes in versus depth in 6 and 18 MV, b) equality of the kerma and dose at $x=d_{max}$ and using these results, coefficients of the obtained analytical relationship were determined. By putting the depth of zero in the relation, amount of PDD and kerma on the surface of the skin, could be obtained. Results: Using the MATLAB software, an exponential binomial function with R-Square >0.9953 was determined for any field size and depth in two energy modes 6 and 18MV, the surface PDD and kerma was obtained and both of them increase due to the increase of the field, but they reduce due to increased energy and from the obtained relation, depth of maximum dose can be determined. Conclusions: Using this analytical formula, one can find the skin surface dose, kerma and thickness of the buildup region.

Toxic detection in mine water based on proteomic analysis of lysosomal enzymes in Saccharomyces cerevisiae

  • Objectives Lysosome is the cell-organelle which is commonly used as biomonitoring tool in environmental pollution. In this study, the lysosomal proteomic of the yeast Saccharomyces cerevisiae was analyzed for utilization in the detection of toxic substances in mine water samples. Methods This work informs the expression of lysosomal proteomic in yeast in response with toxic chemicals, such as sodium meta-arsenite and tetracycline, for screening specific biomarkers. After that, a recombinant yeast contained this biomarker were constructed for toxic detection in pure toxic chemicals and mine water samples. Results Each chemical had an optimal dose at which the fluorescent protein intensity reached the peak. In the case of water samples, the yeast showed the response with sample 1, 3, 4, and 5; whereas there is no response with sample 2, 6, and 7. Conclusions The recombinant yeast showed a high ability of toxic detection in response with several chemicals such as heavy metals and pharmaceuticals. In the case of mine water samples, the response varied depending on the sample content.

Clinical predictive factors of pathologic tumor response after preoperative chemoradiotherapy in rectal cancer

  • Purpose: The aim of this study was to identify clinical predictive factors for tumor response after preoperative chemoradiotherapy (CRT) in rectal cancer. Materials and Methods: The study involved 51 patients who underwent preoperative CRT followed by surgery between January 2005 and February 2012. Radiotherapy was delivered to the whole pelvis at a dose of 45 Gy in 25 fractions, followed by a boost of 5.4 Gy in 3 fractions to the primary tumor with 5 fractions per week. Three different chemotherapy regimens were used (5-fluorouracil and leucovorin, capecitabine, or tegafur/uracil). Tumor responses to preoperative CRT were assessed in terms of tumor downstaging and pathologic complete response (ypCR). Statistical analyses were performed to identify clinical factors associated with pathologic tumor response. Results: Tumor downstaging was observed in 28 patients (54.9%), whereas ypCR was observed in 6 patients (11.8%). Multivariate analysis found that predictors of downstaging was pretreatment relative lymphocyte count (p = 0.023) and that none of clinical factors was significantly associated with ypCR. Conclusion: Pretreatment relative lymphocyte count (%) has a significant impact on the pathologic tumor response (tumor downstaging) after preoperative CRT for locally advanced rectal cancer. Enhancement of lymphocyte-mediated immune reactions may improve the effect of preoperative CRT for rectal cancer.

SPECTRUM WEIGHTED RESPONSES OF SEVERAL DETECTORS IN MIXED FIELDS OF FAST AND THERMAL NEUTRONS

  • The spectrum weighted responses of various detectors were calculated to provide guidance on the proper selection and use of survey instruments on the basis of their energy response characteristics on the neutron fields. To yield the spectrum weighted response, the detector response functions of 17 neutron-measuring devices were numerically folded with each of the produced calibration neutron spectra through the in-house developed software 'K-SWR'. The detectors' response functions were taken from the IAEA Technical Reports Series No. 403 (TRS-403). The reference neutron fields of 21 kinds with 2 spectra groups with different proportions of thermal and fast neutrons have been produced using neutrons from the $^{241}Am$-Be sources held in a graphite pile, a bare $^{241}Am$-Be source, and a DT neutron generator. Fluence-average energy ($E_{ave}$) varied from 3.8 MeV to 16.9 MeV, and the ambient-dose-equivalent rate [$H^*(10)/h$] varied from 0.99 to 16.5 mSv/h.

Simulation and design of individual neutron dosimeter and optimization of energy response using an array of semiconductor sensors

  • Many researches have been done to develop and improve the performance of personal (individual) dosimeter response to cover a wide of neutron energy range (from thermal to fast). Depending on the individual category of the dosimeter, the semiconductor sensor has been used to simplify and lightweight. In this plan, it's very important to have a fairly accurate counting of doses rate in different energies. With a general design and single-sensor simulations, all optimal thicknesses have been extracted. The performance of the simulation scheme has been compared with the commercial and laboratory samples in the world. Due to the deviation of all dosimeters with a flat energy response, in this paper, has been used an idea of one semi-conductor sensor to have the flat energy-response in the entire neutron energy range. Finally, by analyzing of the sensors data as arrays for the first time, we have reached a nearly flat and acceptable energy-response. Also a comparison has been made between Lucite-PMMA ($H_5C_5O_2$) and polyethylene-PE ($CH_2$) as a radiator and $B_4C$ has been studied as absorbent. Moreover, in this paper, the effect of gamma dose in the dosimeter has been investigated and shown around the standard has not been exceeded.
