Omega-3 fatty acids, including eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), are polyunsaturated fatty acids beneficial to human health. A limited number of microalgae have been used for commercial omega-3 production, which necessitates the identification of new microalgae with high omega-3 contents. We explored the fatty acid composition and EPA and DHA contents of the mixotrophic dinoflagellate Gymnodinium smaydae fed with the optimal algal prey species Heterocapsa rotundata. Cells of G. smaydae were found to be rich in omega-3 fatty acids. In particular, the DHA content of G. smaydae was 21 mg g-1 dry weight, accounting for 43% of the total fatty acid content. The percentage of DHA in the total fatty acid content of G. smaydae was the highest among the reported microalgae except for Crypthecodinium cohnii. Moreover, to determine if the prey supply interval affected the growth rate of G. smaydae and its fatty acid content, three different prey supply intervals (daily, once every 2 d, and once for 4 d) were tested. Daily prey supply yielded the highest total fatty acid and DHA contents in G. smaydae. Furthermore, we successfully produced high-density G. smaydae cultures semi-continuously for 43 d with daily prey supply. During the semi-continuous cultivation period, the highest density of G. smaydae was 57,000 cells mL-1, with an average growth rate of 0.7 d-1. Taken together, the percentage of EPA and DHA in the total fatty acid content was maintained in the range of 54.2-56.9%. The results of this study support G. smaydae as a promising microalgal candidate for commercial DHA production and demonstrate that daily supply of prey can efficiently produce high-density G. smaydae cultures for more than a month.
The differential effects of various fatty acids such as n-3 and n-6 types or degrees of unsaturation on the CYP2E1 induction and the production of lipid peroxidation (LPO) were investigated. The CYP2E1-transduced human hepatoma HepG2 cells (E47) were cultured in RPMI 1640 media containing different concentrations of various fatty acids up to 48 h incubation compared to 04 cells and CYP2E1-null cells. Treated fatty acids were linoleic acid (LA:n-6, C18:2), arachidonic acid (AA:n-6, C20:4) and docosahexaenoic acid (DHA:n-3, C22:6). The cell survival rate was decreased corresponding to the degree of unsaturation (LA>AA $\cong$DHA) and to LPO production in E47 and 04 cells. The four or five unsaturation degree of fatty acids, AA and DHA, caused time- and dose-dependent cell death in E47 cells but not as much as in C34 (without CYP2E1), suggesting an important role of CYP2E1 in the DHA mediated damage. In the levels of lipid peroxides (LPO), AA also elevated LPO by 3- and 5- fold compared to DHA or LA treated E47 cells. However, AA did not increase LPO until 48 h incubation in C34 cells. In conclusion, the polyunsaturated fatty acids induced CYP2E1 induction might be changed by the elevated levels of lipid peroxide (LPO) and oxidative stress through the connection of CYP2E1 and degrees of unsaturated fatty acids.
Long chain polyunsaturated fatty acids (LCPUFA) are important components of brain phospholipds and play important role (s) in brain function. In rats, the maximum brain growth occurs during the period of lactation even though it happens during the third trimester of gestation in human. Since milk contained docosahexaenoic acid (DHA) even through the maternal diet had no DHA and/or a very small amount of its precursor, $\alpha$-linolenic acid ($\alpha$-LnA), an emphasis was given to maternal adipose tissue as a reservoir of this fatty acid. We, therefore, investigated the mesenteric and subcutaneous adipose tissues for their fatty acid composition in dams reared with different fat diets. Diets containing various amounts of $\omega$6 and $\omega$3 fatty acids were given to adult female rats (200-250g) throughout the pregnancy and lactation periods. Diets were composed of 10% (wt/wt) corn oil (CO), soybean oil (SO), perilla seed oil (PO) containing about 60% $\alpha$-LnA, or fish oil (FO) rich in eicosapentaenoic acid (EPA) and DHA. The fatty acid ompositions of mesenteric and subcutaneous fat were measured and evaluated at Day-2 and Day-15 after parturition. In general, major characteristics of dietary fatty acid composition was reflected on the fatty acid composition of adipose tissues. Dietary fatty acid composition was reflected more on mesenteric fat as compared to subcutaneous fat. Mesenteric fat was found to contain less arachidonic acid (AA) and mesenteric fats of CO, SO and PO groups contained less DHA than did the subcutaneous fat. The P/M/S ratios of adipose tissues were similar between experimental groups while dietary P/M/S ratios differed significantly. It was noticeable that a small proportion of DHA was found in the adipose tissues of animals of CO, SO and PO groups (Day-2) and in SO and PO groups (Day-15), the groups which do not contain DHA in their diets. The percentage of DHA in mesenteric fat o CO, SO and PO groups decreased as lactation continues, while the proportion of DHA in FO group increased. Adipose tissues of FO group had higher DHA/EPA ratio as compared to the diet. Considering the fact that the body contains a large amount of adipose tissues, our present finding suggests that the adipose tissue can serve as a reservoir of DHA for pregnant and lactating rats.
To predict Daphnia secondary productivity along a trophic gradient indexed as total phosphorus (TP) concentration, we estimated energy transfer efficiencies from food quality for Daphnia such as eicosa-pentaenoic acid (EPA) or docosahexaenoic acid (DHA) content. Eleven flow-through Daphnia magna growth experiments were conducted with seston from 9 lakes, ponds and river waters. Primary productivities were estimated from food supply rates in the flow-through experiments, producing energy transfer efficiencies from seston to D. magna. We found DHA content was the best predictor of energy transfer efficiencies among the essential fatty acids. An asymptotic saturation model explained 79.6% of the variability In energy transfer efficiencies. Based on empirical data in this study and empirical models from literature, we predict that Daphnia productivity would peak in mesotrophic systems by decreasing food quality and Increasing food quantity along trophic gradient.
The purpose of this investigation was to determine the effect of feeding dried mackerel as a means of increasing the intake of these n-3 polyunsaturated fatty acids on fatty acid compositions in liver and nervous tissue. Twenty male mice aged at 4 weeks were fed on the control (5% palm oil, control group) and 5% dried mackerel diets (mackerel group) for four weeks. In fatty acid compositions of liver and cortex, levels of total n-3 fatty acid, specially docosahexaenoic (22:6n-3, DHA) and eicosapentaenoic (20:5n-3, EPA) acids, were increased in the mackerel group compared to the control group, while docosapentaenoic acid (22:5n-6, DPAn-6) levels were decreased (p<0.05). In cerebellum and retina, levels of DHA were not significantly different between the control and mackerel groups, but levels of total n-6 fatty acids and arachidonic acid (20:4n-6, AA) were decreased in the mackerel group. These results indicated that intake of 5% dried mackerel increased levels of n-3 polyunsaturated fatty acids in cortex. Thus, we will investigate the relationship between brain function and cortex fatty acid compositions following intake of mackerel by assessing discrimination leaning ability.
Omega-3 fatty acid, docosahexaenoic acid(DHA) is found in a high proportion in the structural lipids of cell membranes, in particular those of the central nervous system and the retina. Diet-induced changes in fatty acid composition in these tissues may affect physiochemical functions. This study was conducted to investigate whether supplements of DHA in infant formula has an effect on the composition of fatty acids in erythrocytes with regard to brain development. Experimental groups were breastmilk group(n=21), placebo formula group(n=15), and DHA supplemented formula (0.26%) group(n=16). Infants were selected by mothers who deliverecdd at Kyung Hee medical center from February to April, 1996. Infant body weight, length, and head circumference were similar among the experimental groups at 16 weeks of age. The levels of DHA in breastmilk, placebo formula, and DHA supplemented formula were 0.56, 0, and 0.26% of total fatty acids, respectively. There was a significant correlation between dietary DHA intake and erythrocyte DHA levels. The levels of arachidonic acid did not differ among the three expermental groups. The result of flash visual evoke potential(VEP) test was correlated with the erythrocyte levels and dietary DHA levels at 16 weeks of age. No other fatty acid was correlated with VEP test results. No differences were found in Bayley Mental and Psychomotor Development Index scores among the three groups at 20 weeks of age. DHA seems to be an essential nutrient for optimum growth and maturation of term infants. Relatively small amounts of dietary DHA supplementation significantly elevate DHA supplementation significantly elevate DHA content in erythrocytes, which in turn has an implication for better scores for infant's VEP test. Whether supplementation of formula-fed infants with DHA has long-term benefits remains to be elucidated.
Changes of the fatty acid, amino acids and collagen contents in domestic broiler chickens of different marketing standard range from 5ho to 16ho were assessed. There were no significant trend in total ratios of saturated fatty acids and unsaturated fatty acids between breast samples taken from chicken in different weight ranges. The n6 fatty acids/n3 fatty acids decreased as weight ranges decreased and the ratio was decreased down to 5:1. Eicosapentaenoic acids(EPA) were detected from chicken in 10ho and tended to increase thereafter. The contents of EPA were 0.43% in 15ho. Docosahexaenoic acid(DHA) were detected from chicken in 14ho and DHA contents of breast taken from 15ho chicken were 0.94%. The contents of glutamic acids were 2.94∼3.59% and they increased as chicken weight increased. The collagen contents were higher in thigh than breast or wings. The results from this experiments will provide a basic information for establishment of marketing standard of chicken.
Effect of DHA-rich fish oil on brain development and learning ability has been studied in Sprague Dawley rats. Female rats were fed experimental diets containing either corn oil fish oil at 10%(w/w) level throughout the gestation and lactation. Corn oil was added in fish oil diet to supply essential fatty acid at 2.3% of the calories. All male pups were weaned to the same diets of dams at 21-days after birth. Plasma fatty acid composition was analyzed for dams and pups at 21-days, 28-days and 22-weeks after birth. The analysis of DNA and fatty acid profile in the brain were undertaken at birth, 3, 7, 14, 21, 28 days and 22 weeks after birth and learning ability was tested at 18-20 weeks of age. Regardless of dietary fats, arachidonic acid(AA) and docosahexaenoic acid(DHA) were the principal polyunsaturated fatty acids in the brain. Rats fed CO diet showed a continouus increase of AA content in the brain from 10.9%(at birth) to maximum 15.3% level (14-days old), while the rars fed FO diet showed 78-79% of CO group throughout the period. Rats fed FO diet showed higher incorparation of DHA from 15.2% at birth to a maximum level of 18.5% at 140days, while the rats fed CO diet showed only 7.0% incorporation of DHA at birth and a maximum level of 11.1% at 21-days. Compared to CO group, FO group showed lower ratio of chol/PL and higher content of DHA in brain microsomal membrane, resulting in better membrane fluidity. Total amount of DNA per gram of brain was reached maximum level at 21 days in both groups. This would be a period of the cell proliferation during brain development. Overall, the rats fed fish oil diet showed a higher incorporation of DHA and membrane fluidity in the brain and better learning performances (p<0.05).
Supercritical $CO_2$ chromatography was applied for purification of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) from fish oil. Various supercritical $CO_2$ pressures were tested to find out the pressure effects on solubility and selectivity of low fatty acids in the silver nitrate column. The solubility of low fatty acids was increased as the supercntical $CO_2$ Pressure increased. However, the selectiviy between low fatty acids and EPA waw decreased. Stepwise density gradient method was applied to increase the purification efficiency of EPA. Low fatty acids were easily separated at the early elution steps with low $CO_2$ densities. Successive fractions containing 92.1~97.8% of EPA were collected. The average concentration of three purified fractions was 95.6% with the recovery rate of 30%.
This study was conducted to asses the $\omega$-3 fatty acid intake levels and to evaluate the relationship between the levels of $\omega$-3 fatty acids in plasma and erythrocyte membrane and the dietary intake of these fatty acids over the period of two months in 56 young Korean females. Dietary survey was conducted to obtain 7-day weiged records and six 24-hour recalls. Fasting blood sample was collected from each subject after the dietary survey period. Mean daily intakes of energy, fat, and cholesterol were 1,569㎉, 41.8g, and 217mg, respectively. Fat suppled 24% of total energy intake. Mean daily intake levels of eicosapentaenoic acid(EPA), docosahexaenoic acid(DHA), and total $\omega$-3 fatty acids were 0.04g, 0.06g, and 0.48g, respectively. Plasma cholesterol levels of most of the subjects were within normal range, and there was no significant correlation between plasma cholesterol levels and intake levels of any specific fatty acid. Levels of EPA, DHA, EPA+DHA in plasma and erythrocyte membrane, but not the levels of $\alpha$-linolenic acid(LNA), were significantly correlated with dietary in take of respective fatty acids. Such a correlation, however, was not observed in a previous study where dietary intake was assessed for 3 days. The results of this study show that dietary intakes of $\omega$-3 fatty acids are low in the subject and that about two-month period is required to assess dietary intake levels of $\omega$-3 fatty acids with a reasonable accuracy.
