Bioavailability Test of TestoTM Tablets (Methyltestosterone, 25 mg) in Male Healthy Volunteers by a Gas-chromatography/Mass Selective Detector

  • A simple and specific method for determination of methyltestosterone (MT) has been established by a gas chromatography/mass selective detector and applied in plasma of healthy male volunteers received a single oral dose of 50 mg MT $(Testo^{TM}\;tablets,\;25\;mg)$ for bioavailability test. This method involves using liquid-liquid extraction of the sample with diethyl ether and derivatization with MSTFA. MT showed good resolution in this condition. The detection limit of quantitation was 5 ng/ml. A good linearity (r>0.996) was obtained at the range of 5-250 ng/ml of MT. Intra-day precision and accuracy were 2.76-12.56% and 0.39-8.01 %, and inter-day precision and accuracy were 2.29-17.69% and 0.42-7.99%, respectively. The established method was applied on bioavailability test of MT in human volunteers. The value of $AUC_{0\;to\;last}$ to last was $264.5{\pm}123.9\;ng{\cdot}hr/ml$ and that of $AUC_{0\;to\;inf}$ was determined to be $275.2{\pm}126.5\;ng{\cdot}hr/ml$. The values of $C_{max}$ and $T_{max}$ were $95.9{\pm}67.1\;ng/ml$ and $1.13{\pm}0.9\;hr$, respectively. The mean elimination half-life $(t_{1/2})$ was $4.4{\pm}0.9\;hr$. This analytical method is suitable and useful for the pharmacokinetics and bioequivalence studies of MT.

Study on Electrical Properties of X-ray Sensor Based on CsI:Na-Selenium Film

  • In this paper, we have introduced the x-ray detector built with a CsI:Na scintillation layer deposited on amorphous selenium. To determine the thickness of the CsI:Na layer, we have estimated the transmission spectra and the absorption of continuous x-rays in diagnostic range by using computer simulation (MCNP 4C). A x-ray detector with 65 ${\mu}m$-CsI:Na/30 ${\mu}m$-Se layer has been fabricated by a thermal evaporation technique. SEM and PL measurements have been performed. The dark current and x-ray sensitivity of the fabricated detector has been compared with that of the conventional a-Se detector with 100 ${\mu}m$ thickness. Experimental results show that both detectors exhibit a similar dark current, which was of a low value below $400 pA/cm^2$ at 10 V/${\mu}m$. However, the CsI:Na-Se detector indicates high x-ray sensitivity, roughly 1.3 times that of a conventional a-Se detector. Furthermore, a CsI:Na-Se detector with an aluminium reflective layer shows a 1.8 times higher x-ray sensitivity than an a-Se detector. The hybrid type detector proposed in this work exhibits a low dark current and high x-ray sensitivity, and, in particular, excellent linearity to the x-ray exposure dose.

  • An electronic personal dosimeter(EPD) with hybrid type preamplifier adopting a semiconductor detector as a radiation detector has been developed, manufactured and test-evaluated. The radiation detection characteristics of this EPD has been performance-tested by using a reference photon radiation field. After several test-irradiations to a $^{137}Cs$ gamma radiation source the radiation detection sensitivity of this EPD appeared to be $3.8\;cps/Gy{\cdot}h^{-1}$. The linearity of radiation response was kept within 8% of the dose equivalent ranges of $10{\mu}Sv{\sim}4Sv$ and the angular dependence was under less than 4% in angles of ${\pm}60^{\circ}$. It was confirmed that the energy response range was in $60{\sim}1,250keV$ given in the ISO standard. This EPD satisfied the international criteria for the EPD in the mechanical and the environmental performance test for 9 test categories according to IEC 61526.

Thermoluminescence from X-Ray Irradiated Beta-Eucryptite (X-선 조사된 Beta-eucryptite의 열자극 발광)

  • In this study, beta-eucryptite is fabricated and the thermal parameters of this material have been investigated. The thermoluminescence from 4MeV X-ray irradiated beta-eucryptite have been measured over the temperature range of 300K-600K. Thermoluminescence curve from X-ray irradiated beta-eucryptite shows five peaks located at 342K, 392K, 438K, 474K, and 527K. $\tau$, $\delta$ and $\omega$ of peak at 527K are 35K, 39K and 74K, respectively and this peak is found to be 2nd order kinetics. The activation energy of peak shape method is calculated to be 1.03eV and the frequency factor for 527K curve is calculated to be 3.9x10$\^$8/sec$\^$-1/. Based on the various heating rates methods, the activation energy of the peak is computed to be 1.02${\pm}$0.05eV that is similar to 1.19${\pm}$0.03eV of initial rise method. The linearity of thermoluminescence intensity and radiation flux is valid up to 50Gy and beyond higher dose the supralinearity and saturation come out.

Residual Analysis of Insecticides (Lambda-cyhalothrin, Lufenuron, Thiamethoxam and Clothianidin) in Pomegranate Using GC-μECD or HPLC-UVD

  • In this study, the residual levels of four insecticidal compounds (lambda-cyhalothrin, lufenuron, thiamethoxam, and clothianidin) were monitored in the pomegranate, in order to assess the risk to consumers posed by the presence of such residues. The insecticides were applied at the recommended dose rates onto pomegranate trees. The samples were then collected at harvesting time after several treatments (two, three, and four treatments). After sample preparation progressed through the clean-up procedure, lufenuron, thiamethoxam, and clothianidin residues were analyzed via a HPCL-UVD, and the lambda-cyhalothrin residue was analyzed via a GC-${\mu}ECD$. The versatility of this method was evidenced by its excellent linearity (>0.9998 to 1) at broad concentration ranges. The mean recoveries evaluated from the untreated sample spiked with two different fortification levels ranged from 72.45 to 113.90%, and the repeatability (as a relative standard deviation) resulted from triplicate recovery tests was in a range from 0.80 to 11.75%. The residues of all insecticides determined from treated pomegranate samples and their LOD levels (lunfenuron, 0.01; lambda-cyhalothrin, 0.005; thiamethoxam, 0.01; clothianidin, 0.02 mg/kg) were much lower than their MRLs (0.5 mg/kg).

Determination of Ethyl Carbamate in Maesil Extract and Estimated Daily Intake (매실농축액의 에틸카바메이트 분석과 노출량 평가)

  • The analytical method for ethyl carbamate (EC) in maesil (Prunus mume) extract was developed with deuterium-labeled ethyl carbamate as an internal standard. Samples were neutralized with an addition of 1 N sodium hydroxide solution, followed by a solid phase extraction with a Chem Elut cartridge. A standard curve exhibited a good linearity with correlation coefficient of 0.9991. The limit of detection (LOD) and limit of quantification (LOQ) were 2.91 ng/g and 8.83 ng/g, respectively. The recovery rate of EC ranged from 91.40% to 120.90%. The precision never exceeded 12.57% (intra-day) and 11.03% (inter-day). Samples were comprised of 24 home-made and 7 commercially-available maesil extracts. Eight home-made samples contained EC at levels between 3.39 and 75.76 ng/g. Three commercially-available samples had EC at levels between 11.67 and 20.16 ng/g. Average daily intakes of EC from maesil extracts for consumers were 0.23 g/kg of body weight. Based on a benchmark dose confidence limit ($BMDL_{10}$) of 0.25 mg/kg of body weight/day, the margin of exposure (MOE) of EC in maesil extract for consumers was 94,150, which is not of concern. Considering that a daily intake of maesil extract has been increasing, further studies on the formation of EC in maesil extract is needed.

Feasibility study of Hybrid X-ray detecter for Digital X-ray imaging application (디지털 방사선 적용을 위한 Hybrid 방사선 검출기의 Feasibility 연구)

  • In this study, the purpose is to verified the feasibility to develope Hybrid x-ray detector in order to resolve problems of direct and indirect x-ray detectors. The properties of X-ray detector depend on absorption of X-ray, charge generation by x-ray photon, leakage current. In this study, CdS was used as photoconductor, and $Y_2O_2S:Tb$ as x-ray phosphor was formed on CdS in order to embody Hybrid structure. And Screen printing was used to form Muli-layer. Characteristics of this specimen were analyzed by using SEM, and XRD. And Photoluminescence spectrum of $Y_2O_2S:Tb$, leakage current, with respect to applied voltages, output charge with respect to applied voltages, and X-ray sensitivity were measured. Also, linearity with respect to dose was measured. Leakage current was similar with direct digital x-ray detector, but sensitivity of the hybrid structure is much better than the single-layer structure.

Fabrication and Test of a $HgI_2$ Gamma Ray Detector (감마선 검출용 $HgI_2$ 소자 제작 및 특성 평가)

  • The $HgI_2$ single crystal which can be used for the ${\gamma}-ray$ detector at room temperature was grown by Temperature Oscillation Method. The low temperature photoluminescence, specific resistivity and trap concentration of $HgI_2$ single crystal were investigated. Three main luminescence bands were observed at 2.30eV, 2.20eV and 2.00eV at 20K, related to the excitons, I-vacancies and impurities, respectively. The specific resistivity and trap concentration of $HgI_2$ single crystal were $10^{11}{\Omega}\;cm\;and\;1.8{\times}10^{14}/cm^3$ at room temperature, respectively. Also the radiation detecting system was deviced by $HgI_2$ ${\gamma}-ray$ detector, one chip microprocessor, LCD module and personal computer. The prepared $HgI_2$ ${\gamma}-ray$ detector showed a good linearity of ${\gamma}-radiation$ dose for standard ${\gamma}-ray$.

LC-MS Determination and Bioavailability Study of Imidapril Hydrochloride after the Oral Administration of Imidapril Tablets in Human Volunteers

  • The purpose of the present study was to develop a standard protocol for imidapril hydrochloride bioequivalence testing. For this reason, a specific LC-MS method was developed and validated for the determination of imidapril in human plasma. A solid-phase extraction cartridge, $Sep-pak^{R}$ C18, was used to extract imidapril and ramipril (an internal standard) from deproteinized plasma. The compounds were separated using a XTerra $MS^{R}$?C18 column ($3.5 {\mu}m, 2.1\times150 mm$) and $acetonitrile-0.1\%$ formic acid (67:33, v/v) adjusted to pH 2.4 by 2 mmol/L ammonium formic acid, as mobile phase at 0.3 mL/min. Imidapril was detected as m/z 406 at a retention time of ca. 2.3 min, and ramipril as m/z 417 at ca. 3.6 min. The described method showed acceptable specificity, linearity from 0.5 to 100 ng/mL, precision (expressed as a relative standard deviation of less than $15\%$), accuracy, and stability. The plasma concentration-versus-time curves of eight healthy male volunteers administered a single dose of imidapril (10 mg), gave an $AUC_{12hr}$ of imidapril of $121.48\pm35.81 ng mL^{-1} h$, and $C_{max} and T_{max}$ values of $32.59\pm9.76 ng/mL and 1.75\pm0.27 h$. The developed method should be useful for the determination of imidapril in plasma with sufficient sensitivity and specificity in bioequivalence study.

A Study on Characteristics of A Diode Radiation Sensor for Portal Image of Therapy Radiation (치료방사선 Portal Image를 위한 다이오드 방사선 센서의 특성에 관한 연구)

  • In this paper, the characteristics of therapy radiation diode sensors have been studied by using therapy radiation from the MM22 microtron accelerator. The linearity, reproducibility and error ratio were measured for feasibility as a radiation detector. Energy dependence, sensitivity change after a amount of irradiation and output value according to a number of diodes were also measured for same purpose. We have formed pulse shaping of diode signal with nuclear instruments for portal image reconstruction. The percent depth dose ratio according to field size and depth was compared with that of the detector of a ion chamber. Using thirteen silicon diodes, we can directly read diode outputs on a computer monitor after A/D conversion with 16 channels analog to digital conversion board with 12 bit resolution. The possibility for portal image with diodes has been suggested from output comparison between output value with a human phantom and that without a human phantom.
